Recent algorithmic advances have enabled the inference of genome-wide ancestral recombination graphs (ARGs) from large genomic cohorts, providing detailed models of genealogical relatedness along the genome. These inferred ARGs can complement genotype imputation by capturing the effects of unobserved variants, but their use in large-scale linear mixed-model analyses has been computationally prohibitive. Here, we develop methods that leverage the ARG to perform genotype-matrix multiplications in sublinear time and implement scalable randomized algorithms for mixed-model analyses. We introduce ARG-RHE, a randomized Haseman-Elston approach for estimating narrow-sense heritability and performing region-based association testing using ARGs, enabling parallel analysis of multiple quantitative traits. …

Intergroup trust refers to a collective prediction and belief among members of one group regarding those of another in intergroup interactions. It plays an important role in avoiding intergroup conflicts, and thereby sustaining social development. This study was designed to investigate the effects of third-party punishment and compensation, as well as group regulatory focus, on intergroup trust. The findings revealed that, at both individual and group levels, prevention-focused groups exhibited higher levels of intergroup trust than promotion-focused groups under third-party compensation conditions. In contrast, under the third-party punishment conditions, promotion-focused groups displayed greater trust than prevention-focused groups, although this difference …

Adoptive cell therapies (ACT) leverage tumor-immune interactions to cure cancer. Despite promising phase I/II clinical trials of chimeric-antigen-receptor natural killer (CAR-NK) cell therapies, molecular mechanisms and cellular properties required to achieve clinical benefits in broad cancer spectra remain underexplored. While in vitro and in vivo experiments are essential, they are expensive, laborious, and limited to targeted investigations. Background: Digital health interventions (DHIs) can extend the reach of disease prevention interventions; however, few are evidence-based, theoretically grounded, or developed for high-risk youth and families. Co-design approaches engage end users in the design and development of the DHI, which can lead to increased accessibility and engagement.
Objective: This study aimed to describe the adaptation of an evidence-based diabetes prevention program for remote, digital delivery.

BACKGROUND: Sand flies are small hematophagous insects known as leishmaniasis vectors. Similar to most arthropods, they harbor nonobligate endosymbionts that may influence host adaptation and pathogen transmission, but these symbiont communities remain poorly characterized in Leishmania-endemic African sand flies. METHODS: We screened 1700 wild-caught phlebotomine sand flies (1266 females, 434 males) from Kenya's Baringo, Nakuru, and Kajiado counties, and 253 colony Phlebotomus duboscqi, for Rickettsia, Wolbachia, Spiroplasma, Cardinium, Arsenophonus, Microsporidia, and Leishmania by high-resolution melting analysis and sequencing of PCR products. RESULTS: In wild sand flies (Phlebotomus and Sergentomyia spp.), Wolbachia spp. were most common (8.5%, 145/1700), with particularly high …

Green Humanities: A Journal of Ecological Thought in Literature, Philosophy & the Arts
This article offers an ecological analysis of Malika Mokeddem's novel N'Zid (2001), moving beyond postcolonial and anthropocentric interpretations. It centers on the protagonist Nora's individuation, arguing this process is intrinsically linked to her evolving relationship with the aquatic environment from which she emerges. The article aims to situate N'Zid within an ecological perspective, particularly relevant in the Anthropocene, an era compelling a reassessment of humanity's place. Echoing Bruno Latour, addressing inter-relationality is essential as the stable physical framework of the Moderns becomes unstable. This view necessitates rethinking environmental issues and encourages a scientific interpretation of the novel, drawing on neurosciences, …

Snapping shrimp damp the shock waves they produce and use as weapons with a helmet-like structure termed the orbital hood. Here, we ask how structural and material properties contribute to shock wave damping by orbital hoods in Alpheus heterochaelis. Using tensile mechanical testing, we find orbital hoods are approximately half as stiff as carapace and have twice the capacity for viscous energy dissipation. Microstructural features probably contribute to tissue-specific mechanical properties: the endocuticles of orbital hoods have almost twice as many lamellae as those of carapaces despite being half as thick, suggesting a mechanism for enhanced material mobility underlying …

