Advancing Paradigm Of Quantitative Synthetic Biology And Strengthening Mission-Oriented Basic Research, Chenli Liu
Bulletin of Chinese Academy of Sciences (Chinese Version)
Synthetic biology is an important frontier in the life sciences, yet its further development remains constrained by limited capacity for rational design. Quantitative synthetic biology, which iteratively integrates quantitative modeling with the construction and analysis of synthetic systems, offers a promising paradigm for addressing this challenge. In this perspective, the study discusses the background and motivations underlying the emergence of this field and summarizes recent developments and exploratory efforts. Considering the unique characteristics of quantitative synthetic biology, it is suggested that progress in the field may benefit from strategically organized collaborative research that balances mission-oriented goals with investigator-driven exploration. Birch’s book is a remarkable exploration of the nature of sentience, its moral salience, evidence for its presence, and how to arrive at humane policies regarding our treatment of human beings, animals, brain organoids, and AI systems of uncertain sentience. Although Birch endorses the view that sentience is of fundamental moral importance, he endeavors to remain neutral among reasonable moral views and work out how to navigate through uncertainty—both ethical and epistemological—to arrive at defensible policies. While making common cause with Birch’s project, my own recent book advances a developed case in favor of the “sentientism” Birch endorses.

The peripheral nervous system relays signals between the central nervous system and the rest of the body, and peripheral nerve injuries and neuropathies can cause permanent neurological deficits. Schwann cells (SCs) are glial cells that myelinate neurons, which is essential for neuronal function and nerve repair. CAP1 (Cyclase-Associated Protein 1) is best characterized as an actin-regulating protein, while it also regulates other cellular processes, including matrix adhesion, migration, and proliferation. We discovered that cAMP-induced CAP1 dephosphorylation regulates relevant cell functions. Several commentaries on the Oldfield & Bonano (2024) target article proposed that natural behavior should play a larger role in assessing the welfare of fishes in zoos and aquariums. However, the natural-behavior concept of animal welfare has been widely criticized because natural behaviors do not necessarily improve welfare, and unnatural behaviors do not necessarily reduce it. I review the three major concepts of animal welfare — physical health, natural behavior, and affect/feelings — and argue that behavior patterns, whether natural or otherwise, should be evaluated according to their effects on health and subjective experience. Globally, wetland habitats suffered major declines during the 20th century. In the 1990s, the United States government created the Wetland Reserve Program (later consolidated into the Agricultural Conservation Easement Program), to combat the loss of wetlands. Despite success in attracting landowners, minimal monitoring of properties enrolled into the program (easements) has occurred. In this study, we monitored avian communities at easements, compared avian diversity at easements to agriculture and bottomland forests, and quantified microhabitat, site-specific, and surrounding landscape features at easements. Enhancer RNAs (eRNAs) are non-coding transcripts produced at enhancer regions, which appear to be involved in transcriptional regulation. Up to date, these have been primarily investigated using labor- and cost-intensive genomic techniques. However, the precise mechanisms by which eRNA transcription or eRNA transcripts themselves mediate transcriptional regulation remain unclear. Here, we present a novel experimental approach that allows us to analyze characteristics of eRNA transcription in fixed and live whole Drosophila melanogaster embryos. Background: Acute gout arthritis, which is caused by hyperuricemia, is a common inflammatory joint disease, leading to monosodium urate (MSU) crystal deposition. These crystals trigger inflammation, causing pain and significantly impairing the patient’s quality of life. Colchicine is commonly used to treat and prevent acute gout arthritis, but it has several side effects. Cherry tree (Muntingia calabura L.) leaves, which possess anti-inflammatory properties, show promise as an alternative treatment. Wastewater treatment is necessary for environmental conservation and combating water pollution. This study demonstrates real-time dissolved oxygen (DO) monitoring in constructed wetland treatment. The system would ensure an adequate supply of oxygen through automated water circulation whenever the level is below 4 mg/L, and shut down when it reaches the normal level. From experiments conducted, it was observed that within three automated circulation processes, it is possible to increase DO concentrations from 0.5 mg/L to 5.0 mg/L. Additionally, Total Dissolved Solids reduced from 1,380 ppm to 920 ppm. pH also stabilized from 8.38 to 7.9. In this system, data is …

The aim of the present research was to evaluate, for the first time, the effect of ozonated water treatments on the postharvest quality of capia pepper (Capsicum annuum L. ‘Kaptan’). The peppers were harvested at the optimum time and transported to the laboratory immediately. Uniform and quality fruit was selected and precooled. After precooling, the fruit was divided into four groups. The first three groups were immersed in ozonated water at three different concentrations (1.0, 2.0 and 3.0 ppm) for 15 min. Dr. John Hibbing was awarded the ORCA in 1993. John was born in 1953 in Paullina, Iowa. He earned a bachelor's in secondary education from Dana College in Blair, Nebraska, but then realized that teaching high school wasn't for him. Instead, he attended the University of Iowa, where he earned a master's and doctorate in political science. In 1981, he joined the University of Nebraska-Lincoln as an assistant professor of political science and rose quickly through the ranks. Centromeres are critical genomic regions responsible for ensuring accurate chromosome segregation during cell division. Disruptions in the structural or functional integrity of centromeric DNA could impair kinetochore assembly, leading to chromosome missegregation and chromosomal instability (CIN), a hallmark of various human diseases, including systemic sclerosis (SSc). This study investigated the contribution of centromeric DNA damage and deletion to CIN and explored potential pathogenic mechanisms relevant to SSc. Background/aim: Cervical cancer (CC) is a malignant gynecologic tumor. Small RNAs derived from tRNA (tsRNAs) have been reported to play regulatory roles in tumor progression and suppression. However, the functional role of tsRNAs in CC remains largely unclear.
Materials and methods: Small RNA sequencing was performed on normal, cervical intraepithelial neoplasia (CIN), and CC clinical cell samples to identify differentially expressed tsRNAs (DEtsRNAs). qRT-PCR was used to validate the selected DEtsRNAs. The effects of tsRNA-05020 on HeLa cell proliferation, apoptosis, and epithelial-to-mesenchymal transition were assessed.

