Noncontact scanning impedance imaging has been presented as a method to provide high resolution, high contrast images for a variety of material systems. This technique combines electrical impedance measurements with very high resolution scanning. This article reports on efforts to scale this technique down to the very important single micron range and reveals measurements for both thick and thin samples with a measured minimum resolution below 30 µm. A design for a shielded impedance probe applicable to this process is outlined and probes of several different sizes were made and tested. Fabrication of these impedance probes is explained and a …

We report the design, fabrication, and demonstration of single-mode integrated optical waveguides with liquid cores. The principle of the device is based on antiresonant reflecting optical (ARROW) waveguides with hollow cores. We describe design principles for waveguide loss optimization down to 0.1/cm. Using a fabrication process based on conventional silicon microfabrication and sacrificial core layers, waveguides of varying widths and lengths with volumes covering the pico- to nanoliter range were fabricated. We observe confined mode propagation, measure waveguide losses of 2.4/cm, and demonstrate that the waveguides possess tailorable wavelength selectivity. Controlled laboratory microcosms were used to research the phytoremediation potential of lupines (Lupinus chamissonis) for hydrocarbon-contaminated groundwater at a former oil field near Guadalupe, California. During oil production in the Guadalupe Oil Field, a kerosene-like hydrocarbon mixture was used as a diluent to improve the flow of the heavy crude oil. Leaking tanks and pipes resulted in diluent contamination in the soil and groundwater. Native plant species were planted at a pilot-scale field site to investigate the feasibility of using phytoremediation to remediate the groundwater contamination. Thin wall microcapsules were formed via Layer-by-Layer Self-Assembly of alternate adsorption of oppositely charged polyelectrolyte on microcores. After the core dissolution, empty polymeric shells with 20–25 nm thick walls were obtained. These microcapsules were loaded with Myoglobin, Hemoglobin and Glucose Oxidase by opening capsule pores at low pH and closing them at higher pH. The native structure of the enzyme was not affected due to different treatments. Biocompatible nanoshells were also prepared for encasing DNA. Production of recombinant antibodies against botulinum neurotoxin is necessary for the development of a post-exposure treatment. CHO-DG44 cells were transfected with a plasmid encoding the light and heavy chains of a chimeric monoclonal antibody (S25) against botulism neurotoxin serotype A. Stable cell lines were obtained by dilution cloning and clones were shown to produce nearly equivalent levels of light and heavy chain antibody by an enzyme-linked immunosorbent assay (ELISA). In suspension culture, cells produced 35 μg/ml of chimeric antibody after 6 days, corresponding to a specific antibody productivity of 3.1 pg/cell/day. A method for the harvest and recovery of an …

The objective of this work is to investigate the combination of layer-by-layer self-assembly with microfabrication technology and its applications in microelectronics and MEMS.

One can assemble, on a standard silicon wafer, needed multilayers containing different nanoparticles and polymers and then apply various micromanufacturing techniques to form microdevices with nanostructured elements.

Alternate layer-by-layer self-assembly of linear polyions and colloidal silica at elevated temperatures have been firstly studied by QCM and SEM. LbL self-assembly and photolithography were combined to fabricate an indium resistor. The RTA method was employed in the fabrication. The purpose of this study was to analyze the performance effects of gender and regional dialect on air traffic control statement recall. Sixty-one student volunteers participated in the experiment. Thirty-one participants held a pilot’s license and 30 participants had no flight experience. Each participant listened to one CD with 60 ATC statements each representing a male and female voice and New England, Southern, and General American dialect. Participants were asked to recall exactly what they heard. If the participant could not understand what they heard, they requested a repeat. Haemophilia replacement factors, both plasma-derived and recombinant, are in relatively short supply and are high-cost products. This has stymied the study and development of alternative methods of administration of haemophilia therapy even in the most economically advanced countries, owing to the large amounts of material needed because bioabsorption and bioavailability of haemophilic factors can be less than 10% when using non-intravenous routes of delivery. There is therefore a need to increase access to therapy worldwide by decreasing the cost and increasing the abundance so that therapy can be achieved through simplified, alternative delivery methods. An enzyme-catalyzed synthesis of novel polymers within layer-by-layer (LbL) constructed polyelectrolyte microcapsules has been developed. This approach is based on the selective permeability of polyelectrolyte-capsule walls to monomer molecules. Conversely biocatalysts and forming polymeric chains cannot exit the micro-capsule interior because of their characteristic high molecular weight. Horseradish Peroxidase (HRP) was encapsulated into four bilayer PSS (poly-styrenesulfonate)/PAH (poly-allylamine hydrochloride) capsules with an average diameter of 5 μm using pH-driven pore opening. The polymerization of 4-(2-Aminoethyl) phenol hydrochloride (tyramine) catalyzed by HRP produces easily detectable fluorescent polymeric products after the addition of hydrogen peroxide to the system. The focus of this research is the development of computational approaches to understanding the physical basis of layer-by-layer assembly (LBL), a key methodology of nanomanufacturing. The results provided detailed information on structure which cannot be obtained directly by experiments.

