Purpose: This study examined the effects of sustained, isometric forearm flexion tasks anchored to ratings of perceived exertion of 2 (RPE2FT) versus 8 (RPE8FT) on the wavelet-based mechanomyographic (MMG) intensity responses from pretest and posttest maximal voluntary isometric contractions (MVICs) as well as the time course of changes in the MMG intensity responses throughout the sustained tasks. Actively venting high temperature deep-sea hydrothermal vent deposits along tectonic spreading centers and in backarc basins harbor a rich diversity of thermophilic Bacteria and Archaea, many of which have no representatives in cultivation nor any genomic representation in databases. Here, in order to produce a global-scale time series metagenomic resource for studying the microbial functional and genomic diversity in these high temperature ecosystems, we obtained 70 metagenomes from collections across spatial and temporal gradients from 21 different vent fields spanning 16 years (1993-2009). Background: KMT2A partial tandem duplication (PTD) occurs in approximately 5-10% of acute myeloid leukemia (AML) cases and is associated with poor prognosis. While its cytogenetic and molecular features are well described, the immunophenotypic characteristics of AML with KMT2A-PTD remain incompletely defined.
Methods: We identified 47 cases of AML with KMT2A-PTD by optical genome mapping. All cases underwent flow cytometric immunophenotypic analysis and next-generation sequencing using an 81-gene panel.
Results: The cohort included 32 men and 15 women with a median age of 67 years (range, 19-87). Thirty-eight cases were de novo AML, and nine were secondary to myelodysplastic …

Epithelial-to-mesenchymal transition (EMT) is known to induce both stemness and mesenchymal properties, and our findings reveal that these two programs can be uncoupled. During EMT, epithelial cells transition from symmetric divisions producing differentiated daughter cells to self-renewing daughter cells. When we block cell division and induce EMT, cells gain mesenchymal properties but not stemness, suggesting the importance of cell division for gaining stemness. We identified ESRP1 as a key regulator of EMT-driven stemness, which get downregulated during EMT in a cell division-dependent manner. Overexpression of ESRP1 prevents the gain of stemness without affecting the mesenchymal program. Only the stemness and …

Patients with poorly differentiated thyroid cancer (PDTC) and anaplastic thyroid cancer (ATC) face a much poorer prognosis than those with differentiated thyroid cancers. Around 25% of PDTCs and 35% of ATCs carry the BRAFV600E mutation, which constitutively activates the MAPK pathway, a key driver of cell growth. Although combining BRAF and MEK inhibitors can shrink tumors, resistance often develops. The exact cause of this resistance remains unclear. We previously found that in PDTC and ATC cells, the BRAFV600E mutation is strongly linked to the expression of ETV5, a transcription factor downstream of the MAPK pathway. In the current study, we …

Human epidermal growth factor receptor 2 (HER2) is an established therapeutic target in multiple solid tumors, particularly breast and gastric cancers. Significant advancements have been made in the development of HER2-targeted therapies, including monoclonal antibodies, tyrosine kinase inhibitors, antibody-drug conjugates (ADC), and novel bispecific antibodies. These agents have revolutionized the treatment landscape for HER2-positive metastatic cancers, resulting in improved progression-free and overall survival, and quality of life for patients. GLUT1 facilitates a continuous flow of glucose across the inner and outer blood-retinal barriers (BRBs) to support vision. To understand the extent to which photoreceptors rely on glucose transport across the outer BRB, we generated a tamoxifen-inducible conditional knockout of Slc2a1 in the retinal pigment epithelium (RPE) (RPE-iΔGlut1). In the RPE-iΔGlut1 mice, rod photoreceptors exhibited impaired outer segment renewal and decreased the expression of proteins involved in phototransduction and ciliary transport. Proteins regulating the retinal stress response increased. Cone photoreceptors were functional and viable 15 months post-tamoxifen treatment in the RPE-iΔGlut1 mice, while 70% of the rods died. …

In vitro fertilization is a relatively new medical practice that helps people who face infertility be able to reproduce. Reproductive medicine providers face ethical dilemmas often in their field, only some of which have standards of care. There are questions in the medical landscape pertaining to physician autonomy versus patient autonomy, which heavily apply to reproductive medicine providers. Patient autonomy is highly valued in medical ethics; however, physician autonomy should also be considered. [Objective ] This study aimed to investigate the effect of microwave heating on the fructan mass fraction in steamed cakes prepared with Saccharomyces cerevisiae and elucidate the molecular mechanism of the activation of invertase by microwave.[Method ] Steamed cakes were prepared under varying microwave power density (3.40~5.10 W/g ) and heating time (2~5 min ),and the fructan mass fraction was compared between steamed cakes prepared by microwave heating and conventional steaming.The activity of invertase from Saccharomyces cerevisiae was measured under microwave heating and water bath heating conditions.Molecular dynamics simulations were performed to explore the invertase-kestose interactions under microwave fields.[Result ] …

