Rainwater harvesting (RWH) was primarily considered as a source for fresh water supply or a conservation practice for overcoming water shortages in drought prone areas. By retaining storm-water run-off for onsite use, harvesting systems reduce the runoff volumes and pollutant masses entering waterways. Some of the most interesting aspects of RWH are the methods of capture, storage, and the use of this natural resource at the place it occurs.

These specimens were donated by biological and physical sciences faculty prior to the 1960 hire of Dr. John Philley and greatly expanded upon by Drs. Philley, James Chaplin, Jules DuBar, and H.W. Straley III through the 1990’s. Though the first brewing process dates back to ancient Mesopotamia, the modern method still presents a host of flaws. There are both many steps of fermentation and many opportunities for contamination. Most breweries will experience some form of spoilage in their lifetime. These invading microorganisms can alter turbidity, coloration, aroma, taste, and the overall quality of the product. This disruption is devastating for commerce. Eastern Kentucky lags behind the rest of the state regarding proper health, while Kentucky as a whole is doing worse than national averages. Kentucky not only leads the United States in cancer mortality, but sees higher mortality from heart disease, injury, suicide, diabetes, and chronic obstructive pulmonary disease. The health crisis being observed can largely be attributed to high poverty rates throughout the Eastern half of the state. Considering the not-so-broad socioeconomic scale seen throughout much of Kentucky, it is more evident that health has a direct correlation to one’s wealth and resources. Building onto a recent serious of ab initio studies of various acid-gas reactions with metal oxide sorbents, electronic structure methods are being used to study the addition of CS₂ to Group IV (MO₂)ₙ and Group VI (MO₃)ₙ (n = 1, 2, 3) nanoclusters, beginning with the MO₂ and MO₃ monomers according to a “bottom-up” approach. The preliminary density functional theory (DFT) calculations in this study provide structures and vibrational frequency thermodynamic corrections for later expanding upon by way of single point correlated molecular-orbital theory (MO) calculations, mainly CCSD(T) and MP2 to study the structures and energies which could arise from …

Repressor Of The Sos Response Mechanism In Acinetobacter Baumannii Requires Helix-Formation And Dimerization For Its Dna-Binding Ability, Belinda Candra, Deborah Cook, Jenelle Hare

After DNA damage, the multi-drug resistant pathogen Acinetobacter baumannii activates its SOS response, possibly allowing mutagenesis to occur by inducing multiple errorprone polymerases. A. baumannii uses the non-canonical repressor protein UmuDAb to repress operons of these polymerases. It has an N-terminal domain (NTD) with 2 helices (HTH1 & HTH2) whose sequences are needed for repression1. Like other bacterial SOS repressors such as LexA, UmuDAb dimerizes. BACTH analyses2 showed that UmuDAb forms dimers via its LexA-like G124 residue and UmuD-like N100 residue (UmuD is a component of an error-prone polymerase). However, N100 is not needed if the C termini are free. …

