Decreasing The Antibiotic Survival Of Multi-Drug Resistant Gram-Positive Organisms, Areej Malik
Biomedical Sciences Theses & Dissertations
Biofilm production plays a crucial role in bacterial antibiotic tolerance and novel approaches are needed to combat it. The stringent response (SR), activated by nucleotide alarmones (pp)pGpp, is crucial for bacterial transcriptional reprogramming. In Clostridioides difficile, the SR is implicated in antibiotic survival, contributing to infection recurrence. Despite its known roles in sporulation and biofilm production in other bacterial species, the broader influence of the SR on C. difficile physiology remains undiscovered. This study deletes the C. difficile SR gene relQ, revealing the pivotal role of RelQ in regulating SR-dependent phenotypes in this organism. The absence of RelQ …

Optimization Of In-Vitro Regeneration And Agrobacterium-Mediated Transformation Of Lactuca Sativa, Mohamed Omer Elsiddig
Theses
Lettuce (Lactuca sativa L.) is a leafy vegetable consumed worldwide, mainly as a green salad. It is an herbaceous, self-pollinated annual plant of the family Asteraceae. Lettuce is low in calories, fat, and sodium. It is a good source of fiber, iron, folate, and vitamin C. Improving existing lettuce varieties for water use efficiency, photosynthesis efficiency, salt tolerance, increased nutritional content, etc., is the need of the hour, considering that lettuce is a crop ideal for vertical farming in the UAE.
This study aimed to develop a highly efficient in vitro regeneration and Agrobacterium- mediated transformation protocol for a popular …

BACKGROUND: Recent studies have reported the predictive and prognostic value of novel transcriptional factor-based molecular subtypes in small-cell lung cancer (SCLC). We conducted an in-depth analysis pairing multi-omics data with immunohistochemistry (IHC) to elucidate the underlying characteristics associated with differences in clinical outcomes between subtypes.
METHODS: IHC (n = 252), target exome sequencing (n = 422), and whole transcriptome sequencing (WTS, n = 189) data generated from 427 patients (86.4% males, 13.6% females) with SCLC were comprehensively analysed. The differences in the mutation profile, gene expression profile, and inflammed signatures were analysed according to the IHC-based molecular subtype.
FINDINGS: IHC-based …

Purpose: Hypomagnesemia is a common side effect of platinum-based chemotherapy and predicts poor overall survival in some cancers. Standard magnesium replacement strategies are often inadequate for maintaining magnesium levels. We hypothesized that a daily dietary magnesium replacement approach through magnesium-rich foods would help maintain adequate magnesium levels during platinum-based treatment.
Materials and methods: We conducted a prospective feasibility study of magnesium-rich diets in patients 18 years and older with previously untreated ovarian cancer scheduled to receive carboplatin-containing chemotherapy of at least six consecutive cycles. Education about magnesium-rich diets was provided at enrollment and then weekly during chemotherapy. Feasibility was defined …

Postmenopausal osteoporosis is caused by estrogen deficiency, which impairs bone homeostasis, resulting in increased osteoclastic resorption without a corresponding increase in osteoblastic activity. Postbiotics have several therapeutic properties, including anti-obesity, anti-diabetic, anti-inflammatory, and anti-osteoporotic effects. However, the beneficial effects of the postbiotic MD35 of Lactobacillus plantarum on bone have not been studied. In this study, we demonstrated that the postbiotic L. plantarum MD35, isolated from young radish water kimchi, influences osteoclast differentiation in mouse bone marrow-derived macrophage (BMM) culture. In addition, it was effective protecting against estrogen deficiency-induced bone loss in ovariectomized (OVX) mice, an animal model of postmenopausal osteoporosis. …

Introduction: Chronic pancreatitis (CP) is a progressive fibroinflammatory disorder lacking therapies and biomarkers. Neutrophil gelatinase-associated lipocalin (NGAL) is a proinflammatory cytokine elevated during inflammation that binds fatty acids (FAs) such as linoleic acid. We hypothesized that systemic NGAL could serve as a biomarker for CP and, with FAs, provide insights into inflammatory and metabolic alterations.
Methods: NGAL was measured by immunoassay, and FA composition was measured by gas chromatography in plasma (n = 171) from a multicenter study, including controls (n = 50), acute and recurrent acute pancreatitis (AP/RAP) (n = 71), and CP (n = 50). Peripheral blood mononuclear …

Of the “Yamanaka factors” Oct3/4, Sox2, Klf4, and c-Myc (OSKM), the transcription factor c-Myc (Myc) is the most responsive to exercise in skeletal muscle and is enriched within the muscle fiber. We hypothesize that the pulsatile induction of MYC protein after bouts of exercise can serve to epigenetically reprogram skeletal muscle toward a more resilient and functional state.

Background
Observing medication ingestion through self-recorded videos (video directly observed therapy [VDOT]) has been shown to be a cost-effective alternative to in-person directly observed therapy (DOT) for monitoring adherence to treatment for tuberculosis disease. VDOT could be a useful tool to monitor short-course latent tuberculosis infection (LTBI) treatment.
Methods
We conducted a prospective randomized controlled trial comparing VDOT (intervention) and clinic-based DOT (control) among patients newly diagnosed with LTBI who agreed to a once-weekly 3-month treatment regimen of isoniazid and rifapentine. Study outcomes were treatment completion and patient satisfaction. We also assessed costs. Pre- and posttreatment interviews were conducted.

