Hepatocellular Neoplasm In A Wild-Caught Sheepshead Minnow (Cyprinodon Variegatus) From The Northern Gulf Of Mexico, Marcos F.T. Oliveira, William E. Hawkins, Robin M. Overstreet, William W. Walker

Harold W. Manter Laboratory of Parasitology: Faculty Publications

The sheepshead minnow, Cyprinodon variegatus, is a widely distributed small fish species that inhabits estuaries and inshore waters from New England to northern South America. This species has been used extensively in aquatic environmental toxicity and carcinogenicity tests (Couch et al., 1981; Courtney and Couch, 1984). Hepatic neoplasms have been induced in the sheepshead minnow by exposure to several known chemical carcinogens including diethylnitrosamine (Couch and Courtney, 1987), methylazoxymethanolacetate (Hawkins etal., 1985), and dimethylbenz(a)anthracene (Hawkins et al., 1991). Because of its widespread distribution, limited home range, and proven sensitivity to carcinogens, the sheepshead minnow appears to be …

Nitric Oxide Production: A Mechanism For Inhibition Of Chlamydia Trachomatis Replication, Bojun Chen Dec 1993

Nitric Oxide Production: A Mechanism For Inhibition Of Chlamydia Trachomatis Replication, Bojun Chen

Electronic Theses and Dissertations

Chlamydia trachomatis (CT) replicates in macrophages, but is inhibited by IFN-$\gamma$ or LPS. IFN-$\gamma$ and/or LPS induced nitrite production in mouse peritoneal macrophages, macrophage cell lines (RAW264.7 and J774A.1) and McCoy cells. Kinetic studies indicated that peak production occurred 48 hours post-treatment. CT infection itself was insufficient to induce nitrite production, but resulted in enhancement of nitrite production in IFN-$\gamma$-treated cells. Treatment with IFN-$\gamma$ or LPS resulted in significant inhibition of CT replication in these cells. Strong correlation between nitrite production and inhibition of CT replication was observed in RAW264.7 and J774A.1 cells (correlation coefficients: $-$0.93 and $-$0.94, p $<$ 0.001). N$\sp{\rm g}$- monomethyl-L-arginine (L-NMMA) specifically inhibited nitrite production and partially reversed inhibition of CT replication in macrophage cell lines. NOS mRNA was measured in RAW264.7 cells by Northern blot and Dot blot hybridization. Strong correlation between NOS mRNA expression and inhibition of CT replication (correlation coefficient: $-$0.97, p $<$ 0.05) was observed. Anti-TNF-$\alpha$ antibody completely neutralized the biological activity of TNF-$\alpha$ secreted by LPS-treated RAW264.7 cells, yet the antibody neither reduced nitrite production nor restored CT replication. Combination of the antibody and L-NMMA significantly enhanced restoration of CT replication. In peritoneal macrophages, inhibition of CT replication induced by IFN-$\gamma$ was partially restored by L-NMMA or anti-TNF-$\alpha$ antibody. In McCoy cells, inhibition of CT replication induced by IFN-$\gamma$ and LPS was not significantly restored by L-NMMA. Great restoration of CT replication by 1 mM L-NMMA was observed in LPS-treated J774A.1 cells (31%), but not in IFN-$\gamma$-treated cells (5%). Our data indicate that (1) NO production is one of the mechanisms for inhibition of CT replication in IFN-$\gamma$-activated peritoneal macrophages and RAW264.7 cells; (2) NO plays a significant role in CT inhibition in LPS-treated macrophage cell lines, but not peritoneal macrophages; (3) TNF-$\alpha$ may be associated with inhibition, but the mechanism(s) may not involve NO production; (4) NO production may not be the mechanism for CT inhibition in McCoy cells treated with IFN-$\gamma$ and LPS.

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Two cystidicolid nematodes, Ctenascarophis lesteri n. sp. and Prospinitectus exiguus n. sp., are described from the skipjack tuna, Katsuwonus pelamis, from Fiji, New Caledonia, Marquesas Islands, New Zealand, Papua New Guinea, and Puerto Rico. The former species differs from Ctenascarophis gastricus, the only other member of the genus, by having comb rows extending farther posterior than two-thirds the body length, to the anus in the female; it also has differently distributed spines on each comb, with a maximum of 32 rather than 8. The male has 7 rows of precloacal longitudinal crests; 3 pairs of precloacal, 1 pair …

Comparative Study Of Adaptive Radiations With An Example Using Parasitic Flatworms (Platyhelminthes: Cercomeria), Daniel R. Brooks, Deborah A. Mclennan Nov 1993

Comparative Study Of Adaptive Radiations With An Example Using Parasitic Flatworms (Platyhelminthes: Cercomeria), Daniel R. Brooks, Deborah A. Mclennan

