Transgenic Mice Which Overexpress Neurotrophin-3 (Nt-3) And Method Of Use, Kathryn M. Albers, Brian M. Davis

Transgenic mice express increased levels of neurotrophin-3 (NT-3) in epithelium when their ancestors are microinjected with the NT-3 gene. The NT-3 growth factor expressing transgenic mice are useful in the study of neurodegenerative disorders of the brain such as Parkinson's syndrome and Alzheimer's disease, of the spinal cord motor neurons such as amyotrophic lateral sclerosis, and for testing drug candidates for the treatment of these diseases.

Animal research has provided important information about many aspects of the pathophysiology of human disease. Well-performed animal studies can determine the potential benefit of many proposed therapeutic interventions, and experimental results from animal studies have served as the basis for many landmark clinical trials. Many animal research models are described in the research literature, and choosing the appropriate model to answer a research question can be a daunting task. Reciprocal interactions between the hippocampus and the perirhinal and parahippocampal cortices form core components of a proposed temporal lobe memory system. For this reason, the involvement of the hippocampus in event memory is thought to depend on its connections with these cortical areas. Contrary to these predictions, we found that NMDA-induced lesions of the putative rat homologs of these cortical areas (perirhinal plus postrhinal cortices) did not impair performance on two allocentric spatial tasks highly sensitive to hippocampal dysfunction. Remarkably, for one of the tasks there was evidence of a facilitation of performance. The same cortical lesions did, however, disrupt …

Fourteen rats were examined and divided into two groups of 7: the control (Group a) and NGF-treated animals (Group B). All animals received a 50 g clip-compression injury to tthe spinal cord at the T9 level. In the NGF-treated animals, NGF (0.1 mg/kg NGF) was injected subcutaneously for seven days. After spinal cord injury, the mean AChE activity was 41.47±1.7122 activated substrate min/ml/gr. wet tissue and 47.52±1.471 in NGF-treated animals. A review of the use of pindone for rabbit control in Western Australia is timely due to increased public concern over the use of this toxin, and because non-target deaths of some native Australian animals have been known to occur following its use. Pindone is one of the first generation indandione anticoagulant rodenticides which were developed in the late 1940' s. The toxicity of anticoagulants arises from their inhibition of vitamin K-dependent clotting factors in the blood. Thus therapeutic administration of vitamin K provides a reliable antidote against anticoagulant poisoning. Pindone is slow acting and works best with repeated small …

The objective of this study was to compare the anti-inflammatory effects of NG -nitro L-arginine (L-NAME) and corticosteroid in Concanavalin A-induced uveitis in rats. Sixteen male Wistar rats were used. After general anesthesia, intravitreal 0.1 ml Concanavalin A (100mg/ml) was injected into the left eyes of the rats. The protective effect of vitamins E and C on the gastric mucosal barrier in rats irradiated with X-rays was investigated. Thirty-two male Wistar Albino rats were used. The animals were divided into four groups and arranged of follows: the first group (n=8) was the sham group; the second group (n=8) was the control group which was irradiated with X-rays, the third group (n=8) to which vitamin C was administered and the fourth group (n=8) to which vitamin E was given. Semi-thin and thin sections were used to observe the morphogenesis of peripheral nerves regenerating in silicone tubes. The progress of peripheral nerve regeneration across a 10 mm gap within the silicone tubes was examined after 3, 6 and 9 months. The left sciatic nerves of rats were transected. The proximal and distal nerve stumps were inserted into a silicone tube, in which they were separeted by a 10 mm gap. A tissue cable formed inside the silicone tube with regenerated axons. The effects of Rumex patientia and Urtica dioica on levels of blood glucose, plasma amino acids and other parameters, urine excreta, and liver and kidney histology were examined in diabetic rats induced by streptozotocin. Streptozotocin increased blood glucose and changed the levels of amino acids and other parameters, and caused degenerative changes in the liver and kidney. Rumex patientia had some protective effect on these parameters changed by streptozotocin, while Urtica diocia had no protective effects.

We evaluated the effect of nifedip-ine, an L-type voltage sensitive Ca+2 channel (VSCC) blocker, on the onset of neuronal damage induced by transient ischemia of guinea pig retinas in vivo. The animals were divided into two groups. In the first group 2 mg/kg Nifedipine was administered intraperi-toneally, while only 2 cc/kg NaCl 0.9% solution was given to the second group thirty minutes before the ischemic insult. Seven days after the temporary ligation of bilateral common carotid arteries for 45 minutes, the animals were dispatched and the eyes were enucleated for histopathologic analysis. This experiment was conducted to investigate the time-dependent effects of gentamicin (GM) on the neuroepithelial morphology of the posterior crista ampullares (PCA) of the adult chinchilla. The time course of GM-induced HC pathology was documented through measurements of HC density as well as through HC morphologic evaluation. HC morphology was examined under light microscopy and HC density measurements were documented as a function of the time course of GM-induced ototoxicity. In this study we investigated the effect of acetylsalicylic acid (ASA) on tear secretion in rats. The animals were anesthetized intraperitoneally (ip) with urethane (1,2g/kg). The treatment was injected subcutaneously (s.c.). Tear samples were collected by folding a 5 mm section of a Schirmer strip over the lower lid margin to absorb tear fluid from the lower conjunctival sac for 5 min. Biological damage mediated by reactive oxygen species has been increasingly recognized in aging phenomena in recent decades. The increase in catecholamine metabolism is yet another physiological change in aging, predisposing the organism to supranormal hydrogen peroxide produc-tion as a by-product. This study was undertaken to investigate the alterations in oxidant- antioxidant status in rat cardiac tissue during aging. The development of an effective therapy for Duchenne Muscular Dystrophy (DMD) is one of the primary goals of all DMD/Becker Muscular Dystrophy (BMD) research. Golden Retriever Muscular Dystrophy (GRMD), an animal model of DMD is a fatal degenerative myopathy. Unlike the mdx model, the GRMD dog more accurately reflects the phenotype shown by human DMG patients, making the model better suited for the investigation and assessment of potential therapeutic approaches. The GRMD mutation, a base change from A to G in the 3' splice acceptor site of intron 6, results in exon 7 skipping which disrupts the translational reading frame. …

