Heart failure (HF) is a leading cause of morbidity and mortality worldwide, placing substantial clinical and economic burdens on society. Although advances in therapy have improved patient outcomes, the biological mechanisms that drive HF onset and progression remain incompletely understood. Increasing evidence suggests that the gut microbiota, the trillions of microorganisms residing in the gastrointestinal tract, plays a key role in some forms of heart disease, especially cardiovascular disease. Moreover, gut microbial dysbiosis has been associated with systemic inflammation, metabolic imbalance, and immune dysregulation, all of which contribute to the pathophysiology of HF. South-West Freshwater Angling (SWFA) in Western Australia is a licensed fishery primarily targeting the Brown Trout (Salmo trutta) and Rainbow Trout (Oncorhynchus mykiss), which are stocked in selected dams and rivers, as well as the introduced Redfin Perch (Perca fluviatilis) and the native Freshwater Cobbler (Tandanus bostocki). Since 2000/01, annual phone-recall surveys have been undertaken using the SWFA licence database as the sampling frame. This monitoring provides information to understand the fishery and support future management decisions. Herbicide-resistant Palmer amaranth (Amaranthus palmeri S. Watson) pose a severe threat to agronomic crop productivity in the United States, including corn and soybean production in Nebraska. Therefore, there is an urgent need for diversified and sustainable management approaches. Integrating cover crops (CCs) with herbicide programs represents a promising strategy to achieve Palmer amaranth suppression, sustain crop yields, and contribute to long-term soil health in irrigated corn–soybean production systems in Nebraska.

A meta-analysis of 41 studies and 595 paired observations demonstrated that CCs reduced Amaranthus spp. density and biomass by 37–59% depending on the growing season, with grasses and mixtures …

Sustainable crop production in the semi-arid High Plains of Nebraska faces challenges from limited precipitation, high interannual variability, and economic risk. This dissertation investigates the long-term productivity, water use, soil health, and economic sustainability of dryland crop rotations, with emphasis on the role of pulses as alternatives to fallow.

Chapter 1 analyzes nearly three decades (1995–2023) of on-farm data from the High Plains Agricultural Laboratory (Sidney, Nebraska) to compare seven crop rotations, including wheat, corn, millet, sunflower, and field peas. Microbiomes in humans, animals, plants, and other environments are necessary components of the biosphere for global nutrient cycling and maintaining ecosystem balance. In particular, the gut microbiome plays an essential role in modulating human health through glycan utilization. Carbohydrate-active enzymes (CAZymes), CAZyme gene clusters (CGCs), and polysaccharide utilization loci (PULs) are key genomic features that determine microbial capacities for carbohydrate metabolism. Metabolic dysfunction-associated steatotic liver disease (MASLD) is characterized by excessive lipid accumulation in hepatocytes and the development of lipotoxic stress that disrupts cellular homeostasis. Peroxisomal lipid metabolism plays an important role in maintaining acyl-CoA balance during fatty acid overload, yet the contribution of peroxisomal acyl-CoA thioesterases to early lipotoxic responses remains insufficiently understood. Background: Hippobosca camelina (camel ked) is an obligate hematophagous ectoparasite that infests camels. Hematophagy inflicts painful bites leading to myiasis, anemia and pathogen transmission such as Candidatus Anaplasma camelii. A genome assembly for this biting flies is currently unavailable, limiting understanding of its genetics, including the chemosensory system.

Amidst escalating global urbanization and population growth, revolutionizing food systems and redefining waste management strategies has become essential. This study contributes to protein resource diversification in animal feed and offers a sustainable solution for organic waste management using black soldier fly larvae (BSFL) in Bafia, Cameroon. A survey characterized waste production in restaurants and households to assess quantity and quality. Gregarious desert locusts produce stage-specific pheromones that facilitate cohesive behavior in juveniles and synchronize maturation and mating in sexually mature adults. During locust outbreaks, merging populations result in cross-stage interactions, yet their impact on locust biology remains poorly understood. This study tested the hypothesis that cross-stage interactions influence juvenile cohesion and physiological traits. Using behavioral assays and gas chromatography-mass spectrometry, we examined short- and long-term interactions between juvenile and adult desert locusts. Background: Despite significant global effort to control and eradicate malaria, many cases and deaths are still reported yearly. These efforts are hindered by several factors, including the severe underestimation of cases and deaths, especially in Africa. Methods: We used a mathematical model, incorporating the underestimation of cases and seasonality in mosquito biting rate, to study the malaria dynamics in Cameroon. The global microalgae processing industry generates substantial residues rich in ω-3 fatty acids, presenting both a waste management challenge and an untapped resource for innovative food biotechnology. This study investigated the potential of bioconversion of Schizochytrium residue (SCR) by black soldier fly larvae (BSFL) to enrich ω-3 fatty acids and evaluated the industrial applicability of the resulting BSFL products. SCR supplementation enhanced larval growth performance (7.5 % increased weight, 19.9 % higher bioconversion rate) and protein quality, while enabling exclusive accumulation of EPA and DHA in BSFL. The black soldier fly frass fertilizer (BSFFF) has gained global attention as a multipurpose input for soil fertilization and pest and disease management. However, there are limited studies that have examined its effects on insect pest resistance and the underlying mechanisms. We investigated the impact of amending soil with BSFFF on maize growth, defense gene expression and resistance to a polyphagous insect herbivore, Spodoptera frugiperda (Lepidoptera: Noctuidae) through larval feeding assay. Microsporidia MB, a vertically transmitted endosymbiont of Anopheles mosquitoes, shows strong potential as a malaria control agent due to its ability to inhibit Plasmodium development within the mosquito host. To support its deployment in malaria transmission reduction strategies, it is critical to understand how environmental factors, particularly temperature, influence its infection dynamics. In this study, we investigated the impact of four temperature regimes (22 °C, 27 °C, 32 °C, and 37 °C) on Microsporidia MB prevalence and infection intensity by rearing mosquito larvae under controlled laboratory conditions. Accurate pathogen diagnosis is fundamental for effective crop disease management. In Kenya, a pathogen causing significant damage in potato farms was initially misidentified as Phytophthora infestans. This study aimed to correctly identify this pathogen and explore initial control measures using a polyphasic approach. The methodology integrated morphological observation, pathogenicity testing, and molecular analysis using ITS and EF1-α gene sequencing. The results confirmed the pathogen’s identity as Fusarium equiseti, with morphological features consistent with this species and molecular sequencing showing 99.6% identity to reference strains. In this dissertation, I apply a population aware lens to explore how genetic diversity and environmental conditions intersect to shape stress survival. Background: Cryptococcal meningitis remains a leading cause of HIV/AIDS-related mortality, predominantly in Sub-Saharan Africa. Current screening and prevention efforts primarily target antiretroviral therapy (ART)-naïve patients, whereas strategies for ART-experienced individuals are less well defined.

