The Determination Of The Apparent Dissociation Constants Of Arsenic Acid In Seawater, Douglas H. Lowenthal, Michael E.Q. Pilson, Robert H. Byrne

The apparent dissociation constants for arsenic acid in seawater, based on a free molal hydrogen scale, were determined in artificial seawater over a range of salinities and temperatures. A comparison between constants determined in a sodium chloride solution and artificial seawater of the same ionic strength suggests that a significant amount of the reactive arsenate in seawater is ion paired with cations other than sodium. Under typical seawater conditions, arsenic acid appears to be less dissociated than its analogue, phosphoric acid. Protein subunit structure in the nuclear envelope has not been previously described. Covalent crosslinking of polypeptides provides a method for studying the subunit structure of protein-rich systems. In this study, several polypeptide crosslinking methods have been used to determine associations among polypeptides in isolated chicken erythrocyte nuclear envelope. Interchain associations that exist in the isolated membrane were fixed by crosslinking the polypeptides in the isolated nuclear envelope. Crosslinked and uncrosslinked polypeptides were resolved by electrophoresis in a dissociating detergent system. Each agent caused distinctive alterations in the gel pattern. Myosin and myosin light-chain kinase have been isolated and characterized from small quantities of normal and SV40-transformed, murine astrocytic neuroglial cells in culture and from intact normal mouse brain. Sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis of the astrocyte myosins revealed a heavy chain of 200,000 daltons and two light chains of 20,000 and 15,000 daltons. These myosins are similar to other cytoplasmic myosins. The astrocyte 20,000-dalton light chain can be phosphorylated by an endogenous myosin light- chain kinase which has properties similar to those of the myosin light-chain kinase found in human platelets. The feeding ecology of the black sea bass, Centropristis striata, associated with artificial reefs shows seasonal and size-related changes. Monthly collections totaling 520 black sea bass taken from an artificial reef near the Chesapeake Light Tower, Virginia, between August 1975 and May 1976, were analyzed for food habits as revealed by number, volume, frequency of occurrence and a modified Index of Relative Importance.
The black sea bass feeds on the artificial reef as well as the adjacent areas. When food was abundant on the reef, food items from the reef and the surrounding area were consumed. Upon depletion of the …

Duck plague is a hemorrhagic, necrotic disease found exclusively in the order Anseriformes, family Anatidae (ducks, geese, swans). The etiologic agent has been reported to be a herpes-like virus. The Holland strain of duck plague was found to induce cytopathic effects in cell culture (perinuclear vacuolation followed by lysis). A simple reproducible plaque assay for quantitating the virus was developed. Under conditions of the plaque assay, one-hit kinetics were observed. The host range of duck plague virus in cell culture was found to include the order Galliformes, family Phasianidae (pheasants, quails and partridges). Cell-free extracts and whole cell suspensions of Desulfovibrio vulgaris were employed to examine the fixation of 14CO2 into acid stable material. Moderate levels of CO2 fixation occured in the presence of ribulose-1,5-diphosphate, ribose-5-phosphate or pyruvate with only minimal levels of CO2 fixation occuring with formate, malate, or succinate. The fixation of CO2 in the presence of pyruvate was attributed to the pyruvate-CO2 exchange reaction. This exchange reaction was dependent on the presence of an acidic protein fraction but was not stimulated by the additions of ATP to the reaction mixture. The fixation of CO …

Bvr-1, A Restriction Locus Of A Type C Rna Virus In The Feline Cellular Genome: Identification, Location, And Phenotypic Characterization In Cat X Mouse Somatic Cell Hybrids, Stephen J. O'Brien

Somatic cell hybrids were constructed between BALB/c-RAG mouse cells and feline lymphoma cells by the hypoxanthine-aminopterin-thymidine selection scheme. RAG cells spontaneously produce an endogenous B-tropic type C virus. Cat-mouse hybrids preferentially segregate feline chromosomes and retain murine chromosomes,demonstrable by karyotypic and isozyme analyses. Despite the presence of the complete mouse genome, including the viral genome, virus production was diminished to 1-5% of the levels observed in RAG parents based upon particle-associated RNA-dependent DNA polymerase (reverse transcriptase) activity in the culture fluid. With the expanding use of plutonium-239 (239Pu) as a nuclear reactor fuel and increased recycling of spent reactor fuel to recover 239Pu, the possibility of an accidental human exposure exists. Little information is available on the effects of inhaled 239Pu on immune mechanisms. This study was performed in order to examine the effects of 239Pu dioxide (239Pu02) inhalation on the ability of mice to be immunized against a bacterial respiratory infection in which cell-mediated immunity is the primary mechanism of host resistance to infection. Cells enzymatically dispersed from ovine thyroid tissue were cultured in Ham's F-10 medium supplemented with 8.33% Fetal Calf Serum and 8.33% Lamb Serum. Cultures were maintained in a humidified 37° C incubator having a 5% CO2 - 95% air atmosphere. At 48 hours post isolation, cell culture medium was replaced and experiments begun. Altering the serum content (between 4.76 - 16.66%) of the nutrient medium had little effect upon either DNA content or 3H-thyrnidine incorporation into DNA. Thyroid-Stimulating Hormone (TSH) seemed to have a stablizing effect upon DNA content (between 0 and 7 days post TSH addition), and …

