Emery-Dreifuss muscular dystrophy 1 (EDMD1) arises from mutations in EMD. Most EDMD1 patients lack detectable emerin expression. They experience symptoms such as skeletal muscle wasting, joint contractures, and cardiac conduction defects. Currently, physicians rely on treating patient symptoms without addressing the underlying cause-lack of functional emerin protein. Thus, there is a need for therapeutic approaches that restore emerin protein expression to improve patient outcomes. One way would be to deliver emerin mRNA or protein directly to affected tissues to restore tissue homeostasis. Chlamydia trachomatis has a DNA topoisomerase I with a unique C-terminal domain (CTD) homologous to eukaryotic SWIB domains. This study focused on determining the function of the SWIB domain-containing TopA from C. trachomatis (CtTopA). We demonstrated that, despite the lack of sequence similarity at the CTDs between CtTopA and TopA from Escherichia coli (EcTopA), full-length CtTopA removed negative DNA supercoils in vitro and complemented the growth defect of a topA mutant of E. coli. CtTopA is less processive in DNA relaxation than EcTopA in dose-response and time course studies. An antibody generated against the SWIB domain of CtTopA specifically recognized …

Introduction: Running is a common form of exercise that comes with many health benefits; however, many people stop exercising because of the high injury risk. Running injury is connected with biomechanical risk factors. Yet, these risk factors often show inconsistent and conflicting results across studies, largely due to small sample sizes limiting statistical power. Low subject numbers may be partly due to the time requirements of traditional motion capture. Tetralogy of Fallot (TOF) is the most common cyanotic congenital heart defect (CHD). TOF may present in isolation or in conjunction with one or more non-cardiac congenital anomalies or neurodevelopmental disorders (TOF+). Uncertainty regarding the efficacy of various genetic testing strategies, and an incomplete understanding of the genetic causes of TOF+, may lead to hesitancy in recommending genetic testing, particularly, clinical exome sequencing (cES). Here, we analyzed cES data from 131 individuals with TOF+. A definitive or probable diagnosis was made for 31 individuals, yielding a diagnostic rate of 23.6% (31/131). Acenes are a class of polycyclic aromatic hydrocarbons that may hold promise as organic semiconductors (OSCs) in solar cells and electronic devices. Their instability and poor solubility present challenges that may be improved by replacing the hydrogens with phenyl, halogen or alkyl substituents to sterically induce a helical twist to these otherwise planar molecules. This twisted structure also impacts the optical properties of these molecules. We employ time-dependent density functional theory (TD DFT) to investigate acenes spanning from naphthalene to heptacene. "First, I briefly highlight recent discussions of sex and how they contrast with a gametic central understanding of sex in the executive order. I then provide some background on why gametes have been central to defining sex across species in evolutionary biological sciences. Objective: The common gain-of-function variant rs35705950, located in the promoter of MUC5B gene, has been strongly associated with interstitial lung diseases (ILDs) of different aetiology, such as idiopathic pulmonary fibrosis (IPF) and rheumatoid arthritis-associated ILD (RA-ILD). In this study, we aimed to investigate the association of this variant and its nearby single nucleotide polymorphisms (SNPs) in the largest cohort of systemic sclerosis-associated ILD (SSc-ILD) to date.

Extracellular vesicles (EVs) play a crucial role in intercellular communication, signaling pathways, and disease pathogenesis by transporting biomolecules such as DNA, RNA, proteins, and lipids derived from their cells of origin, and they have demonstrated substantial potential in clinical applications. Globally, millions of children suffer from undernutrition, with stunting affecting 150 million and wasting nearly 50 million under five years old. Current interventions have shown limited effectiveness in mitigating long-term consequences such as stunted growth, cognitive deficits, and immune dysfunction. Through this body of work, I investigated the efficacy of a microbiota- directed complementary food (MDCF-2) as a next generation therapeutic food to combat childhood stunting. Across two randomized controlled trials involving Bangladeshi children with either moderate acute malnutrition or post-severe acute malnutrition, MDCF-2 significantly outperformed standard ready-to-use supplementary foods (RUSF), leading to significantly improved ponderal and sustained linear growth …

