The gut microbiome has emerged as a key regulator of response to cancer immunotherapy. However, a better understanding of the underlying mechanisms by which the microbiome influences immunotherapy is needed to identify strategies to optimize outcomes. To this end, we developed a mathematical model to obtain insights into the effect of the microbiome on the immune system and immunotherapy response. This model was based on (i) gut microbiome data derived from preclinical studies, (ii) mathematical modeling of the antitumor immune response, (iii) association analysis of microbiome profiles with model-predicted immune profiles, and (iv) statistical models that correlate model parameters with …

Esophageal adenocarcinoma is increasingly prevalent and is thought to arise from Barrett's esophagus (BE), a metaplastic condition in which chronic acid and bile reflux transforms the esophageal squamous epithelium into a gastric-intestinal glandular mucosa. The molecular determinants driving this metaplasia are poorly understood. We developed a human BE organoid biobank that recapitulates BE's molecular heterogeneity. Bulk and single-cell transcriptomics, supported by patient tissue analysis, revealed that BE differentiation reflects a balance between SOX2 (foregut/esophageal) and CDX2 (hindgut/intestinal) transcription factors. Melanoma, the most lethal form of skin cancer, is commonly associated with mutations in the BRAF gene, particularly BRAFV600E, which drives tumor proliferation via the ERK1/2 signaling cascade. While BRAF inhibitors initially demonstrate efficacy, therapeutic resistance remains a significant challenge. Emerging evidence implicates the cAMP signaling pathway, particularly the cAMP response element-binding protein (CREB) and its repressor, inducible cAMP early repressor (ICER), in melanoma progression and drug resistance. ICER, a transcriptional repressor regulated via Ras/MAPK-mediated phosphorylation and ubiquitination, is degraded in melanoma, undermining its tumor-suppressive role. This study aimed to isolate and identify bacteria from ear swab samples from cats with otitis externa and to detect the antimicrobial resistance profiles of the recovered isolates. For this purpose, ear swab samples from 100 domestic cats that were detected to have otitis externa were used. The samples collected from the animals were inoculated onto 7% defibrinated sheep blood agar and MacConkey agar, and the media were incubated at 37°C for 24-72h under aerobic/microaerobic conditions. Two additives, rosemary oil and a commercial herbal product (HP; AROTEC-G) containing garlic essential oil, carvacrol and thymol were investigated for their potential to reduce the parasitic burden in yellowtail kingfish (YTK; Seriola lalandi) infected with Zeuxapta seriolae. Naturally infected fish were fed diets supplemented with either 2.5 mL.kg−1 rosemary oil or two concentrations of HP (5 and 10 g.kg−1) for 20 days. At 10 and 20 days, the effect of the diets on parasite abundance was evaluated. Feed intake and growth were similar across treatments (p > 0.05). By day 10, both rosemary oil and HP significantly …

Theory and past experimental work suggest that as males age, the strength of their mate preference should decrease. However, the empirical work investigating this question has primarily been conducted in insects that have very short life spans and often live for just a single mating season. This leaves a gap in our understanding of the relationship between male mate preference and age across taxa, as age can conflate with other ecological changes in a single mating season. In this study, the relationship between the Growth Differentiation Factor-9 (GDF9) gene polymorphism and birth weight, as well as weights on days 30, 60, 90, was investigated in Akkaraman lambs, one of the indigenous sheep breeds of Türkiye. For this purpose, a total of 314 lambs were genotyped by PCR-RFLP method for the single nucleotide polymorphism (SNP) c.260G>A in the GDF9 gene. While GG and GA genotypes were observed in the studied breed, the AA genotype was not observed. The results showed that Akkaraman lambs were in Hardy–Weinberg (H–W) equilibrium for the GDF9-c.260G>A polymorphism (p < 0.05). In this study, a significant association was observed between GDF9-HhaI polymorphism and 30th day weight and 60th day weight in Akkaraman lambs. When the animals were analyzed according to birth type, it was observed that this factor was associated with birth weight, as well as weights on days 30, 60, and 90. Optimizing duck production requires understanding growth variability and monitoring serum calcium and magnesium levelsas health