The Occurence Of C4-Photosynthetic Plants In Cedar Glades, Jamie S. Ross Apr 1984

The Occurence Of C4-Photosynthetic Plants In Cedar Glades, Jamie S. Ross

Honors Theses

This project began as a continuation and extension of upper level undergraduate biological study in Plant Taxonomy. It began over a year ago with the collection of plant specimens in the early spring of 1983 and continued throughout that year and into the summer of 1984. The latter half of the project entailed mainly laboratory and library work related to the plant specimens. Collecting was done in Chickamauga Battlefield and was made possible by the granting of a collecting permit by the National Park Service. The collection was restricted to herbaceous vascular plants and most of these were summer annuals. …

Department of Entomology: Faculty Publications

Field trials were conducted in lowland flooded rice in the Philippines to evaluate a number of carbofuran controlled-release formulations in comparison with commercial formulations. The test formulations were based on a biodegradable matrix of pine kraft lignin and were used as granules of different sizes and also in the form of small strips. The release rates were assessed under field conditions by bioassaying rice plants in the field, using adult rice green leafhopper, Nephotettix virescens Distant. The lignin formulations with a high level of active ingredient (15–45% by weight) gave as good or better control than the commercial 3% granules …

Inland Seawalls In Southwestern- Michigan: A Limnological Approach, Jeroen Wagendorp Apr 1984

Inland Seawalls In Southwestern- Michigan: A Limnological Approach, Jeroen Wagendorp

Masters Theses

This research retrospectively evaluates the development of inland lake seawalls in southwestern Michigan. The objective was to look at physically induced phenomena and human induced phenomena, to determine which was most responsible for the need, and large numbers of inland lake seawalls in this region. In addition, the role of the Department of Natural Resources in inland lake seawall development was evaluated.

The findings, obtained through questionnaires, were compared to data from seawalls installed along the eastern shore of Lake Michigan, and showed that man's activity in the littoral zone was the fundamental rationale behind the installation on inland lake …

Isolation Of A Collagenase Cdna Clone And Measurement Of Changing Collagenase Mrna Levels During Induction In Rabbit Synovial Fibroblasts., Robert H. Gross, Lynn A. Sheldon, Colin F. Fletcher, Constance E. Brinckerhoff Apr 1984

Brinckerhoff

Dartmouth Scholarship

To facilitate our studies on the mechanisms controlling collagenase production at a molecular level in rabbit synovial fibroblasts, we have constructed a cDNA library using mRNAs isolated from cells induced with crystals of monosodium urate monohydrate. We have screened this library with cDNA probes made from induced and control mRNA populations. From among 30 clones that hybridized preferentially to the induced-cell probe, 4 contained collagenase sequences. The largest, a clone of 650 base pairs, was identified by its ability to hybrid select a mRNA that could be translated in a cell-free system into a product that was precipitable with monospecific …

Ultrastructure Of The Interface Between Stages Of Eimeria Funduli (Apicomplexa) And Hepatocytes Of The Longnose Killifish, Fundulus Similis, William E. Hawkins, John W. Fournie, Robin M. Overstreet Apr 1984

Ultrastructure Of The Interface Between Stages Of Eimeria Funduli (Apicomplexa) And Hepatocytes Of The Longnose Killifish, Fundulus Similis, William E. Hawkins, John W. Fournie, Robin M. Overstreet

Harold W. Manter Laboratory of Parasitology: Faculty Publications

The interface between stages of Eimeria funduli and hepatocytes of the experimentally infected killifish Fundulus similis was studied ultrastructurally. Parasitophorous vacuoles (PV's) in which meronts, macrogamonts, and microgamonts developed were lined by an inner, smooth membrane and an outer, ribosome-studded membrane. The outer membrane bordered on the cytoplasm of the host cell, whereas the inner one limited the PV. The origins of these membranes have not been determined with certainty, but images were observed in which both membranes appeared to be continuous with the outer nuclear membrane of the host cell. Furthermore, the outer PV membrane was continuous with membranes …

The Fungal Ecology Of The Activated Sludge Process, Douglas William Jaques Apr 1984

The Fungal Ecology Of The Activated Sludge Process, Douglas William Jaques

Masters Theses

Examination of wastewater secondary influent and activated sludge reveal that bacteria and fungi may exist in a negative correlation. Activated sludge may support a resident population of microfungi. All fungi isolated from secondary influent and activated sludge belong to the form-class Deuteromycetes. The density of geofungi in activated sludge may exceed that of bacterial chemolithotrophs and rival that of bacterial heterotrophs.