Background/aim: Polyunsaturated fatty acids (PUFAs) and the hepatic cholinergic axis are critical regulators of lipid metabolism and inflammatory signaling; however, their time-dependent interplay in hepatocytes remains incompletely understood. Clarifying how n-6 and n-3 PUFAs interact with cholinergic pathways may provide mechanistic insight into metabolic dysfunction-associated steatotic liver disease (MASLD).

Pine Grosbeaks (Pinicola enucleator) are boreal finches whose winter ecology and social behavior remain only sporadically documented due to their remote distributions and irruptive dispersions. From 18 December 2023 to 6 March 2024, we conducted repeated daily observations of a flock of up to twelve P. Enucleator occupying a temporary wintering range in an urban neighborhood in Oregon. During these observations, we documented a previously undescribed collective vocalization associated with feeding. Comparison with 2674 archived Pine Grosbeak recordings from major bioacoustic repositories revealed that no clear matches between our recording and archives could be established. The low amplitude …

Background/aim: Epithelial-mesenchymal transition (EMT) is a fundamental process driving tumor plasticity, metastasis, and therapy resistance. Although E26 transformation-specific transcription factor 1 (ETS1) and Sine oculis homeobox homolog 1 (SIX1) are individually implicated in EMT-related transcriptional programs, the regulatory interplay and functional coordination of these proteins across tumor states remain unclear. Background/aim: Visceral leishmaniasis, primarily caused by Leishmania infantum, remains a major global health challenge due to limitations in current chemotherapeutic options, including toxicity and emerging drug resistance. Host-directed therapeutic approaches are increasingly recognized as promising alternatives. All-trans retinoic acid (ATRA) is an immunomodulatory molecule with host-dependent effects on macrophage function; however, its therapeutic use is hindered by instability and poor solubility. Heterochromatin Protein 1 (HP1) family members are highly conserved, non-histone chromosomal proteins essential for heterochromatin formation and transcriptional regulation. In Drosophila melanogaster, HP1a functions both as a transcriptional repressor and activator, as well as other functions, and undergoes post-translational modifications, including phosphorylation. Chronic infection of urogenital schistosomiasis in humans is a major cause of urinary tract morbidity in endemic areas. However, information on the prevalence of schistosomiasisrelated urinary tract morbidity is lacking in most endemic countries including Tanzania. This study was conducted to determine the prevalence of schistosomiasis-related urinary tract morbidity in schoolchildren and adults in Itilima District. Urine samples from 682 residents were examined for Schistosoma haematobium infection using urine filtration technique. Urinary tract morbidity was examined by ultrasound. The prevalence of S. haematobium infection was 16.4% (112/682) and varied significantly between villages (range 3.2% to 29.5%, χ2 = 48.467, p < 0.001). There was no significant variation of S. haematobium infection between male and female (8.3 % vs 8.1%, χ2 = 0.679, p = 0.410). Majority (93.8%) had light infections (

Chromium (Cr+6) toxicity is a significant environmental stress factor that severely hampers plant growth and productivity. This study aimed to evaluate whether the combined application of phosphorus fertilizer and melatonin could alleviate chromium induced stress in maize (Zea mays L.). A foliar spray of melatonin (50 µM) was applied alongside two phosphorus sources—phosphoric acid (PA) and diammonium phosphate (DAP), each at 60% of the recommended rate. Developing resistant chickpea cultivars is essential for managing Fusarium wilt caused by Fusarium oxysporum, a major pathogen responsible for substatial yield losses. In this study, we conducted RNA-seq–based transcriptome analysis to characterize gene expression changes in resistant and susceptible genotypes of Cicer arietinum L. and C. reticulatum following inoculation with Fusarium oxysporum f. sp. ciceris race 0. Susceptible genotypes exhibited markedly higher numbers of differentially expressed genes compared with resistant genotypes. Defense-related genes were induced in all plants upon infection; however, resistant genotypes specifically upregulated genes functioning in the apoplast or extracellular region while downregulating intracellular processes—a pattern not observed …

Arbuscular mycorrhizal fungi (AMF), which exist symbiotically with plant roots, affect plant growth, yield, and fruit quality. The effects of AMF vary depending on the cultivar and mycorrhizal type. In this study, the effects of different AMF types on the Rubygem strawberry cultivar, widely grown in Türkiye, were examined. Gigaspora margarita and Funneliformis mosseae AMF species were applied to plant roots at seedling planting. Fruit quality criteria, including yield, acidity, soluble solid content, phenolic compounds, organic acids, crown and root development, and the number of daughter plants and stolons, were examined. Fluvisols develop in river valleys through the accumulation of alluvial sediments. They are characterised by high fertility and are extensively used for agriculture. This study aimed to identify the core microbiome of fluvisols in the Vistula Valley and to examine its relationship with the basic physicochemical properties of the soils.
