Nerves can regulate tumorigenesis and cancer progression. However, clarification of the role of axon guidance molecules in tumorigenesis, innervation, and metastasis is required to better understand the tumor-promoting functions of nerves. Using murine KrasG12D-mutant pancreatic organoids, we screened axon guidance molecules and identified netrin-1 upregulation. Netrin-1 was also upregulated in vivo during pancreatic tumorigenesis in humans and mice. Mutant KRAS and β-adrenergic signaling upregulated netrin-1 and its receptor NEO1 in epithelial cells in part through the MAPK pathway. Ex vivo culture of celiac ganglia showed that netrin-1 promoted the axonogenesis of sympathetic neurons through nerve NEO1. In the Pdx1-Cre;LSL-KrasG12D/+ model, …

Arts & Sciences Graduate Student Theses and Dissertations
Introduction: Alzheimer’s Disease (AD) remains among the top 10 causes of death in the United States and is projected to affect nearly 14 million people by 2050. To this point, studies of Alzheimer’s genetics and endophenotypes have yielded a bounty of results that culminated in the first disease-modifying drugs, lecanemab and donanemab, in 2022. Most of the genetic studies for Alzheimer’s disease to date have been focused on late-onset AD or Autosomal dominant AD, leaving a critical need for studying sporadic early-onset AD (EOAD; ~10% of cases). Furthermore, the demand for clinical portability has created a need for high-sensitivity proteomic …

Background: The circadian clock is an evolutionarily conserved system that orchestrates 24-h physiological rhythms through transcriptional and translational feedback loops. Mounting evidence suggests a bidirectional relationship between circadian rhythm alteration and disease progression, positioning the circadian clock as a potential therapeutic target. Due to the scarcity of high-resolution temporal omics data, it remains very challenging to elucidate the underlying regulatory mechanisms of the circadian system. As a practical alternative, public untimed transcriptomic datasets offer the potential to infer gene expression oscillations retrospectively. However, existing computational approaches for circadian phase estimation often suffer from limited predictive accuracy, reducing their ability to …

Growing evidence suggests that psychiatric disorders are characterized by a prolonged inflammatory state, which may influence the efficacy of compounds targeting serotonin. Serotonin is a key signaling molecule in neuroplasticity of the adult hippocampus and involved in antidepressant action. Recent in vitro studies indicate the neurotransmitter may also facilitate the response to inflammation and potentially modulate microglial function towards neuroprotection. Using Tph2−/− rats depleted of brain serotonin, we examined microglial expression of various serotonin receptors (5-HTRs) in vivo in both the hippocampus and prefrontal cortex and assessed mRNA levels of cytokines and brain-derived neurotrophic factor (BDNF). We observed age-dependent and …

Effects Of Different Mixing Ratios Of Common Vetch And Oats On Forage Yield And Quality, Meli̇h Okcu, Sedat Severoğlu, İbrahi̇m Kavuncu
Turkish Journal of Agriculture and Forestry
This study investigates the optimal mixing ratios of common vetch (Vicia sativa L., CV) and oats (Avena sativa L., O) for enhanced forage yield and quality in the ecological conditions of Erzurum Province. The research was conducted over two growing seasons with three replications in a randomized complete block design (RCBD). Specifically, three common vetch cultivars (Gülhan, Albayrak, Kubilay) and one oat cultivar (Kupa) were evaluated by sowing as sole crops and in different ratios (25% vetch + 75% oat, 50% vetch + 50% oat, 75% vetch + 25% oat). The analyzed parameters included hay forage yield (HFY), dry matter …
Optimizing Cultivated Land Productivity Through Integrated Fertilization Strategies And Uav-Assisted Management, Kejun Wang
Turkish Journal of Agriculture and Forestry
This study investigates the impact of long-term fertilization modes on crop health and productivity using unmanned aerial vehicle (UAV)-assisted monitoring and a Q-learning-based approach for optimized fertilization strategies. An approximate state-matching Q-learning algorithm was developed to improve the efficiency of agricultural UAVs in fertilization tasks. The spatiotemporal patterns of cultivated land productivity under different fertilization regimes were analyzed in a long-term experiment. An evaluation system for cultivated land health productivity was established. The results indicated that the Q-learning algorithm enabled UAVs to achieve efficient task completion. QLASM outperformed traditional methods on all metrics. QLASM achieved a 92% task completion rate …

Turkish Journal of Biology
Background/aim: The present study investigates the role of Shenfu injection in the treatment of yang-deficient chronic heart failure (CHF). Materials and methods: Sprague-Dawley (SD) rats were modeled for yang-deficient CHF by abdominal aortic coarctation. Echocardiography was performed to detect changes in cardiac function, and serum N-terminal B-type natriuretic peptide proteins (NT-proBNP), cardiac troponin I (cTnI), and ferroptosis-related factors were measured using ELISA kits. Pathological changes in cardiac tissues were observed through hematoxylin-eosin (HE) and Masson’ trichrome staining, cardiomyocyte apoptosis was measured by TUNEL staining, and reactive oxygen species (ROS) production was determined through dihydroethidium (DHE) staining. The expression of nuclear …

Mir-22 Inhibits The Proliferation, Migration, And Invasion Of Human Cd133-Positive Glioblastoma Stem Cells, Sevi̇l Köse, Özlenen Özkan, Ömer Özkan
Turkish Journal of Biology
Background/aim: Glioblastoma multiforme (GBM) is one of the most aggressive and fatal malignancies of the central nervous system. Despite advancements in treatment strategies, effective therapies for GBM remain insufficient, necessitating further improvements. Notably, miR-22 has been found to be significantly downregulated in both glioblastoma tissues and cell lines. In this study, we aim to evaluate miR-22 expression levels in GBM (U87) and CD133-positive (CD133+) GBM stem cells (GSCs) and to investigate its effects on proliferation, colony formation, migration, invasion, and wound-healing in U87 and CD133+ U87 cells in vitro.Materials and methods: We isolated CD133+ U87 cells using magnetic-activated cell sorting …

Efflux Pump-Associated Antimicrobial Resistance Genes In Staphylococcus Spp. From Dairy And Meat Samples, M.Burcu Külahci, Sumru Çitak
Turkish Journal of Biology
Background/aim: This study aimed to investigate the phenotypic resistance and distribution of efflux pump-associated antimicrobial resistance genes in Staphylococcus spp. isolated from dairy and meat samples. Antimicrobial resistance in foodborne bacteria increases with antibiotic exposure and biocides, particularly through efflux mechanisms. Thus, monitoring potential genetic reservoirs in the food chain is very important. Materials and methods: A total of 132 dairy and meat samples were collected for the study, and Staphylococcus spp. were isolated using Mannitol salt phenol red agar. Antimicrobial susceptibility was evaluated using the Clinical and Laboratory Standards Institute’s microdilution method. Twenty-six resistant isolates were identified by 16S …
