Casein, the primary source of protein in ruminant milk, represents a family of proteins that self-assemble into globules composed of finer spherical subunits and stabilized by calcium phosphate (CP) nanoparticles. These globules act as holdase-type chaperones for unfolded or partially folded proteins, preventing their aggregation. Simultaneously, by integrating CP nanoparticles, they enable the bioavailability of calcium and phosphate beyond their solubility limits, thereby enhancing the nutritional value of milk. Using a range of physicochemical and biological characterization techniques, including scanning and transmission electron microscopy, organic and inorganic elemental analyses, SDS-PAGE, and vibrational, UV/Vis and NMR spectroscopies, it is demonstrated that …

Introduction: Small interfering RNA (siRNA) and Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)/CRISPR-associated Protein 9 (Cas9) complexes are effective approaches to temporarily downregulate protein expression via post-transcription RNA interference or permanently altering protein expression via editing genomic DNA, respectively. However, the efficient delivery of these mediators to targeted cells has been challenging, largely due to their anionic and hydrophilic nature, which hinders their interaction with the cell membrane and cellular internalization. Abiotic stress conditions impair plant metabolism partly by inducing the excessive production of reactive oxygen species (ROS). Melatonin, a multifunctional indoleamine, has emerged as an important regulator of plant responses to oxidative stress. This review highlights recent findings on the interactions among phytomelatonin, ROS homeostasis, antioxidant metabolism, photosynthetic protection, stress priming, and abiotic stress tolerance. We discuss how melatonin modulates the antioxidative response under diverse stress conditions and evaluate its effects on photosynthetic performance, ROS signaling, and membrane stability. Special attention is given to priming studies showing that melatonin pretreatment can improve stress tolerance. Emerging evidence from plant tissue culture …

Complete Chloroplast Genome Analysis Of Critically Endangered Endemic Prangos Denticulata And Insights Into Its Evolutionary Dynamics, Ayşenur Bozkurt, Doğa Seli̇n Kayihan, Hali̇s Batuhan Ünal, Ceyhun Kayihan

Prangos denticulata (Apiaceae) is a critically endangered and narrowly endemic species restricted to Ankara, Türkiye. To provide genomic resources for its conservation and evolutionary study, we sequenced, assembled, and annotated its complete chloroplast genome using short-read next-generation sequencing. The circular plastome was 153,486 bp in length, exhibiting the typical quadripartite structure with a large single-copy (86,440 bp), small single-copy (17,894 bp), and two inverted repeats (49,152 bp), and an overall GC content of 37.8%. A total of 116 genes were identified, including 92 protein-coding, 31 tRNA, and 5 rRNA genes, of which 20 contained introns. Comparative analyses with P. trifida, …

Drought Priming Improves Waterlogging Tolerance In Alfalfa, But Less Effectively Than Waterlogging Priming, Gürkan Demi̇rkol, Ni̇lüfer Kara

Waterlogging severely restricts growth and productivity in alfalfa (Medicago sativa L.), necessitating effective strategies to improve stress tolerance. This study investigated whether drought priming or waterlogging priming could enhance tolerance to subsequent waterlogging stress in the alfalfa cultivar “Bilensoy-80”. Plants were subjected to drought priming, waterlogging priming, or no priming prior to progressive waterlogging stress for 0, 4, 8, 12, 16, and 20 days. Waterlogging priming consistently alleviated growth inhibition, maintained chlorophyll content, and reduced oxidative damage under prolonged stress. Adventitious root formation in stem cuttings of Phoebe bournei, an economically and ecologically significant woody species, is a major bottleneck in clonal propagation. To elucidate the impact of spectral quality on early adventitious root development, we characterized the morphological, hormonal, and transcriptomic dynamics of primordium initiation and differentiation under white, red, and blue light. Under red light, cuttings exhibited the earliest and most pronounced formation of white filamentous structures at lenticels, indicative of root primordium initiation, as early as 20 days after cutting, whereas white and blue light treatments showed delayed responses. Focal adhesions (FA) are integrin-based cell-matrix adhesion sites composed of adaptor proteins that facilitate the generation of traction forces important for biological functions such as cell migration, embryo development, wound healing, tissue organization and stability, and immune responses. Mature FA are observed when both autoinhibited multi-domain scaffolding protein talin and membrane-cytoskeletal protein vinculin are activated and interact thus promoting the assembly and function of FA. Nicotine is the active substrate found in tobacco products, acting as a stimulant, which reinforces continued use. The introduction of tobacco-free options such as e-cigarettes and oral pouches has driven an increase in nicotine exposure across the population, especially in young adults. Utah State University Extension partnered with local government and conservation districts to reduce agricultural losses while creating meaningful opportunities for youth. Over three years, 92 participants removed 3,941 rodents, earned $26,620, and gained practical skills through hands-on conservation work. The program shows how Extension-led partnerships can strengthen agricultural profitability, youth development, and community collaboration at the same time.

