This paper starts with a review about Plant Functional Types and then goes deeper into the conceptual foundations of Plant Functional Types (PFT's) with detailed discussion of global trait spectra such as the leaf economics spectrum and the fast-slow continuum. Analysing large population genomic datasets requires an interdisciplinary skillset. Beyond a knowledge base in genetics and population biology, population genomic analyses involve computer science and statistics, representing a barrier for researchers without experience in those fields. PopGenHelpR seeks to lower this barrier by enabling researchers to perform population genomic analyses and generate near-publication quality figures in a streamlined and informed fashion. PopGenHelpR allows users to estimate genetic diversity within populations as well as differentiation among populations and individuals from single nucleotide polymorphism data. OBJECTIVE: Proliferative vitreoretinopathy (PVR) remains a major cause of failure after rhegmatogenous retinal detachment repair and lacks effective pharmacologic therapies. Although epithelial-mesenchymal transition (EMT) is central to PVR pathogenesis, the structural determinants governing the tractability of EMT regulators, particularly those involving intrinsic disorder, remain poorly defined. We developed a disorder-aware, artificial intelligence-enabled computational framework to evaluate EMT-associated proteins in PVR and prioritize structurally tractable regulators for structure-based targeting.
DESIGN: A computational, hypothesis-generating study employing an in silico screening and structural modeling pipeline.

Biogeographical ancestry (BGA) inference is an important tool in forensic science, providing supplemental information when comparison/reference DNA is not available for traditional DNA analyses. Insertion/deletion polymorphisms (INDELs) have been used in the last decade as ancestry-informative markers (AIMs) for BGA inference because they can be easily adapted to forensic-type samples and are determined using low cost and easy processing methods. Noncanonical metabolite 5' caps have recently been identified on RNAs, and the DXO/Rai1 family of enzymes can remove these caps in eukaryotes. While the binding modes of NAD, FAD and dephospho-CoA (dpCoA) caps in the active site of mouse DXO have been determined, how DXO recognizes the UDP-glucose (UDP-Glc) and UDP-N-acetylglucosamine (UDP-GlcNAc) caps is not known. In addition, the molecular mechanism by which DXO catalyzes the decapping reactions is still poorly understood, especially the location of the water/hydroxide that attacks the scissile phosphate to initiate the decapping. Endemism Patterns Of Leaf Beetles (Coleoptera: Chrysomelidae) In Türkiye, İsmai̇l Şen, Ali̇ Gök

In this study, the distribution of endemic leaf beetles in Türkiye was analyzed holistically at the provincial scale using species richness (ENR), corrected weighted endemism (CWE), and range-restricted endemism (RRE) indices. A total of 95 endemic taxa were found in Türkiye and these taxa exhibited significant spatial heterogeneity in terms of richness and extent of distribution. The Western and Central Taurus Mountains and the Eastern Black Sea Region were found to be the main centers of endemism, with high densities of narrowly distributed and geographically restricted species. Background: Global health financing has historically prioritized disease-specific “vertical” programs over broader “horizontal” strengthening of general health systems. Vertical programs in HIV have led to unprecedented reductions in mortality worldwide, but HIV remains a significant public health threat in sub-Saharan Africa, where vertical strategies have long been criticized for creating fragmented health systems. Interest in the anxiolytic-like effects of cannabidiol (CBD), the non-intoxicating phytocannabinoid found within the genus, Cannabis, has grown significantly in recent years. However, preclinical investigations of CBD’s anxiolytic potential often elect to forgo translationally relevant models that maximize external validity in favor of greater internal and mechanistic validity. As a result, the effects of relatively low dose (< 3mg/kg) oral CBD administration on anxiety-like behaviors are not yet fully characterized. The present studies examined the impact of two orally delivered CBD-dominant cannabis products – a “full-spectrum” formulation derived from the whole plant and a CBD isolate derived from the same chemovar – on innate defensive behaviors exhibited by adult male and female rats in various tests of anxiety. Results indicated that acute and chronic low dose oral CBD administration attenuated anxiety-like behavior in rats, regardless of sex or formulation. These findings extend the growing corpus of literature describing anxiolytic-like effects of CBD.

