Protocol For Vibratome Sectioning, Immunofluorescence, And S-Phase Labeling Of Inner Ear Organoids, Jiayi Wu, Stefan Heller, Maggie S. Matern

Inner ear organoids represent a potentially inexhaustible source of otic tissues, including sensory hair cells and supporting cells, for in vitro manipulation. Here, we present a protocol for labeling S-phase entry of cells in inner ear organoids using 5-ethynyl-2′-deoxyuridine (EdU), followed by fixation and vibratome sectioning. Nuclear EdU is then detected alongside protein markers of interest via immunofluorescence. This workflow enables the visualization of cell and tissue morphologies within developing organoids and assessment of how different manipulations affect cell proliferation.

Two additives, rosemary oil and a commercial herbal product (HP; AROTEC-G) containing garlic essential oil, carvacrol and thymol were investigated for their potential to reduce the parasitic burden in yellowtail kingfish (YTK; Seriola lalandi) infected with Zeuxapta seriolae. Naturally infected fish were fed diets supplemented with either 2.5 mL.kg−1 rosemary oil or two concentrations of HP (5 and 10 g.kg−1) for 20 days. At 10 and 20 days, the effect of the diets on parasite abundance was evaluated. Feed intake and growth were similar across treatments (p > 0.05). By day 10, both rosemary oil and HP significantly …

Theory and past experimental work suggest that as males age, the strength of their mate preference should decrease. However, the empirical work investigating this question has primarily been conducted in insects that have very short life spans and often live for just a single mating season. This leaves a gap in our understanding of the relationship between male mate preference and age across taxa, as age can conflate with other ecological changes in a single mating season. Purpose: B7 homolog 3 (B7-H3) is a promising target for antibody-drug conjugates, with ifinatamab deruxtecan demonstrating an objective response rate of 54.8% in previously treated extensive-stage small cell lung cancer (SCLC). This analysis aimed to characterize B7-H3 RNA expression with reference to SCLC molecular subtypes (SCLC-A, SCLC-N, SCLC-P, and SCLC-I) and immune-related parameters.
Experimental design: Tumor RNA expression and mutational burden for 1,721 patients with SCLC were derived from a real-world database (Caris Life Sciences). Understanding how genes influence drug responses is critical for advancing personalized cancer treatments. However, identifying these gene-drug interactions in a physiologically relevant human system remains a challenge, as it requires a model that reflects the complexity and heterogeneity among individuals. Here we show that large-scale CRISPR-based genetic screens, including knockout, interference (CRISPRi), activation (CRISPRa), and single-cell approaches, can be applied in primary human 3D gastric organoids to systematically identify genes that affect sensitivity to cisplatin. Our screens uncover genes that modulate cisplatin response. By combining CRISPR perturbations with single-cell transcriptomics, we resolve how genetic alterations interact with cisplatin at the …

