An Aptamer-Based Microelectrode With Tunable Linear Range For Monitoring Of K+ In The Living Mouse Brain, Yuan-Dong Liu, Jia-Run Li, Li-Min Zhang, Yang Tian
Journal of Electrochemistry
Potassium ion (K+) is widely involved in several physiopathological processes, and its abnormal changes are closely related to the occurrence of brain diseases of cerebral ischemia. In vivo acquirement of K+ variation is significant to understand the roles of K+ playing in brain functions. A microelectrode based on single-stranded DNA aptamers was developed for highly selective detection of K+ in brain, in which the aptamer probes were designed to contain an aptamer part for specific recognition of K+, an alkynyl group used for stable confinement of aptamer probe on the gold surface, and …

Plasmonic nanoparticles such as Au and Ag with localized surface plasmon resonance (LSPR) property exhibit unique scattering and absorption features. The plasmonic scattering and absorption bands are mainly located at visible light region which can be easily applied in visual detections. By modulating the size, shape, and composition of gold and silver colloid solutions, plenty of colorimetric methods have been constructed for the detection of metal ions, biomolecules, and environmental contaminants, etc. For many years, the LSPR-based measurements are implemented in reagent tubes. Since 2000, the plasmon resonance scattering (PRS) light of metal nanoparticles captured by dark-field microscopy enables the …

Intelligen Superpro Designer Files For Economic Simulation Of Batch And Continuous Aqueous Two-Phase Purification For Viral Products, Natalie Nold, Eric Pearson, Caryn L. Heldt
Michigan Tech Research Data
Vaccine manufacturing strategies that lower capital and production costs could improve vaccine access by reducing the cost per dose and encouraging localized manufacturing. Continuous processing is increasingly utilized to drive lower costs in biological manufacturing by requiring fewer capital and operating resources. Aqueous two-phase systems (ATPS) are a liquid-liquid extraction technique that enables continuous processing for viral vectors. To date, no economic comparison between viral vector purifications using traditional methods and ATPS has been published. In this work, economic simulations of traditional chromatography-based virus manufacturing were compared to simulations of ATPS-based virus manufacturing for the same product output in both …
Adsorption Of Long And Short Per- And Polyfluoroalkyl Substances (Pfas) Onto Granular Activated Carbon And Porous Organic Polymers, Yan Zhang
USF Tampa Graduate Theses and Dissertations
Per- and polyfluoroalkyl substances (PFAS) are man-made environmental contaminants causing increasing global concern due to their adverse effect on environmental and human health. Conventional treatment methods are ineffective in removing short-chain PFAS because of their hydrophilicity and resistance to degradation. This study is to design an appropriate adsorption system to remove both long- and short-chain PFSA at environmentally relevant concentrations and conditions. Four primary research tasks were designed to evaluate the performance of a structurally-tunable and chemically-stable porous organic polymers (POPs) for PFAS removal under realistic environmental conditions, including the assessment of POPs’ performance without co-contaminants (Chapter 2), the assessment …
Integration Of Insoluble And Soluble Dopants Into Steel Using Laser Powder Bed Fusion, Mckay Goodman Sperry
Integration Of Insoluble And Soluble Dopants Into Steel Using Laser Powder Bed Fusion, Mckay Goodman Sperry
Theses and Dissertations
Laser Powder Bed Fusion (LPBF) is an Additive Manufacturing (AM) method whereby complex parts can be built to near net geometry in an automated environment. Parts formed by LPBF have excellent properties and require little post-processing. While LPBF allows for creative designs to reduce weight and part count by integrating sub-components of assemblies, designers are limited to one material within any LPBF print. Although various materials can be used in LPBF, metals are of special interest to many industrial customers due to their high strength and toughness. The metals commonly used for LPBF include iron, aluminum, and titanium alloys. These …

USF Tampa Graduate Theses and Dissertations
Coronavirus disease-19 (COVID-19), caused by Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2), is an ongoing pandemic-endemic. In the United States, it remains the third leading cause of death in 2022. SARS-CoV-2, an RNA virus, has a genome prone to rapid mutation, generating variant species capable of resisting therapies and vaccination efforts. However, it has been estimated that more than 40% of SARS-CoV-2 infections may be asymptomatic, which calls into question any biological differences between the asymptomatic and afflicted groups. Additionally, Respiratory Syncytial Virus (RSV) is the most significant cause of bronchiolitis and asthma in neonates, with an increasing burden on the …

Exploratory Data-Driven Models For Water Quality: A Case Study For Tampa Bay Water, Sandra Sekyere
USF Tampa Graduate Theses and Dissertations
Water, a crucial resource for sustaining life, covers approximately 70% of the earth's surface. Nonetheless, the quality of water is deteriorating rapidly due to the rapid growth of urban areas and industries, which is a worrying trend causing harm to human health and the ecosystem. Water quality forecasting has a key role in water resources management by enabling effective pollution control, ecosystem monitoring, and decision-making.
Previously, traditional statistical models were used to forecast water quality, but they were unable to examine the non-linear relationships between water quality parameters, and they assumed that all datasets were distributed normally. This study uses …