This thesis examines the pollinator concept and tests how sampling methods shape bee communities. Chapter 1 provides a conceptual and philosophical analysis, arguing that pollination is a process and that pollinators are context-dependent functional roles rather than fixed taxonomic categories. It traces the concept’s development from natural history to ecological theory, introduces a dual-goal model separating plant reproduction from visitor foraging, and proposes a classification framework based on evidentiary certainty and pollination potential to resolve ambiguity in pollination research.

The One Health framework, which promotes human health and wellness through the understanding that animal, environmental, and human health are interwoven, is critical to understanding pathogen emergence and transmission. Riparian areas exist at the aquatic-terrestrial interface and provide disproportionate ecosystem services, yet multiple stressors have degraded these zones globally. Stream restoration seeks to reverse degradation, but rigorous evaluation of treatment effectiveness remains limited. I conducted a before-after-control-impact (BACI) study in the southern Rocky Mountains to evaluate two stream restoration treatments—beaver dam analogs (BDAs) and plug-and-ponds (P&Ps)—across three dimensions of riparian ecosystem response. Using Bayesian analysis, I quantified treatment effects on functional diversity and composition of riparian plant communities. Germline specification is a fundamental process that ensures the proper development of germ cells. Germline specification occurs either through germ cell determinants maternally inherited through the mechanism of preformation or through induction, where cell interaction and epigenetic modifications direct specification. Hypoxic environments, resulting from decreased atmospheric partial pressure of oxygen, reduce oxygen availability in the blood and tissues, leading to a mismatch between supply and demand. This elicits several physiological adaptations and may influence bone metabolism. Although evidence suggests that hypoxia can modulate bone remodeling through hypoxia-inducible pathways, findings remain mixed, with most data derived from chronic hypoxic exposures in animal and cell culture models. Exercise, an essential stimulus for bone health, may exacerbate hypoxic stress; however, the skeletal response to acute hypoxic exposure combined with exercise remains unclear. Microtubules, composed of a/b-tubulin heterodimers, play a central role in breast cancer tumor growth by polymerizing, leading to metastasis and depolymerizing, contributing to proliferation. Human enzymes protein kinase Ca (PKC-a) and cyclin-dependent kinase 1 (Cdk-1) mediate phosphorylation at sites a:Ser165 and b:Ser172, respectively, influencing the growth of microtubules. It is possible that alternating phosphorylation at these sites contribute to an a/b-tubulin “toggle switch” that mediates microtubule instability and tumor growth.

Vesicoureteral reflux (VUR) is a risk factor for urinary tract infection (UTI). However, data describing the urobiome in children with VUR are limited. Here, we characterized the urobiome in children with and without VUR to better understand its potential role in VUR pathogenesis. Catheterized urine samples were obtained from children who presented for voiding cystourethrogram (VCUG) between 12/1/2020 and 10/3/2023. Children were excluded if they had any surgical procedures of the genitourinary tract in the 30 days prior to VCUG or were on treatment antibiotics at the time of VCUG. 16S rRNA gene sequencing and taxonomical classification identified the urobiome. …

Triple-negative breast cancer (TNBC), an aggressive form of breast cancer, characterized by its lack of estrogen receptors (ER), progesterone receptors (PR), and human epidermal growth factor receptors (HER2) poses a therapeutic challenge as common treatment options are rendered ineffective. Regular exercise and physical therapy are known to increase production of neurotrophins, including brain-derived neurotrophic factor (BDNF), and anti-inflammatory cytokines. These changes combine to theoretically protect and slow the progressive loss of neurons in Parkinson’s disease (PD). The purpose of this study was to examine the effect of exercise and physical activity on various anti-inflammatory and pro-inflammatory cytokines and neurotrophins in people with PD (PwP). Leg stiffness is regulated by reflexive, intrinsic, and passive mechanisms, which are influenced by posture, body weight loading, and motor task demands. After a stroke, the capacity to modulate these mechanisms is often compromised. The H-reflex is a widely used electrophysiological tool to assess the excitability of the Ia afferent pathway within spinal circuitry. Effectiveness Of Cognitive Stimulation Therapy On The Quality Of Life Of Adults With Dementia: A Secondary Analysis, Evelyn Airam

