The common consensus is that owning pets affects human health positively. However, the effects of owning pets on mental health are still controversial. Therefore, the present study aimed to determine the possible associated with depression, anxiety, stress, and owning cats and dogs in Türkiye. Additionally, the potential impact of personality traits on this relationship was examined. This study included 636 participants, of whom 52% were female (Mage = 33.02, SD = 15.78). Using a flow-through laboratory bioassay, experiments investigated the effects of limited feeding, increased temperature, ammonium chloride (NH4Cl), and 17α-ethinylestradiol (EE2) on juvenile mummichog (Fundulus heteroclitus) growth. Weight measurements were taken every three days as the apical growth endpoint. Gene expression was examined by qPCR for genes in the GH (growth hormone)-IGF1 (insulin-like growth factor 1) pathway, including gh, igf1, and their receptors (ghra, ghrb, igf1ra, igf1rb). Introduction: Longitudinal effect of diet-induced obesity on bone is uncertain. Prior work showed both no effect and a decrement in bone density or quality when obesity begins prior to skeletal maturity. We aimed to quantify long-term effects of obesity on bone and bone marrow adipose tissue (BMAT) in adulthood.

Methods: Skeletally mature, female C57BL/6 mice (n = 70) aged 12 weeks were randomly allocated to low-fat diet (LFD; 10% kcal fat; n = 30) or high-fat diet (HFD; 60% kcal fat; n = 30), with analyses at 12, 15, 18, and 24 weeks (n = 10/group). Tibial microarchitecture was analyzed …

Introduction

Growing interest centers on the association between circulating ketone bodies (KB) and cognitive function, notably in aging and neurodegenerative diseases.

Methods

Associations of plasma KB with incident dementia and cognitive performances were examined among Multi‐Ethnic Study of Atherosclerosis (MESA) participants. KB were measured using plasma samples collected following an overnight fasting at Exam 1 (2000–02) and detailed cognitive testing at Exam 5 (2010–2012, N = 4392), Exam 6 (2016–2018, N = 1838), and in MESA‐MIND (2019–2021, N = 2060).

Results

BACKGROUND: The interrelationship of parkinsonism, Parkinson's disease (PD) and other Alzheimer's disease (AD) and Alzheimer's disease and related dementias (ADRD) pathologies is unclear.

OBJECTIVE: We examined the progression of parkinsonian signs in adults with and without parkinsonism, and their underlying brain pathologies.

METHODS: Annual parkinsonian signs were based on a modified Unified Parkinson's Disease Rating Scale. This dissertation addresses critical gaps in dementia research in Native Hawaiian and Pacific Islander (NHPI) communities. Focusing on Samoan and Tongan communities, the study successfully adapted and validated the AD8 informant interview and the Clinical Dementia Rating (CDR) scale in native languages, establishing these instruments as reliable tools for detecting cognitive impairment in these populations. The research process prioritized meaningful community engagement and forged partnerships with indigenous professionals, ensuring alignment with and responsiveness to the specific needs of the community. Telomeres are chromosomal elements that protect and maintain genes as DNA replicates in many organisms. As individuals age and DNA replicates, telomeres often shorten; thus, telomere length often corresponds with an individual’s lifespan, and early-life decreases in telomere length can be predictive of longevity. Here, I investigated the relationship between telomere length and developmental environment in a cooperatively breeding species, the chestnut-crowned babbler (Pomatostomus ruficeps; CCB). This multi-study dissertation examined dual-task performance in young and middle-aged adults using verbal fluency tasks (VFT) during different walking scenarios (baseline, performing a VFT, walking narrowly, and performing a VFT while walking narrowly) as well as the Selection, Optimization, and Compensation (SOC) model. Thirty adults (15 young adults (YA; 19-44 years) and 15 middle-aged adults (MA; 45-65 years) participated in this study. Overall, there were no significant differences between groups when data was examined in short (means of means approach) and long-form (incorporating all data), when analyzing clustering and switching strategies, gait speed variability, or dual-task costs. Aging and excess post-exercise oxygen consumption (EPOC) is something that has not been adequately studied. Studies looking at EPOC mostly are compiled of young college age individuals. Aging in humans is a topic of much research with still more to be learned. The study in this paper attempts to answer the question of whether age affects EPOC when performing a certain exercise. Preliminary findings based on general age-related research suggest the possibility. This thesis will explore the current research related to age and EPOC, analyze the findings, and discuss additional avenues for future research.