Maximal oxygen consumption is consistently lower when measured during swimming than during running or bicycling. A number of factors could contribute to this difference, including size of active muscle mass, body position, heat exchange mechanisms, breathing patterns and adjustments to water immersion.
In this study, the effects of water immersion on submaximal exercise were isolated by having subjects perform identical work in air and water. The exercise consisted of rowing on a rowing machine, with rate of movement controlled so that the subjects always worked at the same relative workload and oxygen consumption. Ultrastructural aspects of interlamellar Henneguya exilis infections in channel catfish are reported. The plasmodium wall of this form differs from that of other species in that it is composed of two outer unit membranes which give rise to a zone of numerous pinocytic canals. Single-membraned canals appeared to be a stable feature of the wall while double-membraned canals are interpreted as those actively carrying out pinocytosis. Evidence suggests that host cellular cytoplasm as well as interstitial material is taken in by plasmodia. This report describes research involving biological and chemical analysis of two undeveloped embayments on Kentucky Lake, namely Anderson and Vickers Bays. Field and laboratory studies were made to assess current biotic standing crops, limnological conditions, levels of inorganic and organic pollutants in the embayments.

Effect Of Altitude On The Peripheral Degradation Of Thyroxine In The Uinta Ground Squirrel (Citellus Armatus), Stuart K. Ware

Various methods were employed to detect changes in the peripheral degradation of thyroxine (T4) in adult Uinta ground squirrels (Citellus armatus) collected at two different altitudes (5400 and 9000 feet.) Plasma half-life of injected T4-125I (tl/2) was significantly decreased with altitude, as was T4-125I distribution space (TDS) and T4 degradation and secretion rate (TSR). In vitro deiodination of T4-125I by heart homogenates was not affected, while urinary excretion of thyroxine derived iodide was increased at high altitude 44-70 hours post-injection. Both total and free serum T4 concentrations were reduced at high altitude, although not significantly. The cause of the reduced …

Population Characterization And Feeding Behavior Of A Subtidal Neogastropod, Amphissa Columbiana, Bruce C. Stone

The neogastropod Amphissa columbiana is found abundantly in subtidal areas of the San Juan Archipelago. Two populations studied were composed of distinct age classes of sexually immature and mature specimens. Reproduction appears to take place in the fall by snails at least two years old. Amphissa is a chemoreceptive scavenger that usually feeds on carrion and on dead and injured animals. Feeding behavior is mediated by chemical attractants from dead and traumatized tissue. An unusual feeding association is formed where Amphissa congregate around feeding predators to share their meals. A specific defense mechanism is deployed by Amphissa especially in response …

The objective of this study was to determine the feasibility of raising carp on pig manure and coli, Brassica campestris. The use of pig feces as a source of fish food has been questioned by researchers (Hickling 1968). Coli was selected for dietary intake by fish since it is used for human consumption by the Indians of Guatemala. Commercial fish food (Silver Cup) was used for a control diet. These diets were both combined and used as a single source of food.

There is at present no recent comprehensive manual of the Great Plains flora. Consequently, students of the flora must utilize monographs, revisions, and other sources in their work. This bibliography was compiled in order to facilitate location of available literature, and will be useful to anyone interested in the systematics of the plants of the Great Plains. This compilation was produced for the Great Plains Flora Association as an initial step in the preparation of a major flora for the region.

Sporocysts of Isospora endocallimici, a parasite of marmosets, were exposed to minimal essentials medium (MEM) or a trypsin-bile salt solution (TBS) and then fixed and prepared for trans- mission electron microscopy. Excystation occurred in TBS but not MEM. The sporocyst wall has 2 layers, a thin outer layer (15 to 110 nm thick) and a thick inner layer (65 to 180 nm thick), which is composed of 4 separate curved plates. The outer layer consists of 1 to 3 membranes interspersed with lipid droplets. In the inner layer, a thin layer of material connects the peripheral margins of 2 …
