Homocysteine (tHcy) has been in routine use in many centers after this amino acid was reported as an independent risk factor for cardiovascular, cerebrovascular and peripheral vascular diseases. Many high-performance liquid chromatographic (HPLC) methods including a commercially available kit (Chromsystems) were reported for measuring tHcy in biologic samples. Although the commercial kits are believed to have higher precision, sensitivity and accuracy as well as practical and fast, it may cause high prices by increased test ordering. Since leukoplakia is a premalignant condition, it is evident that an effective means of control would contribute to reducing the mortality and morbidity of oral cancer. The reactivity of free radicals and singlet oxygen is believed to be an important factor in the pathogenesis of many degenerative diseases including carcinogenesis. Reactive free radicals may damage cells by induction of lipid peroxidation of the polyunsaturated fatty acids. Buege and Aust's method was used for lipid peroxidation analysis. Idiopathic thrombocytopenic purpura (ITP) is an autoimmune disorder. Decreased platelet survival is the main pathogenesis, which is related to platelet antibodies (APA). The levels of these antibodies are correlated with relapse and remission which was shown for the first time by our group. Although APA levels decrease in remission they do not disappear as we have shown. Among the several appraches to ITP treatment, oral megadose methylprednisolone is found to be the cheapest and most effective one. Many studies have established the therapeutic benefits of angiotensin-converting enzyme (ACE) inhibitors such as reducing reperfusion arrhythmias, and angiotensin II type 1 (AT1) blocker may have similar effects to ACE inhibitors. The aim of this study was to develop a human tumor necrosis factor alpha (hTNF-\alpha) ELISA system because specific and sensitive measurement of low levels of circulating TNF-a is very important for enlightening the immunopathologial mechanisms associated with TNF-\alpha. Monoclonal antibodies 6A4c and 8A6 were produced against hTNF-\alpha and were used as the capture antibody and the tracer antibody respectively for the first hTNF-\alpha ELISA system (6A4c/biotin-8A6). Murine polyclonal IgG was used as the tracer antibody in the second hTNF-\alpha ELISA system (6A4c/biotin-polyclonal IgG). Many diagnostic tests exist to assess the salivary and lacrimal involvement in Sjögren's syndrome (SS), but there is still disagreement. The purpose of this study was to examine salivary proteins in patients with primary Sjögren's syndrome and healthy control subjects by the polyacrylamide gel electrophoresis, as well as the applicability of the method. The polyacrylamide gel electrophoresis, applied in our examinations, is a simple and rapid method, requiring only a minute amount of saliva for the analysis. BACKGROUND: Paralemmin (Palm) is a prenyl-palmitoyl anchored membrane protein that can drive membrane and process formation in neurons. Earlier studies have shown brain preferred Palm expression, although this protein is a major water insoluble protein in chicken lens fiber cells and the Palm gene may be regulated by Pax6. METHODS: The expression profile of Palm protein in the embryonic, newborn and adult mouse eye as well as dissociated retinal neurons was determined by confocal immunofluorescence. Bioluminescent imaging (BLI) is particularly well suited for imaging small animals, and it can be readily used by scientists who are already routinely using the luciferase gene as a reporter in cell-based assays. This strategy relies on ATP and an O_2 dependent photochemical reaction between luciferin and luciferase, resulting in the release of photons from only live cells. In BLI, the intensity of the light detected by the device is proportional to the intensity of light emitted and the relationship is quantitative. The purpose of this study was to find out if spontaneously arising point mutations occurred in canine e-kit and if these mutations contributed to the development of mast cell tumors in dogs. Specimens were collected from 11 dogs after tumors had been surgically removed or the dog had died. The e-kit DNA was amplified using PCR, and the previously established point mutations were searched for (London et al., 1999). The point mutations found in human mast cell lines were not detected in the canine e-kit DNA, but new mutations were observed. These new mutations, observed in 50% of the tumors, …

