Efficacy Of A Cell Extract From Actinobacillus (Haemophilus) Pleuropneumoniae Serotype 1 Against Disease In Swine, P. J. Fedorka-Cray, M.J. Huether, D.L. Stine, G. A. Anderson
School of Veterinary and Biomedical Sciences: Faculty Publications
We partially characterized a cell extract (CE) from Actinobacillus pleuropneumoniae serotype 1 and used the CE to test the efficacy of secreted proteins against disease. Secreted products from 4-h culture supernatants were precipitated with 20% polyethylene glycol. Analysis of the CE indicated the presence of protein, endotoxin, and carbohydrate. Hemolytic activity to bovine erythrocytes and cytotoxic activity to porcine mononuclear leukocytes was also demonstrated. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of the CE from a 4-h culture showed a major band at 110 kilodaltons (kDa), while a CE of a 26-h culture indicated the presence of a number of additional …

Adsorption Of The Herbicides Diuron, Terbacil, And Simazine To Blueberry Mulches, Laurie Hodges, Ronald E. Talbert
Adsorption Of The Herbicides Diuron, Terbacil, And Simazine To Blueberry Mulches, Laurie Hodges, Ronald E. Talbert
Department of Agronomy and Horticulture: Faculty Publications
Samples of soil, mulch, and the soil/mulch interface zone were collected from commercial highbush blueberry (Vaccinium corymbosum L.) fields typical in their use of mulch under Arkansas conditions. Mulches included 1-year-old hardwood sawdust, 5-year-old hardwood sawdust, and 1-year-old pine needle mulch. Herbicide adsorption (Kd values) of the samples was determined for diuron, terbacil, and simazine. The soils, mulches, and interfaces adsorbed nearly 10 times as much diuron and more than twice as much simazine as terbacil. Adsorption of the herbicides was three to five times greater to the mulches than to the soils. Adsorption was significantly correlated with the …
Stability Of Iaa And Iba In Nutrient Medium To Several Tissue Culture Procedures, Scott Nissen, Ellen Sutter
Stability Of Iaa And Iba In Nutrient Medium To Several Tissue Culture Procedures, Scott Nissen, Ellen Sutter
Department of Agronomy and Horticulture: Faculty Publications
The relative stabilities of IAA and IBA under various tissue culture procedures were determined. IBA was significantly more stable than IAA to autoclaving. IBA was also found to be more stable than IAA in liquid Murashige and Skoog medium (MS) under growth chamber conditions. The stabilities of IBA and IAA were similar in agar-solidified MS. Light provided by cool-white fluorescent bulbs promoted degradation of IAA and IBA in both liquid and agar media. Activated charcoal in concentrations as high as 5% was found to adsorb more than 97% of IAA and IBA in liquid MS. These results have important implications …
The Role Of Small Peptides In Cancer Physiology And Chemotherapy, Bao-Ling Tsay
The Role Of Small Peptides In Cancer Physiology And Chemotherapy, Bao-Ling Tsay
Theses and Dissertations in Biomedical Sciences
The targeting of proven anticancer drugs specifically to cancer cells would provide a unique opportunity to restrict neoplasms without damaging the cancer patient. The present research utilizes the phenomenon of illicit transport, i.e. the coupling of normally impermeant metabolites to permeant metabolites, in targeting the drug melphalan to mouse Ehrlich ascites tumor cells. The dipeptide beta-alanyl-melphalan was synthesized and tested in vitro for toxicity towards mouse Ehrlich ascites tumor cells, mouse liver cells, and mouse 3T3 embryonic cells. The parent compound, melphalan, was used as a control treatment. In addition, both melphalan and beta-alanyl-melphalan were utilized in in vivo chemotherapeutic …