Purification And Properties Of A Mannitol-Binding Protein From Pseudomonas Aeruginosa, Gregory V. Plano Apr 1984

Purification And Properties Of A Mannitol-Binding Protein From Pseudomonas Aeruginosa, Gregory V. Plano

Masters Theses

Mannitol-binding protein from Pseudomonas aeruginosa strain PAO has been purified 36-fold from magnesium chloride cold-shock supernatants. Sephadex G-100 gel filtration and carboxymethyl Sephadex ion exchange chromotography were utilized in the purification scheme. The position of the mannitol-binding protein band was identified on sodium dodeyl sulfate-polyacrylamide gels. The mannitol-binding protein has an apparent molecular weight of 45,000 as judged by sodium dodecyl sulfate-polyacrylamide gel electrophoresis.

Rhizobium Phaseoli Symbiotic Mutants With Transposon Tn5 Insertions, K. Dale Noel, Araceli Sanchez, Leonor Fernandez, Jan Leemans, Miguel A. Cevallos Apr 1984

Rhizobium Phaseoli Symbiotic Mutants With Transposon Tn5 Insertions, K. Dale Noel, Araceli Sanchez, Leonor Fernandez, Jan Leemans, Miguel A. Cevallos

Biological Sciences Faculty Research and Publications

Rhizobium phaseoli CFN42 DNA was mutated by random insertion of Tn5 from suicide plasmid pJB4JI to obtain independently arising strains that were defective in symbiosis with Phaseolus vulgaris but grew normally outside the plant. When these mutants were incubated with the plant, one did not initiate visible nodule tissue (Nod-), seven led to slow nodule development (Ndv), and two led to superficially normal early nodule development but lacked symbiotic nitrogenase activity (Sna-). The Nod- mutant lacked the large transmissible indigenous plasmid pCFN42d that has homology to Klebsiella pneumoniae nitrogenase (nif) genes. The other mutants had normal plasmid content. In …

Timing Of The Blue Crab Fisheries Of Virginia And Its Application To Harvest Management, J. Dale Shively Apr 1984

Timing Of The Blue Crab Fisheries Of Virginia And Its Application To Harvest Management, J. Dale Shively

OES Theses and Dissertations

The timing of a fishery is a quantitative expression of the composite behaviors of harvesters, markets, animals and climatic events. A quantitative description of the timing of the blue crab (Callinectes sapidus) fisheries of Virginia was established using the time density model of Mundy (1979). Blue crab catch statistics supplied by the National Marine Fisheries Service and the Virginia Marine Resource Commission were divided into two fisheries based on gear type; the summer and winter fisheries. Average performance curves were then constructed using average cumulative proportions of catch. Annual yield estimates were also made and timing in five …

Life History Patterns Of Dytiscidae In A Temporary Pool, Annabel C. Blanco Apr 1984

Life History Patterns Of Dytiscidae In A Temporary Pool, Annabel C. Blanco

Biological Sciences Theses & Dissertations

The Dytiscidae are one of the most distributed families of aquatic Coleoptera. In addition, they comprise one of the largest families in the order. With the exception of some European species, research in the ecology/life history of the Dytiscidae has been limited.

The patterns of appearance and disappearance of Dytiscidae were studied for one growing season, February to June, 1983, in a temporary pool in the Great Dismal Swamp. Fourteen species of larvae and twenty-four species of adults were collected. Definite temporal patterns were evident in larvae of Laccophilus spp., Laccornis difformis, Liodessus affinis, Uvarus granarius, Agabetes acuductus, and Hoperius …

Characterization Of The Dna And Chromosonal Analysis Of Embryonic Dermacentor Variabilis (Say) Cells In Culture, Stanley Nicholas Mason

Biological Sciences Theses & Dissertations

Cultured embryonic Dermacentor variabilis cells were examined by spectrophotometric techniques to determine total cellular DNA concentration and guanine-cytosine content of the DNA and by differential staining procedures to define the karyotype further. The cells were found to have a mean DNA content of 16.98 pg and a DNA guanine-cytosine content of 49%. Chromosome banding attempts resulted in the specific characterization of four chromosomes and differential grouping of the seven remaining chromosomes. A previously unreported submetacentric chromosome was identified.

