Assessment Of Physiological & Biochemical Changes In Tomato Plants Treated With Plant Growth Promoting Rhizobacteria (Pgpr) Under Salinity Stress Conditions, Mohammed Sharif Alhosani

Theses

Plant growth-promoting rhizobacteria (PGPRs) can effectively mitigate the negative impacts of salt stress on plants. The present study reports the isolation of PGPRs from a high-saline mangrove ecosystem in Abu Dhabi, with isolates exhibiting ACC deaminase (ACCD) activity to reduce stress-induced ethylene, along with additional plant growth-promoting (PGP) characteristics, including phosphate solubilization (P+). A total of 57 salt-tolerant bacterial isolates from the mangrove rhizosphere were initially screened for ACCD and P+ activities. Of these, 24 isolates tested positive for PGP characteristics and were further evaluated for their ability to enhance salt stress tolerance in tomato seedlings. In addition, seedlings inoculated …

Evolution Of Mate Guarding Intensity In Crickets As A Possible Adaptation To Sexual Conflict Over Sperm Transfer, Bert Foquet, Jack Thomas Mckermitt, John Hunt, Ben M. Sadd, Scott K. Sakaluk

Faculty Publications – Biological Sciences

Mate guarding is a common behavioral adaptation in males to ensure mating or to hinder current partners from remating with rival males. In the decorated cricket, Gryllodes sigillatus, males transfer a spermatophore to females at mating, comprising a sperm-containing ampulla enveloped in a large gelatinous mass, the spermatophylax, that serves as a nuptial food gift for the female. Males guard the female while she feeds on the nuptial gift and while sperm are evacuated from the ampulla into her reproductive tract. Mate guarding behavior functions to prevent competitors from mating with the female but may also represent a behavioral …

Desert springs are one of the most threatened ecosystems globally. Those in Southwestern North America support high numbers of threatened and endemic species. Human activities present the greatest threats to springs and the organisms they support. Freshwater snails in the genera Pyrgulopsis and Tryonia are two groups of such organisms, representing diverse and threatened groups of springsnails in the Southwestern United States. Desert springs also harbour unique microbial diversity that faces similar threats from water usage and freshwater habitat degradation. Understanding springsnails and the bacteria they host represent potential exemplars for interdisciplinary conservation efforts. We studied the bacterial communities associated …

The Influence Of Historical Climate Cycles On The Diversification And Demography Of Species From The Neotropical Lizard Group Anolis Chrysolepis, Tony Gamble

Biological Sciences Faculty Research and Publications

Historical shifts in climate have significantly influenced the establishment of ecosystems and the evolution of species. Phylogeographical studies integrated with niche modelling can provide insights into divergence events and demography. This study explores the evolutionary history, demographic patterns, and spatiotemporal dynamics of seven South American lizard species of the Anolis chrysolepis species group, which has been proposed to be strongly impacted by Pleistocene climatic fluctuations under the Refugia Hypothesis. We utilized mitochondrial and nuclear DNA sequences to delineate lineages, estimate divergence times, and examine demographic dynamics. Ecological niche models were employed to assess historical range shifts and identify climatic stability …

As awareness of dementia increases, more individuals with minor cognitive complaints are requesting clinical assessment. Neuroimaging studies frequently identify incidental white matter hyperintensities, raising patient concerns about their brain health and future risk for dementia. Moreover, current US demographics indicate that ≈50% of these individuals will be from diverse backgrounds by 2060. Racial and ethnic minority populations bear a disproportionate burden of vascular risk factors magnifying dementia risk. Despite established associations between white matter hyperintensities and cognitive impairment, including dementia, no study has comprehensively and prospectively examined the impact of individual and combined magnetic resonance imaging measures of white matter …

Antigen-presenting cells phagocytose tumor cells and subsequently cross-present tumor-derived antigens. However, these processes are impeded by phagocytosis checkpoints and inefficient cytosolic transport of antigenic peptides from phagolysosomes. Here, using a microbial-inspired strategy, we engineered an antibody-toxin conjugate (ATC) that targets the 'don't eat me' signal CD47 linked to the bacterial toxin listeriolysin O from the intracellular bacterium Listeria monocytogenes via a cleavable linker (CD47-LLO). CD47-LLO promotes cancer cell phagocytosis by macrophages followed by LLO release and activation to form pores on phagolysosomal membranes that enhance antigen cross-presentation of tumor-derived peptides and activate cytosolic immune sensors. CD47-LLO treatment in vivo significantly …

Several reports have demonstrated that CD147, an N-glycosylated protein that is exchanged by cells in soluble form or through small extracellular vesicles (sEVs), can promote cancer progression. However, its activity related to EVs in colorectal cancer (CRC) is still not fully understood. Previously, we showed that sEV secretion during CRC stem cell (CR-CSCs) differentiation is partially controlled by CD147, and that CD147-expressing sEVs (sEVs-CD147) activate a signalling cascade in recipient cells, inducing molecular invasive features in CR-CSCs. In the present study, we demonstrated that sEVs-CD147 increase the expression of myofibroblast and activation markers in cancer-associated fibroblasts (CAF). In sEVs-CD147-activated CAF, …

