Abstract: Crossflow membrane filtration was used to process recombinant Escherichia coli cell lysates containing protein inclusion bodies after high pressure homogenization. The number of passes through the high pressure homogenizer changed the viscosities and average particle sizes of the cell lysates. The different cell lysates were processed with a hollow fiber unit containing microfiltration membranes and a plate and frame unit with either ultrafiltration or microfiltration membranes. There were differences in permeate flux and protein transmission for the various membranes with the best performing membranes giving permeate fluxes greater than 60 L m−2 h−1 and protein transmissions greater than 90%. …

In many real-life situations, e.g., when making an environmental decision, it is important to be able to predict long-term consequences of different decisions. Very often, these predictions must be done in the situation where the only available information consists of expert rules, which are formulated by words from natural language. One possible way to transform these expert words into numerical simulation (leading to prediction) is to use the fuzzy control methodology. This study was conducted to identify sediment movement within a strip intercropping system in southwestern Iowa during the third year of a three-year crop rotation. Soil loss, resulting from the application of simulated rainfall to a Monona silt loam soil, was measured from individual corn (Zea mays L.), soybean (Glycine max (L.) Merr), and winter wheat (Triticum aestivum L.) strips, and from multiple strips which included all three crops. Because of the crop rotation and residue management procedures, used at the study site, a substantial amount of surface cover and vegetative mass was present on each of …

The feasibility and efficiency of simultaneous removal of multiple cations as well as anionic heavy metals from wastewater into a mixed extractant kept stagnant on the shell side of a 2-fiber-set hollow fiber membrane extractor operated as a fixed bed adsorber was explored. The novel extracting solvent was a mixture of an acidic or chelating extracting agent for one or more cations and a basic extracting agent, e.g., trioctylamine (TOA) for extracting anions; both were dissolved in the same diluent kerosene for simultaneous and synergistic extraction of cations and anions. The coupling of a pervaporation membrane unit with a reactor has been investigated as a means of improving the overall process efficiency. As model system, the esterification of salicylic acid with methanol in the presence of a homogeneous catalyst was studied in a unit consisting of a batch reactor externally coupled with a pervaporation module containing a composite poly(vinyl alcohol) membrane. The reaction was carried out at temperatures between 336 and 345 K. The catalyst was sulfuric acid at concentrations varying from 0.5 to 2.0 molar. Kinetic experiments were conducted using Pseudomonas putida (ATCC 17484) to determine the rate of growth when naphthalene was provided as a sole carbon source in suspended biomass systems. Batch and chemostat reactors were used under three sets of conditions: non-aerated, aerated at 29.5° C, and aerated at 25.5° C. A number of concentrations were tested under each set of conditions. Data regression was used to determine parameters for Monod (non-inhibitory), Andrews (inhibitory), and zero-order kinetics. Although the Andrews fit of the data provided a slightly lower sum-of-squares error (SSE), the amount of data scatter and the weak inhibition made the …

( 1) Modify the three-dimensional basewide ground-water flow model for Wurtsmith Air Force Base (WAFS), Michigan (PES, 1997) based on recent investigation by ICF Kaiser (ICF).
(2) Develop contaminant transport models for OT-24 area TCE and DCE plumes. These models will use the same parameters that were used by ICF in their feasibility study ..

Satellite images already produce huge amounts of data, which makes their processing a serious computational challenge. This problem will become even more complicated with the launch of multi-spectral Earth-imaging satellites that will increase the amount of information by at least two orders of magnitude. With such a huge amount of information, it is necessary to come up with data processing methods that are as fast as possible. In particular, we show that for fuzzy processing techniques, this leads to the necessity to use integer-valued fuzzy sets.

If several physical theories are consistent with the same experimental data, which theory should we choose? Physicists often choose the simplest theory; this principle (explicitly formulated by Occam) is one of the basic principles of physical reasoning. However, until recently, this principle was mainly a heuristic because it uses the informal notion of simplicity.

