Cyanogenesis Variation In White Clover Is Decoupled From Regional Climate And Human Footprint Across A Longitudinal Gradient, Philips O. Akinwole, Yangjie Tan, Victor L. Alfonso

Biology Faculty publications

Latitudinal clines in adaptive traits are well documented, yet how such patterns translate across longitudinal gradients, where climatic variation is reduced, remains poorly understood. We investigated spatial variation in hydrogen cyanide (HCN) - a well-characterized anti-herbivore defense polymorphism governed by two independently segregating loci (Ac and Li), in white clover (Trifolium repens) across nine Midwestern U.S. cities. A total of 269 populations (4,035 individuals) were sampled along urban-rural transects, and cyanotype (cyanogenesis phenotypes) frequencies were quantified alongside regional and site-level environmental variables, including vegetation cover (NDVI), land surface temperature (LST), snow cover, impervious surface intensity, aridity, …

In Vitro Reversal Of Abc Transporter Mediated Multidrug Resistance In Human Ovarian And Non-Small Cell Lung Cancer Models, Alison K. Kellom, Pia D. Vogel May 2026

In Vitro Reversal Of Abc Transporter Mediated Multidrug Resistance In Human Ovarian And Non-Small Cell Lung Cancer Models, Alison K. Kellom, Pia D. Vogel

Biological Sciences Theses and Dissertations

One of the major causes of treatment failure in aggressive cancers is multidrug resistance (MDR), which is linked to the overexpression of membrane efflux proteins that export chemotherapeutics from cancer cells. This mechanism prevents chemotherapeutic drugs from reaching cytotoxic concentrations intracellularly, allowing the cancer to survive. Two primary mediators of this mechanism are P-glycoprotein (P-gp) and Breast Cancer Resistance Protein (BCRP). P-gp and BCRP are transmembrane ATP-binding cassette (ABC) transporters that are frequently overexpressed in MDR cancers. They utilize the binding and hydrolysis of ATP to transport a diverse range of amphipathic molecules of varying size (Schinkel and Jonker, 2003), …

Acute And Chronic Oral Administration Of Cannabidiol-Dominant Cannabis Products: Sex- And Formulation-Dependent Effects On Anxiety-Like Behavioral Patterns In Rats, Carlos R. Maestas-Olguin May 2026

Acute And Chronic Oral Administration Of Cannabidiol-Dominant Cannabis Products: Sex- And Formulation-Dependent Effects On Anxiety-Like Behavioral Patterns In Rats, Carlos R. Maestas-Olguin

Psychology ETDs

Interest in the anxiolytic-like effects of cannabidiol (CBD), the non-intoxicating phytocannabinoid found within the genus, Cannabis, has grown significantly in recent years. However, preclinical investigations of CBD’s anxiolytic potential often elect to forgo translationally relevant models that maximize external validity in favor of greater internal and mechanistic validity. As a result, the effects of relatively low dose (< 3mg/kg) oral CBD administration on anxiety-like behaviors are not yet fully characterized. The present studies examined the impact of two orally delivered CBD-dominant cannabis products – a “full-spectrum” formulation derived from the whole plant and a CBD isolate derived from the same chemovar – on innate defensive behaviors exhibited by adult male and female rats in various tests of anxiety. Results indicated that acute and chronic low dose oral CBD administration attenuated anxiety-like behavior in rats, regardless of sex or formulation. These findings extend the growing corpus of literature describing anxiolytic-like effects of CBD.

