Regulation Of Queuine Insertion Into Transfer Rna: Effects Of Tumor Promoters, Bonnie Jean Brooks
Regulation Of Queuine Insertion Into Transfer Rna: Effects Of Tumor Promoters, Bonnie Jean Brooks
Chemistry & Biochemistry Theses & Dissertations
The purpose of this study was to treat normal human fibroblasts with known tumor promoters and observe queuine modification of tRNA. The queuine insertion enzyme, tRNAguanine ribosyltransferase, was studied in vivo and in vitro. Tumor promoter-treated human fibroblast cultures exhibited variable queuine-insertion rates with a transient inhibition that correlated with variable levels of queuine modified tRNA over time in culture from passages 3 — 8. In contrast the in vitro studies showed that phorbol cetera and saccharin actually increased insertion with strong evidence indicating phosphorylation as a positive modulating force of the enzyme activity. It is proposed that chronic stimulation …
Cation Nuclear Magnetic Resonance (Nmr). 7li- And 23na-Nmr Results Obtained With Human Erythrocytes, Jan F. M. Post
Cation Nuclear Magnetic Resonance (Nmr). 7li- And 23na-Nmr Results Obtained With Human Erythrocytes, Jan F. M. Post
Scanning Microscopy
A short discussion of practical results and theoretical aspects of nuclear magnetic resonance (NMR) on intracellular monovalent cations is presented. 7Li- and 23Na-NMR relaxation studies on human erythrocytes are described which indicate that lithium and sodium ions are essentially free inside the cell. However, there is a contribution to transverse relaxation originating from cations diffusing through electric field gradients imposed by the cytoskeleton, which is reflected in the NMR line shape. 39K-NMR results obtained by other authors for intracellular potassium are compared with the 7Li- and 23Na-NMR results presented here. The few 39K-NMR results …
Farmer Attitudes Toward Wild Turkeys In Southwestern Wisconsin, Scott R. Craven
Farmer Attitudes Toward Wild Turkeys In Southwestern Wisconsin, Scott R. Craven
Wildlife Damage Management Conference
The reintroduction of the Wild Turkey (Meleagris gallopavo) represents one of the great success stories for Wisconsin wildlife management. Human settlement, habitat loss, and perhaps disease, combined to eliminate once-common turkeys from Wisconsin by 1900. Periodic attempts to restore a viable turkey population were unsuccessful with the exception of a small flock at Meadow Valley-Necedah in central Wisconsin. However in 1976, the Wisconsin Department of Natural Resources (WDNR) began a new, well conceived cooperative project with the Missouri Department of Conservation. Under the agreement, Missouri received 3 wild Wisconsin ruffed grouse (Bonasa umbellus) in exchange for …

The U.S. Environmental Protection Agency regulates pesticides under the authority of the Federal Insecticide, Fungicide, and Rodenticide Act (FIFRA). Vertebrate pesticides are subsumed under the heading "rodenticides" and, under FIFRA, are regulated similarly to other pesticides.
Animal Damage Control And The Wildlife Profession, Gary J. San Julian
Animal Damage Control And The Wildlife Profession, Gary J. San Julian
Wildlife Damage Management Conference
Conflicts between man and wildlife have always been a part of our history. We have tried to control the damage caused by wildlife and found that this was not always in the best interest of the resource. The role of animal damage control in our profession has changed and so has the public's view of it. As professionals we must strive to explain the need and value of wildlife damage management to our peers, the public and our detractors. This can be accomplished by participation in our professional organizations, the presentation of papers at scientific meetings, and open discussion of …
Dendritic Oxide Growth On The Surface Of Liquid Gallium, Y. L. Wang, A. Raval, R. Levi-Setti
Dendritic Oxide Growth On The Surface Of Liquid Gallium, Y. L. Wang, A. Raval, R. Levi-Setti
Scanning Microscopy
We have studied the oxidation of a liquid gallium surface with a high spatial resolution scanning ion microprobe. A 40 keV focused gallium ion beam, extracted from a liquid metal ion source, was employed, first, to sputter clean a 40 x 40 μm2 area on a drop of liquid gallium, in a ultra high vacuum (UHV) specimen chamber. It was then used to monitor the oxide growth by secondary ion mass spectrometry imaging microanalysis while the chamber was gradually back-filled with oxygen. In the initial stages, gallium oxide grew in a dendritic pattern from the edge of the cleaned …

