Contraceptive Hormone Influences On The Role Of Primary Reinforcement And Reinforcer-Enhancement In Nicotine Self-Administration, Kathleen Robin Mcnealy Mar 2025

Contraceptive Hormone Influences On The Role Of Primary Reinforcement And Reinforcer-Enhancement In Nicotine Self-Administration, Kathleen Robin Mcnealy

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Hormonal contraceptives contain a synthetic estrogen (e.g., ethinyl estradiol/EE) and/or a synthetic progestin (e.g., levonorgestrel/LEVO) and can exacerbate nicotine intake. However, whether altered intake reflects changes in nicotine reinforcement or enhancement by nicotine of co-occurring reinforcers is unknown. This reinforcer-enhancement effect is evidenced in rats when nicotine self-administration is increased by the presence of a visual stimulus (VS) reinforcer. This dissertation examined the effects of EE and LEVO on nicotine reinforcement and reinforcer-enhancement in ovary-intact female Sprague-Dawley rats. Rats were implanted with a jugular catheter and assigned to receive daily injections of EE (Vehicle, 0.125 [Low], or 0.18 [High] μg/day; …

Tac4 Analyses Of Mybl1 Knockdown In Mda Mb231 Cells And Identification And Validation Of E2f4 And Tp53 Differential Gene Expression, Emmanuel Butler

Theses (2016-Present)

Triple-Negative Breast Cancer (TNBC) is known to be the most aggressive subtype of breast cancer, accounting for 15–20% of cases. In TNBC, three receptors commonly associated with breast cancer—estrogen receptors (ER), progesterone receptors (PR), and human epidermal growth factor receptor 2 (HER2neu)—are under-expressed. Earlier studies identified MYBL1 as a major transcriptional activator overexpressed in TNBC, playing a critical role in tumor growth, differentiation and apoptosis. In previous research conducted by our lab, MYBL1 was knocked down in a TNBC cell line and the dataset saved for future use. In the current study, we re-analyzed the dataset and identified E2F4 and …

Neutrophil Exosomes: New Pathogenic Entities In Copd, Tiera L. Isom Mar 2025

Neutrophil Exosomes: New Pathogenic Entities In Copd, Tiera L. Isom

Theses (2016-Present)

Chronic obstructive pulmonary disease (COPD) is a progressive respiratory disease that leads to chronic inflammation and, in turn, a high level of comorbidity and healthcare expense. An increasing body of evidence involves neutrophils, as well as their exosomes, in lung damage exacerbation via the protease-mediated degradation of the extracellular matrix (ECM). We hypothesize that the PMN-exosomes would be significantly increased in the active smoker COPD patient and that they would enhance disease progression by augmenting the proteolytic cascade and tissue damage. This multi-center clinical study included a total of 46 veterans separated into four cohorts: active smoking COPD patients, COPD …

Markey Cancer Center Faculty Publications

The insulin receptor (INSR) has been shown to be hyperactive in hepatic stellate cells (HSCs) in humans and rodents with liver fibrosis. To explore HSC cellular mechanisms that INSR regulates during pro-fibrotic stimulation, we used CRISPR-Cas9 technology. We knocked out a portion of the INSR gene in human LX2 HSC cells (INSRe5- 8 KO) that regulates insulin responsiveness but not the insulin-like growth factor (IGF) or transforming growth factor-β (TGFβ) signaling. The INSRe5- 8 KO HSCs had significantly higher cell growth, BrdU incorporation, and lower TP53 expression that suppresses growth, and they also exhibited increased migration compared to the Scramble …

Exploring Oxylipins In Host–Microbe Interactions And Their Impact On Infection And Immunity, Robert J. Neff, Christopher D. Radka

Markey Cancer Center Faculty Publications

Plasma lipids are essential components of biological systems, transported through interactions with proteins to maintain cellular functions. These lipids exist in various forms, such as fatty acids, glycerolipids, glycerophospholipids, sphingolipids, sterols, and prenol lipids, derived from dietary intake, adipose tissue, and biosynthesis. While the association between certain fatty acids and cardiovascular diseases has been widely recognized, polyunsaturated fatty acids (PUFAs) exhibit cardioprotective effects, reducing risks of arrhythmias and heart-related mortality. This is due to their role in the production of eicosanoids, which modulate inflammation. Chronic inflammation, particularly in obesity, is significantly influenced by fatty acids, with saturated fatty acids promoting …

Predicting The Pathway Involvement Of Compounds Annotated In The Reactome Knowledgebase, Erik D. Huckvale, Hunter N. B. Moseley

Markey Cancer Center Faculty Publications

Background/Objectives: Pathway annotations of non-macromolecular (relatively small) biomolecules facilitate biological and biomedical interpretation of metabolomics datasets. However, low pathway annotation levels of detected biomolecules hinder this type of interpretation. Thus, predicting the pathway involvement of detected but unannotated biomolecules has a high potential to improve metabolomics data analysis and omics integration. Past publications have only made use of the Kyoto Encyclopedia of Genes and Genomes-derived datasets to develop machine learning models to predict pathway involvement. However, to our knowledge, the Reactome knowledgebase has not been utilized to develop these types of predictive models.