Vigorous debate has erupted over the trustworthiness of scientific research findings in a number of domains. The question "what makes research findings trustworthy?" elicits different answers depending on whether the emphasis is on research integrity and ethics, research methods, transparency, inclusion, assessment and peer review, or scholarly communication. Each provides partial insight. We offer a systems approach that focuses on whether the research is accountable, evaluable, well-formulated, has been evaluated, controls for bias, reduces error, and whether the claims are warranted by the evidence. We tie each of these components to measurable indicators of trustworthiness for evaluating the research itself, …

Study objectives: GGIR is an R package for processing raw acceleration data to estimate sleep health parameters. We aimed to (1) assess the performance of three sleep algorithms within GGIR against PSG for detecting sleep/wake in clinically referred, typically-developing children (criterion validity); and (2) describe GGIR-derived sleep estimates from typically developing children enrolled in multiple cohort studies (face validity).
Methods: For criterion evaluation, children (8-16 years, N = 30) wore an actigraphy device for one night during in-lab polysomnography with performance assessed using epoch-by-epoch analyses. Two closely homologous channelrhodopsins from Hyphochytrium catenoides, known as HcKCR1 (kalium channelrhodopsin 1) and HcCCR (cation channelrhodopsin), exhibit >100 times different K+/Na+ relative permeabilities and are emerging optogenetic tools for controlling neurons and cardiomyocytes. Key residue motifs and trimeric organization relate them structurally to bacteriorhodopsin-like cation channelrhodopsins (BCCRs). Here, we demonstrate that they utilize a specific gating mechanism previously suggested for the earlier-discovered BCCR from cryptophytes. Using a comparative analysis of transient absorption changes and photocurrents under single-turnover conditions, we identified an early, far-UV-absorbing photocycle intermediate that precedes channel opening in wild-type HcKCR1 and HcCCR. Avian influenza virus (AIV) remains a major challenge for global poultry health, food production, and agricultural economies. Both low-pathogenic (LPAIV) and highly pathogenic (HPAIV) strains continue to circulate and evolve, creating an ongoing need for vaccines that can be updated quickly and provide broad protection. Although DNA vaccines offer a safe and flexible approach to rapidly incorporate new antigens, their use in poultry has been limited by poor delivery efficiency and the vulnerability of naked DNA to degradation. This study investigated the phytochemical constituents and biological activities of Horsfieldia macrothyrsa leaves to identify their bioactive compounds. Methanol extracts were fractionated using n-hexane and ethyl acetate, followed by silica gel column chromatography with a stepwise polarity gradient. From 100 g of powdered leaves, three major compounds were successfully isolated from the ethyl acetate fraction: 1-(2,4,6-trihydroxyphenyl)dodecan-1-one (1), sesamin (2), and β-sitosterol (3). Their chemical structures were confirmed using UV, IR, LC–MS/MS, and NMR spectroscopy. This study presents the development of a novel multifunctional adsorbent Cu-BTC (HKUST-1)/Zeolite A4 composite functionalized with tannic acid—for the efficient removal of toxic heavy metal ions (Pb²⁺, Cd²⁺, Cr(VI)) from aqueous solutions. The composite was synthesized via a solvothermal process and systematically characterized by XRD, FTIR, SEM-EDS, and BET surface area analysis. The resulting material exhibited a high specific surface area (1050 m²/g), a hierarchical pore structure, and a homogeneous distribution of active sites. Batch adsorption experiments revealed superior adsorption capacities of 220 mg/g for Pb²⁺, 180 mg/g for Cd²⁺, and 150 mg/g for Cr(VI) at pH 6, surpassing those …