indicators. This study aimed to analyze the growth curve, mortality, and serum calcium (Ca) and magnesium (Mg) levels in White Mallard ducks from hatching to the prelaying period in subtropical conditions. A total of 1100 females and 270 males were raised for 146 days. At hatch, day 6 (D6), D13, D23, D34, D50, D64, D80, D94, D111, and D146, ducks were weighed separately to assess body weight (BW). Avians use significant amounts of energy during flight that is supplied by muscles. The Common Pheasant, a member of the Phasianidae family, can fly swiftly over short distances and flies to evade predators. Pheasant species are terrestrial and have a global distribution. This investigation aimed to analyze the anatomical structure of skeletal muscles in pheasants. A cohort of 20 adult pheasants consisting of an equal number of females and males (10 each) was used. The phylum Acanthocephala contains approximately 1300 acanthocephalan species. So far, only nine acanthocephalan species of wild birds have been reported in Türkiye. To study the context of acanthocephalan diversity in wild birds in Türkiye, we examined 193 birds from 69 species belonging to 16 orders. Acanthocephalans were found in seven bird species: Centrorhynchus aluconis in Buteo buteo and Buteo rufinus, Centrorhynchus spinosus in Buteo buteo and Strix aluco, Mediorhynchus papillosus in Iduna icterina, Mediorhynchus tenuis in Luscinia svecica, Plagiorhynchus cylindraceus in Erithacus rubecula and Turdus merula, Polymorphus contortus in Anas platyrhynchos, and Sphaerirostris turdi in Turdus merula. Diffuse mesothelioma (DM) is a rare but highly aggressive and treatment resistant neoplasm with low survival rates. Effective therapeutic strategies are limited, and resistance to treatment is a major obstacle. Myeloid Cell Leukemia (MCL)-1 and B-cell leukemia (BCL)-xL are anti-apoptotic B-cell lymphoma 2 (Bcl-2) family proteins that block cell-intrinsic apoptosis through interactions on the mitochondrial outer membrane which contribute to therapeutic resistance.

The ribosome is the central hub for protein synthesis and is heavily targeted by antibiotics. Ribosomal mutations, antibiotic treatment, and nutrient starvation can alter translational efficiency and lead to stressed cells. Ribosome deficiency plays a critical role in stress responses and disease progression; yet, how it affects bacteria-host interactions remains poorly understood. In this study, we show that a ribosome-deficient strain exhibits a surprising morphological change from rod shape to filamentous in Salmonella cells growing inside host macrophages. Such filamentation depends on an acidic condition within macrophages and in a defined medium mimicking macrophage conditions. Further genetic analyses revealed that …

RET solvent-front G810C/R/S mutations confer resistance to the currently approved RET protein tyrosine kinase inhibitors (TKIs) selpercatinib and pralsetinib. Moreover, RET fusion-positive lung adenocarcinoma frequently metastasizes to the brain. Lighting influences broiler production, health, and behavior. The objective of this study was to examine the effects of three light wavelengths (White [350-780 nm], Blue [450 nm], and Green [560 nm]) on broiler production, activity, fear, stress, blood cell profiles, and tibiotarsi (tibia) morphology. Day-of-hatch male broiler chicks (N = 600) were housed in pens (N = 12) with one lighting treatment for 42 days. Body weight and feed consumption were recorded on day (D) 0, 14, 28, and 42, and the feed conversion ratio was calculated. During a natural history hike at La Selva Biological Station in Costa Rica, we observed a Brown Vine Snake, Oxybelis koehleri, consuming the autotomized tail of a Common Humble Anole, Anolis humilis, which had escaped predation. This event represents a rare documented instance of a vine snake feeding exclusively on a detached lizard tail. Tail autotomy is a widespread anti-predator strategy among lizards, with the shed tail continuing to writhe post-detachment, potentially attracting predators. Using a combination of site visits, document reviews, focus group and key informant surveys of fishermen, we explore the cultural beliefs, taboos and norms that protect Musambwa Islands, Uganda. The islands form part of a large protected wetland with nesting habitat for thousands of water birds and a large population of highly-venomous forest cobras that coexist in and around fishermen dwellings. There are strong taboos against cutting trees, allowing women on the island, and harming cobras. Emery-Dreifuss muscular dystrophy 1 (EDMD1) arises from mutations in EMD. Most EDMD1 patients lack