Methods: We created a dataset ready for …

Prostate cancer (PCa) is one of the leading causes of death among men worldwide. Treatments targeting the androgen receptor pathway remain the standard therapy for PCa patients. Enzalutamide (ENZ), a second-generation androgen receptor inhibitor, was developed to treat castration-resistant prostate cancer. However, while patients initially respond to ENZ, drug resistance typically develops within a few months. Artesunate (ART), a semisynthetic derivative of the Artemisinin plant, is approved for antimalaria treatment. In this study, we conducted an FDA-approved drug screening and identified ART as a potential candidate for overcoming ENZ resistance in PCa. Mechanistically, ART induces the degradation of c-Myc, enhancing …

Department of Pharmacology, Physiology, and Cancer Biology Faculty Papers

Low-dose aspirin is recommended for prevention of hypertensive disorders of pregnancy (HDP) and preterm birth (PTB) in high-risk pregnancies. There is limited data on factors impacting aspirin response in pregnancy. We aimed to evaluate predictors of aspirin response and association with pregnancy outcome with a prospective study of high-risk pregnancies taking 81 mg aspirin daily. Aspirin response was evaluated with Platelet Function Assay-100 (PFA-100) epinephrine closure time at baseline (< 16 weeks' gestation), follow-up 1 (2-4 weeks after aspirin initiation), and follow-up 2 (28-32 weeks gestation). Multivariable regression was used to identify factors associated with PFA-100 at each visit, and results presented with beta coefficient (B) and confidence interval. The median difference (MD) in PFA-100 in those with and without HDP or PTB was compared. Results included 108 who completed follow-up 1 and 96 who completed both visits with > 75% adherence. PFA-100 was increased from baseline at follow-ups 1 and 2 (MD 37 (27-49); MD 26 (15.5-38.5) respectively). At follow-up 1, obesity (B = -30 (-53 to -7) seconds), …

Disrupting Tymp-Mediated Pathways To Block Lymph Node Metastasis In Ovarian Cancer, Emine Bayraktar

Dissertations and Theses (Open Access)

Historically, ovarian cancer (OC) was thought to metastasize by surface-to-surface spread, but recent developments have yielded a new understanding of the paths of metastatic spread. In epithelial ovarian cancer, the mechanisms driving lymph node (LN) metastasis are poorly understood, and it remains unclear whether omental metastasis occurs in parallel with LN metastasis. Utilizing comparative transcriptomic and proteomic profiling of primary tumors, LN metastases, and omental metastases, we identified significant upregulation of thymidine phosphorylase (TYMP) in LN metastases compared to primary and omental tumor sites. Functional studies in murine models demonstrated that TYMP silencing significantly suppressed LN metastasis, underscoring TYMP as …

Evolution Of Mate Guarding Intensity In Crickets As A Possible Adaptation To Sexual Conflict Over Sperm Transfer, Bert Foquet, Jack Thomas Mckermitt, John Hunt, Ben M. Sadd, Scott K. Sakaluk

Faculty Publications – Biological Sciences

Mate guarding is a common behavioral adaptation in males to ensure mating or to hinder current partners from remating with rival males. In the decorated cricket, Gryllodes sigillatus, males transfer a spermatophore to females at mating, comprising a sperm-containing ampulla enveloped in a large gelatinous mass, the spermatophylax, that serves as a nuptial food gift for the female. Males guard the female while she feeds on the nuptial gift and while sperm are evacuated from the ampulla into her reproductive tract. Mate guarding behavior functions to prevent competitors from mating with the female but may also represent a behavioral …

Segregating Sites Enhance Accuracy Of Species Abundance Estimates From Edna, Kara J. Andres Mar 2025

Segregating Sites Enhance Accuracy Of Species Abundance Estimates From Edna, Kara J. Andres

Faculty Publications – Biological Sciences

Species abundance is a fundamental metric in ecology and conservation. Assessing how populations change across space time enables the identification of population trends and informs management and conservation decisions. However, measuring species abundance can be a challenging task, with logistical constraints, sampling biases, and detection limits inhibiting meaningful abundance estimates. Environmental DNA (eDNA) approaches have improved our ability to monitor species presence and biodiversity and may also serve as a tool for measuring species abundance. However, abundance estimates from eDNA typically rely on the correlation between species abundance and the concentration of target species' DNA in a sample, which may …