In this study, the relationship between the Growth Differentiation Factor-9 (GDF9) gene polymorphism and birth weight, as well as weights on days 30, 60, 90, was investigated in Akkaraman lambs, one of the indigenous sheep breeds of Türkiye. For this purpose, a total of 314 lambs were genotyped by PCR-RFLP method for the single nucleotide polymorphism (SNP) c.260G>A in the GDF9 gene. While GG and GA genotypes were observed in the studied breed, the AA genotype was not observed. The results showed that Akkaraman lambs were in Hardy–Weinberg (H–W) equilibrium for the GDF9-c.260G>A polymorphism (p < 0.05). In this study, a significant association was observed between GDF9-HhaI polymorphism and 30th day weight and 60th day weight in Akkaraman lambs. When the animals were analyzed according to birth type, it was observed that this factor was associated with birth weight, as well as weights on days 30, 60, and 90. Optimizing duck production requires understanding growth variability and monitoring serum calcium and magnesium levelsas health indicators. This study aimed to analyze the growth curve, mortality, and serum calcium (Ca) and magnesium (Mg) levels in White Mallard ducks from hatching to the prelaying period in subtropical conditions. A total of 1100 females and 270 males were raised for 146 days. At hatch, day 6 (D6), D13, D23, D34, D50, D64, D80, D94, D111, and D146, ducks were weighed separately to assess body weight (BW). Avians use significant amounts of energy during flight that is supplied by muscles. The Common Pheasant, a member of the Phasianidae family, can fly swiftly over short distances and flies to evade predators. Pheasant species are terrestrial and have a global distribution. The phylum Acanthocephala contains approximately 1300 acanthocephalan species. So far, only nine acanthocephalan species of wild birds have been reported in Türkiye. To study the context of acanthocephalan diversity in wild birds in Türkiye, we examined 193 birds from 69 species belonging to 16 orders. Background: Biological age can be quantified by composite proteomic scores, called proteomics-based aging clocks (PACs). We investigated whether a discrepancy between chronological and biological age in midlife and late-life is associated with cognition and dementia risk.
Methods: We used two longitudinal population-based studies: the Atherosclerosis Risk in Communities (ARIC) Study and the Multi-Ethnic Study of Atherosclerosis (MESA). Appropriate land management has a long-term impact on soil performance and is believed to improve soil fertility. This study investigated the effect of eco-enzymes on soil nutrients and plant productivity of green mustard-peanut in inceptisol. The research was conducted from February to April 2025. Diffuse mesothelioma (DM) is a rare but highly aggressive and treatment resistant neoplasm with low survival rates. Effective therapeutic strategies are limited, and resistance to treatment is a major obstacle. Myeloid Cell Leukemia (MCL)-1 and B-cell leukemia (BCL)-xL are anti-apoptotic B-cell lymphoma 2 (Bcl-2) family proteins that block cell-intrinsic apoptosis through interactions on the mitochondrial outer membrane which contribute to therapeutic resistance.

International Journal of Exercise Science 18(4): 747-756, 2025. The objective was to investigate screen time (ST) and objectively measured sleep quality in college students. Participants were undergraduate students attending a large metropolitan public university in the southeastern U.S. Participants wore accelerometers on their wrist for 24hr/day for 14 days to capture sleep outcomes, and completed a survey reporting ST duration. Analyses explored weight-status, race/ethnicity, and sleep patterns (presence of daytime sleep) differences among students. P. falciparum possesses the most AT-rich genome sequenced to date, with long AAT repeats coding for asparagines (N) in intragenic regions. As a result, nearly a quarter of its proteome is composed of proteins with poly-N repeats. Hematopoietic stem cells (HSC) maintain both a self-renewal capacity to persist throughout life and an ability to differentiate into mature cell lineages. HSC self-renewal programs are often ectopically activated in leukemia cells to sustain developmental arrest and neoplastic proliferation. Recently, the transcriptional regulator Bclaf1 (Bcl2 associated transcription factor 1) was shown to support proliferation and block differentiation of AML blasts. Bclaf1 is ubiquitously expressed in hematopoietic cells but its function in normal hematopoiesis has not been elucidated. Our data demonstrate that BCLAF1 promotes HSC development and function. Small, isolated populations often have low genetic diversity and low potential for adaptation in response to environmental changes. Alternatively, isolated communities are often more likely to display local adaptations and over time speciate. The Great Lakes Region is home to over 50 vascular plant species that are disjunct from the species’ core populations. Agoseris glauca (Asteraceae) is a Great Lakes disjunct and Michigan state threatened species geographically isolated from its core range by 900 km. Ancestry-informative markers (AIMs) are genetic variants that exhibit significant frequency differences between populations, making them valuable tools for inferring biogeographical ancestry in forensic investigations. Insertion/deletion polymorphisms (INDELs) offer particular advantages for degraded forensic samples due to their short amplicon sizes and binary allelic nature. During a natural history hike at La Selva Biological Station in Costa Rica, we observed a Brown Vine Snake, Oxybelis koehleri, consuming the autotomized tail of a Common Humble Anole, Anolis humilis, which had escaped predation. This event represents a rare documented instance of a vine snake feeding exclusively on a detached lizard tail. Tail autotomy is a widespread anti-predator strategy among lizards, with the shed tail continuing to writhe post-detachment, potentially attracting predators. Lizard tails are lipid-rich and offer a convenient energy source, especially in resource-scarce environments. Although such behavior has been noted in other reptiles, it is seldom …