The Concept 'Pollination' And Bee Pan-Trap Color And Uv Reflectance Determine The Taxonomic Compositions Of Bee Taxa Captures Across An Elevation Gradient And Seasons In The Jemez Mountains Of New Mexico, Esther A. The Concept 'Pollination' And Bee Pan-Trap Color And Uv Reflectance Determine The Taxonomic Compositions Of Bee Taxa Captures Across An Elevation Gradient And Seasons In The Jemez Mountains Of New Mexico, Esther A. Burton, Kelly B. Miller, Paul J. Watson, David C. Witherington, Steven Poe, David Lightfoot

Biology ETDs

This thesis examines the pollinator concept and tests how sampling methods shape bee communities. Chapter 1 provides a conceptual and philosophical analysis, arguing that pollination is a process and that pollinators are context-dependent functional roles rather than fixed taxonomic categories. It traces the concept’s development from natural history to ecological theory, introduces a dual-goal model separating plant reproduction from visitor foraging, and proposes a classification framework based on evidentiary certainty and pollination potential to resolve ambiguity in pollination research.

Chapter 2 applies these ideas empirically by evaluating how pan-trap color and ultraviolet (UV) reflectance influence bee assemblages across elevation and …

From Cranes To Clinics: Genomic Epidemiology At The Human-Animal Interface, Sarah Shrum May 2026

From Cranes To Clinics: Genomic Epidemiology At The Human-Animal Interface, Sarah Shrum

Biology ETDs

Emerging infectious diseases represent a significant, increasing threat to human life, economic stability, and global security. The One Health framework, which promotes human health and wellness through the understanding that animal, environmental, and human health are interwoven, is critical to understanding pathogen emergence and transmission. This is underscored by the fact that roughly 60% of all human pathogens and 75% of emerging infectious disease are zoonotic, meaning they are transmitted from animals to humans.

The work presented in this dissertation integrates genomic epidemiology, ecological analysis, and One Health Surveillance approaches to characterize cross-species circulation of opportunistic yeast pathogens (Candida …

Investigating Changes In The Stable Isotopic Composition Of Juniperus Osteosperma Over 34,000 Years Of Climatic Shifts, Lisa S. Garcia May 2026

Investigating Changes In The Stable Isotopic Composition Of Juniperus Osteosperma Over 34,000 Years Of Climatic Shifts, Lisa S. Garcia

Biology ETDs

Isotopic compositions of plants from the past provides empirical evidence for paleoclimatic shifts and plants physiologically response. This study used Neotoma paleomiddens (fossilized debris piles) collected from Death Valley National Park, which contained preserved abundant plant materials. These middens range from modern to >35,000 years and capture different climatic events in Earth history. Employing Juniperus osteosperma (Utah Juniper) leaves found in middens, I investigated variation in carbon and nitrogen stable isotopic composition. To help interpret paleomidden results, we used modern field and herbaria samples of Juniperus from a spatial gradient stretching across their geographic range. Results showed δ 13C …

Reassembling Riparian Ecosystems: Stream Restoration Effects On Vegetation, Arthropods, And Aquatic-Terrestrial Connectivity, Katelyn P. Driscoll May 2026

Reassembling Riparian Ecosystems: Stream Restoration Effects On Vegetation, Arthropods, And Aquatic-Terrestrial Connectivity, Katelyn P. Driscoll

Biology ETDs

Riparian areas exist at the aquatic-terrestrial interface and provide disproportionate ecosystem services, yet multiple stressors have degraded these zones globally. Stream restoration seeks to reverse degradation, but rigorous evaluation of treatment effectiveness remains limited. I conducted a before-after-control-impact (BACI) study in the southern Rocky Mountains to evaluate two stream restoration treatments—beaver dam analogs (BDAs) and plug-and-ponds (P&Ps)—across three dimensions of riparian ecosystem response. Using Bayesian analysis, I quantified treatment effects on functional diversity and composition of riparian plant communities. Using generalized linear mixed models, I evaluated restoration effects on riparian arthropod abundance, richness and guild composition across lateral gradients and …

Rhythms Of Hippocampal Function And Memory After Moderate Prenatal Alcohol Exposure, Lilliana May Sanchez May 2026

Rhythms Of Hippocampal Function And Memory After Moderate Prenatal Alcohol Exposure, Lilliana May Sanchez