The effectiveness of management practices in improving quality of runoff from agricultural land areas has been reported based primarily on results from plot- and field-scale studies. There is limited information available on watershed scales, particularly when the dominant agricultural land use is pasture. Field water balance measurements using monolithic lysimeters were used in validating the Cupid-DPE model for predicting water loss partitioning during sprinkler irrigation from a moving lateral system fitted with impact sprinklers and spray nozzles. The model combines equations governing water droplet evaporation and droplet ballistics with a comprehensive plant-environment energy balance model. Comparisons indicate good agreement between measured and modeled transpiration, and the measured and modeled soil evaporation during the day of irrigation. Total predicted evapotranspiration during the day of irrigation was greater than measured totals using the monolithic lysimeters. However, part of this difference was because the lysimeters could …

This study was conducted to measure runoff, erosion, and soil quality characteristics of a site in southwestern Oklahoma the first year following conversion from the Conservation Reserve Program (CRP). Treatments included undisturbed CRP, Old World bluestem (Bothriochlora ischaemum L.), no-till wheat (Triticum aestivum L.) and conservation-till wheat. Significant differences in surface cover were found between each of the experimental treatments, with values ranging from 100% on the undisturbed CRP site to 42% for the conservation-till treatment. No significant difference in runoff was found among the various experimental treatments. The Old World bluestem and winter wheat treatments had only …

We discuss a variety of adaptive critic designs (ACDs) for neurocontrol. These are suitable for learning in noisy, nonlinear, and nonstationary environments. They have common roots as generalizations of dynamic programming for neural reinforcement learning approaches. Our discussion of these origins leads to an explanation of three design families: heuristic dynamic programming, dual heuristic programming, and globalized dual heuristic programming (GDHP). The main emphasis is on DHP and GDHP as advanced ACDs. We suggest two new modifications of the original GDHP design that are currently the only working implementations of GDHP. They promise to be useful for many engineering applications …

Experimental autoimmune myasthenia gravis (EAMG) is an animal model of human myasthenia gravis (MG). In mice, EAMG is induced by immunization with Torpedo californica acetylcholine receptor (AChR) in complete Freund's adjuvant (CFA). However, the role of cytokines in the pathogenesis of EAMG is not clear. Because EAMG is an antibody-mediated disease, it is of the prevailing notion that Th2 but not Th1 cytokines play a role in the pathogenesis of this disease. Experimental data from a chromatographic system involving the adsorption of bovine serum albumin (BSA) onto porous anion-exchange adsorbent particles packed in a column are presented. The parameters that characterize the mass transfer mechanisms of intraparticle diffusion and convection are estimated by fitting the predictions of dynamic mathematical models describing adsorption in column systems having spherical perfusive and purely diffusive adsorbent Particles to the experimental breakthrough data obtained from the column adsorption system. The ability of microorganisms to selectively adsorb various heavy metal ions has been recognized for over a decade. We have investigated the biosorption of lead by an active culture of the cyanobacterium Anabaena cylindrica. Energy dispersive X-ray microanalysis was used to evaluate the different uptake mechanisms in the various subcellular regions. Three were identified; a very fast adsorption mechanism in the cell envelope; a time- dependent deposition reaction on the cell surface; and an adsorption mechanism, also time dependent, on the polyphosphate body inside the cell. A one-quarter scale model tractor and loader were designed, fabricated, and used in an experiment to evaluate two tractor-loader configurations relative to stability using roll angle as the means of assessment. The nonconventional loader system consisted of a conventional loader attached to a steerable carrier which in turn was attached to the tractor by the front axle and drawbar of the tractor. The stabilizing axle for the non-conventional loader system was the front axle as opposed to the conventional system in which the stabilizing axle was the rear axle. This study explores the feasibility of utilizing an amended vermicomposting treatment approach for detoxifying petroleum hydrocarbon contaminated sandy soil. The bench-scale testing of gasoline and diesel contaminated soil using three test soil vessels and one control soil vessel was performed in a laboratory setting for a six week time period. The control soil received no treatment other than distilled water to maintain soil moisture. Treatment 1 consisted of direct application of liquid municipal biologic sludge on a weekly basis. Treatment 2 consisted of the addition of 30 Eisenia foetida earthworms and distilled water. Treatment 3 consisted of direct application of …

The objective of this study was to develop a ground-water model that can be used in conjunction with an optimization model to optimize the containment and remediation of ground water contaminated with organic compounds at Travis AFB, California (TAFB). Based on studies by WESTON (Roy F. Weston, Inc.), RADIAN (Radian Corporation), and others, the major ground-water quality problem is a plume of TCE that originates at the Oil Spill Area (OSA) and bas migrated to the south and east under the runway. The immediate remediation objective is to stop the movement of the plume.

An apparatus for the monitoring of a column chromatography separation process includes a segmented column with a seal positioned at the joint defined by the segments of the column. A connector is provided for connecting the segments of the column together. The apparatus further includes a sensor for monitoring an analyte in an eluant within a separation zone of the column. The sensor includes a mesh grid made of optical fibers or metal wires which is placed so as to extend through the separation zone of the column.