Here we develop two new social indices: The ADA PARC Absolute Economic Opportunity Index and the ADA PARC Relative Economic Opportunity Index. These indices allow us novel examinations of economic equity between people with and without disabilities within a U.S. State and between people with disabilities in different states using aggregations of multiple component economic indicators. These represent the first efforts to offer U.S. indices of this focus, an important development given the distinct economic needs of people with disabilities and the value in accounting for distinct national policies. The composition, structure and functional properties of Agaricus blazei polysaccharide were summarized. The extraction techniques of polysaccharides and the features of different extraction methods were enumerated and compared. The problems in the research of A. blazei was discussed, such as unsystematic basic research, immature technological conditions, imperfect equipment and facilities, and low degree of specificity in seeds. In the end,the research direction of A. blazei polysaccharide was prospected.

Background and Objectives: Fall incidents from unsafe home environment are frequent in older-adult homes but the literature is ambiguous whether it is the presence/absence, or the interplay of such conditions and physical functioning that is of salience. Background The relationship between mitochondrial DNA copy number (mtDNA CN) and cardiovascular disease remains elusive. Methods and Results We performed cross-sectional and prospective association analyses of blood-derived mtDNA CN and cardiovascular disease outcomes in 27 316 participants in 8 cohorts of multiple racial and ethnic groups with whole-genome sequencing. We also performed Mendelian randomization to explore causal relationships of mtDNA CN with coronary heart disease (CHD) and cardiometabolic risk factors (obesity, diabetes, hypertension, and hyperlipidemia).

The present study tested whether energy-minimizing behaviors evoke reward-related brain activity that promotes the repetition of these behaviors via reinforcement learning processes. Fifty-eight healthy young adults in a standing position performed a task where they could earn a reward either by sitting down or squatting while undergoing electroencephalographic (EEG) recording. Reward-prediction errors were quantified as the amplitude of the EEG-derived reward positivity. Results showed that reward positivity was larger on reward versus no reward trials, confirming the validity of our paradigm to measure evoked reward-related brain activity. Background The relationship between alcohol consumption and ectopic fat distribution, both known factors for cardiovascular disease, remains understudied. Therefore, we aimed to examine the association between alcohol consumption and ectopic adiposity in adults at risk for cardiovascular disease. Methods and Results In this cross-sectional analysis, we categorized alcohol intake among participants in MESA (Multi-Ethnic Study of Atherosclerosis) as follows (drinks/day): <1 >(light drinking), 1 to 2 (moderate drinking), >2 (heavy drinking), former drinking, and lifetime abstention. With this increase in participation, clinicians need better evidenced based strategies to advise those golfers with different pathologies when it is safe to return to the game. This quasi-experimental study evaluated the impact of a 10-week evidence-based falls prevention program (Bingocize®) on self-reported fear of falling, general health, physical activity, social isolation, and avoidance behavior, in community-dwelling older adults in Virginia. Participants > 60 years of age (n= 481) attended Bingocize® group sessions twice per week for 10 weeks. The program combined conventional bingo with periodic strength, balance, flexibility exercises, and fall prevention education. PURPOSE: Germline pathogenic variants in CHEK2 confer moderately elevated breast cancer risk (odds ratio, OR ∼ 2.5), qualifying carriers for enhanced breast cancer screening. Besides pathogenic variants, dozens of missense CHEK2 variants of uncertain significance (VUS) have been identified, hampering the clinical utility of germline genetic testing (GGT).