Psychology ETDs

Fetal Alcohol Spectrum Disorders involve morphologic and neurobiological abnormalities in offspring subjected to prenatal alcohol exposure. Although not as well studied as high exposure, moderate prenatal alcohol exposure represents the most common and undetected form in humans. While learning and memory deficits have been well characterized at the behavioral level in humans with Fetal Alcohol Spectrum Disorders and in animal models of moderate prenatal alcohol exposure, few studies have determined the circuit or systems level mechanisms of the behavioral deficits. A complete understanding of the neural bases of memory deficits after moderate prenatal alcohol exposure is needed to ultimately identify …

What Is Mine, Yours And Ours: Bodily Metaphor And Neuropsychology, Alicia K.E Parham

Art Theses and Dissertations

Mapping the landscape of the brain has taken human lifetimes to navigate, both structurally to understand how our biological functions interact and functionally as our thoughts and emotions shape our existence. Neurological stimulus and response inform our reality, shape our thinking, and affect our bodies in profound physiological ways. Our nervous system acts as a bridge from what we experience physically to what we feel emotionally, through the sending of electrical signals to all parts of our body.

My art practice explores the interactions of neuroscience with outside stimuli. It is rooted in the personal: learning that a neurological condition …

Characterizing A Potential Cbp/P300 Histone Acetyltransferase Homolog In Tardigrade Germline Specification And Development, Brittney Michelle Hartshorn May 2026

Characterizing A Potential Cbp/P300 Histone Acetyltransferase Homolog In Tardigrade Germline Specification And Development, Brittney Michelle Hartshorn

Honors Projects

Germline specification is a fundamental process that ensures the proper development of germ cells. Germline specification occurs either through germ cell determinants maternally inherited through the mechanism of preformation or through induction, where cell interaction and epigenetic modifications direct specification. Determining how these mechanisms evolved and diverged remains an important question in developmental biology. Despite research on germline regulation in model organisms such as Caenorhabditis elegans or Drosophila melanogaster, such regulation remains poorly understood in many other animal groups. Tardigrades are an emerging model system for investigating developmental mechanisms due to their phylogenetic placement and experimentally tractable embryos. CBP/P300 is …

Microtubules In Breast Cancer: Exploring The Α/Β-Tubulin Toggle Switch And Its Implications In Human Breast Cancer, Annemarie Ianos May 2026

Microtubules In Breast Cancer: Exploring The Α/Β-Tubulin Toggle Switch And Its Implications In Human Breast Cancer, Annemarie Ianos

Student Theses and Dissertations

Microtubules, composed of a/b-tubulin heterodimers, play a central role in breast cancer tumor growth by polymerizing, leading to metastasis and depolymerizing, contributing to proliferation. Human enzymes protein kinase Ca (PKC-a) and cyclin-dependent kinase 1 (Cdk-1) mediate phosphorylation at sites a:Ser165 and b:Ser172, respectively, influencing the growth of microtubules. It is possible that alternating phosphorylation at these sites contribute to an a/b-tubulin “toggle switch” that mediates microtubule instability and tumor growth.

The project investigates the influence of the toggle switch model on microtubule stability by determining the impact of mutants (a:S165D, a:S165N, a:S165SP, b:S172SP and a:S165SP/b:S172S …

Department of Medicine Faculty Papers

Vesicoureteral reflux (VUR) is a risk factor for urinary tract infection (UTI). However, data describing the urobiome in children with VUR are limited. Here, we characterized the urobiome in children with and without VUR to better understand its potential role in VUR pathogenesis. Catheterized urine samples were obtained from children who presented for voiding cystourethrogram (VCUG) between 12/1/2020 and 10/3/2023. Children were excluded if they had any surgical procedures of the genitourinary tract in the 30 days prior to VCUG or were on treatment antibiotics at the time of VCUG. 16S rRNA gene sequencing and taxonomical classification identified the urobiome. …

The Effect Of Acute Hypoxia Exposure, With Exercise, On Bone Metabolic Biomarkers, Alyssa Bailly May 2026

The Effect Of Acute Hypoxia Exposure, With Exercise, On Bone Metabolic Biomarkers, Alyssa Bailly