Members of the protein kinase C (PKC) superfamily transduce a myriad of transmembrane signals initiated by the formation of sn-l ,2-diacylglycerol (DAG). As the primary physiological agonist for C] domain containing PKC isoenzymes, DAG functions to regulate the spatial and temporal parameters of PKC activity. It is now recognized that cellular DAG levels can vary from minutes to hours and even days. These observations have suggested a division of PKC functions that correlate to the duration of the cellular DAG signal. Morphine and Δ9-THC are drugs that produce analgesia and rewarding effects. However, chronic treatment with morphine and Δ9-THC produces problematic side-effects including tolerance and physical dependence. The cellular mechanisms underlying opioid and cannabinoid antinociceptive tolerance have been studied for years. Research has demonstrated that the expression of morphine and Δ9-THC antinociceptive tolerance may be mediated through intracellular signaling pathways, such as the adenylyl cyclase /Protein Kinase A (PKA) cascade. Semen cryopreservation has an important role in assisted reproductive technology however, the cooling, freezing and thawing processes often result in a significant loss of sperm motility, viability and nuclear integrity. The destructive effects of cryopreservation are significantly exacerbated in samples exhibiting low sperm number and poor morphological characteristics. Studies of transgenic mice with accelerated accumulation of mtDNA mutations specifically in the heart lead us to propose that apoptotic signaling and cell death is central to the pathogenesis of mtDNA mutations in aging. It is the cellular response to that apoptotic signaling and the organ?s compensatory response to a loss of cells that specify the phenotype of an accumulation of mtDNA mutations. In the heart, cardiomyocytes induce a vigorous anti-apoptotic, pro-survival response to counteract mitochondrial apoptotic signaling. BACKGROUND: Two-photon-excitation fluorescence lifetime imaging (2P-FLIM) was used to investigate the association of protein kinase C alpha (PKCalpha) with caveolin in CHO cells. PKCalpha is found widely in the cytoplasm and nucleus in most cells. Upon activation, as a result of increased intracellular Ca2+ and production of DAG, through G-protein coupled-phospholipase C signalling, PKC translocates to a variety of regions in the cell where it phosphorylates and interacts with many signalling pathways. BACKGROUND: The structural maintenance of chromosome proteins SMC1 and SMC3 play an important role in the maintenance of chromosomal integrity by preventing the premature separation of the sister chromatids at the onset of anaphase. The two proteins are constitutive components of the multimeric complex cohesin and form dimers by interacting at their central globular regions. Colon cancer is the third leading cause of cancer-related death in this country.5 The majority of medical societies recommend some form of colon cancer screening for asymptomatic adult patients over age 50. The evaluation of rectal bleeding is different from screening because the risk of serious disease is higher and it is unclear whether early diagnosis and treatment of serious disease results in improved mortality once gross bleeding has occurred. The optimal evaluation strategy for rectal bleeding is unknown. Neither historical information nor the presence or absence of hemorrhoids has been shown to reliably differentiate benign from serious disease.6–11 Fecal …

BACKGROUND: The Structural Maintenance of Chromosome 3 protein (SMC3) plays an essential role during the sister chromatid separation, is involved in DNA repair and recombination and participates in microtubule-mediated intracellular transport. SMC3 is frequently elevated in human colon carcinoma and overexpression of the protein transforms murine NIH3T3 fibroblasts. In order to gain insight into the mechanism of SMC3-mediated tumorigenesis a gene expression profiling was performed on human 293 cells line stably overexpressing SMC3. Improvements in methodologies to recapitulate and study particular biological functions of the
papillomavirus life cycle have led to great advances in our knowledge of these viruses. Described in
this chapter are techniques that allow low-copy and high-copy replication of full-length human
papillomavirus (HPV) genomes, as well as assembly of virus-like particles, in Saccharomyces
cerevisiae (yeast). This system has several distinct advantages that make it an attractive complement
to the well-established raft-culturing system. First, yeast are inexpensive, rapid, and simple to culture

Genetic variation of human immunodeficiency virus (HIV-1) represents a major obstacle for AIDS vaccine development. To decrease the genetic distances between candidate immunogens and field virus strains, we have designed and synthesized an artificial group M consensus env gene (CON6 gene) to be equidistant from contemporary HIV-1 subtypes and recombinants. This novel envelope gene expresses a glycoprotein that binds soluble CD4, utilizes CCR5 but not CXCR4 as a coreceptor, and mediates HIV-1 entry. Key linear, conformational, and glycan-dependent monoclonal antibody epitopes are preserved in CON6, and the glycoprotein is recognized equally well by sera from individuals infected with different HIV-1 …

The phosphoprotein (P protein) of vesicular stomatitis virus (VSV) is an essential subunit of the viral RNAdependent RNA polymerase and has multiple functions residing in its different domains. In the present study, we examined the role of the hypervariable hinge region of P protein in viral RNA synthesis and recovery of infectious VSV by using transposon-mediated insertion mutagenesis and deletion mutagenesis. We observed that insertions of 19-amino-acid linker sequences at various positions within this region affected replication and transcription functions of the P protein to various degrees. As a spirochete, the genus Treponema is one of the few major bacterial groups whose natural phylogenic relationships are evident at the level of gross phenotypic characteristics such as their morphology. Treponema spp. are highly invasive due to their unique motility in dense media, and their ability to penetrate cell layers. This feature is associated with the helical cell body and the presence of flagellar filaments in the periplasm. Treponema denticola is an oral pathogen involved in endodontic infections and periodontal diseases. The aim of this work was to evaluate changes in lymphocyte subpopulations, especially helper and cytotoxic T cells, in acute brucellosis patients undergoing treatment. Forty-three acute brucellosis patients were included in the study. Twenty healthy subjects served as controls. Total lymphocytes and the CD3^+, CD4^+, CD8^+, CD19^+ and CD (16+56)^+ subpopulations were counted by two-color flow cytometric analysis. The CD4+ counts in patients before and after treatment were not statistically different (p = 0.7), but healthy subjects had significantly more of these cells (p = 0.001 and p = 0.001 compared to pre- and post-treatment patients, respectively).