A Method For The Comparison Of Hdl Containing Apoprotein E And Hdl Cholesterol, Steven A. Knizner Apr 1984

A Method For The Comparison Of Hdl Containing Apoprotein E And Hdl Cholesterol, Steven A. Knizner

Chemistry & Biochemistry Theses & Dissertations

Human serum high density lipoproteins (HDL) were isolated by preparative salt density gradient ultracentrifugation and analyzed further by heparin-sepharose affinity chromatography. Separation of the major classes of serum lipoproteins was achieved after a single ultracentrifugation on a salt density gradient at 272,000 x g for 36 hours in a swinging bucket rotor. After isolation, the total HDL was subdivided into HDL containing apoprotein E and HDL without this apoprotein by heparin-sepharose affinity chromatography. Relative amounts of these subclasses were then calculated.

HDL-cholesterol was measured after precipitation of LDL and VLDL by phosphotungstic acid and magnesium. The statistical relationship between HDL …

Investigation Of The Coenzyme Binding Site Of Lactate Dehydrogenase From Chicken Muscle, Jia-Hwei Lin Apr 1984

Investigation Of The Coenzyme Binding Site Of Lactate Dehydrogenase From Chicken Muscle, Jia-Hwei Lin

Chemistry & Biochemistry Theses & Dissertations

Investigation of the coenzyme binding site of lactate dehydrogenase was approached by using coenzyme analogs. Inhibition studies of this enzyme using n-alkylphosphates as coenzyme analogs were performed with a fluorometric method. The results showed that all of the n-alkylphosphates were competitive inhibitors with respect to NAD+.

The n-alkyl-ADPs were synthesized from AMP and dicycloheaylammonium salts of n-alkylphosphates. The synthetic compounds were subjected to NMR spectroscopic studies, thin layer chromatography and determination of the ratio of adenosine moiety to phosphate groups. The results indicated that the synthetic compounds were n-alkyl-ADPs.

These synthetic compounds were used as coenzyme analogs in …

Phytoplankton Ecology And Dynamics In The James River Estuary Virginia, U. S. A., Margaret J. Filardo Apr 1984

Phytoplankton Ecology And Dynamics In The James River Estuary Virginia, U. S. A., Margaret J. Filardo

OES Theses and Dissertations

Autotrophic biomass and productivity as well as nutrient distributions and phytoplankton cell populations in the James River estuary, Virginia, from Newport News Shipyard to the 0 o/oo isohaline, were sampled on a monthly basis from August of 1981 to December of 1982. Particular emphasis was placed on the very low salinity region (defined as the location where surface salinity measured between 0.0 and 0.75 o/oo based on conductivity) in order to determine the fate of fresh water phytoplankters upon being advected into the estuary because of their suspected role in the biogeochemical cycling that occurs there.

Both chlorophyll a and …

A Description And Analysis Of The Timing Of Virginia's Weakfish Fishery, Bruce William Hill Apr 1984

A Description And Analysis Of The Timing Of Virginia's Weakfish Fishery, Bruce William Hill

OES Theses and Dissertations

The seasonality of Virginia's weakfish fishery is quantitatively defined by providing a mean time of catch and variance of catch for the years 1960 - 1981. On the average. the central half of the population is available for harvest from mid-May through September in all of Virginia's waters and the mean date of harvest is July 20th • The 22 year average variance of catch for Virginia's waters is 3.97. In Virginia's Chesapeake Bay tributaries the mean date of harvest is July 17th and the 22 year average variance of catch is 2.50. There is evidence for an inverse relationship …

A Molecular Phylogeny Of The Grunts (Perciformes: Haemulidae) Inferred From Nuclear Rag1 Gene Sequences, Millicent D. Sanciangco

