What’S The Beef With Agriculture? Evaluating The Effectiveness Of An Agricultural Literacy Intervention On Students’ Attitudes And Knowledge Toward Sustainability And Animal Agriculture, Jenny E. Bennett
All Graduate Theses and Dissertations, Fall 2023 to Present
Agricultural literacy is essential for helping individuals understand how food systems impact their daily lives and society. While most research has focused on students’ knowledge, this study addresses the underexplored role of attitudes—critical predictors of behavior—as outcomes of educational interventions. This research analyzed survey data collected before and after an agricultural literacy intervention delivered to high school students in school-based agricultural education (SBAE) programs. The curriculum focused on sustainability in animal agriculture.
Findings from paired samples t-tests showed statistically significant improvements in students' attitudes (from moderately neutral to more positive) and knowledge scores. Further analyses revealed that demographic factors such …
Electrophysiological Markers Of Emotion Dysregulation In Autistic Youth, Rachel G. Mcdonald
Electrophysiological Markers Of Emotion Dysregulation In Autistic Youth, Rachel G. Mcdonald
Theses, Dissertations and Culminating Projects
Emotion dysregulation (ED) is a transdiagnostic mechanism that is conceptualized as difficultly regulating emotions and deploying appropriate regulation strategies in response to environmental stimuli. Meta-analyses suggest that autistic people, across the lifespan, experience more severe ED as compared to both neurotypical people and those with mental health conditions. Despite elevated risk and evidence linking ED to disruptions in neural mechanisms, no studies to date have examined neural markers of ED in autistic populations. The current project uses multi-method and multi-informant measurement to examine similarities and differences in neural mechanisms of ED in autistic and non-autistic populations. Forty-four youth aged 11—17 …

Stress-Survival Pathway Genes In Colorectal Cancer: Crc Progression And Hispanic Disparities, Highlighting Mcm10 As A Potential Player, Urbashi Basnet
Open Access Theses & Dissertations
Despite advances in screening and treatment, colorectal cancer (CRC) remains a leading cause of cancer-related deaths in the U.S., with notable disparities across racial and ethnic groups. Hispanic populations face a disproportionate burden, underscoring significant differences in incidence and outcomes. This study examines the role of the Stress-Survival Pathway (SSP) in colorectal cancer (CRC), with a focus on addressing health disparities among Hispanic populations. Twenty-eight differentially expressed SSP genes identified from bioinformatic analysis were validated in CRC cell lines and cDNA arrays using qRT-PCR and tissue microarrays by immunohistochemistry. SSP genes were further analyzed in CRC tissues from Hispanic and …

Despite the enthusiasm and advances in the purification of native and engineered full-length membrane proteins, little attention has been paid to their fragments which could serve as attractive inspiration for function, regulation, or targeting of full-length membrane protein with therapeutic antibodies (Abs). Production of recombinant fragments of "therapeutic" membrane proteins for early-stage discovery research requires their purification to near homogeneity. It is important not only for the production of biotherapeutic antibodies but also for structural and functional studies of competitive protein-Abs, protein-protein, and lipid-protein interactions which heavily rely on the purity and quality of the isolated protein fragment of interest. …

In this 5th version of the European LeukemiaNet guidance for adult patients, there are important changes in several areas of management based on evidence available since 2020, including the World Health Organisation's reclassification of CML as a biphasic disease. Previous advice to switch the tyrosine kinase inhibitor (TKI) on failure of molecular milestones, is modified to better account for individual patient circumstances. Our recommendations are summarized in tables designed to be read in conjunction with the text which offers justification and additional advice. We describe decision-making for first-line treatment, both in available drugs and their initial dosing. Similarly we elaborate …

Purpose: Master protocols represent transformations, enabling multiple therapies or diseases under a single protocol. These designs streamline therapeutic development by reducing redundancies. Suited for evolving fields such as oncology and global emergencies such as the COVID-19 pandemic, master protocols have been exemplified by studies such as RECOVERY, Solidarity, and I-SPY 2, which accelerated effective treatment identification (with I-SPY 2 focused on molecular subtypes). Recent oncology examples, such as MORPHEUS, evaluate immunotherapy combinations with shared controls. Despite advantages, their application in early-phase oncology remains underutilized amid growing regulatory emphasis on randomization for robust evidence.

