Kernza® is the trademarked name of the perennial crop intermediate wheatgrass (Thinopyrum intermedium). In addition to its potential for dual-use as a grain and forage crop, Kernza is marketed to provide needed environmental services by increasing crop biodiversity, sequestering soil carbon, and reducing nitrate leaching. However, more work is needed in cultivar selection and crop management to increase total grain yields and prevent long-term grain yield decline. Furthermore, research in quantifying and understanding Kernza’s ability to affect soil properties and water dynamics is limited.

This study, conducted during the 2024 growing season, evaluated short-term effects of soil management practices on greenhouse gas (GHG) emissions, soil microclimate, nitrogen dynamics, weed suppression, and soybean (Glycine max) yield. In addition to conventional tillage and no-till, this research included cereal rye (Secale cereale) cover cropping and a novel sprayable biodegradable mulch (BioWRAP®) to assess emerging strategies for sustainable soybean systems under contrasting moisture regimes.

Recent studies have highlighted the importance of mitochondria in NP cells and articular chondrocyte health. Since the understanding of mechanisms governing mitochondrial dynamics in these tissues is lacking, we investigated the role of OPA1, a mitochondrial fusion protein, in their homeostasis. OPA1 knockdown in NP cells altered mitochondrial size and cristae shape and increased the oxygen consumption rate. OPA1 governed the morphology of multiple organelles, including peroxisomes, early endosomes and cis-Golgi and loss resulted in the dysregulation of autophagy. Metabolic profiling and

The integrated survey of boat-based recreational fishing includes three components: off-site phone surveys (encompassing an initial screening survey, a longitudinal phone-diary survey, and post-enumeration surveys), on-site boat ramp surveys and remote camera surveys. The recent phone-diary survey from September 2023 to August 2024 (inclusive) provides comparisons with previous surveys in 2020/21 (September 2020 to August 2021), 2017/18 (September 2017 to August 2018), 2015/16 (September 2015 to August 2016), 2013/14 (May 2013 to April 2014) and 2011/12 (March 2011 to February 2012).

Background: Generative artificial intelligence (GAI) is transforming health care in a variety of ways; however, the present utility of GAI for supporting clinicians who treat rare disease such as primary immune disorders (PIs) is not well studied. We evaluated the ability of 6 state-of-the-art large language models (LLMs) for providing clinical guidance about PIs.

Objective: To quantitatively and qualitatively measure the utility of current, open-source LLMs for diagnosing and providing helpful clinical decision support about PIs.

Lysimachia vulgaris (garden loosestrife, Primulaceae) is an herbaceous perennial native to Europe that has been identified as an aggressive invasive species in parts of North America. The status of the species in the Mid-Atlantic region of the US, however, is poorly determined. Using recent field observations (from iNaturalist, iMapInvasives, and EDDMapS records) we here report localized incursions of L. vulgaris in the Susquehanna River Valley of central Pennsylvania, along with notes on its ecology, threats to native plant communities, and broader distribution in the state.

Understanding disease progression and sophisticated tumor ecosystems is imperative for investigating tumorigenesis mechanisms and developing novel prevention strategies. Here, we dissected heterogeneous microenvironments during malignant transitions by leveraging data from 1396 samples spanning 13 major tissues. Within transitional stem-like subpopulations highly enriched in precancers and cancers, we identified 30 recurring cellular states strongly linked to malignancy, including hypoxia and epithelial senescence, revealing a high degree of plasticity in epithelial stem cells. By characterizing dynamics in stem-cell crosstalk with the microenvironment along the pseudotime axis, we found differential roles of ANXA1 at different stages of tumor development. This study investigates patterns of stand structure regeneration, growth characteristics, and coarse woody debris (CWD) patterns in Vachellia xanthophloea (fever tree) stands within the Makuleke Contractual Park (MCP), a semi-arid savanna system in northern Kruger National Park (KNP), South Africa. Fieldwork was conducted across two stand types: a monospecific fever tree stand in Rietbok Vlei and a mixed-species stand in the Western Nhlangaluwe Floodplain where fever tree is established with Faidherbia albida (ana tree). Currently, corn (Zea mays) is the predominant feedstock used for bioenergy production in the United States. Proxies such as lake cores, speleothems, dendrochronology, and others help provide high-resolution data on paleoclimatic conditions. Headwater streams specifically are vulnerable to changes in hydrology, water quality, and physical characteristics from anthropogenic practices due to their small watersheds with shallow soils, yet they are often not a priority in conservation (Campbell et al., 2011; Abell et al., 2017). Unlike conventional computing systems based on silicon hardware, biocomputing leverages the parallelism, energy efficiency, and complex dynamics of living systems. Among various cellular mechanisms, calcium (Ca2+) signaling stands out as a central regulator of diverse biological functions, offering a promising basis for programmable logic and control in living cells.

