A method has been developed to deliver an iodinating system into the confines of the phagolysosome, allowing us to study the nature of the phagolysosomal membrane. Lactoperoxidase (LPO) is covalently coupled to carboxylated latex spheres (LPO-latex) in a stable enzymatically active form. The addition of LPO-Iatex to cultured macrophages leads to their rapid attachment, ingestion, and enclosure in a plasma membranederived phagocytic vacuole. These organelles rapidly fuse with preexisting lysosomes and are converted to phagolysosomes (PL) that demonstrate both acid phosphatase and lactoperoxidase activities. Characterization Of Phagosome-Lysosome Fusion In Macrophages, Margaret Carol Kielian

Several approaches were used to study the determinants of phagosome-lysosome (P-L) fusion in intact mouse macrophages. Lysosomes were labeled with the fluorescent vital dye acridine orange and the rate and extent of their fusion with yeast-containing phagosomes was monitored by fluorescence microscopy. Fusion was also assayed by electron microscopy, using horseradish peroxidase or thorium dioxide as markers for secondary lysosomes. Good agreement was found with results obtained from vitally-stained cells, thin section samples using an enzymatic marker, and thorium dioxide-labeled samples evaluated by stereology. Transcriptional And Translational Products Of Avian Erythroblastosis And Avian Myeloblastosis Viruses, Steven Mathias Anderson

The transcription and translation products of two avian acute leukemia viruses, avian erythroblastosisvirus (AEV) and avian myeloblastosisvirus (AMV), were examined. AEV is defective in virus replication and requires the presence of a helper virus to produce infectious progeny. AEV-transformed nonproducer cells were found to contain no proteins participating in virus replication. Instead, nonproducer cells were found to contain a 75,000 Dalton gag-related polyprotein (p75) which contains the p19 region of the gag gene at its amino terminus. Two-dimensional tryptic peptide mapping indicated that p75 contained novel peptides in addition to those derived from p19. p75 was found to be a …

The study of the parasitic protozoa presents a multifaceted opportunity for biological research. These organisms are eucaryotic cells with many of the biological systems described in higher eucaryotic cells. However, they are adapted to highly specialized environments, invoking sometimes subtle and often unique variations of these biological systems. More important, an understanding of the biology of the parasite and the differences between the parasite and mammalian cells can be invaluable in the development of pharmacological and immunological methods of control of the parasite. The subject of this thesis, the parasitic amoeba Entamoeba hystolytica, is an ideal subject for such research. …

Adenovirus-2 messenger RNAs are transcribed from nine separate transcription units which function during early, intermediate and late stages of infection. I have sequenced capped 5' termini of messenger RNAs from each of these transcription units, and located the templates for each capped terminus in the sequence of the Ad2 genome. These results define the putative initiation sites and promoters for all major Ad2 transcripts. By comparing the termini and promoter DNA sequences I conclude that seven of nine initiation sites including EIa, EIb, PIX, major late, EIII, EIV and the late form of region IIa mRNA have clearly defined TATA …

The Ord River Irrigation Area is in the Kimberley Region of Western Australia, 3,200 kms from Perth, the State’s Capital City.
The area is located in the arid tropics with a climate suitable for high plant growth rates provided water supply is adequate.
With only 12,000 ha of land developed out of a potential 72,000 ha of clay soils the area has the potential for further development of a major irrigated agricultural enterprise.