TP53 mutations are common in breast cancer (BC) and are associated with poor prognosis. GD3 synthase (GD3S/ST8SIA1), a gene associated with breast cancer stem cells, is upregulated in tumors with p53 mutations. However, the functional relationship between GD3S and p53 is unknown. Here, we show that GD3S levels are highest in breast tumors with specific p53 mutations. Functional studies revealed that wild-type (WT) p53 inhibits GD3S expression, whereas mutation in p53 enhances GD3S expression by upregulating GD3S promoter activity. Moreover, we found that GD3S inhibits wild-type p53-induced apoptosis in BC cells, while BC cells harboring gain-of-function p53 mutations are dependent …

The combination of the B-cell lymphoma 2 (BCL2) inhibitor venetoclax (VEN) and the hypomethylating agent decitabine (DEC; VEN/DEC) constitutes a primary therapeutic strategy for treating older adults with acute myeloid leukemia (AML). However, a notable subset of patients exhibits resistance to VEN/DEC, demonstrating either no disease response or relapse after initial remission. This study aimed to elucidate the molecular mechanisms underlying this resistance through analyses of gene expression and DNA methylation profiles. We conducted comprehensive RNA sequencing analysis and DNA methylation profiling on AML samples from 35 patients undergoing VEN/DEC therapy. The RNA sequencing analysis revealed that several genes related …

Methylthioadenosine phosphorylase (MTAP) is a key enzyme in purine metabolism that may influence cellular responses to injury. We evaluated the effects of prophylactic MTAP inhibition in mouse models of ischemia-reperfusion and cisplatin-induced acute kidney injury (AKI). MTAP inhibition was confirmed by accumulation of methylthioadenosine (MTA). Treated mice showed reduced renal injury and decreased tubular damage. Transcriptomic analysis revealed protection from inflammatory and stress pathways, while maintaining oxidative phosphorylation, fatty acid metabolism, and epithelial integrity-related genes. Analysis of human single-cell RNA-seq data from the Kidney Precision Medicine Project indicated that MTAP is highly expressed in kidney injury marker-positive adaptive proximal tubule …

Background and objective: NRG/RTOG 0521 randomized men with high-risk localized prostate cancer (PC) to androgen suppression (AS) and definitive radiotherapy (RT) ± docetaxel-based chemotherapy (CT). The overall survival (OS) benefit with CT initially reported was lost on longer follow-up. The Decipher genomic classifier (GC) measures multiple transcripts relevant to docetaxel action. Basal/luminal differentiation portends differential response to AS and CT for high-risk localized and metastatic hormone-sensitive PC. We validated the Decipher GC in pretreatment biopsy samples for risk stratification and examined basal-luminal subtyping to predict docetaxel response.
Methods: Decipher GC scores and basal-luminal cellular subtypes were generated for specimens from …

Introduction: Cancer survivors who quit smoking have improved treatment response and decreased mortality. Although data indicate that greater tobacco cessation treatment engagement leads to better cessation outcomes in cancer survivors, little is known about how different tobacco treatment engagement patterns influence long-term cessation success. To help address this issue, this study examined how tobacco treatment engagement patterns predict cessation outcomes through 18 months and identified baseline participant characteristics predictive of treatment engagement patterns.
Methods: Data from a smoking-cessation RCT among women with a history of cervical intraepithelial neoplasia or cervical cancer comparing a 12-month, 6-session motivation and problem solving phone …

Impaired cerebral circulation, induced by blood vessel constrictions and microthrombi, leads to delayed cerebral ischemia after subarachnoid hemorrhage (SAH). 12/15-Lipooxygenase (12/15-LOX) overexpression has been implicated in worsening early brain injury outcomes following SAH. However, it is unknown if 12/15-LOX is important in delayed pathophysiological events after SAH. Since 12/15-LOX produces metabolites that induce inflammation and vasoconstriction, we hypothesized that 12/15-LOX leads to microvessel constriction and microthrombi formation after SAH, and thus, 12/15-LOX is an important target to prevent delayed cerebral ischemia. SAH was induced in C57BL/6 and 12/15-LOX−/− mice of both sexes by endovascular perforation. Expression of 12/15-LOX was assessed …