Resource Recovery From Crew Metabolic Wastes In Lunar And Martian Habitats: Reuse Of Effluent From An Anaerobic Membrane Bioreactor Treating Fecal Organic Matter As Fertilizer Source For Plants, Alexandra A. Smith
USF Tampa Graduate Theses and Dissertations
As humans set their sights on exploration of the Moon and Mars human focused challenges like providing potable water, supplying nutritious food to the astronaut crew, and managing their waste present themselves. There are no readily available resources on the Moon and Mars, therefore nothing should be considered unusable including the organic wastes (i.e., feces and urine) produced by the crew. Fecal matter is not currently recycled and is treated as solid waste. After collection, it is discarded out of the International Space Station where it is incinerated upon re-entry. This approach will not work on the Moon’s surface. There …

Theses and Dissertations
This work demonstrates how ICP-MS can be employed to evaluate cesium volatilization from LiF-BeF2 (Flibe) salt with ab initio molecular dynamics studies used as corroborating data to better understand cesium behavior. Using mixtures of Flibe with 2 mol% CsF, it was found that cesium was stable within the salt melt at temperatures between 500-650°C over a time span of 8 hours. At 800°C, cesium vaporized from the salt at a rate of 0.83% / hr with a mass flux of 0.0023 g Cs / cm2hr. The atomistic modeling results show poor solvation of Cs at 500°C and 800°C, with stability …
Exploring Alternative Electron Donors For Heterotrophic Denitrification At A Water Reclamation Facility In Tampa Bay, Tejas Athavale
Exploring Alternative Electron Donors For Heterotrophic Denitrification At A Water Reclamation Facility In Tampa Bay, Tejas Athavale
USF Tampa Graduate Theses and Dissertations
An excess of nitrogen in water can pose problems, ranging from algal blooms to creating toxic environments for marine life. Therefore, wastewater treatment plants need to remove nitrogen from wastewater during the treatment process. Biological nitrogen removal is one process these plants may use, which commonly involves nitrification and denitrification, converting nitrogen in the form of ammonia to nitrate, and then ultimately nitrogen gas. The process of denitrification often requires an electron donor, and methanol is a common choice. However, it is flammable, and therefore there is a need to explore alternative electron donors. Alternative donors can include manufactured chemicals …

User Profiling Through Zero-Permission Sensors And Machine Learning, Ahmed Elhussiny
Theses and Dissertations
With the rise of mobile and pervasive computing, users are often ingesting content on the go. Services are constantly competing for attention in a very crowded field. It is only logical that users would allot their attention to the services that are most likely to adapt to their needs and interests. This matter becomes trivial when users create accounts and explicitly inform the services of their demographics and interests. Unfortunately, due to privacy and security concerns, and due to the fast nature of computing today, users see the registration process as an unnecessary hurdle to bypass, effectively refusing to provide …

Creation Of A Hyperplane Device For Horizontal Cellular Migration Assays, Nicholas M. Bittner, Nelofar Nargis, Ghoulem Ifrene, Brent Jeffrey Voels, Colin K. Combs
Biomedical Sciences Posters and Presentations
Cell culture studies routinely seek to monitor cell migration in response to chemoattractant stimuli. Common assays of cell migration employ well inserts and vertical cell migration assessment. This approach does not allow real-time monitoring of cell behavior. To address this need, we sought to develop a horizontal culture platform conducive to time course cell assessment changes in migration, morphology, phenotype etc. Modification of a commercial chamber slide allowed us to quantify cell migration in response to a 20% serum gradient. Based upon this finding, we designed and fabricated a prototype chamber slide for high replicate, real time assessment of cell …

Platinum-containing stents are commonly used in humans with hypercholesterolemia, whereas preclinical stent evaluation has commonly been performed in healthy animal models, providing inadequate information about stent performance under hypercholesterolemic conditions. In this investigation, we used an ApoE−/− mouse model to test the impact of hypercholesterolemia on neointima formation on platinum-containing implants. We implanted 125 μm diameter platinum wires into the abdominal aortas of ApoE−/− and ApoE+/+ mice for 6 months, followed by histological and immunofluorescence examination of neointimal size and composition. It was found that ApoE−/− mice developed neointimas with four times larger area and ten times greater thickness than …

Blood flow is a key regulator of atherosclerosis. Disturbed blood flow promotes atherosclerotic plaque development, whereas normal blood flow protects against plaque development. We hypothesized that normal blood flow is also therapeutic, if it were able to be restored within atherosclerotic arteries. Apolipoprotein E-deficient (ApoE-/-) mice were initially instrumented with a blood flow-modifying cuff to induce plaque development and then five weeks later the cuffwas removed to allowrestoration of normal blood flow. Plaques in decuffed mice exhibited compositional changes that indicated increased stability compared to plaques in mice with the cuff maintained. The therapeutic benefit of decuffingwas comparable …

Gold(I)-Hydroarylation In Polymer Modification – Alkynes And Alkenes With Unexpected Reactivity, Samuel B. Hunt
Gold(I)-Hydroarylation In Polymer Modification – Alkynes And Alkenes With Unexpected Reactivity, Samuel B. Hunt
Dissertations
Polymer modification is crucial for developing new properties and materials from recalcitrant waste polymers. Synthetically, polymers possess a chemical inertness that renders their functionalization challenging. Specifically, the omnipresence of C–H bonds in all commercial polymers endows their chemical resistance. Thus, developments in C–H activation chemistry are crucial to polymer diversification processes and allow for fundamental insight into the role of material processing conditions on novel chemical reactions. Au(I)-chemistry has recently come to the fore owing to a unique chemical reactivity nonexistent with other transition metals. The alkynophilicity of Au(I), in tandem with their insensitivity to common polymer processing conditions (air, …
