Soil bacteria play essential roles in nutrient cycling, yet many abundant groups remain poorly characterized due to challenges in cultivation and genetic manipulation. One such organism is Edaphobacter flagellatus, a member of the Acidobacteriota frequently detected in soil environments but lacking well-developed experimental tools. To address this limitation, our laboratory has generated a genome-wide mutant library using the Tn5 transposon, enabling random disruption of genes across the population.
This project aims to establish culture conditions necessary to support the use of this library for functional genomic studies. In particular, defined minimal media can impose selective pressures that reveal …

Bacterial biomass has emerged as a versatile material with strong potential in environmental, material, and biotechnological applications, as well as broader uses such as bioplastics, packaging materials, and agricultural systems, owing to its rapid growth, controllable production, and surface chemistry enriched with carboxyl, hydroxyl, amino, phosphate, sulfate, and sulfhydryl groups. These features enable multiple interaction mechanisms, including electrostatic attraction, ion exchange, surface complexation, hydrogen bonding, and π –π interactions, supporting applications in pollutant removal, wastewater treatment, composite development, and bioenergy systems. A single-step plate method is described for the screening of transformants of cloned gene xyl-A encoding xylose isomerase. It is rapid, inexpensive, easy to perform, and more specific, as the selection is based on xyl-A expression level. The crucial composition of the plate medium comprised agar, buffer, nutrients, co-factor, substrate, inducer, antibiotic, and an indicator, i.e., 2, 3, 5-triphenyl tetrazolium chloride (TTC). Latar Belakang: Tuberkulosis (TB) merupakan penyakit menular yang masih menjadi masalah kesehatan masyarakat utama di Indonesia. Keberhasilan pengobatan sangat bergantung pada kepatuhan pasien dalam mengonsumsi Obat Antituberkulosis (OAT) .
Tujuan . Untuk mengetahui hubungan antara persepsi kerentanan, keparahan, manfaat, hambatan (stigma), isyarat untuk bertindak (peran tenaga kesehatan), dan efikasi diri dengan kepatuhan minum OAT pada pasien TB di Kota Samarinda.
Metode . Penelitian ini menggunakan desain potong lintang dengan pendekatan kuantitatif yang melibatkan 121 pasien TB dari empat Puskesmas di Samarinda. Sampel dipilih menggunakan teknik multistage sampling. Data dikumpulkan melalui wawancara terstruktur menggunakan kuesioner dan dianalisis menggunakan uji Chi-Square.

The persistent exploitation of the aquatic ecosystem, driven by increasing demand for aquatic resources, including construction-grade sand, is evidenced by hydrological and geomorphological degradation of the aquatic ecosystem, with severe negative consequences on the fauna and flora communities. The rising global demand for sand and inefficient mining regulations have resulted in indiscriminate and illegal sand mining globally. In ecosystems, the harvesting effect is essential to preserving the relationships between prey and predator. This work examines a model of predator-prey in discrete time with harvesting. The Jacobian matrix's eigenvalues are calculated for the associated fixed points. We examine the dynamical and qualitative analysis of this model, look into the criteria for stability and bifurcation, and analyze analytical results that show the rich dynamics and complexity of this model. This study aims to describe the profile of gadget use and learning motivation among Grade XI students in biology learning at SMA Negeri 5 Palu and to explore students’ and teachers’ experiences of gadgets as learning support tools and potential sources of digital distraction. A qualitative descriptive design was employed, supported by descriptive questionnaire profiling. A total of 74 students completed a 50-item questionnaire on gadget use and learning motivation, while 10 students and two biology teachers participated in semi-structured interviews. Biology instruction at SMAN 2 Mori Atas was predominantly conducted through conventional teaching practices. Preliminary observations revealed that students had difficulty understanding biodiversity concepts, and several students did not meet the minimum passing criterion of 70. These conditions highlighted the need for learning activities that promote active investigation, discussion, and conceptual connections. The escalating global ecological crisis highlights the need for new approaches to sustainability education, while the cognitive role of Second Language Acquisition (SLA) remains underexplored. This study employs a critical literature review with a systematic search and qualitative thematic synthesis to develop a proposed Green Language Pedagogy (GLP) model. The review followed a four-stage screening protocol (identification, screening, eligibility, and inclusion) and analyzed 50 theoretical and empirical sources published between 2014 and 2025. This analysis of the selected literature reveals a stark disparity: an estimated 85% of the reviewed environmental education research focuses on factual knowledge transfer, whereas only 15% …