Health, Exercise, and Sports Sciences ETDs

Hypoxic environments, resulting from decreased atmospheric partial pressure of oxygen, reduce oxygen availability in the blood and tissues, leading to a mismatch between supply and demand. This elicits several physiological adaptations and may influence bone metabolism. Although evidence suggests that hypoxia can modulate bone remodeling through hypoxia-inducible pathways, findings remain mixed, with most data derived from chronic hypoxic exposures in animal and cell culture models. Exercise, an essential stimulus for bone health, may exacerbate hypoxic stress; however, the skeletal response to acute hypoxic exposure combined with exercise remains unclear. We propose that intermittent exercise in hypoxia may increase negative markers …

Patterns Of Molecular Evolution Driving Reproductive And Genomic Processes In Sauropsids, Taylor Green May 2026

Patterns Of Molecular Evolution Driving Reproductive And Genomic Processes In Sauropsids, Taylor Green

Theses and Dissertations

Sexual reproduction is an important driver of genetic diversity and speciation through such forces as selection, mutation, and drift. Sauropsids are an appropriate clade to study shared molecular drivers of sexual and genomic evolution due to their genome architecture, sex determination mechanism (SDM) plasticity, and large syntenous genomic regions. Using comparative phylogenetic methods, I identify predictable patterns of selection in meiotic recombination genes and clade-specific drivers of recombination rate evolution. Then, leveraging whole genome sequencing, I demonstrate that American alligator genomes are characterized by an ancient loss of genetic diversity driven by drift and historic inbreeding. I also generate a …

Mllt1 Is Required To Maintain B-Lymphopoiesis, Janani Prakash May 2026

Mllt1 Is Required To Maintain B-Lymphopoiesis, Janani Prakash

Dissertations

MLLT1 (also named ENL) is an epigenetic reader protein that binds to crotonylated or acetylated lysine residues of histone 3 through its N-terminal YEATS domain. Positive and negative regulatory complexes compete for binding to C-terminal MLLT1, and promote transcription elongation (SEC, Super elongation complex; DOT1L) or transcriptional repression (PRC1, Polycomb repressive complex 1). MLLT1 was initially identified as a chromosomal translocation partner to MLL(KMT2A) in a subset of MLL-rearranged leukemias. MLL-ENL fusion protein causes both lymphoid (predominantly B-ALL) and mixed lineage leukemias. MLLT1 has been implicated in leukemic stem cell survival in MLL-rearranged cell lines but its role in normal …

The Extraction And Chemical Characterization Of The Avian Pigments Turacin And Turacoverdin, Sarah R. Bekkali May 2026

The Extraction And Chemical Characterization Of The Avian Pigments Turacin And Turacoverdin, Sarah R. Bekkali

Honors Scholar Theses

Bird coloration is a trait that extends beyond mere aesthetics as it has an extensive range of biological significance. Plumage patterns and hues can influence camouflage, mate choice, social dominance, and physiological performance. Bird fitness, their ability to survive and reproduce, is greatly dependent on color. Melanins, carotenoids, and pterins are well-studied pigment systems that are commonly found across many avian species’. Alternatively, porphyrin-based pigments are rare and less-studied as they only found in turacos a sub-Saharan African bird belonging to the family Musophagidae. This thesis focuses on two pigments of interest: turacin, the deep crimson-red pigment found in …

Seton Hall University Dissertations and Theses (ETDs)

Francisella tularensis is a highly infectious intracellular bacterium and the causative agent of tularemia, with pulmonary infection resulting in severe disease and high mortality if untreated. While protective immunity to F. tularensis has been largely attributed to CD4⁺ T cell–mediated IFN-γ responses, the contribution of γδ T cells to mucosal immunity and vaccine-induced protection remains poorly defined. The objective of this study was to characterize the role of γδ T cells during primary pulmonary infection with F. tularensis live vaccine strain (LVS) and following intranasal immunization with inactivated F. tularensis (iFt) prior to LVS challenge.