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Studies of adaptive radiations require robust phylogenies, estimates of species numbers for monophyletic groups within clades, assessments of the adaptive value of putative key innovations, and estimates of the frequency of speciation modes. Four criteria are necessary to identify an adaptive radiation within the parasitic platyhelminths: (I) a group contains significantly more species than its sister group, (2) species richness is apornorphic , (3) apomorphic traits enhance the potential for adaptively driven modes of speciation (sympatric speciation and speciation by peripheral isolation via host switching), and (4) the frequency of adaptively driven speciation modes is high within the group when …

Developmental Niche Structure In The Gregarine Assemblage Parasitizing Tenebrio Molitor, Richard E. Clopton, John J. Janovy Jr. Oct 1993

Developmental Niche Structure In The Gregarine Assemblage Parasitizing Tenebrio Molitor, Richard E. Clopton, John J. Janovy Jr.

John J. Janovy, Jr. Publications

The exogenous developmental niche spaces of Gregarina cuneata and Gregarina polymorpha, parasites restricted to larval Tenebrio molitor, and Gregarina niphandrodes, a parasite restricted to adult T. molitor, were determined along temperature and humidity axes and associated with parasite niche space on the host resource axis. Gregarine gametocysts were incubated at controlled humidity (0, 10, and 20 mm Hg vapor pressure deficit) at 20, 25, 30, and 35 C to determine the environmental limits to survival and sporulation. Although the environmental conditions that permit gametocyst development overlap, the gregarine parasite restricted to adult T. molitor utilizes a …

Analysis Of Black Point In Wheat, J M. Wilson Oct 1993

Analysis Of Black Point In Wheat, J M. Wilson

Technical Bulletins

Fungal staining (black point) of wheat can reduce the quality of grain. The most serious problem is a discolouration of products. Discolouration is a consequence of infection by microorganisms. The most likely cause in Western Australia is a species of the common fungus Alternaria. Infection and discolouration occur between flowering and grain maturity, and the optimum environmental conditions are probably consecutive days of high relative humidity together with warm temperatures.

Patterns Of Morphological Variation Of Salsuginus Yutanensis (Monogenea: Ancyrocephalidae) Over Space And Time, Michael T. Ferdig, Mary Ann Mcdowell, John J. Janovy Jr., Richard E. Clopton

John J. Janovy, Jr. Publications

Salsuginus yutanensis occurs on the gills of the Plains topminnow Fundulus sciadicus Cope. The fish of this species have been found to vary morphologically and biochemically among disjunct populations. Morphological characteristics of the sclerotized parts of S. yutanensis were examined from three localities in Nebraska, over a 2-yr collecting period. Analysis of variance was used to assess morphological variation with respect to site and date. Worms from two localities, Keith and Saunders counties, differed significantly for most characters considered. A third site, also in Keith County, contained worms for which measurement means tended to be intermediate between those in the …

Host Specificity Of Calyptospora Funduli (Apicomplexa: Calyptosporidae) In Atheriniform Fishes, John W. Fournie, Robin M. Overstreet Oct 1993

Host Specificity Of Calyptospora Funduli (Apicomplexa: Calyptosporidae) In Atheriniform Fishes, John W. Fournie, Robin M. Overstreet

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Calyptospora funduli has a broad host specificity, infecting at least 7 natural and 10 additional experimental definitive hosts, all atheriniform fishes within 5 families, but most in the genus Fundulus. Barriers, apparently innate ones, prevent any development of C. funduli in perciform fishes but allow incomplete or abnormal development of the parasite in a few unnatural atheriniform hosts. In the freshwater species Fundulus olivaceus and Fundulus notti, these abnormalities consisted of asynchronous development, degeneration of the parasite in early stages of development, and the formation of numerous macrophage aggregates. Rivulus marmoratus has the ability to eliminate infections with …

Twenty-two of 26 white-throated sparrows, Zonotrichia albicollis, from Algonquin Park, Ontario, Canada, hosted one or two species of platyhelminths. One species of cestode and three of digeneans were collected. Anonchotaenia quiscali inhabited 50% of the sparrows. This is the third report of A. quiscali; Z. albicollis is a new host. Brachylecithum nanum inhabited 43%, Zonorchis alveyi 15%, and Prosthogonimus macrorchis 8% of the sparrows examined. Ontario is a new geographic distribution record for all four parasite species. The proportion of hosts infected with two species was not significantly different from expected based on single species prevalences, indicating that …

