Familial hypercholesterolemia (FH) is a genetic condition characterized by elevated levels of low-density lipoprotein cholesterol (LDL-C) that leads to an increased risk of developing cardiac disease early in life (Shah et al., 2020). Current treatments such as statins and PCSK9 inhibitors have helped lower LDL-C levels, however they require repeated administration every 4-6 weeks to remain effective (Raal et al., 2018). Angiopoietin-like 3 (ANGPTL3) is an inhibitor of plasma lipid metabolism that has become a promising molecular target for the treatment of FH. Optofluidic lasers are flexible, reconfigurable, easy to fabricate and their emission spectrum can be dynamically tuned. Peripheral arterial disease (PAD) is a severe impairment of arterial vessels resulting in obstruction of normal blood flow in the legs, leading to acute or chronic lower limb ischemia and subsequently high morbidity and mortality rates. Common treatments for PAD, such as medications and surgical revascularization, have several limitations. For instance, medications used to lower cholesterol, reduce high blood pressure, control blood sugar, prevent blood clots, and reduce/delay symptoms such as leg pains. Still, they cannot treat the established disease directly and often cause side-effects, including bleeding, headache, and diarrhea. ABSTRACT: Landfill leachates are considered now as a reusable source of water. Even irrigation projects are now considering use of non-traditional water sources, including treated wastewater, to augment supplies. Treated wastewater or leachate, however, may still contain many inorganic and organic pollutants of emerging concern, because traditional wastewater treatment plants are unable to remove all of them. Adsorbents have been demonstrated to effectively remove heavy metals and recalcitrant organics from leachate/wastewater more effectively, at lower cost, and using less chemicals and energy than many competing techniques. This dissertation presents a collision cone/rendezvous cone-based approach for reactive motion planning in three-dimensional dynamic environments. Collision avoidance is fundamental to robot motion planning. In dynamic environments, the path and velocities of obstacles are not known a-priori, and hence it is a common practice to use reactive planners. Reactive planners should be computationally inexpensive because they need to act fast to avoid potential collisions. Current biological strategies for remediating trichloroethylene (TCE) in low pH aquifers (i.e., pH14C-TCE assay was developed to determine pseudo first-order rate constants for the degradation of TCE in microcosms containing soil and groundwater from the Boeing Michigan Aeronautical Research Center (BOMARC) superfund site, where the pH ranges from 4.1 to 4.9. The 14C-TCE assay was also adapted to calculate soil-normalized rate constants for data from this site, as well as data that Szwast21 collected from the Beltsville Agricultural Research Center (BARC). Membrane distillation (MD) is a rapidly emerging water treatment technology used to combat the global water crisis. Membrane pore wetting is a primary barrier to widespread industrial use of MD. The primary causes of membrane wetting are membrane fouling and an exceedance of liquid entry pressure. The development of different types of polymer membranes and the use of pretreatment have led to significant movement towards the prevention of wetting in MD. Armillaria root rot (ARR), primarily caused by the soilborne fungus Desarmillaria tabescens, has become the number one cause for peach tree decline in the Southeastern United States. Research has shown that planting peach trees on shallow berms and excavating the soil around the root collar two years after planting lessens the effects of ARR. This dissertation describes the design and implementation of Adaptive Current Tomograph 5 (ACT5) with a focus on the digital processing and data ow in the instrument. ACT5 is an electrical impedance tomography (EIT) instrument that produces images of the complex impedivity distribution within the body by injecting currents through and measuring voltages on electrodes applied to the skin. ACT5 is a parallel-drive EIT system with a dedicated current source for each electrode, capable of driving all electrodes simultaneously and measuring the induced voltages on them. Electrical Impedance Tomography (EIT) is a promising medical imaging technique used to detect the body's internal electrical characteristics based on electrical measurements made on its surface. EIT systems, particularly those that use parallel, multiple source architectures, require current sources with very high output impedance. This is a usual challenge for EIT as the practical current sources come with finite output impedance, which is highly degraded as the operation frequency gets wider. To overcome this challenge and maintain the high output impedance requirement, sources often use complex analog circuits which require manual or electronically-controlled adjustments.

Arsenic is a ubiquitous trace metalloid. However, increasing concentrations of arsenic in soil and water is a problem the world faces due to inputs from anthropogenic sources such as mining, milling, agriculture, and coal ash. One method of remediation for these arsenic-contaminated sites is bioleaching. While it is most often used to extract metals from raw ore, it is a viable and environmentally friendly method to remove the same metals from soil. The most complex heart valve is the mitral valve (MV). Many pathologies can affect the MV, including stenosis, regurgitation, prolapse, and mitral annulus calcification (MAC). MAC is chronic degeneration of the annulus, which is the fibrous, saddle-shaped “ring” that can contract and relax with the myocardium. The prevalence of MAC is around 15% but increases in patients with other cardiovascular diseases and risk factors. This thesis project describes the modification and evaluation of a new affinity membrane for rapid chromatographic purification of non-antibody proteins. The affinity membrane utilizes Im7/CL7 coupling technology developed by Dr. Vassylyev's lab at the University of Alabama Birmingham (UAB), licensed by TriAltus Bioscience. The behavior of the membrane was evaluated using purified CL7-tagged Cas9 as my model protein for static and dynamic binding capacity analysis.

Hydrogel adhesives are a new class of materials with excellent biocompatibility, which makes them very attractive for biomaterial applications. It has been previously shown that Tetronic T1107, a four-arm poly (propylene oxide)-poly (ethylene oxide) (PPO-PEO) block copolymer, is useful as a chemical crosslinking thermo-responsive hydrogel for bioadhesive applications. The end groups of this polymer are modified with acrylate and N-hydroxysuccinimide (NHS) functional groups. The acrylate end group gives the polymer cohesive properties with long-range chemical crosslinking using dithiothreitol (DTT), while the NHS end group gives the polymer adhesive properties through bonding with amines found in organic tissue. The second chapter of this report addresses the experimental challenges of this getter system by demonstrating the utility and experimental strengths of a custom magnetic suspension balance (MSB) based gravimetric sorption system. Sustainable intensification of rice farming is crucial to meeting human food needs while reducing environmental impacts. Rice produces 8% of all anthropogenic CH4, which is a potent greenhouse gas. CH4 emissions can potentially be reduced by cultivation practices that minimize the number of days the fields are saturated, such as dry-seeding instead of water-seeding and irrigation using the alternate wetting and drying (AWD) technique instead of delayed, continuous flooding (DF). In the search for a sustainable method to meet increasing energy needs, solar energy emerges as an underutilized, plentiful resource. Solar intermittency and requirements for transportation necessitate storing solar energy in the form of chemical bonds via artificial photosynthesis. Photoelectrochemical (PEC) water splitting generates hydrogen fuel from solar energy and water. A semiconducting material that successfully meets the complex requirements for building an industrially scalable PEC device has yet to emerge.