EXPERIMENTAL DESIGN: We collected 460 CHEK2 missense VUS identified by the ENIGMA consortium in 15 countries. Their functional characterization was performed using CHEK2-complementation assays quantifying KAP1 phosphorylation and CHK2 autophosphorylation in human RPE1-CHEK2-knockout cells. INTRODUCTION: Comparative epigenomics is an emerging field that combines epigenetic signatures with phylogenetic relationships to elucidate species characteristics such as maximum life span. For this study, we generated cytosine DNA methylation (DNAm) profiles (n = 15,456) from 348mammalian species using amethylation array platform that targets highly conserved cytosines.

RATIONALE: Nature has evolved mammalian species of greatly differing life spans. To resolve the relationship ofDNAmwith maximum life span and phylogeny, we performed a largescale cross-species unsupervised analysis. Comparative studies in many species enables the identification of epigenetic correlates of maximum life span and other traits.

This research focuses on FOXO4 that is differentially localized in U87MG glioblastoma cells where FOXO4 is targeted to the nucleus. Mitochondrial disorders are complex diseases that involve multiple systems and result from mutations and deletions in the mitochondrial genome. Leigh syndrome (LS) is a rare and devastating primary mitochondrial disorder characterized by progressive neuromuscular degeneration. Despite its severe consequences, the clinical genotype pathophysiological underlying LS remains elusive, posing significant challenges for the development of effective therapies. Importance: Sarcopenia is an established prognostic factor in patients with head and neck squamous cell carcinoma (HNSCC); the quantification of sarcopenia assessed by imaging is typically achieved through the skeletal muscle index (SMI), which can be derived from cervical skeletal muscle segmentation and cross-sectional area. However, manual muscle segmentation is labor intensive, prone to interobserver variability, and impractical for large-scale clinical use.

Introduction: This study investigated the impact of the multistage ultra-marathon event ‘‘Marathon des Sables’’ (MdS) performed in the Sahara Desert on the psychophysical capacity of an athlete. Methods: We collected and analyzed environmental, physiological, and behavioral data from a 39-year-old athlete who participated in the MdS. Specifically, we collected environmental temperature (Tenv), upper inguinal skin temperature (Tsk), heart rate, and running speed data. An epidemic of obesity has affected large portions of the world, increasing the risk of developing many different age-associated diseases, including cancer, cardiovascular disease, and diabetes. In contrast with the prevailing notion that "a calorie is just a calorie," there are clear differences, within and between individuals, in the metabolic response to different macronutrient sources. Recent findings challenge this oversimplification; calories from different macronutrient sources or consumed at different times of day have metabolic effects beyond their value as fuel. Genome-wide polygenic risk scores (GW-PRSs) have been reported to have better predictive ability than PRSs based on genome-wide significance thresholds across numerous traits. We compared the predictive ability of several GW-PRS approaches to a recently developed PRS of 269 established prostate cancer-risk variants from multi-ancestry GWASs and fine-mapping studies (PRS269). GW-PRS models were trained with a large and diverse prostate cancer GWAS of 107,247 cases and 127,006 controls that we previously used to develop the multi-ancestry PRS269. Driving is a complex skill involving the interaction of various brain functions, making it challenging to determine the independent contributions of each process to driving proficiency. While past studies have identified assessments such as the Useful Field of View Test (UFOV) and the Trial Making Test (TMT) as strong predictors for driving skills, little research has explored the impact of visual search and movement inhibition on driving after stroke. In recent years, several new classes of therapies have been investigated with their potential for restoring or improving physical functioning in older adults. These have included Mas receptor agonists, regulators of mitophagy, skeletal muscle troponin activators, anti-inflammatory compounds, and targets of orphan nuclear receptors. The present article summarizes recent developments of the function-promoting effects of these exciting new compounds and shares relevant preclinical and clinical data related to their safety and efficacy. The development of novel compounds in this area is expanding and likely will need the advent of a new treatment paradigm for age-associated mobility loss and disability.

Standardised bomb calorimetry methods are essential to accurately quantify the gross energy within food and beverages, yet no accepted protocols exist. The objective of this review was to synthesise literature on food and beverage sample preparation methods used for conducting bomb calorimetry. This synthesis enhances our understanding of the extent to which methodological variances may currently affect estimates of the caloric values of dietary items.