Nubenocephalus nebraskensis n. gen., n. sp. (Apicomplexa: Actinocephalidae) is described from trophozoites, sporonts, gamonts, and oocysts collected from adult Argia bipunctulata (Odonata: Zygoptera). The new genus is distinguished from existing acanthosporine genera by elongate dodecahedral oocysts, without equatorial faces, that are hexagonal in equatorial cross section with equatorial and terminal spines. The epimerite is very broadly ovoid and truncated posteriorly with equatorial lobiform tumidi that are posteriorly bilobate and do not form hooks, spines, or digitiform processes. The epimerite is borne on a long, slender stalk. The new species is also reported from naiads ofArgia bipunctulata and adults and naiads …

Extending The Symbiotype Concept To Host Voucher Specimens, Daniel R. Brooks Aug 1993

Extending The Symbiotype Concept To Host Voucher Specimens, Daniel R. Brooks

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Recently, Frey, Yates, Duszynski, Gannon, and Gardner (1992) suggested that parasitologists deposit specimens of hosts from which type specimens of new parasite species are collected. Their proposal was directed especially at systematic and evolutionary biologists. Frey et al. (1992) pointed out that, whether one emphasizes microevolutionary (e.g., Price, 1980) or macroevolutionary (e.g., Brooks and Mc- Lennan, 1993) aspects of parasite evolutionary biology, much of the context of evolution involves hosts. Robust estimates of the selective effects and the patterns of diversification of host associations require the best possible estimates of the host species involved. This is …

Suppressive Effects Of Transforming Factor-Β And Interleukin-10 On The Cytolytic Activity Of Murine Macrophages And Reversal By Cytokines, Chin-Hung Lin

Loma Linda University Electronic Theses, Dissertations & Projects

In this study, the suppressive effects of transforming growth factor-β (TGF-β) and interleukin-10 (IL-10) on peritoneal macrophage killing of H238 target cells and the potential for reversal of the immunosuppressive effect by IL-4 and interferon-γ (IFN-γ) were investigated. The responsiveness of naive and peptone-activated macrophages was compared. The cytolytic activity for tumor cells of these effector cells was measured by percent lysis of 3H-thymidine labeled Herpes simplex virus type 2-transformed tumor cells (H238). After 18-24 hours of incubation with TGF-β or IL-10, the cytolytic activity of macro-medium alone. The immunosuppressive effect of TGF-β or IL-10 on non-activated macrophages was …

The Niche Of The Gill Parasite Dactylogyrus Banghami (Monogenea: Dactylogyridae) On Notropis Stramineus (Pisces: Cyprinidae), Jill A. Anderson, Kimberly J. Blazek, Tamara J. Percival Cook, John J. Janovy Jr. Jun 1993

John J. Janovy, Jr. Publications

Distribution of the monogenean Dactylogyrus banghami on the gills of the fish Notropis stramineus (Cyprinidae) was described by calculation of mean relative positions and Levins' niche breadths on the linear spatial resource gradients gill filament length and gill arch length. Thirty fish with 276 worms were examined; only one of the fish had an additional gill parasite species (Trichodina sp). Worms were more broadly and evenly distributed across the length of the gill arch than they were on the filament (breadths of 0.91 and 0.67, respectively). Mean worm positions were near the center of both resources: 54% of the …

Mechanisms Of T Cell-Mediated Macrophage Activation: Role Of Antigen Specific And Antigen Nonspecific Cognate Interactions, Xiang Tao Jun 1993

Mechanisms Of T Cell-Mediated Macrophage Activation: Role Of Antigen Specific And Antigen Nonspecific Cognate Interactions, Xiang Tao

Electronic Theses and Dissertations

Macrophages play an important role in host antimicrobial immunity and in non-septic inflammatory reactions. Most studies on macrophage activation have focused on the roles of the T cell-produced cytokine, interferon-$\gamma$ (IFN$\gamma)$ and bacterial product, lipopolysaccharide (LPS). T cell-macrophage interaction is a critical step in initiating both specific and nonspecific immune responses to antigenic stimulation. The current study examines the role of cognate T cell-macrophage interaction in activation of macrophage effector functions and induction of macrophage early activation gene expression. Viable resting T$\sb{\rm H}$2 clone cells can activate IFN$\gamma$-primed macrophages to produce reactive nitrogen intermediates (RNI) or express cytostatic activity. The …

Reversal Of Age-Associated Decline In Immune Response To Pnu-Imune Vaccine By Supplementation With The Steroid Hormone Dehydroepiandrosterone, Manju Garg, Subbarao Bondada

Microbiology, Immunology, and Molecular Genetics Faculty Publications

Recently, we reported that murine antibody responses to the 23-valent pneumococcal polysaccharide (Pnu-Imune) vaccine declined with age. Here we present data to support the concept that age-associated immune defects are not only due to intrinsic defects in immune cells but are also due to extrinsic factors emanating from the neuroendocrine system. We found that supplementation with dehydroepiandrosterone, a steroid hormone known to be reduced in the aged, corrects the immune deficiency of aged mice and significantly enhanced their splenic immune responses to the Pnu-Imune vaccine.

