Chondrosarcoma (CS) are common bone cancers that produce cartilaginous tumors and are extremely refractive to chemo-and-radiation therapies, leaving very limited treatment options. They contain mutations in the isocitrate dehydrogenase genes, IDH1 or IDH2. Either IDH1 or IDH2 mutations are present in individual tumors, but not both. They both convert alphaketoglutarate (αKG) into the potent oncometabolite D-2-hydroxyglutarate (D2HG). D2HG can be transported from the tumors to cells of the tumor microenvironment (TME) where it can alter metabolic and epigenetic profiles in recipient cells. The process of α-KG to D2HG conversion occurs in the cytoplasm of IDH1 mutant cells, and in the …

This dataset supports the production, characterization, and biological validation of a novel recombinant hagfish intermediate filament (rHIF) protein functionalized with spider silk motifs (rHIF-alpha-GRGGL). The data demonstrates a scalable bioprocess resulting in a 7.76 g/L yield, high structural alpha-helicity (77.2%), and significant support for N2a cells.

The broadly conserved ParB protein performs crucial functions in bacterial chromosome segregation and replication regulation. The cellular function of ParB requires it to dimerize, recognize parS DNA sequences, clamp on DNA, then slide to adjacent sequences through nonspecific DNA binding. How ParB coordinates nonspecific DNA binding and sliding remains elusive. Here, we combine multiple in vitro biophysical and computational tools and in vivo approaches to address this question. We found that five conserved lysine residues in the C-terminal domain (CTD) of ParB play distinct roles. While the two central rigid lysine residues provide the structural platform for the CTD, the …

The nexus between food security and armed conflict, with particular focus on developing countries, has been analyzed by a few studies in the literature. Every country under armed conflict is obligated to solve related food security issues. Therefore, the overall objective of this study was to analyze the nexus between households’ food security and armed conflicts in the context of climate change in Mali. To achieve this objective, first, the study used an ordinary least squares model to determine the factors influencing households’ food security. This review offers a comprehensive analysis of the expanding role of artificial intelligence (AI) in mycological research. It highlights how AI supports the identification of fungal taxa at both microscopic and macroscopic levels, facilitates the detection of toxic species, contributes to ecological mapping, aids in discovering new fungal bioproducts, and enhances precision agriculture practices. Advanced techniques such as convolutional neural networks, you only look once, U-shaped network (UNet), and residual network have demonstrated strong performance in classification, segmentation, and object detection tasks. Boron (B) toxicity disrupts multiple metabolic and signaling processes in plants, yet its interaction with post-translational regulators remains poorly understood. The E3 ubiquitin ligase NITROGEN LIMITATION ADAPTATION (NLA) is known to control nutrient transporter turnover, but its role in B toxicity responses has not been previously characterized. Here, we present the first genome-wide transcriptomic analysis of nla mutant Arabidopsis thaliana exposed to mild (1 mM) and moderate (2 mM) B toxicity. This research was conducted between 2020 and 2022 in 21 villages located on the Kozak Plateau and its surroundings, in the Bergama district of İzmir province, Türkiye. The main aim of the study is to document the medicinal uses of plants by the local population and to contribute to the preservation and transmission of ethnobotanical knowledge—passed down through generations— to future generations before it disappears. Interviews were conducted with 159 informants, and the traditional medicinal uses of 126 plant taxa belonging to 54 families were identified. Among these, the therapeutic uses of 37 taxa were recorded for the first time. …

However, only a small percentage of CHDs has a known genetic cause, highlighting the need for research surrounding heart development and the pathogenesis of CHDs. Heart development research aims to elucidate the precise coordination of gene expression and cell signaling with the goal of advancing prevention and treatment methods. Rising global temperatures due to climate change threaten optimal strawberry growth and sustainable yields. Limited data exist on the physiological and biochemical responses of strawberry cultivars to high-temperature stress and their tolerance mechanisms. T his study evaluated the responses of 21 day-neutral and short-day strawberry cultivars to high-temperature stress and classified their tolerance levels using a novel weighted rating method. Plants were grown in 15-cm plastic pots (3:1:1 garden soil, peat, perlite) under open-field conditions until the 3–4 leaf stage and then transferred to climate chambers. Exploring the trait structure of grassland plant communities enhances our understanding of how these communities will respond to climate change and disturbances such as herbivory. The trait structure of a plant community was investigated in the Central Anatolian steppe, an ecoregion with high biodiversity that has been largely unexplored in terms of plant functional ecology. Conservation science and practice mirror racist notions of colonialism, prolonging a cycle in which policies and programs disregard land access. Many conservation leaders operate from a culture of land exclusion, focusing on nature to generate wealth, deeming the number of individuals removed from the commons as progress. By the turn of the 21st Century, government conservation policies compensated private actors and groups for voluntarily adopting market-based mechanisms aimed at land conservation. Despite the emergence of various conservation programs, the data show that White Americans primarily benefit from federal conservation payments. Heavy metals are natural components of the environment and can be relatively non-impactful in low concentrations. However, high concentrations of heavy metal toxicity can result in air, water, and soil contamination that adversely impacts environmental, human, and wildlife health. This study focuses on detecting the presence of heavy metals in soil, Loblolly Pine tree bark, and wood core samples from sites near a landfill in the Tallassee, Alabama, community and a post-agricultural site in Tuskegee, Alabama. The system of rice intensification (SRI) was implemented in Mali to boost rice productivity. The objectives of this paper are to identify the constraints faced by SRI adopters, estimate the food security index, and assess the impact of in-depth adoption of SRI on smallholder farm households’ food security. The study uses Kendall’s ranking to rank the six main constraints identified from the literature. “Clemson Spineless” and “Jambalaya” okra were grown in a Tunnel House and field to compare yield and other growth parameters. A split-plot design with three replications/treatment was used; the Tunnel House and field were main plots, and varieties split plot. Plant height was significantly taller, and pod harvesting began two weeks earlier in the Tunnel House. Yield showed no significant differences between varieties, and Tunnel House vs. field-produced okra. However, “Clemson Spineless” produced 9% higher pod yield in the Tunnel House than “Jambalaya,” which had 23% higher field yield than “Clemson Spineless.” An enterprise budget analysis showed that Tunnel House …

