[Objective ] The effects of the transcription regulation system on menaquinone- 7 (MK- 7) production in Bacillus subtilis (B.subtilis ) 168 were studied by excavating transcription regulators.[Method ] With the signal molecule PhrG pentapeptide,RapG,and the transcription regulator DegU,a PhrG-RapG-DegU regulatory system was successfully constructed in B.subtilis 168 to fine-tune the expression of key genes,so that the expression of key genes in the MK- 7 synthesis pathway can respond to the changes in cell density.[Result ] The production of MK- 7 in strain BS 06 reached 102.47 mg/L through this system.[Conclusion ] Fine tuning of quorum sensing can effectively improve the …

[Objective ] This study aims to investigate the immunoregulatory mechanisms of lactoferrin formula and its four nutritional components (α-lactalbumin,N-acetylneuraminic acid,yeast β-glucan,and lactoferrin ) in RAW 264.7 macrophages.[Method ] M1 and M 2 macrophage polarization were induced by lipopolysaccharides (LPS) and interleukin (IL)-4,respectively.Quantitative real-time PCR,flow cytometry,and Western blot were employed to analyze macrophage polarization and related signaling pathways.[Result ] Lactoferrin formula enhanced the phagocytic activity of macrophages and dose-dependently inhibited M 1 polarization while promoting M 2 polarization within the range of 10~50 µg/mL.A combination of four nutritional components exhibited similar effects,with greater effects than the three-component combination.Additionally,the lactoferrin formula upregulated …

[Objective ] This study aims to evaluate the antibiotic resistance and transmission risk of Enterococcus faecalis (E.faecalis ) in the pork production chain.[Method ] A total of 300 samples were collected from the surface of live pigs,carcasses,and the environment in 2 pig farms,1 slaughterhouse,and 2 markets in Hangzhou,Zhejiang.The strain identification,antibiotic resistance phenotypes,resistance genotypes,and virulence genotypes were performed by 16S rDNA sequences,broth microdilution method (MIC ),and PCR.In addition,multilocus sequencing typing (MLST ) and conjugational transfer tests were conducted.[Result ] The 125 isolates of E.faecalis showed high resistance to antibiotics such as tiamulin,tetracycline,and doxycycline,and the isolates from pig farms showed higher antibiotic …

Water deficit is a critical challenge to global agricultural productivity and food security, which is exacerbated by climate change and increasing water scarcity. To withstand drought stress, plants employ a variety of molecular strategies driven by complex regulatory networks. This review examines the central role of transcription factors (TFs) as master regulators that activate drought responsive genes, which in turn mediate key adaptive processes such as osmoregulation, antioxidative defense, and membrane stability. Microbial consortia of compatible microorganisms play a key role in enhancing agricultural sustainability by optimizing nutrient uptake by promoting plant growth more effectively. This study aimed to identify local isolates with the most effective plant growth-promoting (PGP) properties and to form a consortium of isolates showing the optimal synergistic effect between plant and bacterial components to improve the growth of wheat plants. For this purpose, 48 isolates identified from 145 isolates were screened for their PGP activities. Salix section Subviminales, including only three species, is endemic to East Asia. In this study, we revised three species, S. gracilistyla, S. graciliglans, and S. blinii in Korea based on their plastomes and extensive morphological analyses to provide a comprehensive understanding of their taxonomic relationships. Based on our findings, we identified the morphological characteristics of S. blinii endemic to Korea, including its small size and distinct leaf shapes, which are well distinguished from both S. gracilistyla and S. graciliglans. Erigeron akaydinii Günal & Semiz sp. nov., belonging to the Trimorpha Cass. (DC) section (Asteraceae), is described as a new species from Uludağ Mountain (Bursa, Türkiye). This study examines the diagnostic traits of E. akaydinii and related species, including morphological, palynological, and molecular analyses. Notes on the ecology, distribution, and conservation status of the new species are also provided, along with a diagnostic key for relevant taxa.

