Despite the advances made in the area of kidney transplantation, the disparity between the demand and available donated organs remains a dominant and unresolved issue. Given the paucity of available renal allografts the preservation of existing grafts is vital. One factor that has negatively impacted renal allograft survival is acute rejection (AR). Traditionally, kidney transplant centers have used elevations in serum creatinine as a screening tool for detecting AR. However, …

The purpose of this research was to test the hypothesis that starch-entrapped microspheres would produce favourable fermentation profiles and microbial shifts during in vitro fermentation with the faecal microbiota from patients with inflammatory bowel disease (IBD). In vitro fermentation was carried out using a validated, dynamic, computer-controlled model of the human colon (Toegepast Natuurwetenschappelijk Onderzoek gastrointestinal model-2) after inoculation with pooled faeces from healthy individuals, patients with inactive IBD (Crohn’s disease (CD)) or patients with active IBD (ulcerative colitis (UC)). Starch-entrapped microspheres fermented more slowly and produced more butyrate than fructooligosaccharides (FOS) when fermented with the faecal microbiota from patients …

In vertebrates, including humans, individuals harbor gut microbial communities whose species composition and relative proportions of dominant microbial groups are tremendously varied. Although external and stochastic factors clearly contribute to the individuality of the microbiota, the fundamental principles dictating how environmental factors and host genetic factors combine to shape this complex ecosystem are largely unknown and require systematic study. Here we examined factors that affect microbiota composition in a large (n = 645) mouse advanced intercross line originating from a cross between C57BL/6J and an ICR-derived outbred line (HR). Quantitative pyrosequencing of the microbiota defined a core measurable microbiota …

Effect Of Dietary Folate Restriction On Colon Carcinogenesis In Dna Polymerase Β Haploinsufficient Mice, Lisa F. Ventrella Lucente
Wayne State University Dissertations
The data presented in this research is central to establishing the role that the base excision repair pathway (BER) plays in the development and progression of colon cancer when dietary folate is deficient. Both cellular folate restriction and BER deficiencies have been shown to result in the accumulation of endogenous damage and lesions that could eventually develop into carcinogenesis. In this study, a dietary folate deficiency (FD) resulted in a significant increase in aberrant crypt foci (ACF) formation and triggered liver tumorogenesis in wildtype (WT) animals, as did a BER deficiency in DNA polymerase Β haploinsufficient (Β-pol+/-) mice exposed to …

High-grain adaptation programs are widely used with feedlot cattle to balance enhanced growth performance against the risk of acidosis. This adaptation to a high-grain diet from a high-forage diet is known to change the rumen microbial population structure and help establish a stable microbial population within the rumen. Therefore, to evaluate bacterial population dynamics during adaptation to a high-grain diet, 4 ruminally cannulated beef steers were adapted to a high-grain diet using a step-up diet regimen containing grain and hay at ratios of 20:80, 40:60, 60:40, and 80:20. The rumen bacterial populations were evaluated at each stage of the step-up …
Epigenetics: Blurring The Line Between Nature And Nurture, Elizabeth H. Rose
Epigenetics: Blurring The Line Between Nature And Nurture, Elizabeth H. Rose
CMC Senior Theses
This long-standing nature versus nurture debate is cited in behavioral and physical expressions of disease dysfunctions, resiliencies, and recovery. Their purposes are noted both in scientific pursuits as well as literature. This discourse has been particularly intense in the fields of psychology, psychiatry, and biology where there is a long history of scientists’ attempts to disprove or discredit others’ intellectual and professional measures. Interestingly, recent advances in the neurosciences and genetic technologies have brought these fields closer together with a new focus – the interactional relationship between nature and nurture – epigenetics.