While solar power is one of the leading renewable energy sources today, current solar farms produce an amount of byproduct heat that is contributing to rising global temperatures and the deaths of many birds. Furthermore, solar panels that use water as a coolant through a sprinkler system use up a great amount of water that could be allocated somewhere else. Our project seeks to address both of these issues through a biomimetic design. Deoxyribonucleic acid (DNA) codes for proteins using a unique order of four major nitrogenous bases adenine, thymine, cytosine, and guanine. In several biological applications, such as the determination of DNA stability, production of biologic based drugs, and even taxonomic reorganization of species, it becomes necessary to understand the relative amounts of each nucleotide in a DNA sample, regardless of their sequence. Sewer overflows discharge 850 billion gallons of untreated sewage into U.S. lakes and rivers every year during high-intensity precipitation events. Common approaches include storage tanks or rapid solids removal followed by chlorine disinfection. These approaches can be capital intensive and only achieve partial treatment. A decentralized, end-of-pipe sewer overflow treatment system would eliminate detrimental overflow effects while handling highly variable flow and pollutant loading. Specifically, the rapid reaction rate of chemical oxidation can achieve soluble organics removal and disinfection of chlorine-resistant pathogens in a small footprint. Technologies are needed to treat contaminants such as disinfection byproducts (DBPs), trace organic compounds (TOrCs), and per- and polyfluoroalkyl substances (PFAS) in water to improve consumer safety and mitigate chronic risk. This dissertation focused on evaluating electrocoagulation (EC) and hydrogen peroxide (H2O2)-enhanced EC (peroxi-electrocoagulation; EC:H2O2) for treatment of these contaminants. The impacts of source water quality (such as natural organic matter [NOM]) on overall performance was assessed and the non-destructive and destructive pathways involved in treatment were evaluated. The extracellular microenvironment regulates many of the mechanical and biochemical cues that direct musculoskeletal development and are involved in musculoskeletal disease. The extracellular matrix (ECM) is a main component of this microenvironment. Tissue engineered approaches towards regenerating muscle, cartilage, tendon, and bone target the ECM because it supplies critical signals for regenerating musculoskeletal tissues. Engineered ECM–material scaffolds that mimic key mechanical and biochemical components of the ECM are of particular interest in musculoskeletal tissue engineering. Background: The removal of hair and ruler marks is critical in handcrafted image analysis of dermoscopic skin lesions. No other dermoscopic artifacts cause more problems in segmentation and structure detection. Purpose: The aim of the work is to detect both white and black hair, artifacts and finally inpaint correctly the image. Method: We introduce a new algorithm: SharpRazor, to detect hair and ruler marks and remove them from the image. Our multiple-filter approach detects hairs of varying widths within varying backgrounds, while avoiding detection of vessels and bubbles. Carbon disulfide is a toxic gas emitted from industrial plants and a common greenhouse gas. Acute poisonings from carbon disulfide are rare, but recurring exposure to low doses can have long term health effects. Currently, there are two approaches being considered for the removal of acid gas pollutants: (1) sequester these gases from the atmosphere or (2) remove these gases directly upon combustion (post-combustion). Potential energy surfaces (PES) for a series of CS2 capture reactions by NH3 in the presence H2O were calculated at the MP2/a(D+d) evel in the gas phase. G3(MP2) and FPD calculations are in progress. This thesis presents a study on the optical characteristics of hollow-core photonic crystal fibers (HC-PCFs) with a band gap cladding structure and their applications in optical fiber sensing. This 800B HC-PCF exhibited excellent optical properties and has a flexible structure, which makes them suitable for a wide range of industrial applications. Finite element simulations and structural optimization designs were conducted using the surface plasmon resonance (SPR) technique to determine the optimal performance parameters of the 800B HC-PCF. This study aims to advance the understanding and application of spectroscopic sensor data fusion for improving soil nutrient content predictions. In addition to presenting an extensive review of studies on the spectroscopic sensor data fusion, a research investigation was conducted to assess the effectiveness of five fusion algorithms in predicting three primary nutrients (Nitrogen, Phosphorous, and Potassium) and two secondary nutrients (Calcium and Magnesium) in soil using Visible and Near-Infrared, Mid-Infrared, and X-ray Fluorescence data. During photosynthesis and transpiration, crops exchange carbon dioxide and water with the atmosphere through stomata. When a crop experiences water stress, stomata are closed to reducing water loss. However, the closing of stomata also negatively affects the photosynthetic efficiency of the crop and leads to lower yields. Stomatal conductance (gs) quantifies the degree of stomatal opening and closing by using the rate of gas exchange between the crop and the atmosphere, which helps to understand the water status of the crop for better irrigation management. The increase in the use of carbon fiber-reinforced polymers (CFRPs) composites in the aerospace industry generated the need of improving the properties and capabilities of these composites by adding nano-reinforcements to the carbon fibers, also called hybrid fiber reinforced polymer composites. In this study, the energy absorption due to impact at low speed will be tested and simulated in four configurations of CFRPs utilizing the same [0/90]S layout throughout them.

Alzheimer’s disease (AD) is characterized symptoms of cognitive impairment resulting from blood-brain barrier (BBB) breakdown and neuronal cell death. AD is associated with aggregated amyloid-β (Aβ) plaques deposited around brain vasculature and within the brain. Changes in the brain occur up to 20 years before symptoms arise; however, there is currently no pre-symptomatic detection method available for AD. An intact BBB requires tight junction proteins (TJs) that render relatively high transendothelial electrical resistance (TEER). AD-associated breakdown of the BBB correlates with vascular Aβ deposition, suggesting that aggregated, pathological Aβ can modulate TEER. We explore leveraging this cellular response for early …

The US Air Force has gone through many aeromedical patient isolation transport system designs. The first designs were developed in response to the Ebola outbreak in 2014 and, more recently, the COVID-19 pandemic. The trade study analysis part of the system engineering design method was used to analyze the historic and current aeromedical patient contamination control transport systems. Recent mounting nuclear threats and postures from adversary nation-states, such as Russia, China, North Korea, and Iran, represent a clear danger to the interests and security of the United States of America and its Allies. To meet these threats, the 2022 Nuclear Posture Review requires the Department of Defense (DoD) to design, develop, and manage a combat-credible U.S. military which, among other prioritizations, is survivable. A survivable force can generate combat power despite adversary attacks. A Seawater Photovoltaic-Hydrogen Generation System is developed with the goal to produce a safe, user friendly, cost effective, and realistically scaled hydrogen generation system using seawater, particularly for remote regions as well as storage alternatives for renewable energy. An iterative design process was implemented within three prototypes, the first primarily focused on proving the concept of electrolysis, the second to create a robust hydrogen generation system resistant to corrosion using forward osmosis, and the third to scale up our system while also aiming for safety, user friendliness, and cost effectiveness. Activated sludge is a conventional treatment process for biochemical oxygen demand (BOD) and total suspended solids (TSS) removal at water resource recovery facilities (WRRFs). Foaming events are a common operational issue in activated sludge and can lead to decreased treatment efficiency, maintenance issues, and potential environmental health risks. Stable foaming events are caused by biological and chemical drivers (i.e., microbes and surfactants) during the aeration process. However, foaming events are difficult to predict and quantify. Urban water quality impairments have long burdened urban aquatic ecosystems in a phenomenon termed “urban stream syndrome”. The symptoms of this “syndrome” include physical changes in stream morphology and water level, biologically stressed environments, and perturbations in ecosystem processes. Despite years of research and costly investments in restoration and rehabilitation, urban waterways are still plagued by degradation. Improvement in urban water quality will require multi-faceted efforts, including progress on 2 key fronts 1) increased understanding of current impairments, and 2) increased knowledge about urban stormwater infrastructure like best management practices (BMPs).