The model systems chosen for study are polypeptide chains. Reasons for this are that polypeptides are no less polyelectrolytes than the more usual polyions, and one can control the primary structure of a polypeptide on a residue-by-residue basis using modern synthetic methods. Several non-linear models were studied, the one-dimensional logistic map, two-dimensional coupled logistic map, three dimensional Lorenz systems with and without time delay. In areas of the country where the U.S. Department of Energy has classified the available wind resources as Class 3 or greater, the use of wind turbines to provide power to relatively small remediation systems such as groundwater circulation wells may be technically and economically feasible. Groundwater circulation wells are a good candidate technology to couple with renewable energy, because the remediation system removes contamination from the subject aquifer with no net loss of the groundwater resource, while the wind turbine does not create potentially harmful air emissions. Over 90 percent of all microprocessors are now used for real-time and embedded applications. The behavior of these applications is often constrained by the physical world. It is therefore important to devise higher-level languages and middleware that meet conventional functional requirements, as well as dependably and productively enforce real-time constraints. Our lab has used an optical coherence microscope (OCM) to assess both the structure of tissue-engineered corneal constructs and their transparency. Currently, we are not able to resolve cells versus collagen matrix material in the images produced. We would like to distinguish cells in order to determine if they are viable while growing in culture and also if they are significantly contributing to the light scattering in the tissue. A study was conducted to estimate the degree of load asymmetry in a bin with non-axial internal inserts. Internal inserts in the form of an annulus segment were attached to the wall, and their influence on vertical wall loads during centric filling and discharge in a model bin were measured. Wall and floor loads were measured in a corrugated-wall model grain bin with a diameter of 2.44 m and a height of 7.3 m filled with soft red winter wheat to a depth of 6.7 m (height-to-diameter ratio of 2.75). When a prerecorded cross-beam hologram is reconstructed (so-called edge-lit readout) with a uniform plane wave and a point source, the resulting exact solutions reveal Bessel-function-type diffracted beam profiles, which are fundamentally modified under weak propagational diffraction. The case of a profiled beam readout with propagational diffraction may be analyzed using a transfer function approach based on 2-D Laplace transforms. In a second series of investigations, dynamic readout from a cross-beam volume hologram recorded with two orthogonal uniform plane waves is considered for various dependences of the refractive index modulation with intensity. Mel frequency cepstral coefficients (MFCC) are the most widely used speech features in automatic speech recognition systems, primarily because the coefficients fit well with the assumptions used in hidden Markov models and because of the superior noise robustness of MFCC over alternative feature sets such as linear prediction-based coefficients. The authors have recently introduced human factor cepstral coefficients (HFCC), a modification of MFCC that uses the known relationship between center frequency and critical bandwidth from human psychoacoustics to decouple filter bandwidth from filter spacing. To accurately identify the site of origin of a tumor is crucial to cancer diagnosis and treatment. With the emergence of DNA microarray technologies, constructing gene expression profiles for different cancer types has already become a promising means for cancer classification. In addition to binary classification, the discrimination of multiple tumor types is also important semi-supervised ellipsoid ARTMAP (ssEAM) is a novel neural network architecture rooted in adaptive resonance theory suitable for classification tasks. ssEAM can achieve fast, stable and finite learning and create hyper-ellipsoidal clusters inducing complex nonlinear decision boundaries.