This secondary analysis study evaluated the effectiveness of Cognitive Stimulation Therapy (CST) on quality of life (QoL), depressive symptoms, and cognitive function in 108 community-dwelling older adults with cognitive impairments, including dementia. Outcomes involved Quality of Life (QoL) measured by the Quality of Life in Alzheimer’s Disease (QoL-AD), symptoms of depression by the Cornell Scale for Depression in Dementia (CSDD), and cognitive function using the Saint Louis University Mental Status (SLUMS). After the CST, significant improvements were observed in QoL (p = .004), depressive symptoms (p = .002), and cognitive function (p < .001), with small effect sizes. Additionally, a significant interaction between sex and dementia diagnosis status was found in predicting cognitive changes, suggesting that the relationship between dementia diagnosis and cognitive improvement differs by sex. No between-group differences were found for QoL or depression. Findings support CST as a multidimensional, non-pharmacological intervention that improves cognitive, psychological, and social outcomes. Although effects were small, consistent improvements are clinically meaningful given the progressive nature of dementia. This study extends existing literature by demonstrating CST’s effectiveness in a community-based setting and reinforces its alignment with occupational therapy’s emphasis on participation and holistic care. Results highlight the need for early intervention and expanded access to CST programs to enhance quality of life in this population.

Apical constriction is a conserved morphogenetic mechanism that drives epithelial folding during development. During ventral furrow formation in Drosophila melanogaster, contractile actomyosin networks generate forces that reduce apical cell area resulting in tissue invagination. For these cell-scale forces to produce coordinated tissue-scale changes, contractile networks must be mechanically integrated across neighboring cells. Although adherens junctions are known to mediate this coupling, it is unclear how junctions are integrated into cortical actin cytoskeleton, which directly determines cell, and consequently tissue, shape. Trustworthy prediction of enzyme function from protein sequences remains a central challenge in computational biology, particularly when annotated data are limited, imbalanced, or incomplete. This dissertation develops interpretable deep learning methods for enzyme discovery and enzyme function prediction from amino acid sequences. First, it introduces PEPIC, an interpretable convolutional neural network for substrate-level prediction of hydrolytic plastic-degrading enzymes. Using curated and expanded sequence datasets, PEPIC improved predictive performance over benchmark methods, identified sequence regions aligned with catalytic and substrate-binding residues, and supported the discovery and experimental validation of a previously uncharacterized PET-degrading enzyme. Most modern mammals maintain stable body temperatures (Tb) ~37ºC and high metabolic rates. Hibernators, like ground squirrels, are homeothermic during the active summer season, but are heterothermic during the hibernation season. During the hibernation season, ground squirrels follow a predictable pattern of torpor bouts interrupted by interbout arousals. Tenrecs are unusual because they are heterothermic and heterometabolic even during the active season. Previously, ground squirrel bile constituents were shown to be predictive of whether a squirrel is in the summer active or winter hibernation season. The SRY (Sex-determining Region Y) protein is a transcription factor encoded on the Y chromosome and is the key regulator responsible for initiating male sex determination in mammals. During early embryonic development, SRY activates the genetic program that leads to testis formation by promoting the expression of downstream genes involved in male gonadal differentiation. Mutations or dysregulation of SRY can lead to disorders of sex development such as male-to-female sex conversion and hermaphroditism, highlighting its critical role in sex determination.