Objectives: This study primarily aimed at determining the prevalence of asymptomatic cryptococcal antigenemia (ACA) among ART-naïve and ART-experienced patients, along with associated factors and predictors of three-month outcomes among HIV patients in three HIV clinics in Dar es Salaam, Tanzania. Knowledge of novel and revised bacterial taxonomy facilitates routine operations in the medical microbiology laboratory and communication with important stakeholders. Notable new gram-positive taxa accepted in 2024 include the clinically significant Staphylococcus brunensis sp. nov., as well as Streptococcus suis subsp. hashimotonensis subsp. nov., which can be delineated from other Streptococcus suis subsp. by possession of the Lancefield group A antigen. Four novel Enterobacterales species are discussed, plus a novel eighth family Gallaecimonadaceae. Background: Over 55 million people worldwide live with dementia, with more than60% residing in low- and middle-income countries. Alzheimer’s disease, the mostcommon form of dementia, accounts for 60–70% of cases. Early and accurate diagnosisremains a global challenge, necessitating novel approaches to mitigate the diseaseburden. Biomarkers hold significant promise in improving diagnostic accuracy andpredicting disease progression. Validated biomarkers for the preclinical stages ofdementia are crucial for advancing diagnosis and therapeutic strategies. One hundred and fourteen calves (average BW= 224kg ± 89kg) were utilized to evaluate the effects of injectable trace mineral (ITM) administration on calf performance and health when administered pre-weaning and at weaning. Calves were blocked by sex, stratified by age, and randomly assigned to one of the following treatment groups: 1) no ITM injection (CON), 2) ITM administered 28 d before weaning (PW), or 3) ITM administration at weaning (WEAN). The purpose of this study was to compare the loading rates of shod and unshod conditions in treadmill walking. It was hypothesized that walking barefoot would demonstrate higher loading rates than walking with shoes. It was also hypothesized that walking barefoot on a treadmill would result in greater stride-to-stride variability than walking with shoes. Sixteen healthy adult females volunteered and provided their consent to participate in this study. Transcription factors (TFs) function as precision switches that integrate signaling inputs, chromatin state, and protein homeostasis to control gene expression. This thesis unites three interconnected research themes: (1) mechanistic studies of the ubiquitin-proteasome system (UPS) in regulating eukaryotic TFs, focusing on polymerase-associated Factor 1 (Paf1) and TATA-box binding protein-associated factor 2 (Taf2); (2) identification of biomarker genes within odor-related TF networks; and (3) integration of published undergraduate research linking molecular regulation, bioinformatics, and synthetic biology. Method: We utilized a cross-sectional sample (n = 161,981) comprising countriesfrom Latin America, Europe, Asia, and Africa. This thesis investigates the therapeutic potential of Avaren-Fc (AvFc), a novel lectin fusion protein that integrates the Fc region of human IgG1 with the engineered lectin Avaren, specifically targeting lymphoma. The study delineates the in vitro binding capabilities of Avaren-Fc with a human B-cell lymphoma cell line, a canine B-cell lymphoma cell line, healthy canine peripheral blood mononuclear cells (PBMCs), and healthy human PBMCs. AvFc was shown to exhibit significant binding to human lymphoma cells. Fear plays a crucial role in survival but can lead to psychological disorders when dysregulated. Fears can be innate due to evolutionary relevance or learned through associative processes, where predictive accuracy of threat is pivotal. Disruptions in fear neurocircuitry contribute to anxiety disorders, characterized by heightened activation and impaired learning. Fear conditioning and extinction paradigms based on classical Pavlovian conditioning have been used to study mechanisms underlying fear learning.