The genus Aethionema in Türkiye was investigated through a comprehensive taxonomic study including morphological, seed micromorphological, and molecular methods. This study led to the synonymization of 11 taxa: A. annuum, A. dincii subsp. baytopiae, A. alidaghenicum, A. bingoelicum, A. dumelicum, A. ertughrulii, A. fruticulosum, A. gypsicola, A. huber-morathii, A. maraschicum, and A. turanicum. Conversely, seven species were resurrected: A. levandowskyi, A. sintenisii, A. acarii, A. pallidiflorum, A. pseudoarmenum, A. pulchellum, and A. recurvum. Small cell carcinoma is a highly lethal cancer variant often found with neuroendocrine (NE) features, as exemplified by small cell lung cancer and small cell NE prostate cancer (SCPC). A genome-wide CRISPR dependency screen using SCPC models generated through human prostate cell transformation identifies a requirement for the transcription factor E2F3. E2F3 dependency is linked to RB inactivation, a near universal occurrence across small cell cancers. The requirement for E2F3 is shared by RB-deficient cells originating from the prostate, lung, and adnexa. In RB-deficient cancer cells, E2F3 inhibition restrains cell cycle progression, proliferation, and tumor growth in vivo. Inhibition of …

Radish is a widely cultivated vegetable with known medicinal value and produces siliques that differ significantly in number and size. The present study compares normal Duanye-13 radish (CK-type) siliques with mutant radish siliques induced by chemical mutagenesis at the young (YS), short (SS), and long (LS) silique stages. Differences in long noncoding ribonucleic acid (lncRNA) expression were investigated using high-throughput RNA sequencing and bioinformatic analyses, revealing 45 324 new lncRNAs in radish siliques. lncRNA expression was higher in YS and LS mutant lines than in CK-type lines, and expression in SS mutants tended to be lower than in CK-type lines. …

Lower gastrointestinal graft-versus-host disease (LGI-GVHD) carries morbidity and mortality for patients undergoing allogeneic hematopoietic stem cell transplantation (allo-HSCT), with critical contributions from the intestinal microbiome. In a retrospective cohort of metagenomic sequencing of stool from patients with allo-HSCT (N = 90), we found that a reduction in specific Parabacteroides and Bacteroides species around the time of engraftment contributes to LGI-GVHD risk. Given the known diverse carbohydrate-degrading functionality of these bacteria, we investigated gene abundances for carbohydrate-active enzymes (CAZymes) and found that Parabacteroides merdae, P distasonis, and Bacteroides ovatus abundances were significantly correlated with CAZymes in patients who did not develop …

Resistance to HIF-2α inhibitors such as Belzutifan underscores the need to better understand how HIF-2α is transcriptionally regulated in clear cell renal cell carcinoma (ccRCC). Here, we uncover a cytokine-driven enhancer mechanism that sustains HIF-2α expression through the JAK1-STAT3 signaling pathway. Using a genome-wide CRISPR screen in VHL-deficient ccRCC cells, we identified SOCS3 as a key negative regulator of HIF-2α. Mechanistically, loss of SOCS3 activates JAK1-STAT3 signaling, leading to the recruitment of STAT3 to distal enhancers upstream of EPAS1 that physically loop to its promoter to drive HIF-2α transcription. Background: The present study aimed to systematically develop and validate a robust, rapid, and stability-indicating reversed-phase high-performance liquid chromatography (RP-HPLC) method for the simultaneous estimation of metformin and glimepiride in fixeddose combination tablets. Method development was guided by a design of experiments (DoE) approach using the Box-Behnken design (BBD) to ensure optimal analytical performance.
