Mineralization Of 2-Chlorophenol Using A Hollow Fiber Ultrafiltration System In Combination With A Reactor Containing An Immobilized White Rot Fungus, Sheau-Ming Tung

Theses

Phanerochaete chrysosporium, a white rot fungus was immobilized on a packed-bed reactor, and was used to mineralize an aqueous stream contaminated with 50 ppm of 2-chlorophenol. Hollow fiber ultrafiltration membranes (for 5,000 daltons and 10,000 daltons molecular weight cut-oft) were utilized to retain the extracellular enzyme resulting from the immobilized fungus in a recirculation loop outside of the reactor.

It was found here that the concentration of proteins (roughly proportional to the extracellular enzyme releasd by the fungus) does not correlate with the rate of degradation of 2-chlorophenol in the system. The protein was concentrated 1.5 fold in the recycle …

Over the last decade, interest has grown in the use of the scanning acoustic microscope (SAM), with a single lens being used for both transmitting and receiving the acoustic signal. A major objective of this thesis was to determine if the transmission and ultrasonic techniques yield similar "quantitative" results, when measuring the elastic constants of human femoral bone. The transmission technique has been used since 1970 to yield values for the elastic properties of calcified tissue. A simple mathematical model and simulation of the progression of cancer with particular emphasis on Cutaneous T Cell Lymphoma was investigated. The mathematical model consists of a set of differential equations that first satisfies the homeostasis levels of the different components present and second predicts the dynamics during a diseased or unhealthy condition. The simulation was done using a FORTRAN program for integrating the differential equations.

A novel heterogeneous catalytic-oxidation technology where activated carbon is used as a catalyst-adsorbent, ozone serves as the oxidant and UV light as activation agent, is developed in a lab scale study. We demonstrate this process for destruction of recalcitrant organics in water i.e: combined oxidation by ozone, UV light plus activated carbon. The investigation includes evalution of various operating conditions: ozone levels, activated carbon levels; time for complete destruction of the individual target pollutants, plus evalution of activity of the continuously recativated carbon surface in-situ. Artificial neural networks with such characteristics as learning, graceful degradation, and speed inherent to parallel distributed architectures might provide a flexible and cost solution to the real time control of robotics systems. In this investigation artificial neural networks are presented for the coordinate transformation mapping of a two-axis robot modeled with Fischertechnik physical modeling components. The results indicate that artificial neural systems could be utilized for practical situations and that extended research in these neural structures could provide adaptive architectures for dynamic robotics control.

The purpose of this project was to explore the advantages and/or limitations of high-energy radiation treatment as a method for degrading organic pollutants, particularly aryl halides, in an aqueous medium. We have done analyses of 60Co-irradiated samples and kinetic studies using pulsed electron beams. For aryl halides containing no more than two fused rings, the main products detected are those of simple halogen replacement by hydrogen, although the amount of aryl halide destroyed was always greater than the total amount of products detected. Retention Of Cellular Viability And Growth Potential In Cryopreserved Bovine Arterial Tissue: Implications For Clinical Use Of Cryopreserved Vascular Allografts, Kip G. Gardner

Segments of bovine calf aortas and pulmonary arteries were cryopreserved at -196° C for two weeks to 18 months and subsequently thawed. Cellular morphology, viability, and growth potential in these segments were then compared with the same attributes in segments from fresh (control) arteries. Scanning electron microscopy revealed no disruption of intimal surfaces attributable to cryopreservation. Endothelial cells were successfully cultured from all cryopreserved arterial segments. Endothelial monolayers grown from cryopreserved arteries had a cobblestone appearance and expressed Factor VIII antigen but not smooth muscle alpha-actin. Plating efficiency in primary cultures of endothelial cells was lower for cryopreserved arteries than …

Fucoidan is a polysaccharide commonly found in brown algae. It is a sulfated polymer of the monosaccharide L-fucose. Fucoidan is thought to function in water retention by algae. Commercially available fucoidan is extracted from a brown algae called Eurus vesiculosus.

This polysaccharide has immense potential as a biologically active agent: (i) it exhibits anticoagulant action, (ii) it has also been reported to block retroviral replication and (iii) it is capable of blocking sperm-egg binding in sea urchin, mouse and rabbit. It was hypothesized that nutrient removal from wastewater could be achieved by using methane oxidizing bacteria (methanotrophs). Because methane is inexpensive. it can be used as an energy source to encourage bacterial growth to assimilate nitrogen and phosphorus and other trace elements. This initial feasibility study used synthetic nutrient mixtures and secondary sewage effluent as feed to a laboratory-scale methanotrophic attached-film expanded bed (MAFEB) reactor operated at 35°C and 20°C. The MAFEB system operated successfully at low nutrient concentrations under a variety of nutrient-limited conditions. A transient model to predict temperature within a tunnel ventilated broiler house during misting is developed. The model is calibrated with field data to obtain steady-state constants; transient predictions are compared to measured temperatures during cyclic misting for two different size birds. Measured temperatures during cyclic misting are shown to swing between steady-state asymptotes predicted from the model. Transient response of the model was faster than measured temperature data, in part due to temperature sensor dynamic response. The model predicts the length-wise temperature profile within the building during misting, and can be used to investigate alternate misting strategies and designs. …