The Secondary Blood Vessel System Of Segmental Arteries And Dorsal Aorta In Blennius Pavo And Zosterisessor Ophiocephalus. Histology, Fine Structure And Sem Of Vascular Corrosion Casts., F. Lahnsteiner, A. Lametschwandtner, R. A. Patzner
Scanning Microscopy
The secondary blood vessel system of the segmental arteries and of the dorsal aorta of the teleost fish Blennius pavo and Zosterisessor ophiocephalus are examined by light microscopy, transmission electron microscopy and by scanning electron microscopy of appropriately processed tissue and of vascular corrosion casts. Dorsal, lateral and ventral segmental arteries and the caudal portions of the dorsal aorta have a secondary vessel system. The secondary vessels are formed by capillaries which arise from the proximal portions of the segmental arteries and from the caudal parts of the dorsal aorta. In Z. ophiocephalus these capillaries are strongly dilated at their …

Binding Of Ions To Nuclear Chromatin, Thomas Von Zglinicki, Heiko Ziervogel, Martina Bimmler
Binding Of Ions To Nuclear Chromatin, Thomas Von Zglinicki, Heiko Ziervogel, Martina Bimmler
Scanning Microscopy
Ion concentrations in isolated lymphocyte nuclei subjected to KCl or MgCl2 media of varying ionic strength were measured by X-ray microanalysis. Values were corrected for the contribution of free ions by estimating the volume fraction of the water space morphometrically. The amount of bound cations and Cl was constant and independent of the widely varying free ion concentration.
It is concluded that the mechanism of binding is counterion condensation but with limited cooperativity. In contrast to classical counterion condensation theory, the binding of ions occurs at oppositely charged clusters at the surface of the chromatin. Therefore, both cations and …
Electron Microscopy Of Endothelial Cell - Biopolymer Interaction, Stuart K. Williams, Timothy Schneider, Bruce E. Jarrell
Electron Microscopy Of Endothelial Cell - Biopolymer Interaction, Stuart K. Williams, Timothy Schneider, Bruce E. Jarrell
Scanning Microscopy
Vascular endothelial cells form a natural antithrombogenic lining on all blood vessels. Replacement or bypass of small diameter blood vessels with artificial polymeric grafts has not been clinically acceptable due to the thrombogenic nature of polymeric material. One approach to improving the patency of vascular prosthetic devices has been the establishment of endothelial monolayers on the blood flow surface using the technique known as seeding. Scanning electron microscopy has been a major tool in evaluating the interaction of endothelial cells with polymeric surfaces resulting in a basic understanding of forces and structures regulating endothelium-polymer interactions. In-vitro and in-vivo studies have …
Comparison Of Backscattered Scanning Electron Microscopy And Microradiography Of Secondary Caries, Grayson W. Marshall Jr., Michal Staninec, Yasuhiro Torii, Sally J. Marshall
Comparison Of Backscattered Scanning Electron Microscopy And Microradiography Of Secondary Caries, Grayson W. Marshall Jr., Michal Staninec, Yasuhiro Torii, Sally J. Marshall
Scanning Microscopy
Carious lesions are usually studied using light microscopy and/or microradiography which require preparation of thin sections. Backscattered scanning electron microscopy (BSEM) has received little attention although it provides information similar to that obtained with microradiographs, with the potential for higher resolution. Recently, microscopes have been introduced that can be used to study wet or nonconducting specimens, offering techniques for studying specimens without desiccation or preparation of thin sections. This investigation sought to determine if secondary carious lesions have the same characteristics when studied by microradiography as when using the "wet" BSEM mode. Microradiographs were made of thin sections from restored …