Biological Sciences Theses & Dissertations

Species and genera of Haemulidae have undergone various taxonomic revisions, however, there is no study that infers the phylogeny of the haemulid genera using morphological or molecular data. The purpose of this study was to use approximately 1386 base pairs of the nuclear Recombination Activation Gene-I (RAG1) from 35 haemulid species representing 13 genera, one species of the closely related Inermiidae, and two species of the outgroup Sparidae to infer an intrafamilial phylogeny of Haemulidae. This analysis is corroborated using approximately 650 base pairs of the mitochondrial Cytochrome Oxidase I (COI) gene and RAG I-COI combined gene analyses of27 haemulids, …

Bioassay Of Solubilized Bacillus Thuringiensis Var. Israelensis Crystals By Attachment To Latex Beads, Danny J. Schnell, Mary Ann Pfannenstiel, Kenneth W. Nickerson Mar 1984

Bioassay Of Solubilized Bacillus Thuringiensis Var. Israelensis Crystals By Attachment To Latex Beads, Danny J. Schnell, Mary Ann Pfannenstiel, Kenneth W. Nickerson

Papers in Microbiology

Solubilized crystals of Bacillus thuringiensis var. israelensis were 7,000 times less toxic to Aedes aegypti larvae than intact crystals, presumably because mosquito larvae are filter feeders and selectively concentrate particles while excluding water and soluble molecules. A procedure is described whereby soluble toxins are adsorbed to 0.8- micrometer latex beads, with retention of toxicity. The latex bead assay should make it possible to analyze the structure and mode of action of the mosquito toxin.

A Survey Of Larval Caddisflies (Insecta: Trichoptera) From Riffles Of Three Streams Of The Drake's Creek Drainage, Kentucky, With A Preliminary Investigation Of The Adult Trichoptera Of The Region, Michael Beiser

Masters Theses & Specialist Projects

A total of 1665 larval caddisflies, representing four families and eight genera, was collected from seven stations in the Drake's Creek drainage from 1 November, 1982 through 15 October, 1983. The pattern of longitudinal distribution of the larvae, based on feeding relationships, in the West and Middle Forks of the system was consistent with studies by Wiggins and Mackay (1978), Andrews and Minshall (1979), and Ross and Wallace (1982) and the River Continuum Concept (Vannote et al. 1980). The Trammel Fork, a spring fed stream, showed a pattern of distribution contrary to the predictions of the Concept, suggesting that the …

Cushing's Disease Due To Pituitary Carcinoma, M. Saeed Zafar, R. C. Mellinger, J. L. Chason Mar 1984

Cushing's Disease Due To Pituitary Carcinoma, M. Saeed Zafar, R. C. Mellinger, J. L. Chason

Henry Ford Hospital Medical Journal

Criteria for the diagnosis of pituitary carcinoma are subject to controversy. Some authors do not accept the demonstration of cellular anaplasia alone as sufficient to establish the diagnosis but require evidence of metastatic disease in order to label a pituitary tumor as malignant. However, pituitary tumors can metastasize even in the absence of anaplasia. The present case of Cushing's disease and pituitary carcinoma had no clinical evidence of metastases when first reported. Partial surgical removal of the tumor followed by irradiation resulted in clinical remission of the Cushing's syndrome, but two years later progressive uncontrollable recurrence of the disease ended …

Activation Of 2-Aminoanthracene By Cell-Free Microbial Extracts, Thomas Hamill Mar 1984

Activation Of 2-Aminoanthracene By Cell-Free Microbial Extracts, Thomas Hamill

Masters Theses & Specialist Projects

The activation of the precarcinogen 2-aminoanthracene was investigated employing cell-free extracts of Escherichia coil, Pseudomonas aeruginosa, Streptococcus faecalis, Bacteroides fragilis, Clostridium perfringens and the eukaryote Candida albicans. A modification of the Salmonella typhimurium/mammalian microsomal mutagenicity assay was employed to determine activation. Of the species tested, only cell-free extracts of Escherichia coli and Bacteroides fragilis failed to induce the reversion of the Salmonella typhimurium his- mutants TA-98 and/or TA-100 to his+ revertants.

Separation of the cytosol and membrane pellet enzymes was completed. Of the organisms tested only the membrane pellet enzymes of Clostridium perfringens showed activation of …