The Neural Tactile Sensor (NTS) is a high resolution, easily manufactured tactile sensor consisting of electrodes, a thin resistive 'skin', and pattern recognition circuitry that is capable of resolving dynamic and static contact location, force, and slip throughout the continuum of the sensor's active region. The sensor operates by means of a resistive 'skin' harboring the electric field generated when a current is injected into it, and a plurality of electrodes for taking measurements of said electric field. Professor Shrader-Frechette discusses factors responsible for acute disagreement between the federal government and Nevada citizens over potential Risks at Yucca Mountain and focuses on the use of expert judgment, concluding that some of them appear to exemplify "bad science." That aside, she argues that 1,000 year predictions cannot be made from current knowledge of geology or, e.g., institutional behavior and concludes that permanent disposal of radioactive waste is currently impossible.

The purpose of this study was to implement an autonomous face segmentor as the front end to a face recognition system on a Sun SPARCStation2. Face recognition performance criteria, specifically, the capabilities to isolate and resize faces in an image to a consistent scale, were analyzed to determine current practical limitations. Face images were acquired using a S-VHS camcorder. Segmentation was accomplished using motion detection and predefined rules. Section 319 of the 1987 Water Quality Act requires states to develop management programs for reduction of NPS pollution via Best Management Practices (BMPs). The Air Force's Pollution Prevention Program Mandates composting as an opportunity to reduce landfill disposal and save future environmental costs. There are more than 4,000 Air Force (AF) sites requiring restoration due to hazardous waste contamination. The AF goal is to completely restore all sites by 2000. One method of achieving this ambitious goal is to use in-house capabilities. This study examined the potential use of RED HORSE capabilities and training activities to assist with cleanup of contaminated sites. RED HORSE capabilities and training requirements were compared to the primary cleanup needs of the AF. The study of left ventricular filling in both the normal and the pathological heart is of great importance. A computer model simulation of the heart with emphasis on mitral flow was designed by Meisner in 1986 and implemented in QuickBASIC. This paper discusses how the original program was altered to be able to perform "Parameter vs. Parameter" runs in which it can be observed how varying an input parameter affects output parameters. The output data is then formatted in such a way that it can be used by QUATTRO PRO, a standard spreadsheet package, to yield graphical outputs.

Conservation tillage systems help to maintain residue materials from the previous crop on the soil surface. The potential for serious erosion may exist if crop residues are removed by overland flow. This study is conducted to identify the hydraulic conditions required to initiate residue movement by overland flow. Corn, cotton, peanut, pine needles, sorghum, sunflower, and wheat residue are placed in a flume on smooth and sand surfaces, and flow is then introduced in progressive increments. The effect of pronuclear microinjection of DNA and culture in excised mouse oviducts on the development of porcine zygotes was assessed in this study. Precocious ovulation was induced in prepubertal gilts with pregnant mare's serum gonadotrophin and hCG. Zygotes received either pronuclear microinjection of buffer alone, buffer containing a DNA construct, or no microinjection. Zygotes were cultured in vitro in either modified Krebs-Ringer bicarbonate medium (KRBI for 144 h or in mouse oviduct (MO) explant culture with KRB for 48, 72, 96, or 120 h. Pronuclear microinjection of DNA resulted in a lower (P < .05) cleavage index (CII than did buffer or no microinjection (CI 2.16 k .10 vs 2.80 f .13 and 2.93 f .lo). The CI loss was greatest for DNA-injected zygotes at the two-cell stage of development. Coculture of zygotes in MO resulted in a higher CI (P < .01) than did culture in KRB. Culture in MO for 72 h was the most beneficial system compared with MO for 48, 96, or 120 h (P < .05; CI 3.25 k .12 vs 2.66 k .18, 2.79 f .14, and 2.40 .14, respectively). Microinjection of DNA, not merely the mechanical procedure, was detrimental to early zygote development and may be the cause of low pregnancy rates.

This thesis presents an application of artificial neural networks in real-time resource reallocation, a methodology used in the implementation of an intelligent interface node in the Computer Integrated Manufacturing (CIM) environment. In particular, the problem is formulated using a Hopfield neural network model. The real-time reallocation problem is mapped into a two-dimensional matrix of neurons similar to Hopfield and Tank's approach to the traveling salesman problem (TSP) .
