Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants that pose ecological and human health risks due to their resistance to degradation and potential for bioaccumulation. Phytoremediation has emerged as a potential strategy for mitigating PFAS contamination, and plant-microbe interactions may influence contaminant uptake and transport within plant systems. These studies investigated microbial inoculation methods and examined whether Bacillus subtilis could influence the uptake and distribution of perfluorooctane sulfonate (PFOS) in hydroponically grown hemp (Cannabis sativa L.). Plant height and stem diameter growth rates and biomass accumulation were compared across treatments with and without PFOS exposure and microbial inoculation. PFOS …

Importance: Whether bipolar II disorder (BD-II) is associated with increased long-term mortality remains uncertain because most studies have not distinguished BD-II from bipolar I disorder (BD-I).

Objective: To examine whether BD-II is associated with elevated all-cause and cause-specific mortality compared with population controls, unaffected siblings, and those with BD-I.

Design, setting, and participants: This population-based, retrospective cohort study used data from Taiwan's National Health Insurance Database from January 1, 2000, to December 31, 2022. Individuals 12 years or older with 2 or more psychiatrist-assigned BD-II diagnoses were individually matched to 4 controls without BD-II by sex and birthdate. Additional comparisons …

In the neuroscience intensive care unit (NSICU), nurses rely on advanced monitoring tools to detect neurological deterioration. Two noninvasive measures, heart rate variability (HRV) and pupillary light reflex (PLR), are used to assess autonomic nervous system function; however, it remains unclear whether these indicators provide overlapping or distinct information. The purpose of this observational prospective study is to evaluate the relationship between HRV and quantitative pupillometry measurements to determine whether these noninvasive indicators provide congruent information. Fish have a system of sensory organs known as the lateral line system. This system contains numerous mechanoreceptive neuromasts and electroreceptive ampullary organs (Northcutt et al, 2000). Neuromasts are small, hair-like structures located within canals in the lateral line system just under the fish’s skin and are open to the environment through small pores visible on its skin (Parker, 2025). Water flows into these canals through the pores which stimulates the sensitive hair cells, resulting in electrical signals sent to the fish’s brain. Mitochondria Dynamics In Microglia During Health, Inflammation, And Aging, Alicia Nicole Pietramale

Microglia continually surveil the brain, allowing for rapid detection of tissue damage or infection. Microglial metabolism is linked to tissue homeostasis, yet how mitochondria are subcellularly partitioned in microglia and dynamically reorganize during surveillance, injury responses, and phagocytic engulfment in the intact brain has remained unclear. In this dissertation, I combine intravital imaging, ultrastructural analyses, and metabolic and cytoskeletal markers to investigate how mitochondrial organization relates to microglial function across health, inflammation, and aging.

I first show that microglial processes diverge in their mitochondrial content, with some containing multiple mitochondria while others are completely void. This heterogeneous partitioning is conserved …

The lack of effective preventive measures for tick-borne diseases represents a major public health challenge. Transmission-blocking vaccines targeting tick molecules essential for feeding or reproduction could provide a sustainable and broad-spectrum solution. In this study, we evaluated two tick-derived proteins as potential vaccine antigens, subolesin, a key regulator of blood-feeding and cellular processes, and vitellogenin, a protein involved in egg development. Using bioinformatic tools, we identified immunogenic peptide sequences from each protein and chemically conjugated them to virus-like particles (VLPs) to enhance antigen presentation. C57BL/6 mice were immunized intradermally with VLP-peptide conjugates, followed by a booster dose. Blood and spleen …

Glioblastoma multiforme is the most common adult primary tumor with a grim median survival time of 14.6 months. It is characterized by rapid progression, diffuse infiltration, and rather poor clinical outcomes. The WHO classifies GBM as a grade 4 diffuse astrocytic tumor with distinctive histopathological features including high mitotic activity, marked cellular atypia, microvascular proliferation, and necrosis. Advances in molecular diagnostics have further classified GMB into IDH-wild type and IDH-mutant subtypes, with important therapeutic and prognostic implications. Extracellular vesicles (EVs) mediate intercellular communication by transferring molecular signals between tissues. Liver-derived EVs contribute to metabolic regulation and have been implicated in tissue cross-talk during health and disease. Skeletal muscle is a highly metabolic tissue that depends on mitochondrial respiration for energy production. However, whether liver-secreted EVs influence myotube mitochondrial function remains unclear. Here, we investigate the impact of liver-derived EVs on myotube mitochondrial respiration.

• The reflex reaction time measures how quickly the nervous system can respond to a stimulus. • In the experiment, both visual and auditory reflexes were measured to evaluate differences in reaction time after the consumption of sour and spicy foods. • Spices and herbs produce different signals that interact with the nervous system. (McMullen 2024) • Comparing the response times from before and after consumption, this experiment allowed us to determine if different tastes affect reflex reaction times.

Purpose: To investigate contrast-enhanced T1-weighted MRI and diffusion-weighted magnetic resonance imaging (DWI) for early prediction of tumor response to combined irreversible electroporation (IRE) and anti-PD-1 immunotherapy.