A Calculation Method Of X-Ray Emitted Intensity In Multi-Layer Films By Monte Carlo Simulation, Yen-Cai Ho, Jia-Guang Chen, Xin-Lei Wang
Scanning Microscopy
A calculation method of X-ray emitted intensity in multi-layer films is proposed in this paper. The method is based on the work developed by us: (1) a simplified physical model of electron scattering and Monte Carlo evaluations in a single medium and in multi-layer media and (2) the theories and the formulae for excitation, absorption and fluorescence of characteristic X-rays. The intensity ratio of X-rays for the known thickness films, Au/Cu/Si and Cr/Ni/Si, were calculated at 20, 25 and 30 keV. Calculated results are compared with experimental values of electron microprobe analysis for the multi-layer film specimens, and the correspondence …

Inbred strain 2 guinea pigs were vaccinated with Mycobacterium bovis BCG or were left unvaccinated. They were maintained for 6 weeks on defined, isocaloric diets containing either 30% (control animals) or 10% (animals receiving low protein) ovalbumin as the sole protein source. Animals were challenged by the respiratory route with a low dose of virulent M. tuberculosis H37Rv and killed 4 weeks later. Protein-malnourished animals were not protected by previous vaccination with BCG. Lymphocytes isolated from various tissues were tested in vitro for proliferative responses to mitogen (concanavalin A) and antigen (purified protein derivative [PPD]), production of interleukin-2 (IL-2), and …

Transcriptional Regulation Of The Human Calcitonin Gene: A Progress Report, Sara Peleg, Gilbert J. Cote, Ronald V. Abruzzese, Robert F. Gagel
Henry Ford Hospital Medical Journal
We have applied DNA transfer techniques lo study the transcriptional regulation of the calcitonin (CT) gene in a C-cell line (TT) derived from a human medullary thyroid carcinoma. TT cells were transfected with a fusion gene containing the CT gene promoter and 5' -flanking DNA attached to the promoter-less growth hormone gene (reporter). We quantitated the reporter gene product to monitor transcriptional activation by the CT promoter and deletion mutants of the 5' -flanking DNA. We found that the proximal CT promoter which includes the DNA sequence from +1 to -129 bp upstream from the CT transcription start site did …

Screening For Pheochromocytoma In The Men 2 Syndrome, J. Kotzerke, C. Stibane, H. Dralle, H. Wiese, W. Burchert
Henry Ford Hospital Medical Journal
Twenty-two patients with the multiple endocrine neoplasia type 2 (MEN 2) syndrome were screened for pheochromocytoma since it is a major cause of morbidity in MEN 2 families. Clinical symptoms, biochemical parameters, ultrasound, computed tomography or magnetic resonance imaging, and meta-iodo-benzylguanidine (MIBG) scintigraphy were evaluated for detection of adrenal tumors. Clinical symptoms and plasma or urine catecholamines appeared to be nonspecific, whereas MIBG scintigraphy was highly specific and the most sensitive parameter. Patients older than age 30 should be scintigraphically screened at least once despite the radiation exposure. Demonstration of only slight uptake is not an indication for surgery but …

The Voltage Sensor Of Excitation-Contraction Coupling In Skeletal Muscle. Ion Dependence And Selectivity, Gonzalo Pizarro, Robert H. Fitts, Ismael Uribe, Eduardo Rios
Biological Sciences Faculty Research and Publications
Manifestations of excitation-contraction (EC) coupling of skeletal muscle were studied in the presence of metal ions of the alkaline and alkaline-earth groups in the extracellular medium. Single cut fibers of frog skeletal muscle were voltage clamped in a double Vaseline gap apparatus, and intramembrane charge movement and myoplasmic Ca2+ transients were simultaneously measured. In metal-free extracellular media both charge movement of the charge 1 type and Ca transients were suppressed. Under metal-free conditions the nonlinear charge distribution was the same in depolarized (holding potential of 0 mV) and normally polarized fibers (holding potentials between -80 and -90 mV). The manifestations …