The current genetic study aimed to screen out the genetic basis of ß-thalassemia in five unrelated consanguineous Pakistani families. Blood samples were collected, and targeted Sanger Sequencing of the HBB gene was done. Serum Interleukin-6 (IL-6) and quantitative measurements of T Helper and T Suppressor Lymphocytes were also carried out. Additionally, in silico analysis, including protein modeling, Protein-Protein docking, and Multiple sequence alignment, was also performed. This study aimed to evaluate the antifungal activity of Piper aduncum secondary metabolites along with additive oils against Sclerotium rolfsii, causing stem rot on patchouli cuttings. It was conducted in two stages—an in vitro experiment using a completely randomized design with seven treatments: P. aduncum oil, combinations of P. aduncum oil with citronella grass, lemongrass, cinnamon leaf, and cloves leaf oils, a synthetic pesticide (propineb 70%), and without essential oils. Recent environmental change has altered migration timing and winter distribution of many bird species, including Sandhill Cranes. Historically, the Central Platte River Valley in Nebraska functioned primarily as a spring staging area for the midcontinent population, with limited fall use and rare overwintering events. I examined records of Sandhill Crane occurrences during October to January in the Central Platte River Valley from 2015-2026 using incidental observations and aerial surveys starting in 2024. Overwintering Sandhill Cranes were documented in 7 of 11 Dec/Jan, with unprecedented increases observed since 2023, peaking with approximately 42,000 Sandhill Cranes in January of 2026, the largest …

An integrated strategy was developed to promote conversion of a sugarcane bagasse cellulose-rich fraction into water-soluble cellulosic oligomers (WCOs) through phosphoric acid-mediated regeneration and enzymatic hydrolysis. Alkaline pretreatment on sugarcane bagasse powder increased glucan content from 57.6 to 93.4 g/100 g sample and lowered Klason lignin to 6.1 g/100 g sample. Phosphoric acid-mediated regeneration was subsequently capable of reorganizing the structure of cellulose from sugarcane bagasse, resulting in a decrease of crystallinity from 72.9 to 64.0% and introducing cellulose II-like features. [Methods] Arabinoxylan (AX) was extracted from distillers dried grains with solubles and modified to prepare AX with different molecular structures. [Methods] EMP-K is extracted by the water extraction and alcohol precipitation method, and modified by the acetic anhydride method. Then, A-EMP-K preparation conditions are optimized using Box–Behnken response surface methodology. With the growing demand and variety of meat analogs on the commercial market, there is a need to revisit testing methods for foodborne pathogens in these products. The U.S. Food and Drug Administration (FDA) regulatory methods for Salmonella enterica detection in meat analogs follow the Bacteriological Analytical Manual and are relatively time-consuming. The purpose of this qualitative mini ethnography adapted from traditional ethnography with limited time and scope, was to explore and understand perceptions of barriers and facilitators to health literacy among Black Americans with overweight and obesity in three South Carolina counties. Population data showed that approximately 40.3% of U.S. adults are obese. Moreover, Non-Hispanic Black communities are disproportionately affected by overweight and obesity despite efforts to combat the weight-related health risk. The intraspecific variation in otolith wavelet coefficients, contours, and morphometric variables (six size and nine shape indices) was analyzed to discriminate stock variation in Pethia ticto collected from four selected sites along the River Ganga (Narora, Kanpur, Varanasi, and Bhagalpur). The wavelet coefficients extracted revealed symmetrical yet site-specific contour variations, particularly along the rostral, dorsal, and posterior margins, with the fourth-level wavelet explaining 98% of the cumulative accuracy and being selected for contour analysis. The combined effects of pharmaceuticals and nanoparticles (NPs) on aquatic organisms remain insufficiently characterized. The present study examined toxicokinetics and multibiomarker responses of Cyprinus carpio (common carp) following exposure to sulfamethoxazole (SMX) and titanium dioxide (TiO₂) NPs. Juvenile fish were treated with SMX (25, 50, and 100 μg/L) alone and in combination with 1.5 mg/L TiO₂ NPs (96 h). TiO₂ NPs had a significant effect on the bioaccumulation of SMX in muscle tissues, with SMX reaching peak concentrations of 2286.54 ng/g in the 100 μg/L SMX + 1.5 mg/L TiO₂ NPs group after 72 h, compared to 1477.2 ng/g with …

This study examined the extent to which UAE audiences rely on social media as a source of information on complementary and alternative medicine (CAM) and the resulting effects of this reliance. Grounded in Media System Dependency Theory, the study employed an audience survey using a questionnaire administered to a sample of (132) Emirati citizens in Al Ain. The findings revealed that the combined medium and high reliance reached (53%). Regarding the effects, cognitive effects recorded the highest arithmetic means, followed by affective and behavioral effects at comparable levels. This study explores the weather resistance, mechanical properties and corrosion resistance of polyurea coatings in marine environments. The relevant analysis results can be used as a reliability assessment of offshore wind turbine structural coatings during their service life under the influence of marine environments. The long-term experiment uses seawater and 70°C thermal aging conditions as coating reliability assessment items. Casein, the primary source of protein in ruminant milk, represents a family of proteins that self-assemble into globules composed of finer spherical subunits and stabilized by calcium phosphate (CP) nanoparticles. These globules act as holdase-type chaperones for unfolded or partially folded proteins, preventing their aggregation. Simultaneously, by integrating CP nanoparticles, they enable the bioavailability of calcium and phosphate beyond their solubility limits, thereby enhancing the nutritional value of milk.
