The present study analyses the influence of various parameters on the accuracy and reliability of ROH identification procedures, as well as their relationship with inbreeding and genetic diversity in Duroc pig populations. The genotyping data of 70 Duroc pigs obtained using PorcineSNP80v1 chips were used, and the effects of LD pruning and ROH calling parameters were evaluated. It was found that the use of strict basic filters during the execution of the --homozyg command reduces the likelihood of false positives and contributes to the more accurate identification of ROH. In veterinary medicine, various diseases and surgical procedures may require the isolation and hospitalization of dogs, allowing for more effective individual patient monitoring and increasing the chance of successful treatment by preventing unnecessary drug use. However, this process can be stressful. Our study aimed to determine the changes in cortisol levels, one of the primary stress hormones responsible for stress responses, in individually hospitalized dogs. Bacterial Chondronecrosis with Osteomyelitis (BCO) lameness is an infection of opportunistic bacteria in the structural skeletal bones impacting multiple animal species, particularly poultry species. BCO lameness results in significant financial losses to industrial poultry production and increases the risk of foodborne illnesses, posing a major threat to consumers' food safety. As BCO lameness is an inherent risk of fast body weight gain in poultry species, especially broiler chickens, abundant studies have been conducted in broilers and turkeys. This study assessed the feasibility of integrating two pre-existing aquaculture activities in Akaroa Harbour, Aotearoa New Zealand into an integrated multi-trophic aquaculture (IMTA) system. The cultivation of giant kelp (Macrocystis pyrifera) presents an opportunity to improve the sustainability of Chinook salmon (Oncorhynchus tshawytscha) farming by introducing alternative food sources for salmon, which can be used to produce fish feed locally, enhancing the self-sufficiency of the community and boosting the holistic efficiency of the system. Selective autophagy of mitochondria is known to promote cancer cell survival and progression, including in triple-negative breast cancer (TNBC). Here, we apply an integrated multi-omics approach together with functional experimental analyses to investigate metabolic adaptations that support mitochondrial quality control in TNBC. We detail a mitochondrial quality control mechanism, complementary to mitophagy, that is enabled by a program of heightened extracellular sphingomyelin salvaging in TNBC coupled with extracellular vesicle-mediated intracellular clearance of mitochondrial damage. Treatment of lung adenocarcinomas (LUADs) that exhibit activated epidermal growth factor receptor (EGFR) with EGFR tyrosine kinase inhibitors (TKIs) has limited efficacy. Assessment of the impact of EGFR TKI on the LUAD surfaceome remodeling reveals potential therapeutic targets. We identify placental type alkaline phosphatase (ALPP), which has restricted expression in normal tissues, among upregulated surface proteins following EGFR TKI treatment of both TKI sensitive as well as resistant cells. EGF treatment represses ALPP expression, whereas EGFR TKIs upregulate its expression through dephosphorylation and activation of FoxO3a, a transcriptional regulator that binds to the promoter region of ALPP. The importance of this study is to perform antibody discovery as a means to derive novel preventative and therapeutic antibody countermeasures against the Junín virus, to alleviate the potential risk of a global pandemic. Lung cancer is the leading cause of cancer-related deaths worldwide, driven by carcinogens such as tobacco-derived nitrosamines, dioxins, and urethane. This study evaluated the chemopreventive effects of two monoterpenes, α-pinene and D-limonene, using NNK/TCDD- and urethane-induced lung tumor models and A549 lung cancer cells. Both compounds significantly reduce tumor number and size, with enhanced effects when combined. Mechanistic studies revealed suppression of PRC1, β-catenin, and c-Myc, alongside activation of p53, Bax, and caspase-3, indicating inhibition of the PRC1—Wnt/β-catenin pathway and induction of apoptosis. These findings suggest α-pinene and D-limonene as safe, promising agents for lung cancer chemoprevention.