Using a murine intranasal infection …

Exploring Health Insurance’S Role In Chronic Disease Management Among Hispanics, Romelyn Dones May 2026

Exploring Health Insurance’S Role In Chronic Disease Management Among Hispanics, Romelyn Dones

Seton Hall University Dissertations and Theses (ETDs)

This qualitative study examined how insured non-English- and English-speaking Hispanic adults perceive the role of health insurance in the management of chronic disease. Chronic conditions remain a leading cause of morbidity in the United States, and although prior research suggests that insurance coverage improves access to care, it does not always enhance how individuals understand or utilize support for chronic disease management. Limited research has explored how individuals with chronic conditions interpret the role of insurance in their daily self-management practices, particularly among Hispanic populations. Guided by Andersen’s Behavioral Model of Health Services Use, this study explored how contextual, enabling, …

Contemporary U.S. Anthromes As Defined By Hanpp Regimes, Aishwarya Chandrasekaran, Kat F. Fowler, Christopher Lant

Environment and Society Faculty Publications

The concepts of anthromes and human appropriation of net primary production (HANPP) are both valuable in understanding our human-dominated planet, yet they have never been integrated theoretically or empirically. Here we utilize an extensive county-level dataset on HANPP and its product-level components to derive, through cluster analysis, ten contemporary US anthromes. From highest to lowest density of harvested HANPP, the anthromes are: rainfed corn–soy, dairy fodder, spring wheat–small grain, dryland winter wheat, subtropical soy–cotton, commercial timber, mixed hardwood and pasture, recovered eastern forest, prairie–sagebrush rangeland, and arid and alpine sparse grazing. Expanding to thirteen anthromes maintains these, while bifurcating the …

Accuracy Of Parameter Estimation For A Simple Gene Regulatory Network Model Is Sensitive To Network Motif, Number Of Parameters Estimated, And Magnitude And Direction Of Regulatory Relationships, Nikki C. Chun, Kam Dahlquist

Honors Thesis

A gene regulatory network (GRN) is a set of transcription factors that regulate the expression of genes encoding other transcription factors. The dynamics of a GRN explain how gene expression changes over time. GRNmap is a MATLAB software package that uses ordinary differential equations to model dynamics of small-scale GRNs. We used the program to estimate production rates, expression thresholds, and regulatory weights for each transcription factor in three related literature-derived GRNs based on yeast cold shock microarray data previously collected in the Dahlquist Lab. We noticed large differences in estimated weight values when 1-2% of the expression values were …

Biology, Chemistry, and Environmental Sciences Faculty Articles and Research

Eukaryotic pre-mRNA processing steps, including splicing and 3′ processing, are tightly coordinated, yet the underlying mechanisms remain incompletely understood. U1 snRNP has been proposed to inhibit 3′ processing at intronic polyadenylation (IPA) sites through a splicing-independent mechanism termed telescripting. In contrast, we discovered that disrupting splicing by using six different methods—targeting various key components such as U1 snRNP, U2 snRNP, U2AF, and SF3b—activates 3′ processing at thousands of IPA sites. Notably, splicing inhibition, especially of U1 snRNP, induced widespread premature transcription termination within gene bodies through both IPA-coupled and IPA-independent mechanisms. Inhibition of different splicing factors activated overlapping and distinct …

Department of Pharmacology, Physiology, and Cancer Biology Faculty Papers

Acute myeloid leukemia (AML) is a fatal blood cancer with cytotoxic chemotherapy offering at best 25% 5-year survival. While targeted BCL2 and FLT3 inhibitors venetoclax and gilteritinib are used upfront in the treatment of a subset of adult patients with AML and help to extend the survival of some patients, a curative treatment combination with minimal side effects has yet to be discovered. We find that use of the dual histone acetyltransferase p300/CBP bromodomain inhibitor CCS1477 (inobrodib), together with venetoclax and gilteritinib, virtually eliminates leukemia stem cells in an aggressive preclinical model of DNMT3A/FLT3-mutant AML by impairing pro-oncogenic survival and …