1. Data shortages fuel controversy about an ongoing insect biodiversity crisis. Insects are immensely diverse and functionally critical for ecosystems, yet data on their trends remain patchy and biased. Sensors, ranging from camera traps and acoustic recorders to weather radar stations, are set to transform data collection in entomol- ogy. Processing bodies (P-bodies) are cytoplasmic ribonucleoprotein condensates that regulate mRNA storage, translational repression, and decay. While the biochemical principles governing condensate assembly have been extensively studied in vitro, how P-body organization is established and dynamically regulated in vivo to control specific mRNA populations remains poorly understood. This dissertation investigated the molecular and cellular mechanisms that govern P-body architecture and function in the Drosophila melanogaster female germline. First, I identified the conserved LSm protein Trailer Hitch (Tral) as a central organizer of P-body architecture. In Wisconsin, no published literature species records for the keroplatid genera Keroplatus and Proceroplatus exist (Diptera: Bibionomorpha: Keroplatidae). New state records are presented for Keroplatus carbonarius Bosc, Keroplatus clausus Coquillett, Keroplatus militaris Johannsen and Proceroplatus elegans (Coquillett). The new records are associated largely with Malaise trap samples from Hemlock Draw (The Nature Conservancy), in the Baraboo Hills of Sauk County, and a Malaise trap at Kemp Natural Resources Station, in Oneida County, Wisconsin. Two specimens of K. clausus and a single specimen of K. militaris discovered within the research collection at the University of Wisconsin-Madison are also reported. The specimens …

Lead (Pb) is a persistent, toxic pollutant with widespread but inequitably distributed impacts, especially for humans in urban areas. The impacts of lead pollution on other organisms is largely unknown, even as they commonly experience exposure to lead alongside humans. We tested hypotheses regarding the lead concentrations of paper wasps across a gradient of pollution relative to soil and human lead levels, and correlations with wasp body condition. Background: The murine double minute 2 (MDM2) gene encodes an E3 ubiquitin ligase that targets p53 for degradation, thereby influencing the pathogenesis of oral squamous cell carcinoma (OSCC).
Objective. The present study investigates MDM2 mRNA expression and its association with clinical and pathological characteristics in OSCC patients.
Methods: We conducted an observational-analytical study using a cross-sectional design. The study involved 39 OSCC patients. RNA extraction, cDNA synthesis, and quantitative PCR (qPCR) were performed. We quantified relative MDM2 mRNA expression using the Livak method (2-ΔΔCT) and GenEX MultiD software version 6.0. Statistical significance was assessed using independent t-tests.

Photodynamic therapy (PDT) is a minimally invasive treatment that uses light-activated photosensitizers to selectively destroy diseased tissue through the generation of reactive oxygen species. However, conventional in vitro models often fail to accurately reproduce the pharmacokinetics of photosensitizer accumulation and clearance observed in vivo. This study investigated protoporphyrin IX (PpIX) accumulation following 5-aminolevulinic acid (ALA) exposure under physiologically relevant conditions using two-dimensional (2D) monolayers and three-dimensional (3D) spheroids. By comparing continuous ALA exposure with transient exposure followed by ALA washout, this work provides a more biologically relevant framework for studying PpIX kinetics and optimizing photodynamic therapy.

Real-time detection of large whales at sea is critical for mitigating serious injury and mortality from ship strikes and other human activities such as seismic surveys, especially for endangered species such as the critically endangered North Atlantic Right Whale (Eubalaena glacialis). Traditional marine mammal observers are limited by environmental conditions and human factors, necessitating a robust, 24-hour solution. Environmental pollutants can disrupt normal developmental processes, leading to neurobehavioral and developmental abnormalities. Toxicity tests are important for evaluating environmental contaminants; however, standard assays are often time- and cost-intensive and typically require large numbers of vertebrate animals. These limitations highlight the need to reduce reliance on vertebrate testing and to develop New Approach Methodologies that provide comparable results while improving efficiency and cost-effectiveness. Caveolae are 50-100nm cholesterol rich flask shaped invaginations found in many differentiated cells. Namely, these structures have been seen to be abundant in endothelial cells where they are involved in many signaling contexts. However, under mechanical stress, the characteristics of caveolae alter, flattening to increase membrane compliance, while displaced proteins can shift from structural roles to mechanosensitive signaling functions. Caveolin-1 (Cav1) is the principal protein required for Caveolar formation while Eps15 Homology Domain Containing Protein 2 (EHD2) is a membrane ATPase that acts to stabilize caveolae at the surface. The most ancient eukaryotic immune defense mechanisms evolved in unicellular species. Multicellular eukaryotes, however, expanded these mechanisms to meet challenges unique to multicellular life, including barrier formation, long-distance cell signaling, and coordinated responses that eliminate microbial threats while minimizing damage to the host. As such, animal immune signaling pathways are exceptionally intricate and diverse. Though, most of what is understood about animal host–microbe interactions stems from a small handful of model systems. The United States is a dominant power in the global bioeconomy. An in-depth analysis of its policy system characteristics holds significant reference value for China to further formulate China’s bioeconomy strategy and drive the development of China’s bioeconomy. This study employs the “3Ms” policy analysis framework to conduct a comprehensive analysis of the US bioeconomy policy from three dimensions: macro strategy, meso architecture, and micro structure. The research reveals that the US bioeconomy policy demonstrates the following characteristics in the macro strategy: being systematic, comprehensive, and scientifically decision-making; in the meso architecture: forming government-congressional cooperation and multi-party collaboration; and in …

Resources are spent toward fortification and mission success over convenience. The study examined the relationships between lower-body anthropometric characteristics and vertical force-velocity profiles in collegiate American football players. Although the use of force-velocity profiling has increased significantly in recent years, there has been limited research on the association between structural features and key mechanical outputs. A sample of 16 NAIA football players was used to examine how skeletal breadths, girths, and derived ratios relate to key force-velocity metrics. Anthropometric variables were collected using ISAK standards, while force-velocity profiles were obtained through a series of loaded squat jumps on a force platform.