Cryofixation Of Heart Tissue For X-Ray Microanalysis, Alice Warley
Cryofixation Of Heart Tissue For X-Ray Microanalysis, Alice Warley
Scanning Microscopy
Cryofixation of tissues is necessary to be able to study the concentrations of elements by X-ray microanalysis. Simple dissection of heart tissue fragments of the size needed for optimum cryofixation by the conventional methods of plunge or slam freezing leads to the development of ischaemia in the tissue fragments and a consequent redistribution of the diffusible elements. Heart tissue can be frozen in vivo using liquid nitrogen cooled Cu clad pliers, but the morphological detail is preserved better if the cooled pliers are exposed to liquefied propane immediately before freezing, Concentrations of Na are lower and concentrations of K are …
An Analytical Method Of Determining Thickness Of Multi-Layer Films With Electron Microprobe, Yen-Cai Ho, Jia-Guang Chen, Xin-Lei Wang
An Analytical Method Of Determining Thickness Of Multi-Layer Films With Electron Microprobe, Yen-Cai Ho, Jia-Guang Chen, Xin-Lei Wang
Scanning Microscopy
In the previous work we have developed a series of theoretical corrections for calculating the emitted X-ray intensity in multi-layer films. By the use of these theories, along with careful experimental operation of the electron probe microanalysis (EPMA) and Monte Carlo iteration calculation, the thickness of each layer in multi-layer films can be determined.
To test the reliability of this method, the multi-layer film specimens Au/Cu/Si, Cu/Au/Si and Ag/Cr/Si of known thicknesses were analyzed at 20, 25, 30 and 35 keV. The percentage relative errors between the thicknesses determined using the correction procedures and those measured using nuclear backscattering are …
Attribute Relationships: An Impediment In Automating Schema Integration, Amit P. Sheth, Sunit K. Gala
Attribute Relationships: An Impediment In Automating Schema Integration, Amit P. Sheth, Sunit K. Gala
Kno.e.sis Publications
Success in schema integration depends on understanding the semantics of schema components (e.g., entity sets, relationship sets, attributes), and the ability to capture and reason about these semantics. An important objective of our work on schema integration is to automate the reasoning as much as possible, and when not possible, depend on the human input and guidance. A key results of comparing the semantics associated with schema objects is that of determining attribute relationship. Once attribute relationships are determined, the task of object class(e.g., entity sets and relationship sets) integration becomes simpler and can e automated to a great extent. …
Site-Directed Mutagenesis Of The Putative Processing Site Of Recombinant Human Immunodeficiency Virus Reverse Transcriptase, Susan M. Poppe
Site-Directed Mutagenesis Of The Putative Processing Site Of Recombinant Human Immunodeficiency Virus Reverse Transcriptase, Susan M. Poppe
Masters Theses
The human immunodeficiency virus (HIV) is the etiological agent of the acquired immune deficiency syndrome (AIDS). This retrovirus, encodes a RNA dependent DNA polymerase, reverse transcriptase (RT), which is essential to virus replication. Biochemical analysis indicated that RT is composed of 2 polypeptides of 66,000 and 51,000 daltons (p66 and p51) which combine to form a heterodimer. This heterodimer is thought to arise from the post-translational processing where p66 is made and either autocatalytic or proteolytic cleavage events result in a heterogeneous p66/p51. Site-directed mutagenesis was used at the proposed processing site to probe HIV-RT processing.
Three prokaryotic expression vectors …