he fish community of a large artificial reef made In 1960 of 800 concrete blocks in a seagrass bed on the south coast of St. John, U.S. Virgin Islands, was studied by comparing a series of visual censuses (1975-1979) with each other and with a 1962 total-poison station collection (Randall, 1963). Seit mehr als 20 Jahren führen Biologen der Staatlichen Mongolischen Universität Ulan-Bator und von Akademie-Instituten der Mongolischen Volksrepublik gemeinsam mit Zoologen und Botanikern befreundeter wissenschaftlicher Partnereinrichtungen insbesondere der DDR, der UdSSR, der CSSR sowie der Volksrepublik Ungarn in großem Umfang Expeditionen zur Erforschung der biologischen Ressourcen der MVR durch. Im Rahmen dieser Expeditionen wurde unter anderem ein umfangreiches Arthropodenmaterial gesammelt, dessen Bearbeitung einen wesentlichen Beitrag zur Erschließung der mongolischen Entomofauna darstellt. Mit zwei großen komplexen biologischen Sammelexpeditionen 1962 und 1964 der Akademien der Wissenschaften der DDR und MVR in die Süd- und Westmongolei wurde der Grundstein für die kontinuierliche Entwicklung der Wissenschaftskooperation auf biologischem Gebiet zwischen beiden Staaten gelegt. Die Kollektionen wurden zum Grundstock eines aufzubauenden botanischen und zoologischen Sammlungsfundus in Ulan-Bator. Durch den Freundschaftsvertrag zwischen den Universitäiten Halle und Ulan-Bator im Jahre 1967 wurden die Kontakte neu aktiviert. An evaluation of the economics of supplemental irrigation when using a surface water supply must be site specific in order to account for variations in soil moisture holding capacity, watershed area supplying the runoff, climatic conditions, and proposed irrigation management procedures.
With the use of farm specific simulation models to determine grain yields, availability of irrigation water, and economic expenditures involved in irrigation, an economic evaluation of supplemental irrigation can be performed, In the model presented in this report, the Duncan SIMAIZ model is used to predict grain yields using long-term daily weather information. Various patterns of resource utilization in the feeding strategy of Valenciennellus tripunctulatus, a small mesopelagic fish of tropical-subtropical oceans, were explored. Ivlev electivity indices suggest that copepods in general and the genus Pleuromamma in particular are being actively selected. Prey-size selection was apparent and strong negative indices were calculated for all plankton smaller than 1 mm. Non-crustacean prey in all size classes also gave negative indices. Strong positive indices were indicated for prey size classes greater than 2 mm. A model was developed to estimate grazing rates and efficiencies which is linearly influenced by swimming speed and plankton abundance, …

A model was constructed which relates energy in diet to metabolic requirements for the mesopelagic zooplanktivore fish Valenciennellus tripunctulatus. Linear and Von Bertalanffy growth models were used to explore the energetic consequences of a number of feasible age/growth strategies. Maxlmum efficiencies were obtained with linear growth to about 30 mm in 1 y. Efficiencies declined rapidly for slower growth strategies. It was hypothesized that an 'annual' life history strategy was optimum based on energetic considerations. Copepod prey in the 1-2 mm size class were an order of magnitude more abundant and over 4 times higher in caloric content than …

Seventy-two 4-week-old pigs were randomly assigned by sex (gilts, barrows, boars) to two dietary treatments and four age periods for determinations of the effects of age, sex and Ca and P levels on the mechanical properties of various bones in swine.

Lack of standardized test procedures has resulted in considerable variation in reported values for bone strength. Such variation can be attributed in part to the type of instruments used to determine physical properties of bone, procedures used to prepare the bones for testing and equations used to calculate strength. If bone strength is to be used as a major criterion of response in mineral nutrition research, standardization of procedures for measuring and reporting bone strength is essential. One-hundred and ninety-two crossbred weanling gilts (7.25 kg) were assigned to one of two treatments - (A) .65% Ca, .50% P or (B) .975% Ca, .75% P - and fed to an average weight of 92.8 kilograms. Average daily gain (.65 and .67 kg, respectively) and feed/gain were not affected by dietary treatment, but average daily feed intake was lower (P<.05) for gilts on treatment A (1.87 vs 1.95 kg).

River otter (Lutra canadensis) management in Arkansas is hampered by a lack of information on population parameters. This initial study on the biology of Arkansas river otter is concerned with present distribution and harvest trends. Otter occur throughout Arkansas, except in the upper Ozark region. A distributional shift, apparently along the Arkansas River, has led to an increase in otter harvest in the Ouachita Mountain region. A dramatic increase in otter harvest over the past four years (1976-1979) is attributable, in part, to a pelt price increase. Sweet dry whey was used to replace 25% and 50% of nonfat dry milk solids in ice cream mix. Forty-five batches experimental and control lots of vanilla ice cream were manufactured. A commercial lactase was used to hydrolyze lactose to over 90% conversion and the lactase was inactivated after hydrolysis. The extent of lactose hydrolysis in the mix was adjusted to 50% or 75% by blending non-hydrolyzed nonfat dry milk and whey solids with hydrolyzed preparations. Extra sweetness of the conversion products permitted reduction of sucrose by 6.25% and 12.5%, respectively, in the 50% and 75% hydrolyzed lactose ice cream …
