The determination of long non-coding RNA (lncRNA) function is a major challenge in RNA biology with applications to basic, translational, and medical research. We developed BigHorn to computationally infer lncRNA-DNA interactions that mediate transcription and chromatin-remodeling factor activity. Its accurate inference enabled the identification of lncRNAs that coordinately regulate both the transcriptional and post-transcriptional processing of their targets. These lncRNAs may act as molecular chaperones, regulating the stability and translation of mRNAs they helped transcribe, leading to tightly coupled expression profiles. Lysosomes are cytoplasmic organelles central for the degradation of macromolecules to maintain cellular homoeostasis and health. However, how lysosomal activity can be boosted to counteract ageing and ageing-related diseases remains elusive. Here we reveal that silencing specific vacuolar H+-ATPase subunits (for example, vha-6), which are essential for intestinal lumen acidification in Caenorhabditis elegans, extends lifespan by ~60%. This longevity phenotype can be explained by an adaptive transcriptional response typified by induction of a set of transcripts involved in lysosomal function and proteolysis, which we termed the lysosomal surveillance response (LySR). Metastasis is the predominant cause of cancer mortality, primarily driven by complex tumor-host interactions within specialized metastatic niches. Recent advances in single-cell technologies have provided unprecedented insights into metastatic niche formation, evolution and function, including how primary tumors precondition distant organs for metastases and how disseminated tumor cells dynamically interact with host cells to modulate their environments. Integrated single-cell studies across multiple cancer types have also revealed divergent and convergent metastatic adaptation strategies. These findings collectively highlight metastasis as a dynamic, cooperative process shaped by intricate tumor-host interactions, and provide a foundation for novel therapeutic strategies targeting components of the …

Synthetic biology has enabled the development of biosensors to detect intestinal inflammation, yet few target the clinically validated biomarker of intestinal inflammation calprotectin with both diagnostic and therapeutic capabilities. Here, an optimized calprotectin biosensor is presented that leverages a zinc uptake regulator (Zur) controlled promoter coupled with a memory circuit to detect and record intestinal inflammation in vivo. The level of biosensor activation strongly correlates with calprotectin levels in the colon of two independent mouse models of colitis. Introduction: Network hyperexcitability (NH) is observed in patients with early-stage Alzheimer's disease (AD), emerging decades before cognitive decline. A key molecular determinant of NH is voltage-gated Na+ channel 1.6 (Nav1.6), which mediates action potential firing in CA1 hippocampal neurons. Ameliorating NH through inhibition of the glycogen-synthase kinase 3β (GSK3β/Nav1.6 complex may provide immediate benefits to cognition and memory and slow AD progression.
Methods: Hight-throughput virtual screening and multiple in vitro biological assays were utilized to identify compound 1063. Patch-clamp electrophysiology and electroencephalogram recordings were utilized to functionally assess 1063 in models of AD neuropathology.

The impact of exogenous stressors, such as cancer chemotherapies, on the genomic integrity and clonal dynamics of normal hematopoiesis is not well defined. We conducted whole-genome sequencing on 1,276 single-cell-derived hematopoietic stem and progenitor cell (HSPC) colonies from ten patients with multiple myeloma treated with chemotherapies and six normal donors. Melphalan treatment significantly increased the mutational burden, producing a distinctive mutation signature, whereas other chemotherapeutic agents had minimal effects. Consequently, the clonal diversity and architecture of post-treatment HSPCs resemble those observed in normal elderly individuals, particularly through the progression of oligoclonal hematopoiesis, thereby suggesting that chemotherapy accelerates clonal aging. In 2025, the National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK) at the National Institutes of Health celebrates 75 years of leadership in diabetes research. The NIDDK serves people of the U.S. affected by or at risk for many chronic diseases, including diabetes and other endocrine, metabolic, and digestive disorders, by funding innovative research to develop better treatment and prevention and a cure for these conditions. Initial custodians of the National Forests of the United States were primarily concerned about protecting what were perceived as deteriorating timber stands and watersheds. Considerable progress was made in correcting these problems in the first half of this Century. As the population of the United States grew, more people became interested in management of all resources on these public lands. Fringed sagebruch (Artemisia frigida will) is one of the common plants in inner mongolia plateau. It was recognized as an excellent perennial herbaceous plants accunting for a large proportion in wild vegetation. Fringed sagebruch belongs to composites, Semi-frutex, tomentellate in whole plant, tap root may be 100 cm deep in the soil. Fringed sagebrush distributed to the north of the Yellow River in China, Fareast and Caucasus in USSR, Mongolia and Canada. Fringed sagebruch is an excellent forage for horses, cattle, sheep, goats and Camels. The animals will get gain if fringed sagebruch is fed.