Research outputs 2022 to 2026
Sulfur containing compounds including glucosinolates (GLS), sulforaphane (SFN) and S-methyl-L-cysteine sulfoxide (SMCSO) have been proposed to be partly responsible for the beneficial health effects of cruciferous vegetables. As such, greater understanding of their measurements within foods is important to estimate intake in humans and to inform dietary intervention studies. Herein is described a simple and sensitive method for simultaneous analysis of 20 GLS, SFN and SMCSO by liquid chromatography mass spectrometry. Analytes were effectively retained and resolved on an Xbridge C18 column. Detection can be achieved using high resolution or unit resolution mass spectrometry; the latter making the method more …

Soil is an important ecosystem component; it serves as a habitat for many microscopic organisms that play an important role in the ecosystem's maintenance and crops grown in soil. Unfortunately, human activities have adversely affected not only the environment but also the soil health. Soil has been contaminated with heavy metals hydrocarbons and hydrocarbon-based products that are affecting soil health and the growth of plants. In this review two methods of heavy metal remediation are discussed; one is the role of plant growth-promoting bacteria (PGPB) in the enhancement of phytoremediation and the other is composting. PGPB help plants to grow …

The research involving the beneficial aspects of amino acids being added to poultry feed pertaining to performance, growth, feed intake, and feed conversion ratio is extensive. Yet currently the effects of amino acids on the gut microbiota aren’t fully understood nor have there been many studies executed in poultry to explain the relationship between amino acids and the gut microbiota. The overall outcome of health has been linked to bird gut health due to the functionality of gastrointestinal tract (GIT) for digestion/absorption of nutrients as well as immune response. These essential functions of the GI are greatly driven by the …

Open water swimming (OWS) has rapidly grown in popularity, driven by the purported health benefits of cold-water immersion. A paucity of research remains specifically considering the notable risks inherent in OWS participation, and a lack of qualitative research on freshwater swimming experiences, and safety-related issues therein. This rapid ethnographic study, based at a dedicated OWS lake in the UK, conducted semi-structured interviews with OWS participants (n=17; female=11, male=6). Two core themes emerged: environmental issues impacting OWS experiences and behaviours; and knowledge and education of OWS which highlighted safe/unsafe practices, levels of education for managing risks, personal preparedness, swimming solo/with others, …

Brown adipose tissue (BAT)-mediated thermogenesis plays an important role in the regulation of metabolism, and its morphology and function can be greatly impacted by environmental stimuli in mice and humans. Currently, murine interscapular BAT (iBAT), which is located between two scapulae in the upper dorsal flank of mice, is the main BAT depot used by research laboratories to study BAT function. Recently, a few previously unknown BAT depots were identified in mice, including one analogous to human supraclavicular brown adipose tissue. Unlike iBAT, murine supraclavicular brown adipose tissue (scBAT) is situated in the intermediate layer of the neck and thus …

Diverse patterns of climate and edaphic factors challenge detection of soil property change in the US Great Plains. Because detectable soil change can take decades, insights into the trajectory of soil properties frequently require long-term site monitoring and, where available, associated soil archives to enable comparisons with initial or baseline states. Unfortunately, few multi-decadal soil change investigations have been conducted in this region. Here, we document effects of dryland cropping on a suite of soil properties by comparing matched historic (1947) and contemporary (2018) soil samples from the Haas Soil Archive at three sites in the US Great Plains: Moccasin, …

Mead, an ancient alcoholic beverage, relies on the indigenous microbial community in honey for alcohol fermentation. Nevertheless, data on the use of bee indigenous flora for mead production in Thailand are scarce. This investigation involved the isolation of alcohol-producing yeast strains from the Thai stingless bee species Heterotrigona itama. Subsequently, these isolated yeast strains were employed in mead fermentation with honey sourced from the same stingless bee species. Findings revealed that the yeast strain denoted as P03b, which was identified molecularly as Saccharomyces cerevisiae, exhibited the highest alcohol production of 13.53% ± 0.76%. Furthermore, the yeast strain P03b …

The prevailing climate is an important factor affecting the development of the physical quality and flavor of cacao (Theobroma cacao L.) beans. The influence of harvest seasons on the physical, chemical, and flavor characteristics of cacao beans from Nakhon Si Thammarat Province, southern Thailand, was assessed. Among cacao beans, those harvested in the rainy season (R) had the highest fresh and dry weights and the lowest bean count per 100 g (87.00 ± 2.83). Although the overall fat content remained relatively constant across harvest seasons, cacao beans harvested in R exhibited a significantly higher fatty acid (C16:0, C18:1, and …

Fruit growing, especially in family farming, has a significant income potential in small areas, but climate change is a major challenge. This study aimed to quantify the irrigation requirements for citrus, papaya, mango and passion fruit, in the Vão do Paranã region, Goiás state, Brazil. The climate data encompassed the observed periods from 1961 to 2020 and future scenarios from 2021 to 2100. The irrigation demand was obtained from the daily water balance, while the reference evapotranspiration (ETo) was estimated using the Penman-Monteith method and then compared with an artificial intelligence tool. The future scenarios indicated a higher increase for …