The Phylogenetic Component Of Cooperative Breeding In Perching Birds: A Commentary, Deborah A. Mclennan, Daniel R. Brooks May 1993

The Phylogenetic Component Of Cooperative Breeding In Perching Birds: A Commentary, Deborah A. Mclennan, Daniel R. Brooks

Harold W. Manter Laboratory of Parasitology: Faculty Publications

The study of behavioral and ecological evolution within a phylogenetic context (historical ecology; Brooks and McLennan 1991) is an important component of comparative studies in evolutionary biology. Although the number of historical ecological studies is growing rapidly, this research field is still in its infancy--an infancy whose maturation is hampered by the absence of rigorous phylogenetic hypotheses for many of the groups that have traditionally fascinated behavioral ecologists. In the absence of such critical information, behavioral ecologists are faced with the options either of forming cooperative groups with phylogenetic systematists or of investigating their ecological data based on "trees" reconstructed …

Probing Protein-Protein Interactions Among Proteins Of A Nonaggregated Fatty Acid Synthetase From Euglena Gracilis Variety Bacillaris, Sande G. Williams

Electronic Theses and Dissertations

Enoyl-acyl carrier protein (ACP) reductase from chloroplast nonaggregated fatty acid synthetase (FAS) of Euglena gracilis variety bacillaris was purified to a single band on a denaturing polyacrylamide gel. The enzyme was partially characterized with respect to substrate specificity, reduced nucleotide requirement, and the effect of ACP and Ca$\sp{++}$ on enzyme activity. Antibodies against the purified protein were raised in hens and isolated from eggs. ACP was purified from Euglena in yields of about 1mg/100g (wet weight) of cells. Antibodies were raised against the purified protein. ACP antibodies inhibited the Euglena chloroplast FAS using Euglena or E. coli ACP as a …

An Update On The Biological Control Of Rabbits, Stuart Wheeler Jan 1993

An Update On The Biological Control Of Rabbits, Stuart Wheeler

Journal of the Department of Agriculture, Western Australia, Series 4

When myxomatosis was introduced into Western Australia in the early 1950s, farmers, pastoralists and government alike thought it was an answer to the rabbit problem.

The disease was devastatingly effective in the short term and initially had a 99 per cent kill rate. With time, the virus declined in strength, and the surviving rabbits have multiplied.

There have been many recent reports about new forms of biological control for rabbits and potential improvements in old ones.

This article summarises each of the prospective methods and improvements, and assesses the potential usefulness of each.

Control Of Cheesy Gland In Sheep, Michael Paton Jan 1993

Control Of Cheesy Gland In Sheep, Michael Paton

Journal of the Department of Agriculture, Western Australia, Series 4

Cheesy gland is a widespread problem in Western Australian sheep flocks. Less than 1 per cent of flocks are free of this disease, and all sheep in one line of 550 cull ewes slaughtered at Katanning Abattoir were infected.

A study of what affects new cheesy gland . infection found that shower dipping and keeping sheep under cover after shearing increased new infections. Farmers who shower dip sheep should consider vaccinating them against cheesy gland. Sheep should be let out into the open as soon as possible after shearing.

The total cost of the disease in Australia is about $30 …

Parasite-specific antibody responses were detected using an indirect fluorescent antibody (IFA) test in cattle that were naturally or experimentally infected with Neospora parasites. The test was developed using Neospora tachyzoites isolated from an aborted bovine fetus and grown in bovine cell cultures (isolate BPA1). In all cases, infections were confirmed by the identification of Neospora tachyzoites and/or bradyzoite cysts in fetal or calf tissues using an immunoperoxidase test procedure. Fifty-five naturally infected cows that aborted Neospora-infected fetuses had titers of 320-5,120 at the time of abortion. The titer of 6 cows that were serologically monitored over a prolonged period decreased …

Sporulated oocysts (mean dimensions = 13.0 x 10.8 μm) and sporocysts (11.3 x 5.5 μm) of a coccidian resembling Frenkelia sp. or Sarcocystis sp. were present in the lamina propria of the small intestine of a naturally-infected northern spotted owl (Strix occidentalis caurina) collected near Medford, Oregon, USA. Dimensions of these oocysts and sporocysts appear to be considerably smaller than those from other sarcocystid species with avian definitive hosts. Additionally, numerous developmental stages and unsporulated oocysts (mean dimensions 22.8 x 17.8 μm) of a possible species of Isospora also were observed in the intestinal epithelium. This constitutes the …