Increasing exploitation of water resources has led to declining water quality in aquatic environments. Freshwater ecosystems face various environmental pressures, such as the impacts of climate change and intensifying land-use practices. This study aimed to assess the ecological preferences of phytoplankton communities and determine the ecological status (ES) of several reservoirs and lakes in Ceyhan River Basin across different seasons. A canonical correspondence analysis was conducted to understand the associations between phytoplankton assemblages and stressors in freshwater ecosystems, revealing a strong correlation of 97%. Actin proteins play an important role in microfilaments, one of the three components of the cytoskeleton of the cell. The actin protein is responsible for cellular functions such as with their role in neuromuscular junctions. The dynamics of these proteins can be further explored with manipulation of polymerization through the treatment of a biochemical drug, Cytochalasin D. This fungal toxin caps the ends of actin filaments, destabilizing them by inhibiting G- and F-actin monomers from binding. Sickle cell disease (SCD) is a genetic disorder characterized by sickle red blood cells (RBCs). Sickle RBCs cause cerebral vasculopathies including vaso-occlusive events, leading to ischemia-reperfusion injury and hypoxic tissue environment. To date, the physiological blood flow velocities in cerebral vessels of preclinical SCD models has not been evaluated under hypoxic-reoxygenation. In our study, we used transcranial ultrasound techniques to measure abnormal blood flow velocities in the internal carotid (ICA) and middle cerebral arteries (MCA) of transgenic sickle cell mice (SS) challenged with hypoxia-reoxygenation. Meiotic chromosome segregation errors in human oocytes are the leading cause of miscarriages and aneuploid pregnancies, and these errors increase dramatically as women age. Using Drosophila oocytes as a model system, this dissertation identifies the NAD⁺-dependent deacetylase Sirt1 as a key regulator of chromosome segregation and meiotic arm cohesion. Loss of Sirt1 activity during meiotic prophase leads to premature loss of arm cohesion and increased segregation errors in Drosophila oocytes. In addition, elevated acetylation of its substrate, histone H4K16, indicates that Sirt1 activity declines in oocytes during aging. Strikingly, dietary administration of the Sirt1 activator SRT1720 preserves Sirt1 deacetylase activity …

Many bacteria form multicellular communities known as biofilms. Biofilms contain bacterial cells and a protective extracellular biofilm matrix. This matrix contains exopolysaccharides, extracellular DNA (eDNA), membrane vesicles, and proteins. The matrix proteins are far less characterized than other extracellular biofilm components. In biofilms formed by the model organism Pseudomonas aeruginosa, the protein OprF is abundant in the cellular outer membrane and the extracellular matrix. In my dissertation research, I have presented data supporting the hypothesis that the OprF protein significantly affects P. aeruginosa biofilms. Transcript isoforms arising from alternative splicing events can contribute to the development of a cell’s identity and function and overall protein diversity. Peripheral blood is a useful model to study and evaluate computational methods for alternative splicing detection due to its abundant data across various sequencing protocols. Many health benefits are associated with Vitamin D (VitD), although deficiency is associated with poor health outcomes and increased risk for cancer development. For example, many tissue specific enzymes are involved in VitD metabolism, and mutations or deletions within Vitamin D receptor (VDR) genes are known to increase cancer risk by altering their functions or bioavailability, although less is known about these phenomena in oral cancers. Alzheimer’s disease (AD) is an irreversible and devastating neurodegenerative disease characterized by progressive synaptic, dendritic, glial, and neuronal loss, collectively leading to impairments in learning, memory, and deterioration of cognitive and behavioral functions. AD is the most common form of dementia accounting for 60-80% of all cases. AD affects approximately 7 million Americans and is the 6th leading cause of death in the US. Fungal exposure can cause acute and chronic illnesses or exacerbate existing illnesses. Some fungi are opportunistic pathogens and can lead to serious systemic fungal infections. In 2020, more than 150 million cases of severe fungal infections were reported globally resulting in an estimated 1.7 million deaths. Coccidioidomycosis, or Valley fever, is classified as a serious fungal infection caused by the fungi Coccidioides immitis and C. posadasii. Walnut (Juglans regia L.) is one of the most economically important species. It is of great importance to the food industry due to its nutritional value, and high content of many beneficial components such as macro and microelements. In the current study, the aim was to determine markers of single nucleotide polymorphism (SNP) associated with the gene(s) controlling the accumulation of Selenium (Se) and Magnesium (Mg) in walnuts. Se and Mg concentrations were measured in 154 genotypes of Juglans regia in 2021 and 2022. A new fossil-species of coralloid palm root mat under the organ genus Rhizopalmoxylon is reported from the latest Maastrichtian (Late Cretaceous) to earliest Danian (Early Palaeocene) sediments (c. 66–65Ma) of the Deccan Intertrappean Beds of Madhya Pradesh, Central India. The significant anatomical attributes, specifically an exodermis comprising 2–4 layered parenchymatous cells; a 5–8 layered homogeneous outer cortex, a highly lacunate inner cortex central diffuse layer of aerenchyma with well-developed radially elongated cells arranged in 2–4 rows, and a stele consisting of radial and exarch vascular bundles with proto and metaxylem strands reveal its close resemblance to root mats of extant …