detectable emerin expression. They experience symptoms such as skeletal muscle wasting, joint contractures, and cardiac conduction defects. Currently, physicians rely on treating patient symptoms without addressing the underlying cause-lack of functional emerin protein. Thus, there is a need for therapeutic approaches that restore emerin protein expression to improve patient outcomes. One way would be to deliver emerin mRNA or protein directly to affected tissues to restore tissue homeostasis. Tetralogy of Fallot (TOF) is the most common cyanotic congenital heart defect (CHD). TOF may present in isolation or in conjunction with one or more non-cardiac congenital anomalies or neurodevelopmental disorders (TOF+). Uncertainty regarding the efficacy of various genetic testing strategies, and an incomplete understanding of the genetic causes of TOF+, may lead to hesitancy in recommending genetic testing, particularly, clinical exome sequencing (cES). Here, we analyzed cES data from 131 individuals with TOF+. A definitive or probable diagnosis was made for 31 individuals, yielding a diagnostic rate of 23.6% (31/131). Nest-building in social insects requires layered communication to coordinate complex, functional structures for housing, protection, storage, and homeostasis. This is especially striking in mound-building termites, which construct and repair large mounds collectively despite lacking image-forming eyes. This thesis investigates how Odontotermes obesus achieves such coordination through chemical communication embedded in soil.
Using controlled two-choice assays with manipulated soils, I separated airborne volatile signals from contact non-volatile signals by baking to remove volatiles, a mesh assay that presented volatile cues without contact, and solvent extraction with reapplication of contact cues. Air pollution, particularly exposure to fine particulate matter (PM2.5), poses a growing threat to cardiopulmonary health. PM2.5 consists of microscopic particles smaller than 2.5 microns in diameter and originates from industrial combustion, transportation, and urban dust. These particles are classified as xenobiotics and are capable of bypassing respiratory defenses to enter systemic circulation, where they cause oxidative stress, inflammation, and cardiovascular dysfunction. One-third of the world’s freshwater crayfish species face extinction risk, necessitating extreme and urgent conservation measures such as captive rearing. Little information exists on these measures for most threatened species. The hairy marron, Cherax tenuimanus (Parastacidae), is a critically endangered freshwater crayfish endemic to the Margaret River in south-western Western Australia. C. tenuimanus is being rapidly replaced by the smooth marron, Cherax cainii, a translocated congener, causing its progressive decline in range, and now it faces imminent extinction in the wild. Purpose: Hippocampal avoidance (HA) during therapeutic whole-brain radiotherapy reduces the risk of neurocognitive function (NCF) toxicity in patients with brain metastasis. This trial hypothesized that HA during prophylactic cranial irradiation (PCI) in patients with small cell lung cancer (SCLC) leads to noninferior intracranial relapse (ICR) and reduction in NCF toxicity.
Methods: This randomized phase II/III trial enrolled patients with SCLC, no brain metastases, and response to chemotherapy. The primary end points were 12-month ICR (noninferiority design, randomized phase II) and 6-month Hopkins Verbal Learning Test-Revised (HVLT-R) Delayed Recall (DR) failure (phase III). KRAS mutations frequently co-occur with alterations in STK11/LKB1 and/or KEAP1, defining an aggressive subset of lung cancers resistant to immuno- and chemotherapy. While LKB1 loss is associated with vulnerability to DNA damage response-based therapies, the impact of KEAP1 alterations remains unknown. We demonstrate that KEAP1-NRF2 pathway drives a compensatory modulation of ATR-CHK1 signaling, enhancing vulnerability to ATR inhibitors (ATRi), particularly in the setting of increased replication stress associated with LKB1 loss. ATRi shows enhanced anti-tumor activity in LKB1 and/or KEAP1-deficient non-small cell lung cancer (NSCLC) models and synergizes with gemcitabine. Seven jurisdictions from around the world have issued judicial opinions that address fundamental issues about the governance and regulatory systems of agricultural biotechnology. This article summarizes these legal proceedings and describes their impact upon agricultural biotechnology. The article then provides a commentary and critique of the legal proceedings and resulting judicial opinions.
