Submerged And Solid-State Phytase Fermentation By Aspergillus Niger: Effects Of Agitation And Medium Viscosity On Phytase Production, Fungal Morphology And Inoculum Performance, Maria Papagianni, Sue E. Nokes, Keith Filer

Qualitative relationships between agitation levels and medium viscosity, Aspergillus niger morphology and phytase production were investigated in submerged and solid-state fermentations. Overall phytase production increased with increasing shaker speeds from 150 to 300 rpm, although specific growth rates and phytase production rates were higher at 150 rpm for 72 h from inoculation. Fungal morphology was greatly influenced by agitation with the morphological forms of small pellets and entangled mycelia predominating at 150 rpm, while the free filamentous form was obtained at 300 rpm. Upon inoculation of SSF, increased productivities were obtained from inocula grown at 150 rpm. A shift towards …

Extensive research into the management of soil acidity in Western Australia is increasing farmer awareness that lime use can reduce soil acidity and improve crop yields. However, as lime use increases, the question of lime performance and particle size is becoming more critical. Chris Gazey from the Department of Agriculture is heading up the soil acidity project, with the team comprising members from the department, CSIRO and the University of Western Australia. Mark Whitten reports on project outcomes relating to lime particle size.

The hybridization of DNA by sequential electrostatic and hydrogen-bonding immobilization of single-stranded complementary oligonucleotides at the air-water interface with cationic Langmuir monolayers is demonstrated. The complexation of the single-stranded DNA molecules with octadecylamine (ODA) Langmuir monolayers was followed in time by monitoring the pressure-area isotherms. A large (and slow) expansion of the ODA monolayer was observed during each stage of complexation in the following sequence: primary single-stranded DNA followed by complementary single-stranded DNA followed by the intercalator, ethidium bromide. Langmuir-Blodgett (LB) films of the ODA-DNA complex were formed on different substrates and characterized using quartz-crystal microgravimetry (QCM), Fourier transform infrared …

Modeling the scheduling of patient appointments is an important issue in simulating a health care delivery facility. A simulation model must include the control logic of appointment scheduling software and the explicit and implicit decision rules used by the human scheduler in selecting an appointment time. Expert systems provide one way of modeling such control logic and decision rules. We describe a structure for an expert system that models patient appointment scheduling and the integration of such an expert system within a simulation model. An example expert system for a small animal veterinary clinic is presented.

A finite deformation mixture theory is used to quantify the mechanical properties of the annulus fibrosus using experimental data obtained from a confined compression protocol. Certain constitutive assumptions are introduced to derive a special mixture of an elastic solid and an inviscid fluid, and the constraint of intrinsic incompressibility is introduced in a manner that is consistent with results obtained for the special theory. Thirty-two annulus fibrosus specimens oriented in axial (n = 16) and radial (n = 16) directions were obtained from the middle-lateral portion of intact intervertebral discs from human lumbar spines and tested in a stress-relaxation protocol. …

The Upflow Anaerobic Sludge Blanket (UASB) reactor is widely used for the anaerobic treatment of concentrated municipal / industrial wastewaters and sludges. Ability to handle high organic loading rates and cost effectiveness are often the most cited merits of the UASB process. Numerous mathematical models have been developed to describe the process and mechanistic phenomena in these systems. However, evidence in the literature of these models having been applied, either as control or diagnostic tools is limited. This thesis presents a study on the effects of antioxidants on the oxidative induction time (OIT), thermal behavior, and hardness property of ultra high molecular weight polyethylene (UHMWPE). The antioxidants were dissolved into methanol alcohol before the solution was mixed with UHMWPE powder. The mixture was degassed under vacuum, and then compression-molded into disks 1.25 in. in diameter.
Different antioxidants were evaluated at concentrations up to 0.6 percent. A 0.6 percent concentration was used to screen the effectiveness of various antioxidants. Effect of antioxidant concentration was investigated in the range between 0 and 0.6 percent. OIT was evaluated on UHMWPE …

Phytoremediation of metal-contaminated soils requires an understanding of the interactions between metal-tolerant plant species and soil chemical properties controlling the bioavailability of metals. We conducted controlled laboratory studies to investigate the effects that river birch (Betula occidentalis Hook.) and smallwing sedge (Carex microptera Mack.) had on the fate and behavior of Pb in a contaminated soil (3000 mg Pb/kg) and tailings (13 000 mg Pb/kg) collected from an abandoned mining site in Utah. Significant Pb accumulation in aboveground tissue was observed in smallwing sedge (≥1000 mg/kg dry wt.) in both the soil and tailings, but Pb was primarily excluded by …