This study investigated whether the timing of a 24-hour fast influences indicators of glycemic control throughout the duration of the fast or upon refeeding. Twenty-four adults who have overweight or obesity (13 female, 11 male) completed this randomized crossover study involving three 24-hour fasts initiated at either 8:00 am, 1:00 pm, or 6:00 pm followed by a 75 g oral glucose tolerance test (OGTT) which lasted 2 hours. Continuous glucose monitors (CGMs) were employed to track interstitial glucose throughout each fast and OGTT. Materials with cylindrical pores are optimal for electrochemical devices and filtration due to their straight paths with minimal tortuosity when aligned. Polymer templates have led to many individual cylindrical architectures however the independent tailoring of pore and wall dimensions has remained elusive. Here short cylindrical micelle templates (PEO-b-PS) with glassy cores enabled the fabrication of sample series with constant cylindrical pore diameter (∼63 nm) and tailored wall thickness (TiO2, ∼45–100 nm). Rabbit urine liquid organic fertilizer (LOF) can be used as an alternative fertilizer to reduce the use of inorganic fertilizers, especially for shallots, as a national food commodity. This study aims to determine the best concentration of rabbit urine LOF, the best dose of NPK fertilizer dose reduction, and a combination of rabbit urine LOF and NPK fertilizer dose reduction on the physiological characters of shallot plants. Roses are known as a high-value commodity frequently used in various important events. However, they are susceptible to postharvest quality deterioration, which can affect their vase life and appearance. In this study, roses with a blooming stage of approximately 25–50% were immersed in melatonin and 6-benzylaminopurine (BAP) solutions at different concentrations. This research aims to analyze the effect of melatonin and BAP application on the freshness of cut roses. Nest-building in social insects requires layered communication to coordinate complex, functional structures for housing, protection, storage, and homeostasis. This is especially striking in mound-building termites, which construct and repair large mounds collectively despite lacking image-forming eyes. This thesis investigates how Odontotermes obesus achieves such coordination through chemical communication embedded in soil.
Using controlled two-choice assays with manipulated soils, I separated airborne volatile signals from contact non-volatile signals by baking to remove volatiles, a mesh assay that presented volatile cues without contact, and solvent extraction with reapplication of contact cues. Reintroducing native plants into restoration and revegetation projects could increase plant species diversity, increase floral resources for pollinators, and positively affect trophic-level interactions in the community. Direct seeding in restoration projects has generally had poor success rates due to biotic and abiotic factors, leading to significant seed wastage of the already insufficient native seed supply. Seed balls (aka seed aggregates, seed bombs, and seed pellets) are a low-cost, low-tech seed enhancement technology. Long intergenic non-coding RNA 00,511 (LINC00511) is considered an oncogene for various cancers. However, the association between LINC00511 single nucleotide polymorphisms (SNPs) and colorectal cancer (CRC) remains unclear. Our study aims to study whether LINC00511 SNPs could predict CRC susceptibility or prognosis, an important step-toward precision-health, based on an Egyptian CRC patient cohort. A total of 200 CRC patients and 200 cancer-free controls were genotyped for three LINC00511 SNPs − rs9906859, rs1558535, and rs17780195 using qRT-PCR. Air pollution, particularly exposure to fine particulate matter (PM2.5), poses a growing threat to cardiopulmonary health. PM2.5 consists of microscopic particles smaller than 2.5 microns in diameter and originates from industrial combustion, transportation, and urban dust. These particles are classified as xenobiotics and are capable of bypassing respiratory defenses to enter systemic circulation, where they cause oxidative stress, inflammation, and cardiovascular dysfunction. RNA post-transcriptional modifications act by stabilizing the functional conformations of RNA. While their role in messenger RNA (mRNA) decoding is well established, it is less clear how transfer RNA (tRNA) modifications outside the anticodon contribute to tRNA stability and accurate protein synthesis. Absence of such modifications causes translation errors, including mRNA frameshifting. By integrating single-molecule fluorescence resonance energy transfer and cryogenic electron microscopy, we demonstrate that the N1-methylguanosine (m1G) modification at position 37 of Escherichia coli tRNAProL is necessary and sufficient for modulating the conformational energy of this tRNA on the ribosome so as to suppress +1 frameshifting otherwise induced …