Endothelial Injury In Human Atherosclerosis, G. Pasquinelli, A. Cavazza, P. Preda, A. Stella, B. I. Cifiello, M. Gargiulo, M. D'Addato, R. Laschi
Scanning Microscopy
A light and electron microscopic investigation (scanning and transmission electron microscopy) was performed on 51 human atherosclerotic carotid lesions. The purpose of this study was to establish whether features of endothelial injury such as those described in animals occur in man and whether these features can be related to specific stages of human atherosclerosis.
Irrespective of their histological appearance the atherosclerotic lesions were covered with endothelium which showed non-specific changes in cell shape and size. However, all complicated lesions appeared denuded. Moreover, a peculiar interaction of endothelium with monocytes and lymphocytes as well as blood components (e.g., fibrin and lipoproteins) …

Effects Of Extracellular Matrix On Cytoskeletal And Myofibrillar Organization In Vitro, Lula L. Hilenski, Louis Terracio, Roger Sawyer, Thomas K. Borg
Scanning Microscopy
The distribution and three-dimensional relationship of myofibrillar and cytoskeletal components during myofibrillogenesis were examined in preparations of neonatal rat cardiac myocytes processed in parallel for scanning electron microscopy (SEM), intermediate voltage transmission electron microscopy (IVEM) and immunofluorescence (IF). Of the various methods used for processing, optimal results were achieved by pre-extraction with Triton X-100 in an actin-stabilizing buffer. This procedure effectively removed the surface membrane, as viewed by SEM images, while preserving myofibrillar and cytoskeletal structure, as evidenced by IF for actin, 𝝰-actinin and vinculin. Cytoskeletons in SEM images consisted of a cortex of anastomosing filaments through which ran parallel …

Correlated Microscopic Observations Of Arterial Responses To Intravascular Stenting, K. A. Robinson, G. Roubin, S. King, R. Siegel, G. Rodgers, R. P. Apkarian
Scanning Microscopy
Percutaneous catheter implantation of intravascular stent prostheses has emerged as a novel clinical adjunct to balloon angioplasty in the treatment of obstructive atherosclerotic vascular disease. We have examined the cellular and subcellular responses to stenting in the coronary arteries of the dog and pig (both normal and atherosclerotic), and in the iliac arteries and aorta of the atherosclerotic rabbit, using scanning electron, transmission electron, and light microscopies. Stenting in these models resulted in a thrombotic reaction ranging from mild to severe, depending on species and antithrombotic therapy. Subsequent organization of thrombotic material with hyperplasia of smooth muscle and inflammatory cells, …
A Review On The Use Of Bulk Specimen X-Ray Microanalysis In Cancer Research, I. Zs.-Nagy
A Review On The Use Of Bulk Specimen X-Ray Microanalysis In Cancer Research, I. Zs.-Nagy
Scanning Microscopy
The freeze-fracture, freeze-drying ( FFFD) method of biological bulk specimen preparation combined with quantitative X-ray microanalysis is suitable for the measurement of intracellular concentrations of biologically relevant elements in human biopsy or experimental animal materials. Especially useful information can be obtained regarding the intracellular Na+/K+ ratios being independent of the actual (and unknown) water content of the cytoplasm. The sustained increase of this ratio indicates a sustained depolarization of the cell membrane. These data are of importance from the point of view of the membrane hypothesis of mitogenesis (MHM). It has been revealed that the distribution histograms …

The Chorioallantoic Capillary Plexus Of The Chicken Egg: A Microvascular Corrosion Casting Study, G. J. Burton, M. E. Palmer
Scanning Microscopy
The chorioallantoic membrane of the avian egg serves as the principal organ of respiratory gaseous exchange for the embryo until close to hatching. It lies closely apposed to the inner shell membrane and contains an extremely dense capillary plexus supplied by the allantoic blood vessels. This study applied the microvascular corrosion casting technique to investigate the three-dimensional arrangement of the plexus at various stages of incubation. Casts were produced between days 6 and 14 of incubation, and their appearances were compared with those obtained from traditionally sectioned material and from freeze-cleaved specimens.
By day 6 the capillary network was remarkably …