A plasmid bearing a cDNA copy of the genome of bovine viral diarrhea virus (BVDV), chimeric derivatives of the plasmid and a method of producing an infectious bovine viral diarrhea virus using the plasmid are disclosed. The invention relates to a plasmid DNA molecule that replicates easily in E. coli and contains a sufficient portion of the genome of BVDV, cloned as cDNA, to be a suitable template to produce RNA in vitro which, upon transfection into bovine cells, gives rise to infectious BVDV. Gnotobiotic piglets were inoculated intralaryngeally with swine Chlamydia trachomatis strain R33 or orally with swine C. trachmatis strain R27. Archived formalin-fixed, paraffin-embedded tissues from piglets euthanatized 4–7 days postinoculation were examined by in situ hybridization for C. trachomatis nucleic acid using a nonradioactive digoxigenin-labeled DNA probes that targeted specific ribosomal RNA or omp1 mRNA molecules of the swine C. trachomatis strains. Positive hybridization signals were detected in bronchial epithelial cells, bronchiolar epithelial cells, pneumocytes, alveolar and interstitial macrophages, and jejunal and ileal enterocytes. Alcoholism is transmitted in families. The complexity and heterogeneity of this disorder has made it difficult to identify specific genetic correlates. One design with the potential to do so is the family-based association study, in which the frequencies of genetic polymorphisms are compared between affected and nonaffected members. Reduced central serotonin neurotransmission is associated with features of an antisocial subtype of alcoholism, although a primary deficit has not been traced to a particular component. Genetic markers related to the serotonergic system have been identified, located, and cloned. If associations can be discovered, the development process for pharmacotherapy could be facilitated. …

The question how fish oil (FO) and soybean oil (SO) change the state of antioxidant and the lipid peroxidation in liver and plasma of rats and the protective role of vitamin E were investigated. This experiment was conducted to investigate the dose-dependent effects of gentamicin (GM) on the neuroepithelial morphology of the posterior crista ampullares (PCA) of the adult chinchilla. The experiment focused on hair cell (HC) morphology to monitor GM's dose-dependent effects and also to define an ototoxically-efficient and safe dose what would subsequently be used in a time course study. Chinchillas (n=13) were treated with subcutaneous (SC) daily doses of 120 mg/kg, 60 mg/kg, and 30 mg/kg GM, respectively, for six days. The acousticolateralis sensory system is characterized by a specific receptor cell type called the sensory hair cell and is found in all vertebrates. There are two types of hair cell sensory epithelia based on location: those of the inner ear, such as the organs for hearing and balance, and the lateral line system located within the epidermis. In mammals, including humans, loss or damage of the hair cells of the auditorysystem results in permanent hearing loss. However, this is not the case with birds and amphibians. Therefore, since the structure of polymerase-beta shows significant evolutionary conservation in both plant and animal species and given strong functional correlations among species, polymerase-beta performs a vital role in plants in the repair of newly synthesized DNA just as it does in animal systems.

Superantigens stimulate T-lymphocyte proliferation and cytokine production, but the effects of superantigen exposure on cell function within a complex, highly regulated immune response remain to be determined. In this study, we demonstrate that superantigen exposure significantly alters the murine host response to bacterial antigens in an in vitro coculture system. Two days after exposure to the superantigen staphylococcal enterotoxin B, splenocytes cultured with Streptococcus mutans produced significantly greater amounts of gamma interferon (IFN-gamma) and interleukin-12 than did sham-injected controls. We evaluated the side effects induced by injection of Freund's adjuvant (FA)and alternative adjuvants combined with different antigens. Rabbits and mice were injected subcutaneously, intramuscularly (rabbits) and intraperitoneally (mice)with different adjuvants (FA, Specol, RIBI,TiterMax, Montanide ISA50)in combination with several types of antigens (synthetic peptides, autoantigen, glycolipid, protein, mycoplasma or viruses). The effects of treatment on the animals' well-being were assessed by clinical and behavioural changes (POTand LABORASassays) and gross and histopathological changes. In rabbits, treatment did not appear to induce acute or prolonged pain and distress. Mice showed behavioural changes immediately after (predominantly secondary) immunization.