Excessive fructose intake leads to fatty liver and kidney dysfunction, with associated inflammation and metabolic disturbances. This study evaluated the effects of fish oil rich in ω-3 fatty acid on these conditions in rats fed a high- fructose diet. Male Sprague Dawley rats were divided into three groups: normal diet, high-fructose (HF) diet, and high- fructose diet with 5% fish oil supplementation. The experiment lasted for 21 weeks. Fish oil supplementation significantly reduced blood levels of total cholesterol, triglycerides, and advanced glycation end products (AGEs) in HF-fed rat. Rapid quantitative detection methods enable producers to identify contamination early, implement corrective actions, and enhance food safety. Neuroinflammation has emerged as a critical modulator of cognitive function and neural circuit dynamics, yet the distinct contributions of acute and chronic neuroinflammation to brain function remain poorly defined. This dissertation examines how inflammation independently alters large-scale neural activity and behavior, focusing on interactions between the hippocampus (HPC) and anterior cingulate cortex (ACC), regions essential for memory and executive processes. Homeostasis, the process of maintaining stable biochemical and biophysical conditions, is a fundamental facet of cellular viability. Biochemical homeostasis is critical to support the biological functions that sustain living organisms. Protein ubiquitylation is one of the primary cellular pathways that regulate protein homeostasis and turnover. Ubiquitylation is initiated through the conjugation of a small globular protein known as ubiquitin, onto a given substrate. This research explores early college science majors’ conceptions of the Mojave Desert ecosystem as a complex system. A multi-level, explanatory sequential mixed methods design was employed to study college science majors’ understanding of the Mojave Desert ecosystem to better inform science pedagogy. The first level, the quantitative phase of the study, consisted of developing a mathematical representation of twelve ecosystem variables present within the Mojave Desert. The second level, the qualitative phase of the study, consisted of phenomenological semi-structured interviews with 23 early college science majors. Global demand for animal protein is increasing rapidly, underscoring the need for sustainable meat production. Cultured meat is a promising alternative to traditional meat production techniques, but it requires optimization before it can be implemented on a large scale. Scaffolds are an important component for making cultured meat, as they provide structural support for cell attachment and mimic the texture of meat. Plant preservation, whether through disease research or endemic extinction analyses, plays an important role in the relationship between humans and our surrounding environment. In these analyses, both the role of mobile elements in the evolution of pathogenic Xanthomonas sp. bacteria, and the demographic and reproductive analysis of Castilleja parvula, an imperiled alpine plant, are evaluated. Xanthomonas species are one of the top agricultural threats, and are known to evolve rapidly. Operant conditioning is a form of learning in which a behavior is reinforced by reward. Operant conditioning has multiple temporal domains, ranging from short-term, lasting a few minutes, to long-term, persisting for at least 24 h. The extent to which short- and long-term operant conditioning memories rely on shared or separate neural mechanisms is poorly understood. Voltage-sensitive dye (VSD) imaging has been used previously to record the activity of a large number of neurons simultaneously in the buccal ganglion to measure changes in neuronal activity during short-term operant conditioning. Spring-fed aquatic ecosystems of the North American Southwest and U.S. Great Basin hosts a plethora of endemic fish species due to the effects of hydrologic fragmentation that resulted from Pleistocene aridification. Threats to endemic fishes worldwide include human intentional or unintentional release of exotic species into their habitats. Early detection of invasive species, while corrective actions are still possible, is critical for management, yet traditional survey methods are often disruptive to sensitive ecosystems, costly, and limited in sensitivity. Triple-negative breast cancer (TNBC), being both aggressive and highly lethal, poses a major clinical challenge in terms of treatment. Its heterogeneity and lack of hormone receptors or HER2 expression further restrict the availability of targeted therapy. Breast cancer stem cells (BCSCs), known to fuel TNBC malignancy, are now being exploited as a vulnerability for TNBC treatment. Here, we dissected the transcriptome of BCSCs and identified kinesin family member 20A (KIF20A) as a key regulator of BCSC survival and TNBC tumorigenesis. Genetic depletion or pharmacological inhibition of KIF20A impairs BCSC viability and tumor initiation and development in vitro and in vivo. …