In gut microbiome research, carbohydrate-active enzyme gene clusters (CGCs) have emerged as key functional units for understanding microbial glycan degradation. Unlike taxonomic or broad pathway annotations, CGCs offer gene-cluster-level resolution and capture substrate-specific microbial functions. However, their diversity and distribution in relation to host metabolic phenotypes, such as obesity, remain poorly characterized. This study tests the hypothesis that the composition and abundance of fiber-targeting CGCs vary between obese and healthy human gut microbiomes, reflecting distinct microbial carbohydrate utilization strategies. AquaCrop was calibrated and validated for soybean [Glycine max (L.) Merr.] using 19 irrigation treatments from five years using data from field experiments. The model accurately simulated canopy cover (CC), soil water content (SWC), and grain yield, with overall validation nRMSE values of 12%, 7%, and 7%, respectively. The overall validation nRMSE for SWC in individual 30-cm soil layers was comparatively higher, at 19%. The model was subsequently applied for long-term simulations (2010–2024) to estimate soybean yield and water requirements for two dominant soil types, Yutan silty clay loam and Tomek silt loam under three irrigation levels - rainfed, …

Maternally derived antibodies (MDAs) play a vital role in protecting neonates from infectious diseases, but their presence at the time of vaccination can interfere with vaccine-induced immune responses, thereby reducing vaccine effectiveness. MDA interference has been documented in pigs vaccinated with whole inactivated virus (WIV) vaccines against swine influenza A virus (IAV). In this thesis, we evaluated the efficacy of a lipid nanoparticle encapsulated DNA (LNP-DNA) vaccine against swine IAV in the presence and absence of MDAs, comparing it to a WIV vaccine. The retinal determination genetic network controls the development of the visual system in all seeing animals through the molecular regulation of cells to adopt an eye tissue fate. The compound eye of the fruit fly, Drosophila melanogaster, is an excellent model system to study the complex mechanisms within the network that regulate specification of cellular identity during embryogenesis. Maintenance of mitochondrial transport and clearance at the pre- and post-synaptic terminal is an essential function of neuromuscular junctions to meet their high energy demands. Damaged or dysfunctional mitochondria can put stress on these synaptic junctions, ultimately leading to the degradation of the synapse and subsequent cell death. Dysfunctional mitochondria are also often present in neuromuscular diseases such as ALS. One possible mechanism by which neurons may eliminate dysfunctional mitochondria is through the muscle-synthesized BDNF (msBDNF) /TrkB-mediated Bcl-2 nineteen-kD interacting protein (BNIP3) mitophagy pathway, although the relationship is not well-defined. This study explores the regulatory role of microRNA-10 (miR-10) in larval zebrafish caudal fin regeneration, a model with broad implications for the future of regenerative medicine. Building on prior research from the lab, which indicated that miR-10 knockdown impairs regeneration, we verified this finding through rescue experiments, assessed its influence on healing projections, and examined its role in the innate immune response during the wound healing process. To address these questions, we employed a multi-faceted approach, including timed matings, microinjections, immunohistochemistry, fixed sample microscopy, and live imaging. Introduction: Network hyperexcitability (NH) is observed in patients with early-stage Alzheimer's disease (AD), emerging decades before cognitive decline. A key molecular determinant of NH is voltage-gated Na+ channel 1.6 (Nav1.6), which mediates action potential firing in CA1 hippocampal neurons. Ameliorating NH through inhibition of the glycogen-synthase kinase 3β (GSK3β/Nav1.6 complex may provide immediate benefits to cognition and memory and slow AD progression.

Methods: Hight-throughput virtual screening and multiple in vitro biological assays were utilized to identify compound 1063. Patch-clamp electrophysiology and electroencephalogram recordings were utilized to functionally assess 1063 in models of AD neuropathology.

ParB is a central player in bacterial chromosome segregation, and cytidine triphosphate (CTP) is known to modulate its DNA compaction activity. This study explored whether CTP’s regulatory effect is due to specific molecular interactions beyond its known CTP-binding pocket. Using single-molecule flow-stretching and TIRF microscopy, we showed that CTP significantly reduces DNA compaction by wild-type Bacillus subtilis ParB, while ATP has no effect, supporting the hypothesis that CTP acts through specific recognition rather than general charge. CTP’s modulatory effect persisted across C-terminal domain (CTD) lysine mutants. Compaction was reduced in wild-type and K255A but abolished in K252A and K259A. The …

Mitochondrial dynamics occurring under stress-induced conditions in SHSY5Y neuroblastoma cells provide insight into the OPA1-mediated mitochondrial fusion threshold in cells of varying energetic demands. Mitochondria are dynamic structures that are continuously balancing fusion and fission, depending on cellular needs. The ability to fuse and divide relies on the maintenance of the mitochondrial transmembrane potential. Citizen science-provided insect data inherently exhibits variability in accuracy. In the Lower Rio Grande Valley (LRGV), butterfly occurrence reporting is predominantly reliant on citizen science contributions. My research compared fieldwork conducted monthly over a year to occurrence records submitted by visitors at the National Butterfly Center and in Mission, Hidalgo County, Texas. I hypothesized that butterfly occurrences at the Center would be maximized during the fall and minimized during winter and summer months. Diabetic retinopathy (DR) is a major complication of diabetes and a leading cause of vision loss largely driven by the overexpression of vascular endothelial growth factor (VEGF), which promotes neovascularization and vascular permeability in the retina. Current treatments, primarily anti-VEGF injections, inhibit the effects of VEGF once it has been secreted but can be ineffective in a certain subset of patients. Recent studies suggest that oxytocin (OXT), a neuropeptide mainly known for its roles in reproduction and social bonding, may downregulate VEGF expression.
