Using mice as a model system, this study aimed to compare the effect of a high fat diet (HFD), high sugar diet (HSD) and a control diet (CD) on fetal growth and development at the time of birth (post-natal day 0 or PND 0), post-natal day 7 (PND 7) and post-natal day 14 (PND 14). Published literature has documented the effects of a negative in-utero environment on fetal development, but currently there is limited research regarding the continued effects on development after birth. A new book on the production of biofuels by Gupta and Demirbas provides technical insight into the conversion processes that could provide a range of domestic fuels from plant materials to substitute for foreign oil. It focuses primarily on conversion processes for production of cellulosic ethanol, Fischer-Tropsch diesel, pyrolysis bio-oil, and hydrothermal biocrude from biomass resources, as well as "first-generation" grain ethanol and biodiesel from vegetable oil. Silicon membranes with highly uniform nanopore sizes fabricated using microelectromechanical systems (MEMS) technology allow for the development of miniaturized implants such as those needed for renal replacement therapies. However, the blood compatibility of silicon has thus far been an unresolved issue in the use of these substrates in implantable biomedical devices. We report the results of hemocompatibility studies using bare silicon, polysilicon, and modified silicon substrates. The surface modifications tested have been shown to reduce protein and/or platelet adhesion, thus potentially improving biocompatibility of silicon. This thesis considers methods developed based on ultrasonic reflections from interfaces and stereo-optical surface strain measurements to study the mechanical characterization of materials and bodies. The ultrasonic method is used to characterize wave speed and attenuation for highly attenuating samples. The stereo-optical strain measurements are used to characterize the effects of genes on mechanical properties of bone, and the dynamic characterization of blast waves in the UNL Shock Wave Trauma Mechanics facility.

2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) inhibits antibody secretion and immunoglobulin heavy chain (IgH) expression. Our previous work has shown that a possible mechanism for inhibition of IgH expression could be inhibition of the 3’IgH regulatory region (3’IgHRR). The 3’IgHRR has four enhancer regions (hs3a; hs1,2; hs3b; hs4) which are purported to control IgH gene expression. Previously we demonstrated a sensitive inhibition by TCDD of LPS-induced 3’IgHRR activity which is correlated with an inhibition of LPS-induced hs1,2 enhancer activity; however, the hs4 enhancer showed TCDD-induced activation following the co-treatment of TCDD and LPS. This thesis presents a micromachined differential scanning calorimeter (DSC) for cellular differentiation and metabolism monitoring. The misregulation of cell physiology due to disease increases the metabolic rate of the cell and therefore its heat output. Observing or monitoring the cell's heat output will lead to a method to detect diseased cells and distinguish them from normal cells.

The calorimetric chambers of the DSC were fabricated on a silicon nitride thin film, which allows for rapid thermal equilibrium and insulation. However, new technologies need to be developed to combat an emerging generation of infectious diseases. DNA vaccination has been demonstrated to have great potential for use with a wide variety of diseases. Alone, this technology does not generate a significant immune response for vaccination, but combined with delivery by electroporation (EP), can enhance plasmid expression and immunity. Arterial restenosis limits the effectiveness of coronary stenting. Restenosis is caused by excessive tissue growth which is stimulated by arterial injury and alterations to the arterial WSS. The altered WSS results from stent-induced disturbances to the natural haelnodynamics of the artery. Recent numerical studies have predcted only minor digerences in altered WSS between different stent designs using a commonly employed threshold assessment technique. Dynamically driven microcantilevers are normally excited into resonance in the out-of-plane flexural mode. The beam's resonant frequency and quality factor are used to characterize the devices. The devices are well suited for operation in air, but are limited in viscous liquid media due to the increased viscous damping. Biogeography-based optimization (BBO) is a new evolutionary optimization method that is based on the science of biogeography. We propose two extensions to BBO. First, we propose a blended migration operator. Benchmark results show that blended BBO outperforms standard BBO. Second, we employ blended BBO to solve constrained optimization problems. Constraints are handled by modifying the BBO immigration and emigration procedures. The approach that we use does not require any additional tuning parameters beyond those that are required for unconstrained problems. This paper reports on the integration of an optical protein with single electron transistors to form a nano-bio-hybrid device for sensing. Bacteriorhodopsin (bR) is an optoelectric protein that translocates a proton across a distance of several nanometers in response to an absorbed photon of incident light. This charge gradient results in a measurable voltage in the dried state. Single electron transistors (SETs) have active regions consisting of one or more quantum islands with a size typically 10 nanometers or less. In this work, a class of DNA sequence characterization, namely computational genomics signatures, which capture global features of these sequences is used to address emerging computational biology challenges. This paper describes an ongoing collaboration between the Hope College student chapter of Engineers Without Borders – USA and the rural community of Nkuv in the Northwest Province of Cameroon related to improving drinking water quality using Manz biosand filters. The collaboration began in 2006 and focused on developing a community-based construction and distribution model for household water treatment units. The magnitude of current through a conical nanopore filled with an electrolyte solution depends on the polarity of the applied bias, indicating an asymmetric diode-like current-voltage (I-V) curve. This kind of phenomenon refers to ionic current rectification (ICR), which is of interest because many ion-channel proteins in cellular membranes are rectifying. In addition, ICR in nanopores can be used to control ion concentrations in nano- and microfluidic systems. Heat conduction between two parallel solid walls separated by liquid argon is investigated using three-dimensional molecular dynamics (MD) simulations. Liquid argon molecules confined in silver and graphite nano-channels are examined separately. Heat flux and temperature distribution within the nano-channels are calculated by maintaining a fixed temperature difference between the two solid surfaces. Temperature profiles are linear sufficiently away from the walls, and heat transfer in liquid argon obeys the Fourier law.