Scanning Electron Microscopy Of Chromosomes And Chromosome Fragments In Transgenic Rainbow Trout, Jane E. Disney, Christine M. Davitt, Gary H. Thorgaard
Scanning Electron Microscopy Of Chromosomes And Chromosome Fragments In Transgenic Rainbow Trout, Jane E. Disney, Christine M. Davitt, Gary H. Thorgaard
Scanning Microscopy
Chromosomes and chromosome fragments from embryonic offspring of a transgenic rainbow trout were examined using scanning electron microscopy (SEM). SEM is an extremely useful technique for studying the structure of chromosome fragments since little morphological detail is revealed by conventional staining methodologies and light microscopy. The chromosome preparations were processed for SEM by combining an osmium-thiocarbohydrazide-osmium (OTO) technique with 2-4 nm of gold deposition. This technique revealed the organization of individual chromatin fibers in chromosome fragments and intact chromosomes. Both a linear chromosome fragment with a width similar to that of an intact chromatid (approximately 0.60 micrometers) and a spherical …

Identification And Characterization Of Peptidoglycan-Associated Proteins In Neisseria-Gonorrhoeae, Stuart A. Hill, Ralph C. Judd
Biological Sciences Faculty Publications
The principal proteins associated with Neisseria gonorrhoeae peptidoglycan (PG), as identified by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, are the following: two proteins at approximately 90 kilodaltons (kDa), single major species at both 60 and 44 kDa, a 34- to 36-kDa protein, and three proteins between 28 and 32 kDa. A protein analogous to Escherichia coli Braun lipoprotein was not detected with gonococcal cell wall preparations. The identity of the PG-associated proteins was confirmed immunologically with antibody generated against purified cell walls. Two types of protein species, dithiothreitol extractable (the majority) and alkylation dependent (primarily the 34- to 36-kDa protein), appeared …

Two Pseudomonas strains were engineered to contain the nptII gene and plasmid vector sequences in their chromosomes. After incubation of these strains in nonsterile soil, total bacterial DNA was isolated and analyzed by Southern blot hybridization with the nptII gene and the plasmid vector as probes. In addition to the expected bands of hybridization, a new band corresponding to the loss of vector sequences from the chromosome while retaining the nptII gene was observed for one of the strains. The more stressful conditions encountered in soil appeared to increase the frequency of loss of the vector sequences …

Hourly Fluctuation Of Middle Ear Pressure As A Function Of Age In School-Age Children, Susan Hogue Henry
Hourly Fluctuation Of Middle Ear Pressure As A Function Of Age In School-Age Children, Susan Hogue Henry
Dissertations and Theses
Tympanometry is a useful means of evaluating the status of the middle ear. For the pediatric population, tympanometry is particularly valuable for determining the presence of middle ear effusion. The test has been incorporated in many school hearing conservation programs because of its ease of administration, objectivity, and diagnostic value.
In a study by deJonge and Cummings (1985), the hourly fluctuation of middle ear pressure was reported in a group of kindergarten-age children. The variability of middle ear pressure for that group of children averaged 150 daPa. In the present study, a maturational effect of this hourly fluctuation was observed …

Quantitative Measurement From Vascular Casts, Ralph G. Kratky, Christine M. Zeindler, Dorian K. C. Lo, Margot R. Roach
Scanning Microscopy
A review of quantitative measurements show casting materials shrink from 0.2 - 20% and have viscosities ranging from 1.4 - 100,000 centipoise. Blood vessels have highly variable mechanical properties. Some microvessels are very stiff having little change in dimensions with pressure. Larger vessels generally change diameter significantly but show highly variable changes in length with pressure. Perfusion fixation does not fix the dimensions of blood vessels. Dog carotid arteries well fixed with glutaraldehyde at physiologic dimensions retain ≈20% of their elastic recoil circumferentially and ≈30% longitudinally. We recommend vascular casting as a method of accurately measuring the vasculature if care …
The Physical State Of Potassium In The Human Lymphocyte: A Review, William Negendank
The Physical State Of Potassium In The Human Lymphocyte: A Review, William Negendank
Scanning Microscopy
Studies of the effects of chemical potential, temperature, and metabolic perturbation on static ion contents, kinetics of the approach to equilibrium, and kinetics of ionic self-exchange in human lymphocytes are reviewed. The results contradict the classical concept of cell ion and water physiology, the membrane-osmotic, pump-leak theory, and are re-interpreted by an adsorption model of the cell. In this model, most of cell water exists in a physical state sufficiently ordered to reduce the partition function of dissolved ions, and most of cell potassium is associated with fixed charges on macromolecules. Competing adsorption of potassium and sodium is cooperative and …

Audubon/Au-Du-Bon: Man And Artist, Walter Sutton
The Courier
This article highlights some of the works of the legendary work of John James Audubon, drawn from the collection located in Syracuse University's Special Collections. The author gives special attention to the 1820-21 journal of his voyage down the Ohio and Mississippi (which has been preserved intact), the English and Scottish journal of 1826 (also in its original form), and the descriptive sketches of early pioneer life in the Ornithological Biography. These early journal sources dramatically reveal, at first hand, Audubon's long struggle through many failures and obstacles to win the success and recognition he craved and also enduring status …
