Rates of lactate production and consumption reflect the metabolic state of many cell types, including neurons. Here, we investigated the effects of nutrient deprivation on lactate dynamics in Drosophila neurons by leveraging the limiting effects of the unstirred layer, a diffusion barrier that forms around cells in culture. We found that neurons constitutively consume lactate when the availability of trehalose, a glucose disaccharide preferred by insects, is limited by the unstirred layer. Acute mechanical disruption of the unstirred layer reduced this reliance on lactate. Through kinetic modeling and experimental validation, we demonstrate that neuronal lactate consumption rates under unstirred layer …

Trauma remains a leading cause of morbidity and mortality in part due to complex pathophysiological responses. Yet our abilities to predict these changes are limited. Plasma miRNAs have been proposed as DAMPs that drive immune response and organ injury. Here, we test a panel of plasma miRNAs-selected based on next-generation sequencing and nucleotide motifs identified via a machine-learning algorithm-for their abilities to predict subclinical pathophysiological injuries. We find marked and severity-dependent increases in the miRNA biomarkers following trauma, which are closely associated with various injury markers. AUROC indicates that these biomarkers possess strong diagnostic and predictive abilities in overall trauma …

Lysosomes are essential organelles for cellular homeostasis and signaling, with dysfunction linked to neurological disorders, lysosomal storage diseases, and cancer. While proteomics has advanced our understanding of lysosomal composition, the structural characterization of lysosomal membrane proteins in their native environment remains a significant challenge. Here, we developed a cryo electron tomography workflow to visualize lysosomal membrane proteins within intact, native lysosomal membranes. We isolated endolysosomes by independently targeting two lysosomal membrane proteins, transient receptor potential mucolipin 1 and transmembrane protein 192, enriching organelles that exhibited the expected morphology and proteomic composition of the endolysosomal system. Sub-tomogram averaging enabled the structural …

Comparative biology seeks to unlock the power of cross-species trait variation to learn the rules of life. In this venture, modern studies increasingly leverage large datasets spanning many traits and levels of biological organization and complexity. To analyze these complex data in a statistically-sound manner, researchers must choose a phylogeny that is assumed to model the mean trait values across species-an assumption that may be tenuous depending on the true evolutionary architecture of the traits. Yet the consequences of this decision remain poorly understood, particularly for modern studies seeking to analyze multiple, distinct traits within the same framework. Here, we …

Limb Regeneration In Damselflies Based On Developmental Stage, Haven O. Clements
Limb Regeneration In Damselflies Based On Developmental Stage, Haven O. Clements
Inquiry: The University of Arkansas Undergraduate Research Journal
Injury is common in wild animals, yet some species possess an adaptive advantage: regeneration. Enallagma vesperum, a species of damselfly, exhibits this regenerative ability, though its persistence and effectiveness across developmental stages remain poorly understood. While previous studies indicate that regeneration ceases following adult emergence, it is unclear whether this ability also diminishes prior to the final larval molt. This study investigated the effectiveness of limb regeneration in E. vesperum across early and late larval instars. I hypothesized that earlier instars would exhibit greater regenerative success due to fewer competing energetic demands compared to later instars, which may prioritize …

This project focused on the ecological restoration of a kudzu-invaded section of the Oconee River Greenway while integrating high-impact learning into the Environmental Methods and Measurements (ENSC 2000) course at GCSU. Kudzu (Pueraria montana), a Category I invasive species, has significantly disrupted native plant communities at the site, making it an ideal setting for applied learning and ecological intervention. The project engaged students in authentic environmental research by involving them in every stage of the process including problem assessment, methodology design, field implementation, data collection, and reflection.
Students initiated the first phase of invasive plant removal using mechanical …

This article captures an experience in conflict management that happened during the summer of 2024 as a recreational therapy intern with an adaptive sport and recreation organization located in the Southeastern United States. The backdrop to the case is that a recreational therapy intern worked with various special needs populations – mainly with paraplegic (adult and youth) athletes and veterans. In many organizations, user conflicts and poor communication regarding service provision and scheduling hinder the implementation of programs for special populations, such as veteran groups. In this case, a problem occurred with the scheduling of a water aerobics program for …

Drowning remains a significant public health problem in the United States, with The Centers for Disease Control and Prevention (CDC) reporting approximately 4,000 drowning deaths annually. Children aged 1-4 face the highest risk, and African American children are disproportionately affected–7.6 times more likely to drown than white children (CDC, 2024). In a city on the East Coast that is predominantly African American, limited access to swimming lessons heightens the risk. To reduce drowning rates, a collaborative swimming program was established between a city high school and a public school enrichment program. The initiative provided essential water safety and swimming skills …