Methods: Murine pancreatic ductal adenocarcinoma (PDAC) cells KRAS* were inoculated into the pancreas of C57BL/6 mice. IRE was performed when tumors became palpable. After IRE, mice received six intraperitoneal injections of anti-PD-1 over 2 weeks. T2-weighted MRI, T1-weighted MRI with macromolecular contrast agent poly(L-glutamic acid)-conjugated gadolinium (PG-Gd), and DWI were performed before and after IRE. Survival was analyzed using Kaplan-Meier curves. Mice surviving at least 100 days without recurrence after IRE were considered complete …

Neutrophils and neutrophil extracellular traps (NETs) contribute to early neuromyelitis optica (NMO) histopathology initiated by IgG targeting astrocytic aquaporin-4 (AQP4) water channels. Yet, the mechanisms underlying neutrophil recruitment and their pathogenic roles in disease progression remain unclear. To investigate molecular-cellular events preceding classical complement cascade activation in a mouse NMO model, we continuously infused, via spinal subarachnoid route, a non-complement-activating mouse monoclonal AQP4-IgG. Parenchymal infiltration of netting neutrophils containing C5a ensued with microglial activation and motor impairment but no blood-brain barrier leakage. Background: Cancer is a systemic disease with most deaths attributed to metastatic burden. Primary and metastatic tumors, albeit at different anatomic locations, are interconnected through multiple biological processes. Pre-clinical and clinical observations of growth acceleration of metastases after surgery, or abscopal effects outside the radiation field are widely reported, yet reliably triggering favorable and avoiding unfavorable systemic responses remains an unmet clinical need. Understanding local and systemic tumor interaction dynamics will help guide future treatments.

Huntington's disease (HD) is a neurodegenerative disorder caused by an abnormal CAG expansion in the Huntingtin (HTT) gene. Given its simple genetic cause but complex pathogenic mechanisms, interest in targeting HTT for HD treatment is growing, necessitating a clear understanding of HTT regulation. PURPOSE: The MTXPK.org webtool facilitates model-informed supportive care and glucarpidase use in patients receiving high-dose methotrexate, but its reliance on manual data entry limits workflow integration. We aimed to highlight how translational informatics can advance, automate, and enhance the usability of clinical decision support tools by embedding MTXPK.org within the electronic health record (EHR).

METHODS: A human factors study with 6 clinical providers guided iterative prototype development. MTXPK.org was rebuilt within the EHR using a 3-tier architecture with Fast Healthcare Interoperability Resources-based data retrieval, automated pharmacokinetic modeling, and interactive visualization.

Background: PD-L1 expression and tumor mutational burden (TMB) are biomarkers for immune checkpoint inhibitor (ICI) therapy in non-small cell lung cancer (NSCLC); however, patients harboring oncogenic alterations have limited benefit from ICIs. The impact of oncogenic alterations on TMB and PD-L1 tumor proportion score in lung cytology specimens is poorly understood. Herein, the association between oncogenic alterations, TMB, and PD-L1 in NSCLC cytology specimens is explored.

Methods: Next-generation sequencing results from 312 NSCLC cytology specimens were retrospectively reviewed that interrogate 610 genes and select immuno-oncology signatures. TMB and PD-L1 immunohistochemical expression across oncogenic alterations were analyzed to explore associations.

Purpose: Inhibitors of poly(ADP-ribose) polymerase (PARP), an enzyme with numerous roles in DNA damage response signaling, represent a class of anti-cancer drugs approved for treating solid tumors with defects in the DNA damage response. However, additional methods for identifying patients who would benefit from the use of PARP inhibitors are urgently needed. We evaluated a novel radiotracer, 18F-FluorThanatrace ([18F]-FTT), to noninvasively assess PARP activity with PET/CT before treatment and determine associations between uptake, tumor mutation status, and prior receipt of therapy including PARP inhibitors.

Background & aims: Sorafenib has been a cornerstone of hepatocellular carcinoma (HCC) therapy; however, its efficacy is limited, and identifying patients who will benefit from sorafenib is challenging. We aimed to identify predictive biomarkers of sorafenib benefit in patients with HCC.

Methods: Gene expression data from 33 HCC tumors treated with sorafenib were analyzed to construct a prediction model aimed at identifying patients with greater benefit from sorafenib treatment. The robustness of the predictor was validated using gene expression data from two phase III clinical trials, IMbrave150 and STORM.

The central research question of this thesis asks: How does nutrition influence ocular health overall and the impact of nutrition on the development or progression of common eye diseases? Building upon existing research in optometry/ophthalmology, nutrition science, and public health, this project examines the physiological mechanisms linking diet to eye health and evaluates how specific nutrients and dietary patterns contribute to disease prevention and visual function. While podcasts may not be the most popular form of mass media, they are growing in their accessibility and prevalence in educational discourse. My creative work, in the form of a podcast, unmasks scientific knowledge in an untraditional, mass-media form and will use podcast techniques, such as music transitions and storytelling techniques, to maintain engagement.