The ribosome is the central hub for protein synthesis and is heavily targeted by antibiotics. Ribosomal mutations, antibiotic treatment, and nutrient starvation can alter translational efficiency and lead to stressed cells. Ribosome deficiency plays a critical role in stress responses and disease progression; yet, how it affects bacteria-host interactions remains poorly understood. In this study, we show that a ribosome-deficient strain exhibits a surprising morphological change from rod shape to filamentous in Salmonella cells growing inside host macrophages. Background: Aneurysmal subarachnoid hemorrhage (aSAH) is notoriously known for its high mortality and morbidity. Approximately one-third of the patients who survive aneurysm rupture are reported to develop delayed cerebral ischemia (DCI), which contributes to a poor clinical outcome. Currently, there are no biomarkers for identifying which aSAH patients are at risk of developing DCI. We aimed to determine the feasibility of cerebrospinal fluid (CSF) exosomal microRNAs (miRNAs) for predicting DCI post-aSAH.
Methods: aSAH patients were prospectively enrolled, and CSF samples were collected at two time points (< 24 h and 72 h post-aSAH) from individuals undergoing external ventricular drainage. Exosomal miRNAs were isolated from the CSF for analysis. In the initial group of patients (discovery cohort), an exploratory analysis was conducted using a CSF panel containing 84 miRNAs, assessed by quantitative real-time PCR (RT-qPCR). Based on this analysis, 27 miRNAs were selected for further evaluation in a second group of patients (validation cohort). Among these, 10 miRNAs had previously been reported in SAH-related CSF studies, supporting their relevance for continued investigation.

Objectives: We investigated the performance of a slow computed tomography (CT) protocol to reduce alignment errors arising from motion when using CT-on-rail (CTOR) for image guidance for patients receiving thoracic stereotactic body radiation therapy (SBRT).
Methods: A Quasar lung phantom with a moving tumor was programmed with three breathing rates and three motion amplitudes. MIP and average 4DCT images were used for contouring and alignment, respectively. Ten CTOR images were obtained for each of the breathing rates and amplitudes, under both CT protocols. RET solvent-front G810C/R/S mutations confer resistance to the currently approved RET protein tyrosine kinase inhibitors (TKIs) selpercatinib and pralsetinib. Moreover, RET fusion-positive lung adenocarcinoma frequently metastasizes to the brain. To address these challenges, it is imperative to develop a RET TKI that is effective against solvent-front mutations and exhibits intracranial activity. We synthesized alkynyl nicotinamide-based RET TKIs and tested their efficacy in cell cultures in inhibiting selpercatinib/pralsetinib-resistant RET solvent-front mutants G810C/R/S found in cancer patients, and in BaF3/KIF5B-RET(G810C) cell-derived subcutaneous and intracranial tumors in vivo. Objective: Despite the availability of approved treatments, a substantial proportion of patients with bipolar disorder experience treatment-resistant bipolar depression (TRBD), characterized by persistent depressive symptoms unresponsive to standard therapies. However, a universally accepted definition of TRBD is lacking. This consensus document, developed by the International Society for Bipolar Disorders (ISBD) Task Force on TRBD, aims to provide a standardized definition of TRBD to facilitate clinical trials, research, and treatment strategies.

Continuous advances in technologies ranging from deep sequencing and genetic manipulation to mass spectrometry, single cell imaging, and structural biology have led to previously unimaginable advances in our understanding of microbial physiology and the molecular mechanisms underlying microbial responses in a multitude of environments.