Activation of cGAS-STING signaling in cancer cells requires cytosolic DNA produced by intrinsic or treatment-induced DNA damage. However, clinical efforts to exploit this pathway to improve immunotherapy have yielded limited success, highlighting gaps in understanding the link between DNA damage and immunotherapy. Here, we identify ubiquitination-directed cytosolic DNA degradation as a critical determinant for cGAS-STING activation following DNA damage. Mechanistically, the cytosolic DNA exonuclease TREX1 is degraded by the E3 ubiquitin ligase SPOP but is reversely stabilized by the deubiquitinase USP7. The co-evolution of different cell subsets in the progression of precursor lesions to lung adenocarcinoma (LUAD) is incompletely understood. We generated spatial transcriptomic maps of 56 human precursor lesions and LUADs from 25 patients and of an independent cohort of 36 lesions from 19 patients, analyzing a total of 486,519 spots and 5.4 million cells. We identify region-specific programs that distinguish precursors from LUADs. Spatially resolved clonal architectures reveal patient-specific heterogeneity in evolution of precursors to LUADs. We find epithelial alveolar progenitors expressing tumor-associated meta-programs and residing in niches enriched with proinflammatory subsets including IL1B high macrophages. Proper regulation of ribosome biogenesis is essential for stem cell function and tissue homeostasis, yet its upstream control in adult intestinal stem cells (ISCs) remains unclear. Here, we identify the WD repeat protein Wdr4 as a key regulator of ISC homeostasis in the Drosophila midgut. Wdr4 cooperates with the methyltransferase Mettl1 to catalyze N⁷-methylguanosine (m⁷G) modification of let-7 miRNA. Wdr4 or Mettl1 depletion disrupts this modification, reducing let-7 levels and aberrantly activating TOR-JNK-dMyc signaling. This drives elevated ribosome biogenesis, ISC overproliferation, misdifferentiation, and intestinal dysplasia. Overexpression of let-7, inhibition of TOR, or suppression of JNK rescues these defects. HER2-low breast cancer (IHC 1+ or 2+ without HER2 gene amplification) is a distinct and understudied subtype. In a cohort of 14,593 early-stage breast cancer patients treated at MD Anderson from 2006 to 2019, 60.4% were HER2-low. Multivariable analysis showed HER2-low status was independently associated with race, histologic subtype, higher nuclear grade and stage, and estrogen receptor (ER) positivity. Among 2464 patients receiving neoadjuvant chemotherapy, HER2-low status was not linked to pathologic complete response (pCR), overall survival (OS), or disease-free survival (DFS) compared to HER2-0 tumors. Rapid and comprehensive analysis of complex proteomes across large sample sets is vital for unlocking the potential of systems biology. We present a high-throughput mass spectrometry (MS) proteomics method that integrates narrow-window data-independent acquisition (nDIA) with short-gradient micro-flow chromatography, enabling profiling of >240 samples per day. This optimized MS approach identifies 6,201 and 7,466 human proteins with 1- and 2-min gradients, respectively. As a practical application, we analyzed 507 samples composed of 13 different tissues from mice treated with the enzyme-drug L-asparaginase (ASNase) or its glutaminase-free Q59L mutant, generating a quantitative profile of 11,472 proteins following drug treatment. This study presents an optimized design and operation strategy for a stand-alone solar photovoltaic power system to drive a 4.5 kW elevator at Mansoura University Hospital, Egypt. The research aims to reduce the battery bank capacity and overall system cost while maximizing the utilization of solar panels. A novel "Optimum Operator System" is proposed, incorporating three power switches controlled by an Arduino microcontroller. The system's performance is analyzed through extensive modeling, simulation, and experimental validation. Results show a significant reduction in battery bank capacity, with 7 fewer 100 Ah batteries required compared to conventional systems. The proposed method significantly enhances …

This study examines the effects of suction chamber baffle configurations on both hydraulic efficiency and cavitation characteristics in a low specific speed centrifugal pump. Four baffle configurations (zero, one, two, and three baffles) were tested at three different heights using refined computational fluid dynamics simulations. The Zwart-Gerber-Belamri cavitation model was enhanced by adjusting nucleation site radius and evaporation coefficient values to better align with previous research findings. Results revealed that the optimal baffle height is approximately one-third of the impeller suction eye radius, significantly improving cavitation resistance. Conventional foam concrete suffers from low strength and heterogeneous pore structure due to its porous structure and incomplete hydration, limiting its structural applications. This study investigates the use of magnetized water (MW) as a mixing medium to enhance hydration efficiency and improve both fresh and hardened properties of foam concrete. Three density grades 620, 1300, and 1900 kg/m³, designated as Groups A, B, and C were prepared and tested. The p53 tumor suppressor protein plays a central role in maintaining genomic stability by regulating cell cycle arrest, apoptosis, and DNA repair under cellular stress. Mouse double minute 2 (MDM2), an E3 ubiquitin ligase, negatively regulates p53 via direct binding and proteasomal degradation. Overexpression or amplification of MDM2 can disrupt this pathway and promote tumorigenesis, even in cancers with wild-type p53. This review outlines the structural features of MDM2, particularly its N-terminal hydrophobic pocket and C-terminal RING domain, and their roles in p53 regulation.