Proteasome Augmentation Mitigates Age-Related Cognitive Decline In Mice, Danitra Parker, Kanisa Davidson, Pawel A Osmulski, Maria Gaczynska, Andrew M Pickering

Faculty, Staff and Student Publications

The aging brain experiences a significant decline in proteasome function. The proteasome is critical for many key neuronal functions including neuronal plasticity, and memory formation/retention. Treatment with proteasome inhibitors impairs these processes. Our study reveals a marked reduction in 20S and 26S proteasome activities in aged mice brains, including in the hippocampus, this is driven by reduced functionality of aged proteasome. The decline in proteasome activity is matched by a decline in 20S proteasome assembly. In contrast, 26S proteasome assembly was found to increase with age, though 26S proteasome activity was still found to decline. Our data suggests that age-related …

Kinesiology

Regular exercise is a practical non-pharmacological approach to maintaining physical and mental health through rehabilitation and prevention of chronic diseases due to its cardiovascular, cardiorespiratory, neurological, and neuromuscular benefits. Despite awareness of the benefits of exercise, a “lack of time” has proven to be the most common impediment to regular activity. Thus, a time-efficient and potentially enjoyable training modality with growing popularity is high-intensity interval training (HIIT). HIIT incorporates intermittent bouts of work and recovery intervals performed at an intensity close to maximal oxygen consumption (VO2max). HIIT is considered to have equivalent or superior benefits compared to moderate-intensity continuous training …

Conventional plug-in charging faces numerous challenges due to radiation hazards, inconvenience, etc. To mitigate the challenges conventional charging methods face, wireless power transfer (WPT) systems emerged as an efficient method for wireless energy transmission across various applications such as electric vehicles, biomedical implants, smartphones, and network sensors. A robust WPT system needs to perform exceptionally well under varying conditions, however, the WPT performance is reduced due to the leakage inductance between the coils. To address this challenge, compensation networks are often employed to minimize the losses and enhance the overall performance of the WPT system. This paper focuses on designing …

Protein Serine Kinase H1 (PSKH1) was recently identified as a crucial factor in kidney development and is overexpressed in prostate, lung, and kidney cancers. However, little is known about PSKH1 regulatory mechanisms, leading to its classification as a “dark” kinase. Here, we used biochemistry and mass spectrometry to define PSKH1’s consensus substrate motif, protein interactors, and how interactors, including Ca2+ sensor proteins, promote or suppress activity. Intriguingly, despite the absence of a canonical Calmodulin binding motif, Ca2+-Calmodulin activated PSKH1 while, in contrast, the ER-resident Ca2+ sensor of the Cab45, Reticulocalbin, Erc55, Calumenin (CREC) family, Reticulocalbin-3, suppressed PSKH1 catalytic activity. Brain tumors are commonly treated with radiotherapy, but the efficacy of the treatment is limited by its toxicity to the normal tissue including post-irradiation contrast enhanced lesions often linked to necrosis. The poorly understood mechanisms behind such brain lesions were studied using cerebral organoids. Here we show that irradiation of such organoids leads to dose-dependent growth retardation and formation of liquid-filled cavities but is not correlated with necrosis. Instead, the radiation-induced changes comprise of an enhancement of cortical hem markers, altered neuroepithelial stem cell differentiation, and an increase of ZO1+/AQP1+/CLDN3+-choroid plexus (CP)-like structures accompanied by an upregulation of IGF2 mRNA, …

The Great Lakes Entomologist

Pennycress is being developed in the Midwestern USA as a new oilseed crop that provides numerous ecosystem services. These services include being an early spring source of pollen and nectar for flower-visiting insects. Most past research on pennycress flower visitors used wild genotypes of pennycress. All wild genotypes have seed coats that are black, which occurs because of high levels of proanthocyanadins (PACs). Concentrations of PACs are minimal in newly developed genotypes with yellow-colored seed coats. However, little is known regarding flower abundance and timing nor insect visitation preferences for black-seeded genotypes (BSG) or yellow-seeded genotypes (YSG) of pennycress. Flower …

The twolined chestnut borer, Agrilus bilineatus (Weber), is a native North American buprestid beetle that normally infests oak (Quercus) trees stressed by drought or defoliation. However, during the 1980s and early 1990s, A. bilineatus preferentially infested English oak (Quercus robur L.) trees that initially appeared healthy at two experimental plantings in southern Michigan as well as on the Michigan State University (MSU) campus in East Lansing, Michigan. At both experimental plantings the English oaks were intermixed with native white oaks (Quercus alba L.) and had a border of native northern red oaks (Quercus rubra L.). …