Purpose: Hippocampal avoidance (HA) during therapeutic whole-brain radiotherapy reduces the risk of neurocognitive function (NCF) toxicity in patients with brain metastasis. This trial hypothesized that HA during prophylactic cranial irradiation (PCI) in patients with small cell lung cancer (SCLC) leads to noninferior intracranial relapse (ICR) and reduction in NCF toxicity.
Methods: This randomized phase II/III trial enrolled patients with SCLC, no brain metastases, and response to chemotherapy. The primary end points were 12-month ICR (noninferiority design, randomized phase II) and 6-month Hopkins Verbal Learning Test-Revised (HVLT-R) Delayed Recall (DR) failure (phase III). Purpose: There are limited data on the risk for mortality and health outcomes among the increasing population of older (age >50 years) survivors of childhood cancer during this later stage in life when there is an expected increase in aging-related morbidities.
Methods: We assessed cause-specific mortality, incident new cancers, chronic health conditions (CHCs), frailty, and health status among survivors from the Childhood Cancer Survivor Study, conditional on surviving to 50 years. We calculated conditional survival rates, standardized mortality ratios (SMRs), and, for incident new cancers, cumulative burden, standardized incidence ratios (SIRs), and relative rates (RRs), compared with the general US …

Seven jurisdictions from around the world have issued judicial opinions that address fundamental issues about the governance and regulatory systems of agricultural biotechnology. This article summarizes these legal proceedings and describes their impact upon agricultural biotechnology. Climate change, with global temperatures rising by 1.09°C from 1850–1900 to 2011–2020, threatens potato production, a critical staple crop, by exceeding the optimal temperature range of 15–20°C. This review synthesizes over 45 peer-reviewed studies published between 2015 and 2025 from Google Scholar and ScienceDirect to evaluate the physiological, morphological, and tuber quality responses of potatoes to high temperatures and to identify adaptive cultivation strategies for sustainable production. High temperatures reduce photosynthetic efficiency through chlorophyll degradation and stomatal closure, increase respiration, and divert photosynthates to vegetative growth, leading to 18–32% yield losses globally by the 2050s. PI3K inhibition causes apoptosis selectively in NOTCH1-mutant head and neck squamous cell carcinoma (HNSCC), but modest single-agent responses and acquired resistance (AR) limit the clinical efficacy of targeted agents. To address these limitations, we investigated novel combination therapies. We tested the efficacy of 5768 compounds as single agents and 139 in combination with PI3K inhibitors in sensitive and AR NOTCH1-mutant HNSCC cell lines. We generated synergy/efficacy classifications for the combinations using multiple metrics of statistical drug synergy and growth rate indices. Black Soldier Fly (BSF) larvae offer a cost-effective, nutrient-rich alternative to synthetic fertilizers and conventional feed inputs such as soy, fishmeal, and maize bran in sub-Saharan Africa. In Malawi, where smallholder farmers grapple with declining soil fertility, high input prices, chronic food insecurity, and livestock nutritional challenges, BSF farming is emerging as a promising circular agricultural innovation. This study examines the potential for neuroaesthetic design and adaptive neurofeedback principles to enhance brain-on-chip (BoC) research, with an emphasis on optimizing neural engagement and cognitive accessibility for diverse populations. Existing BoC platforms have transformed neuroscience through high-fidelity modeling of neural circuits, yet cognitive accessibility and sensory inclusion remain comparatively underexplored. Drawing upon recent studies in neuroinclusion, multisensory interface design, and adaptive feedback, this work synthesizes findings from biophilic interface architecture, neurofeedback literature, and embodied cognition frameworks. Frost and heat events at critical growth stages could cause large yield losses. These temperature extremes are increasing in frequency and intensity due to climate change in many parts of the broadacre cropping regions globally, presenting challenges to food production. For cool-season grain-growing regions, where summers are already too hot, heat and frost risks can limit adaptation options. Capturing these stresses in crop models accurately is increasingly important for evaluating the timing, severity, and yield consequences of extreme events. Quaking aspen (Populus tremuloides) forests in Northern Arizona provide critical habitat for endangered bird species and support regional tourism and recreation. However, aspen populations are declining due to a combination of abiotic and biotic stressors. In response, land managers implemented strategies, such as ungulate exclusion fencing, prescribed burns, and stand thinning, but efforts are constrained by limited data on where and when to intervene. This project worked in collaboration with the U.S. Forest Service, the National Park Service, the Arizona Department of Forestry and Fire Management, and Northern Arizona University School of Forestry. Coral reefs are biodiverse coastal ecosystems at risk of exposure to spilled oil; spills reaching shorelines can damage corals, mangroves, and many other species in nearshore shallow-water tropical environments. Efforts to remediate such spills have evolved over the years through the introduction of various chemical response agents that efficiently and effectively control the spread of oil. Chemical herding agents and dispersants, which can contain and control the oil spread, are promising additions to the spill response toolbox as potential alternatives to other response methods.