[Objective ] This study aims to elucidate the transformation process of black ginseng saponins and their immunomodulatory mechanisms.[Method ] An ultrasound-assisted enzymatic method was employed for the transformation of black ginseng saponins,and the mass concentration of black ginseng saponins was determined.Furthermore,the immunomodulatory effects of black ginseng saponins were investigated through animal experiments.[Result ] Under the conditions of an enzymatic hydrolysis temperature of 40 ℃,an enzyme mass fraction of 8%,enzymatic hydrolysis time of 60 min,and ultrasonic treatment time of 25 min,the mass concentration of black ginseng saponins increased from 1 615.40 μg/mL to 3 351.38 μg/mL,which represented an increase of 107.47%.The …

[Objective ] This study aims to screen the strains of lactic acid bacteria (LAB ) suitable for Suancai fermentation.[Method ] Traditional fermented Suancai from the Dai ethnic region of Yunnan was used as the research samples for the isolation and purification of LAB.The purified isolates were evaluated for fermentation characteristics,including nitrite degradation,acid production,acid resistance,and salt tolerance.Additionally,the antimicrobial activity,antibiotic susceptibility,hemolytic activity,and amino acid decarboxylase activity were assessed to identify dominant strains.[Result ] A total of 208 LAB strains were isolated from 40 Suancai samples.Preliminary screening based on nitrite degradation identified 20 strains with high efficiency,and 16S rDNA sequencing identified 15 strains …

[Objective ] This study aimed to explore the antioxidant and anti-fatigue properties of Malus spectabilis fruit polysaccharides (MPPs ) and analyze the structure-activity relationships.[Method ] Two types of MPPs were prepared via acidic (AC-MPC ) and alkaline (AL-MPC ) extraction methods.The polysaccharide composition was identified by high-performance anion-exchange chromatography (HPAEC ).The changes in mouse body mass,serum biochemical indicators,and exhaustive swimming time were measured to assess the in vivo antioxidant and anti-fatigue effects of MPPs.[Result ] Galacturonic acid was the main component in both types of MMPs,while AL-MPC had higher mass fraction of total polysaccharides.The in vivo experiments demonstrated that both …

Pasture-based ruminant systems link nitrogen (N) nutrition with ecosystem N cycling. Grazing ruminants convert fibrous forages into milk and meat but excrete 65 to 80% of ingested N, creating excreta hotspots that drive ammonia volatilization, nitrate leaching, and nitrous oxide (N2O) emissions. While disinfection byproducts (DBPs) are typically measured at drinking water treatment plants, levels can change dramatically within the distribution system before reaching the consumer. In this study, the spatio-temporal trends of 66 DBPs across 9 different classes were examined in two drinking water distribution systems with similar source waters, but different pretreatments and residual disinfectants. One system uses residual chlorine in the distribution system, and the other uses chloramine, allowing for examination of how DBP concentrations change over time in distribution systems with different residual disinfectants. Agricultural technologies are widely seen as a driver for sustainability, resilience, and food-system transformation. Yet in 2025, agtech startup across North America, Europe, Asia, and Africa experienced multiple shutdowns. Although each company closed for its own reasons, the failures highlight consistent underlying patterns. We analyze 18 publicly reported shutdowns across controlled-environment agriculture (CEA), robotics, insect protein, digital platforms, sensors, and ag-biotechnology. We find that most ventures struggled not with scientific feasibility but with economic and adoption dynamics at the farm level. Epigenetic modifications such as DNA methylation play a fundamental role in oncogenesis and the progression of neoplasms neoplasias. DNA methyltransferase inhibitors (DNMTi) constitute a family of therapeutic agents that impede the methylation at the 5-position on cytosine nucleotides, thereby modulating the epigenetic regulation of tumor suppressor genes, oncogenes, and other key regulatory genes. The first-generation DNMTi azacitidine and decitabine have demonstrated substantial efficacy in the treatment of medically non-fit, older patients with acute myeloid leukemia (AML) or myelodysplastic syndrome (MDS) ineligible for intensive chemotherapy (IC), by virtue of their favorable safety profile. Hypoxic eye diseases represent a pivotal yet often underappreciated contributor to the onset and progression of many retinal disorders. When hypoxia persists or exceeds the tissue's compensatory capacity, it triggers pathological retinal neovascularization, blood-retinal barrier disruption, and neuronal apoptosis, ultimately resulting in irreversible visual impairment. Connexins (Cxs) form gap junction channels and hemichannels and regulate retinal cell proliferation, differentiation, and survival, thereby playing a central regulatory role in the pathogenesis of hypoxic ocular diseases. In addition to gap junctions, Cx hemichannels promote transmission of molecules between intra- and extracellular environments, further influencing retinal homeostasis under hypoxic stress. Microbial communities associated with desert plants play a pivotal role in enhancing host survival under extreme environmental stressors, including drought, salinity, and nutrient limitation. The Desert Plant Endophyte Microbial Collection is one of the largest curated repositories of 2500 cultivable endophytic bacteria isolated from 23 native desert plant species across Saudi Arabia, Jordan, and Pakistan. The purpose of this research was to examine if there are any variances in a female athlete’s ability to produce power, that could be correlated with the menstrual cycle phases. The study observed three elite female athletes throughout 13 weeks, collecting relative propulsive power and relative braking power data from their vertical jump performances which occurred multiple times a week. At the end of the 13 weeks, participating athletes were asked to download their personal Oura Ring data.