Trace Elements And Physical Activity In Children And Adolescents With Depression, M. Meti̇n Donma, Orki̇de Donma
Turkish Journal of Medical Sciences
Depression is a common mental health problem among adolescents. Depressive symptoms are also important and may appear even during the preschool period. Physical activity, which may improve some mental health problems, is inversely associated with depression. Due to the presence of some clinical evidence about the relation between the use of antidepressants and suicide, there is a need for supportive agents during antidepressant therapy. Within this context, essential trace elements gain importance for further consideration. Protection of the developing brain from the negative effects of iron deficiency is important because of iron deficiency's association with poor mental development. Reactions between …

Freshwater planarians, Plathelminthes, have been an intriguing model animal of regeneration studies for more than 100 years. Their robust regenerative ability is one of asexual reproductive capacity, in which complete animals develop from tiny body fragments within a week. Pluripotent adult somatic stem cells, called neoblasts, assure this regenerative ability. Neoblasts give rise to not only all types of somatic cells, but also germline cells. During the last decade, several experimental techniques for the analysis of planarian neoblasts at the molecular level, such as in situ hybridization, RNAi and fluorescence activated cell sorting, have been established. Moreover, information about genes …

To investigate the effects of ingesting Fructus vitis minuta (black grape) on plasma and erythrocyte antioxidant parameters in elderly subjects. Materials and methods: Thirteen subjects (mean age 74.67 ± 0.58) participated in the study. They consumed 1 g/kg body weight per day of dried black Maraş grapes for 1 month. Before and after these periods, fasting blood samples were obtained, and oxidant (as MDA) and antioxidant (as SOD) parameters were studied in erythrocytes. Also, plasma samples were obtained from the subjects. Results: In the erythrocyte hemolysate, MDA (malondialdehydes) levels and SOD (superoxide dismutase) activities were found to be lower (P < 0.01) in the second samples compared to the first ones. Conclusion: We think that because free radical production was decreased SOD activities were not increased. Our results show that reduced peroxidation processes caused by Maraş grape consumption suggest that grape consumption has beneficial effects in elderly subjects.

To investigate the association between the 3 most frequently observed single nucleotide polymorphisms of the NAT2 gene and the risk of developing prostate cancer in the Turkish population. Materials and methods: A total of 110 unrelated patients with prostate cancer were included in this case-control association study and constituted the study group. The control group also consisted of 150 unrelated but healthy men. Genomic DNA was isolated from peripheral blood leukocytes of all patients and analyzed with a sensitive real-time PCR method. After melting curve analysis genotypes were identified for the NAT2*5A, NAT2*6A, and NAT2*7A/B polymorphisms. Results: Prostate cancer patients …

The aim of this study was to investigate the level of serum adenosine deaminase (ADA) and the total antioxidant capacity (TAC) in pulmonary tuberculosis (PTB), non-tuberculosis pulmonary disease (non-PTB) and healthy subjects. Materials and methods: Serum ADA activity was measured using the Giusti and Galanti method, and the total antioxidant capacity of plasma was determined by the ferric reducing ability of plasma (FRAP) test. Results: The serum ADA levels were significantly higher (P < 0.001) in pulmonary TB (PTB, 19.78 ± 7.09 U/L), as well as in non-PTB patients (14.78 ± 4.65 U/L) when compared to healthy controls (10.02 ± 1.99 U/L). The sensitivity and specificity were found to be 71.7% and 63.3%, respectively, in distinguishing PTB from non-PTB. In distinguishing PTB from healthy subjects, the sensitivity and specificity were 87% and 93.3%, respectively. The TAC was significantly lower in PTB (485.2 ± 190.0 µM) and non-PTB patients (588.3 ± 195.8 µM), when compared to the controls (784.3 ± 190.0 µM; P < 0.001). Plasma antioxidative activity decreased in PTB and non-PTB patients when compared to the controls. Conclusion: We concluded that serum ADA activity is not a useful test to differentiate pulmonary TB from other respiratory diseases. The TAC is low in pulmonary TB, therefore supplementation with a suitable anti-oxidant may be useful.

To examine the prevalence of anaemia in Meghalaya, India, by exploring the 2005-2006 National Family Health Survey (NFHS -3) data. Materials and methods: The study population consisted of 3934 ever-married women of reproductive age (15-49 years) from the state Meghalaya, India, which were taken from the NFHS-3 survey, to explore the predictors responsible for the prevalence of anaemia by using different background characteristics such as age, place of residence, nutritional status, number of children ever born, pregnancy status, educational achievement, and economic status. As a response variable, anaemia levels were categorised as a dichotomous variable, and the predicted probabilities were …
