A Novel Microfluidic Device For Preclinical Testing Of Chemotherapeutics, Kaija Saleh Ogan

This work demonstrates the feasibility of a novel microfluidic tumor model that reproduces physiologically relevant nutrient gradients and endothelium–tumor interactions, validating its potential as a preclinical test platform. Finite element simulations of glutamine diffusion and fluid dynamics aligned with experimental tracer studies, and co-cultures supported directed endothelial migration toward tumor spheroids, confirming the device’s ability to mimic in vivo–like behavior.

Molecular dynamics (MD) simulation is an important tool in biochemical sciences for studying complex molecular processes and interfacial interactions at the molecular level. Biological interfaces play key roles in processes such as ocular lubrication and biomolecular adsorption. This dissertation studies interfacial phenomena involving two distinct yet conceptually related systems: the tear film lipid layer (TFLL) at the air–water interface at the surface of the human eye, and RNA adsorption on silica surfaces. Flooding has profound social, economic, and environmental impacts on local communities, and zoning plays a critical role in mitigating these risks. The 2019 Nebraska floods, among the most severe in the state’s history, highlighted the urgent need to strengthen local preparedness and mitigation strategies. This study assesses the extent to which local zoning regulations in Nebraska’s most affected jurisdiction, including rural areas, address flood risk reduction. Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype with limited treatment options due to the absence of estrogen (ER), progesterone (PR), and human epidermal growth factor receptor 2 (HER2) receptors. We previously developed highly adaptable polyamidoamine (PAMAM) dendrimer-based gel nanoparticles (GDP-uPA/GTI) with dual-targeting capabilities against urokinase-type plasminogen activator receptor (uPAR) and ribonucleotide reductase R2 (R2) to achieve selective antitumor effects. In this study, we investigated the underlying mechanisms of GDP-uPA/GTI-induced tumor inhibition. In vivo, GDP-uPA/GTI significantly downregulated R2 expression and activated caspase 8 and caspase 3, indicating caspase-dependent apoptosis. Histological evaluation of major organs revealed no observable toxicity. …

Two sets of model performance based on the application are selected namely a low resource and a high resource setting. High-friction surface treatments (HFSTs) are widely applied to improve pavement safety by enhancing long-term skid resistance. Although epoxy resins are commonly used due to their strength and durability, their high cost, susceptibility to delamination, incompatibility with substrates of flexible pavements, and adverse environmental concerns limit their long-term performance. This study presents crumb rubber modified (CRM) asphalt as a sustainable alternative binder for HFST applications. Farmers face the critical challenge of monitoring cattle well-being for both ethical and economic success, relying on signals like body temperature and facial expressions to assess their animals' health. However, these indicators have traditionally relied on human observation with manual measurement, which is time-consuming and subjective. This thesis focuses on using Multi-Criteria Decision-Making (MCDM) methods to support sustainable product design by addressing the trade-offs between environmental, economic, and social goals. The research is presented through two studies that demonstrate how both subjective and objective decision-making approaches can guide sustainability evaluations.

The first study applies the Analytic Hierarchy Process (AHP) to assess residential solar panels by incorporating stakeholder perspectives from sustainability experts, end users, and industry professionals. In Chapter 1, we consider a network maintenance problem which focuses on time-based redeployment of multi-class nodes for reliable wireless sensor network coverage. Hispanic workers face higher injury and fatality rates, with miscommunication being a major contributing factor. Existing translation tools lack the accuracy, cultural context, and jobsite relevance needed for effective safety communication.

Significance: Accurate transfer of annotations from histological images to fluorescence images is essential in developing deep learning (DL)-based optical imaging systems for intraoperative assessment of tumor margins. Manual annotation is time-consuming, prone to interobserver variability, and impractical for large-scale datasets.

Aim: We present a semi-automated method that can effectively transfer tumor annotations from pathologist-annotated hematoxylin and eosin (H&E) images to fluorescence images captured using microscopy with ultraviolet surface excitation (MUSE). This thesis investigates multi-port solid-state circuit breaker (M-SSCB) architectures that reduce steady-state conduction losses while preserving microsecond-class DC fault interruption and per-port selectivity. The proposed M-SSCB consolidates protection for several node interfaces into a single assembly with shared sensing, control, and a common energy-absorption branch. In normal operation, two electrically symmetric parallel semiconductor paths are established so each port’s current divides approximately in half; because conduction loss scales with current squared, the M-SSCB achieves ≈75% reduction in on-state loss relative to a conventional anti-series SiC path without increasing device count. The expansion of solar photovoltaic energy infrastructure is transforming landscapes worldwide. Increasingly, facilities are planted with native vegetation, yet how vegetation characteristics and landscape variables shape wildlife communities in solar facilities remains poorly understood. Because avian populations have been in decline and are sensitive to habitat changes, understanding how solar facilities influence avian communities is important. In Chapter 1, we used autonomous recording units, local vegetation measurements, and land use and land cover data to quantify how avian community occupancy is influenced by solar cover in solar facilities and comparable reference sites. Volume Status Detection With Integral Pulse Frequency Modulation Model Of Peripheral Venous Pressure, Jeremiah Rhys Wimer

This thesis studies the effectiveness of utilizing a modified integral pulse frequency modulation (IPFM) algorithm to synthesize peripheral venous pressure (PVP) waveforms for the detection of several physiological conditions using logistic regression. PVP waveforms are collected from 18 human patients and 4 porcine subjects. Human data are used to train models for determining the hypovolemic status of the patient, while the porcine data are used to train models for determining level of anesthesia and detecting internal hemorrhaging.