Alzheimer's disease (AD) is a complex neurodegenerative proteinopathy in which Aβ and tau misfold and aggregate into entities that structurally unsettle native proteins, mimicking a prion-like or "seeding" process. These Aβ and tau "seeds" can arrange in different conformations or strains that might display distinct pathogenic properties. Furthermore, recent evidence suggests that microglia play a key role in the amyloidogenic event and can modulate the propagation and aggregation processes. Here, we employed histological and molecular approaches to determine whether seeds from human AD brains compared to those from transgenic mice (3xTg-AD) are more prone to induce Aβ and tau aggregates …

Using Herpetofauna To Determine Wetland Health And Function In The Red River Watershed, Shelby Rodriguez-Edwards
Biology Theses
Reptiles and amphibians are commonly used indicator species in wetlands because of their increased sensitivity to environmental conditions and use of both aquatic and terrestrial habitats. Herpetofauna can reflect wetland ecological functionality through their community composition and responsiveness to environmental stressors. This study evaluated reptile and amphibian communities in natural and ACEP-WRE wetlands within the Red River Watershed of northeastern Texas and Northwestern Louisiana to assess whether restored wetlands support herpetofaunal assemblages compares to their natural counterparts. Fifteen wetlands were surveyed from February to June 2024 using visual encounter surveys, call surveys, and standardized trapping methods to document species richness …
Characterizing Host And Viral Factors Relevant To Turkey Hemorrhagic Enteritis Virus-Induced Immunosuppression, Abraham Quaye
Characterizing Host And Viral Factors Relevant To Turkey Hemorrhagic Enteritis Virus-Induced Immunosuppression, Abraham Quaye
Theses and Dissertations
Turkey hemorrhagic enteritis virus (THEV) is an adenovirus (AdV) that causes hemorrhagic enteritis (HE) in turkeys, a debilitating disease affecting predominantly 6-12-week-old turkey poults characterized by immunosuppression (IMS) and intestinal lesions leading to bloody diarrhea. Although a naturally-occurring avirulent THEV strain has been adopted as live vaccine and has proven effective at preventing HE in turkeys, it still retains its immunosuppressive ability. Thus, vaccinated birds are rendered more susceptible to opportunistic infections and death than unvaccinated cohorts, leading to substantial economic losses. This dissertation focuses on elucidating both host and viral factors mediating THEV-induced IMS in turkeys. As THEV gene …

Characterizing Gastric Cancer Tumor Immune Microenvironment Through Extracellular Vesicles, Claire Elizabeth Bruno
Theses and Dissertations
The tumor immune microenvironment (TIME) critically influences cancer progression and determining therapeutic response. As immunotherapies continue to improve and reshape cancer treatments, there is a need for tools that can more efficiently and accurately assess and monitor the TIME. Extracellular vesicles (EVs), especially small EVs (sEVs), are lipid-bound nanoparticles that carry proteins, DNA, and RNA reflective of their cell of origin, offering a novel, non-invasive biomarker of the TIME. This makes them a promising candidate for immune profiling and treatment monitoring in cancer. This thesis investigates the potential of EVs as biomarkers for TIME composition and immune activity. An analysis …

Stakeholder Perspectives On Extended Producer Responsibility (Epr) And E-Waste Management: A Case Study Of China, Viola Dheena Pongajow, Yufei Qin, Kaihua Xu, Siliang Liu, Zongliang Zhang, Fangyang Liu
Journal of Environmental Science and Sustainable Development
Electronic waste (e-waste) is increasing at an unprecedented rate, with China as the world’s largest contributor. Extended Producer Responsibility (EPR) has emerged as a key policy tool in China's effort to control the environmental and health impacts of e-waste. However, existing research on EPR tends to focus narrowly on government perspectives and policy design, overlooking how industries, associations, and researchers perceive and implement the system. This study addresses this gap by exploring the stakeholder dynamics within China’s formal e-waste management ecosystem. It hypothesizes that stakeholder perceptions diverge according to their roles, financial incentives, and policy understanding, influencing EPR’s effectiveness in …

Given global environmental challenges, developing sustainable behavior among pre-service teachers is crucial, as they will be responsible to educate future generations who must contribute to solving environmental problems. Offline pro-environmental identity refers as the individual's environmental commitment in real-life setting. Previous studies have primarily focused on online identity and behavioral intentions, while real-life behavioral context have not been fully examined. The goal of this study was to investigate the effects of the online pro-environmental identity, the perceived behavioral control, and the moral identity on the behavioral intention and the offline pro-environmental behavior. The study employed a quantitative research design and …