Effects Of Inter-Specific Density And Food Supply On Survivorship And Growth Of Newly Settled Benthos, Roman Zajac, Robert B. Whitlatch, Richard W. Osman
Biology and Environmental Science Faculty Publications
Using a laboratory model system comprised of newly settled oysters Crassostrea virginica and established fouling species (Botrylloides sp. initially, and others including Styela clava and Ciona intestinalis as the experiment progressed), we tested how differences in food supply and competitor density may affect post-settlement surivorship and growth of sessile marine invertebrates over a 44 d period. After 15 d, results were mixed but indicated that both food and density conditions affected growth and survivorship significantly, with some suggestion of high food levels ameliorating high density effects However, 44 d after settlement, oysters had reduced survivorship and growth when competitors …

Immunosuppressive Activity Derived From The Uc729-6 Lymphoblastoid And M21-Hpb Melanoma Cell Lines, Charlene J. Repique
Loma Linda University Electronic Theses, Dissertations & Projects
Previous work conducted in our laboratories, using a murine model, suggested that soluble factors secreted by tumor cells suppress lymphocyte responses. To apply this premise to human tumors, we studied the effects of UC729-6 (lymphoblastoid B-cell) and M21-HPB (malignant melanoma) conditioned media (CM) on lymphocyte proliferation assays, as well as on the growth of other cells. The CM was collected at 2-5 day intervals from cultures of UC729-6 and M21- HPB cells in serum-free media. In assays using phytohemagglutinin (PHA) and concanavalin A (ConA), mononuclear peripheral blood cells from healthy human donors showed decreased [3H]-thymidine ([3H]-Tdr) …

Effect Of Photoperiod On Developmental Morphology And Enolase Isoenzyme Immunohistochemistry In Rat And Djungarian Hamster Superficial Pineal Glands, Chalmer D. Mcclure
Loma Linda University Electronic Theses, Dissertations & Projects
The best understood functional activity of the pineal gland is its diurnal production of melatonin in response to environmental lighting cues. Several enzymes of the melatonin pathway respond to daily photoperiod changes, for example hydroxyindole-O-methyltransferase (HIOMT) and serotonin N-acetyltransferase (SNAT). Increased levels of the glycolytic enzyme neuron-specific enolase (NSE) are thought to reflect increased physiological demands placed on neurons and neuroendocrine tissues. Homodimer non-neuronal enolase isoenzyme (NNE) is immunolocalized to cells, and the hybrid enolase (consisting of subunits from NSE and NNE) has been seen in cerebellar stellate and basket cells. Although not rate limiting, concentrations of both NSE and …

The Composition And Concentrations Of Cyanobacteria In The North Landing River, Virginia And Upper Currituck Sound, North Carolina, Sonja E. Midgette
Biological Sciences Theses & Dissertations
A 12 month study of cyanobacteria composition and concentrations in the North Landing River and upper Currituck Sound was conducted from March 1986 to February 1987. Epifluorescence microscopy and the fluorochrome proflavin were used to determine cell concentrations and vertical, station to station and seasonal distribution of these cells. Cyanobacterial assemblage patterns were coupled with pH, temperature, salinity and water transparency readings.
Cyanobacteria were found to be a persistent and prolific phytoplankton component of both the river and upper sound. For a comparison of vertical distribution for each station, an analysis of variance for. two groups of equal size was …

Mounting Sex Pheromone: A Novel Pheromone Responsible For Mate Recognition In The Ixodidae, Jgc Hamilton
Theses and Dissertations in Biomedical Sciences
Cholesterol oleate was demonstrated to be the cuticular contact sex pheromone of Dermacentor variabilis. This pheromone which has been termed the Mounting Sex Pheromone (MSP) was also demonstrated to be present on the surface of D. andersoni, Amblyomma maculatum and A. americanum. This contact sex pheromone enables males excited and attracted by 2,6-dichlorophenol to identify the female as a potential mating partner. The MSP is the second in the series of three sex pheromones guiding the hierarchy of behavioral responses which constitute tick courtship behavior. Tests with D. variabilis and D. andersoni showed that this pheromone …
