BACKGROUND: Glioblastoma (GBM) is an aggressive brain tumor with universal recurrence and poor prognosis. The recurrence is largely driven by chemoradiation resistant cancer stem cells (CSCs). Epidermal growth factor receptor (EGFR) and its mutant EGFRvIII are amplified in ~ 60% and ~ 30% of GBM patients, respectively; however, therapies targeting EGFR have failed to improve disease outcome. EGFRvIII-mediated cross-activation of tyrosine kinase receptor, cMET, regulates GBM CSC maintenance and promote tumor recurrence. Here, we evaluated the efficacy of pan-EGFR inhibitor afatinib and Temozolomide (TMZ) combination on GBM in vitro and in vivo.
METHODS: We analyzed the effect of afatinib and …

BACKGROUND: Trefoil factors (TFF1, TFF2, and TFF3) are small secretory molecules that recently have gained significant attention in multiple studies as an integral component of pancreatic cancer (PC) subtype-specific gene signature. Here, we comprehensively investigated the diagnostic potential of all the member of trefoil family, i.e., TFF1, TFF2, and TFF3 in combination with CA19.9 for detection of PC.
METHODS: Trefoil factors (TFFs) gene expression was analyzed in publicly available cancer genome datasets, followed by assessment of their expression in genetically engineered spontaneous mouse model (GEM) of PC (KrasG12D; Pdx1-Cre (KC)) and in human tissue microarray consisting of normal pancreas adjacent …

Modulation Of Klotho Affects Dendritic Spine Remodeling And Neuronal Network Activity, Hai T. Vo
All ETDs from UAB
Klotho protein expression has profound effects on lifespan where klotho-deficient mice exhibit premature aging phenotypes and live only to ~8 weeks of age while klotho-overexpressing mice have lifespans that are at least 20% longer than wild-type mice. Klotho expression also has similar effects on cognitive function as klotho-deficient mice develop cognitive impairment by 7 weeks of age and klotho-overexpressing mice show enhanced cognitive function. These effects also extend to humans as polymorphisms that alter circulating levels of klotho have likewise effects on lifespan and brain function. Despite the fact that modulation of klotho expression has reciprocal effects on cognitive function, …

Drugs of abuse increase dopamine concentrations in the nucleus accumbens, a key reward structure that integrates contextual and cue-related information and regulates motivated behavior. This surge of dopamine triggers cell signaling cascades that converge in the nucleus to cause changes in gene expression, which are thought to lead to the observed functional and structural alterations in the reward circuit after exposure to drugs of abuse. However, while various drugs of abuse cause transcriptional changes, previously available tools have not had the capacity to deeply characterize these gene programs. Therefore, we optimized a dual lentivirus CRISPR system for targeted gene modulation …
Intracellular Compartment-Specific Abnormalities In Proteasome Expression And Activity In The Superior Temporal Gyrus In Schizophrenia, Madeline R. Scott
Intracellular Compartment-Specific Abnormalities In Proteasome Expression And Activity In The Superior Temporal Gyrus In Schizophrenia, Madeline R. Scott
All ETDs from UAB
Protein homeostasis is an emerging component of schizophrenia (SZ) pathophysiology. Proteomic alterations in SZ are well-documented; however the underlying mechanism driving these changes remains unknown. The ubiquitin proteasome system (UPS) is fundamental to protein homeostasis regulation, and UPS transcript abnormalities in both blood and brain have been observed in SZ. Supporting a role for UPS dysfunction in SZ, we have reported decreased protein expression of ubiquitin-associated proteins in SZ brain. However, previous work on the proteasome has been limited to transcript expression. In the following studies we sought to address this gap in knowledge by characterizing proteasome dysfunction in the …

Mycobacterium avium subspecies paratuberculosis (Map) is the etiologic agent of Johne’s disease in ruminants and has been associated with Crohn’s disease in humans. An effective control of Map by either vaccines or chemoprophylaxis is a paramount need for veterinary and possibly human medicine. Given the importance of fatty acids in the biosynthesis of mycolic acids and the mycobacterial cell wall, we tested novel amphiphilic C10 and C18 cyclobutene and cyclobutane fatty acid derivatives for Map inhibition. Microdilution minimal inhibitory concentrations (MIC) with 5 or 7 week endpoints were measured in Middlebrook 7H9 base broth media. We compared the …

Right-sided congestive heart failure (brisket disease) commonly occurs in cattle raised at elevations >2,500– 3,500 m. We investigated clinical cases resembling brisket disease at a western Nebraska feedyard at a moderate altitude (1,369 m). Over a 15-mo period (2009–2010), we examined 17 cases (16 steers and 1 heifer), all purebred Angus. All animals had clinical right-sided heart failure: brisket and ventral abdominal edema, and severe chronic passive congestion of the liver. Gross examination confirmed right ventricular hypertrophy (left ventricle plus septum: right ventricle weight ratio mean: 1.33 vs. 2.8–4.0 reference interval). Microscopically, all 17 cases had interstitial fibrosis (mean score: …

Innate CD8 + T cells express a memory-like phenotype and demonstrate a strong cytotoxic capacity that is critical during the early phase of the host response to certain bacterial and viral infections. These cells arise in the thymus and depend on IL-4 and IL-15 for their development. Even though innate CD8 + T cells exist in the thymus of WT mice in low numbers, they are highly enriched in KO mice that lack certain kinases, leading to an increase in IL-4 production by thymic NKT cells. Our work describes that in C57BL/6 WT mice undergoing a Th1 biased infectious disease, …

Background/aim: Cyclosporine A (CsA), a traditional immunosuppressive compound, has been reported to specifically prevent isch-emia reperfusion tissue injury via apoptosis pathway. This study aimed to explore the renoprotective effects of CsA on the kidneys of rabbits undergoing renal pelvic perfusion.Materials and methods: A total of 30 rabbits were randomly assigned into a control group (n = 6) and an experimental group (n = 24). The experimental group underwent a surgical procedure that induced severe hydronephrosis and was then stochastically divided into 4 groups (S1, S1?, S2, and S2?), consisting of 6 rabbits each. Groups S1 and S1? were perfused with …

Periodontal disease, also known as periodontitis, is a dental disease that is estimated to affect 2.4 billion people worldwide. This disease is characterized by chronic inflammation and bleeding of gums, as well as loss of tooth bone density. Currently, treatments for the disease include dental surgeries and various types of antibiotics, but few are targeted specifically at the Red Complex bacteria which are strongly associated with chronic periodontitis. The Red Complex consists of three bacteria: Tannerella forsythia, Porphyromonas gingivalis and Treponema denticola. Each of these bacteria, are anaerobic oral pathogens that reside in the subgingival pocket adjacent to …

In addiction, an individual’s ability to inhibit drug seeking and drug taking is thought to reflect a pathological strengthening of drug-seeking behaviors or impairments in the capacity to control maladaptive behavior. These processes are not mutually exclusive and reflect drug-induced modifications within prefrontal cortical and nucleus accumbens circuits, however unlike psychostimulants such as cocaine, far less is known about the temporal, anatomical, and cellular dynamics of these changes. We discuss what is known regarding opioid-induced adaptations in intrinsic membrane physiology and pre-/postsynaptic neurotransmission in principle pyramidal and medium spiny neurons in the medial prefrontal cortex and nucleus accumbens from electrophysiological …