The Air Force has instituted Barrier Reef to protect its networks. The Air Force medical community operates network connections that are incompatible with Barrier Reef. To overcome this problem, OASD(HA) directed the Tri-Service Management Program Office (TIMPO) to develop an architecture that protects all military health systems and allows them to link with all three services and outside partners. This research studied the underlying networking issues and formed a framework based on data from network experts from the Air Force's medical centers and their base network organizations. Managing the environmental risk associated with livestock production is a significant challenge. Nitrogen and phosphorus are commonly implicated as the sources of ground and surface water quality problems associated with livestock production. The degree of imbalance between these nutrient inputs and the managed nutrient outputs for a livestock operation defines the magnitude of potential environmental risk and provides insight as to the underlying causes of these challenges. A nitrogen and phosphorus balance was constructed for 33 Nebraska confinement livestock operations. Twenty-five and 17 of these operations experienced significant nitrogen and phosphorus imbalances, respectively (50% more nutrient inputs than outputs). Nutrient …

This paper highlights some of the major findings of the Alkali Deposits Investigation, a collaborative effort to understand the causes of unmanageable ash deposits in biomass-fired electric power boilers. A group of interested industrial institutions and the US DOE Energy Efficiency and Renewable Energy Office's Biomass Power Program through the National Renewable Energy Laboratory jointly sponsored the project. Properties of biomass relevant to combustion are briefly reviewed. The compositions of biomass among fuel types are variable, especially with respect to inorganic constituents important to the critical problems of fouling and slagging. Alkali and alkaline earth metals, in combination with other fuel elements such as silica and sulfur, and facilitated by the presence of chlorine, are responsible for many undesirable reactions in combustion furnaces and power boilers. Reductions in the concentrations of alkali metals and chlorine, created by leaching the elements from the fuel with water, yield remarkable improvements in ash fusion temperatures and confirm much of what is …

Biogenic Mn oxides were produced by the bacterium Leptothrix discophora SS-1 (= ATCC 3182) in a chemically defined mineral salts medium, and the Pb binding and specific surface area of these oxides were characterized. Growth of SS-1 in the defined medium with pyruvate as a carbon and energy source required the addition of vitamin B12. Complete oxidation of Mn(II) within 60 h required the addition of ≥0.1 μM FeSO4. The research described in this paper represents a part of a much broader research project with the general objective of describing the effects of elevated [CO2] and temperature on tree growth, physiological processes, and ecosystem-level processes. The specific objective of this research was to examine the below-ground respiratory responses of sugar maple (Acer saccharum Marsh.) and red maple (Acer rubrum L.) seedlings to elevated atmospheric [CO2] and temperature. Fertigation is widely practiced with drip irrigation, yet remains very unsophisticated. Major areas which need improvement are (i) making certain the irrigation system applies water with a high DU (uniformity), (ii) using proper injection pumps and protection hardware, (iii) recognizing that chemicals can interact with each other and with water to form precipitates which will clog emitters, (iv) understanding of nutrient ratios in plants and soil, (v) recognizing that nitrogen fertilization requires attention to the type of nitrogen, and (vi) developing programs for spoon-feeding chemicals. This paper covers the first 4 topics, plus discusses some specific fertilizers.

Abstract—Toxic trace metals have been implicated as a potential cause of recent flamingo kills at Lake Nakuru, Kenya. Chromium (Cr), copper (Cu), lead (Pb), and zinc (Zn) have accumulated in the lake sediments as a result of unregulated discharges and because this alkaline lake has no natural outlet. Lesser flamingos (Phoeniconaias minor) at Lake Nakuru feed predominantly on the cyanobacterium Spirulina platensis, and because of their filter-feeding mechanism, they are susceptible to exposure to particle-bound metals. Members of a panel of stress-responsive biosensors have been used to study the effect of megahertz frequency ultrasound onEscherichia coli. Insonification causes acoustic cavitation, the collapse of oscillating microbubbles in solution, which can damage bacterial cells. A focused 1-MHz ultrasound transducer, capable of generating a spatial peak pulse average intensity of 500 W/cm2, was used to treat liquid bacterial cultures. Stress-responsive promoters fused to luxCDABE allowed the continuous measurement of light produced as a result of protein damage, DNA damage, oxidative stress, and membrane perturbation. Solid-state fermentation has the potential to produce inexpensive enzymes for use in high-volume industrial applications. Process parameters such as substrate moisture content and length of fermentation can have a significant effect on the amount and timing of enzyme production. This study was conducted in two stages, a screening stage and an optimization stage, to determine the effects of moisture content of the substrate, surfactant addition upon inoculation, depth of the substrate, and duration of fermentation on xylanase activity produced by Trichoderma longibrachiatum.