In the continuing search for superior germplasm of tropical legumes adapted to Oxisol savanna conditions, a study of the perennial wild species of groundnut (peanut), Arachis pintoi, was initiated at Carimagua Research Station on the eastern plains (Llanos Orientales) of Colombia. A pintoiis an agronomically little-known legume pasture species indigenous to South America. Its performance in grazed association with four different Brachiaria spp. was evaluated in terms of dry matter yield and animal weight gain. Previous experience of test animals has been mentioned as a factor which may affect grazing preference of cattle. Palatability trials have usually been conducted without allowing animals to previously graze individual forage species under evaluation. A cafeteria trial was designed to study the effect of previous grazing of individual species in the palatability ranking of six tropical legumes. Results indicated that short-term previous experience of the test animals affected the magnitude of the palatability index of the most palatable legume on the first days of the trial, but did not affect the overall palatability ranking of species. Aeschynomene spp. have been identified as a promising group of legumes adapted to seasonally waterlogged soils on the central Queensland wet coast. Near Mackay (1470 mm M.A.R.) the legume component of Macroptilium atropurpureum cv. Siratro/Setaria sphacelala var. sericea cv. Kazungula pastures on adequately fertilized solodic soils, did not persist under heavy grazing (2.5 and 2.0 steers/ha), whereas the native legume Aeschynomene indica persisted and gave yields up to 1401 kg/ha. Climate in Louisiana is influenced by tropical air masses in summer and alternately by tropical and continental air masses in winter. From northwest to southeast, the number of frost-free growing days increases from 230 to 350. Rainfall increase from 1200 mm to 1500 mm and is evenly distributed winter through summer with a generally drier fall. Livestock production is an important economic activity throughout the state. Seagrasses are a valuable component of global blue carbon ecosystems yet are also endangered due to natural and man-made pressures. This study assessed the seagrass resources regarding species community structure and diversity using a transect-quadrat design along the intertidal coastlines of selected towns of Surigao del Sur. Of the 19 recorded species in the Philippines, seven common species were observed: Cymodocea rotundata, Enhalus acoroides, Halodule pinifolia, Halodule uninervis, Thalassia hemprichii, Syringodium isoetifolium, and Halophila ovalis. Shark and ray species are both limited in number and distribution and require time to mature and reproduce, making these species extremely vulnerable to mass exploitation as a resource. Based on the identification results, Muncar Port Banyuwangi had 26 species from 490 ray samples, and Brondong Port Lamongan had 28 species from 1830 ray samples. Fifteen species of rays were found only in Brondong, 17 species of rays were found only in Muncar, and as many as 13 were found in the two sample sites. This study aims to analyze students' perceptions of the implementation of science laboratory practicums at public middle school 020 Pekanbaru and public middle school 7 Pangkalan Kuras, Riau Province. Using a quantitative survey method, this study involved 75 student respondents. The instrument used was a questionnaire consisting of 26 statements, analyzed descriptively. The results show that students' perceptions of the preparation, implementation, and conclusion stages of the practicum fall into the "good" category, with urban schools having a higher percentage. This study aims to develop digital comic learning media that is effective in increasing students' interest in learning about reaction rate material, focusing on the media's validity, practicality, and overall effectiveness. This research utilizes the research and development method with the ADDIE model. Research instruments were used, including validity sheets to assess the media's validity, student activity observations, and student response questionnaires to evaluate the media's practicality, as well as student interest questionnaires and learning outcomes to measure the effectiveness of the learning media. Regulatory toxicology depends primarily on animal testing, which limits throughput, mechanistic interpretability, and applicability to human-relevant endpoints. This dissertation presents computational approaches that integrate publicly available bioactivity data with machine learning to support chemical safety evaluation. Oral carcinogenicity was modeled using curated chemicals from the US Environmental Protection Agency’s Integrated Risk Information System (IRIS) and activity data from over 1,900 PubChem assays. Quantitative structure-activity relationship (QSAR) models trained on statistically prioritized assays demonstrated strong predictive performance and were applied to rank the carcinogenic potential of more than 16,000 compounds. Amylases are one of the important enzymes with tremendous biotechnological applications. In this study, the amylase enzyme producing bacterial strains associated with the soft coral Sarcophyton trocheliophorum was isolated and identified. The amylases were produced in a specific growth medium and the stability was tested under different pH and temperature conditions. The crude amylases were tested for the antibiofilm activity against two biofilm-forming bacterial strains. The amylase enzyme extracted from two bacterial strains (KAU-AS1 and KAU-AS3) showed good activity in the antibiofilm activity assays. This study produced biodiesel from Malapari seed oil (Pongamia pinnata L.) by optimizing the esterification and transesterification processes. Background: Body mass index (BMI) has a complex association with a variety of chronic pain conditions, which may be influenced by sleep apnea syndrome (SAS). However, the role of SAS within the causal pathway linking BMI to chronic pain remains unconfirmed. Organelle dynamics lie at the heart of cellular signaling and metabolic regulation, orchestrating physiological processes such as reproduction and aging across the lifespan. In this dissertation, I investigate how organelle-based signaling coordinates sperm activation and modulates organismal aging in the nematode Caenorhabditis elegans. I identify SPIN-4, a previously uncharacterized Spinster family transporter, as a key regulator of spermiogenesis. SPIN-4 is strongly expressed in developing sperm, localizes to the plasma membrane, and promotes sperm activation by facilitating the export of sphingosine-1-phosphate (S1P). Loss of spin-4 results in defective pseudopod formation, reduced motility, and compromised fertility, phenotypes that can be rescued …

Background: Vitamin D3 over-supplementation can induce toxicity through the development of hypercalcemia and may disturb immune modulation. This study examines the relationship between hypercalcemia resulting from Vitamin D3 Overdose and serum Interleukin-6 (IL-6) levels in patients diagnosed with staphylococcal arthritis.

Ungulate browsing represents a contemporary issue for forest development, influencing forest regeneration, composition, and management practices across the world, especially in North America. This review synthesizes findings from 101 studies conducted between 1980 and 2025 to examine patterns of herbivory damage through browsing by moose, elk, white-tailed deer, black-tailed deer, and bison. Despite regional variation, high ungulate density consistently emerges as the primary factor driving browsing intensity and ecological and economic impact, leading to decreased social acceptance of coexistence with ungulates.
