DNA is a re-configurable, biological information-storage unit, and much remains to be learned about its heterogeneous structural dynamics. For example, while it is known that molecular dyes templated onto DNA exhibit increased photostability, the mechanism by which the structural dynamics of DNA affect the dye photophysics remains unknown. Here, we use femtosecond, two-dimensional electronic spectroscopy measurements of a cyanine dye, Cy5, to probe local conformations in samples of single-stranded DNA (ssDNA–Cy5), double-stranded DNA (dsDNA–Cy5), and Holliday junction DNA (HJ–DNA–Cy5). Background

This work aims to present a fast, affordable, and reproducible three-cell co-culture system that could represent the different cellular mechanisms of atherosclerosis, extending from atherogenesis to pathological intimal thickening.

Methods and results

Airports operating under the Code of Federal Regulations Part 139 should conduct a wildlife hazard assessment (WHA) when some wildlife-strike events have occurred at or near the airport. The WHA must be conducted by a Qualified Airport Wildlife Biologist (QAWB). The required elements in a WHA include the identification of the wildlife species observed and their numbers, and the location of features on and near the airport that could attract wildlife. The proposed presentation will include the following project and results from implementation in an aircraft hydraulic systems course.

In 2020 we developed a digital hydraulics system schematic sandbox for use in an FAA Part 147 airframe maintenance technician training program (See Figure 1). The sandbox allows students to drag-drop-connect components used in systems such as those found in aircraft. It allows them to ‘test-connect’ the components to get feedback on correct and incorrect ‘construction’. Using energy harvesting to convert ambient vibrations efficiently to electrical energy has become a worthy concept in recent years. Nevertheless, the low frequencies of the ambient vibrations cannot be effectively converted to power using traditional harvesters. Therefore, a frequency up-conversion harvester is presented to convert the low-frequency vibrations to high-frequency vibrations utilizing magnetic coupling. The presented harvester consists of a low-frequency beam (LFB) and a high-frequency beam (HFB) with identical tip magnets facing each other at the same polarity. Imagery from drones is becoming common in wildlife research and management, but processing data efficiently remains a challenge. We developed a methodology for training a convolutional neural network model on large-scale mosaic imagery to detect and count caribou (Rangifer tarandus), compare model performance with an experienced observer and a group of naïve observers, and discuss the use of aerial imagery and automated methods for large mammal surveys. During a move to the new drilling location, two braces of the upper mast section on a 550 HP oil and gas rig was damaged. When damage is discovered, the rig operating company should conduct a risk analysis related to the damage using the standard used to determine rejection criteria and follow-up repairs. The strength of the rig mast is calculated using Finite Element Analysis (FEA) with initial conditions (before damage occurs) where all braces are installed and extreme conditions where neither brace is repaired. Membrane flux was observed and the two membrane sizes that achieved stable flux were chosen to be used in the later stages of the study. The chosen membranes were of 300 kDa and 400 kDa Molecular Weight Cutoff (MWCO).

The third phase of the study involved running the full system on synthetic greywater then on real greywater. The parameters investigated were COD, TSS, Turbidity and BOD5. In this study, copper-loaded activated alumina (Cu/AA) was synthesized and used for the CWPO of catechol (a representative refractory organic pollutant). Various characterization techniques were deployed to characterize the catalysts, e.g., activated alumina (AA), as well as pristine and spent 1% Cu/AA. The innovative 1% Cu/AA catalyst exhibited good thermal stability up to 1173 K with a marginal weight loss of 13%. The Cu species were well dispersed on the activated alumina framework with no significant cluster formation. Typically, the average copper particle size of 5 nm was dispersed on the AA framework. Curcumin is a natural polyphenol that exhibits remarkable antioxidant and anti-inflammatory activities; however, its clinical application is limited in part by its physiological instability. Here, we report the synthesis of curcumin-derived polyesters that release curcumin upon hydrolytic degradation to improve curcumin stability and solubility in physiological conditions. Curcumin was incorporated in the polymer backbone by a one-pot condensation polymerization in the presence of sebacoyl chloride and polyethylene glycol (PEG, Mn = 1 kDa). The thermal and mechanical properties, surface wettability, self-assembly behavior, and drug-release kinetics all depend sensitively on the mole percentage of curcumin incorporated in these statistical copolymers. Diagnostic blood tests can guide the administration of healthcare to save and improve lives. Most clinical biosensing blood tests require a trained technician and specialized equipment to process samples and interpret results, which greatly limits test accessibility. Colorimetric paper-based diagnostics have an equipment-free readout, but raw blood obscures a colorimetric response which has motivated diverse efforts to develop blood sample processing techniques. This thesis presents a path planning framework for a very-large-scale robotic (VLSR) system in an known obstacle environment, where the time-varying distributions of agents are applied to represent the multi-agent robotic system (MARS). A novel family of the multivariate skew-normal (MVSN) distributions is proposed based on the Bernoulli random field (BRF) referred to as the Bernoulli-random-field based skew-normal (BRF-SN) distribution. Myotonia congenita is an inherited skeletal muscle disorder caused by loss-of-function mutation in the CLCN1 gene. This gene encodes the ClC-1 chloride channel, which is almost exclusively expressed in skeletal muscle where it acts to stabilize the resting membrane potential. Loss of this chloride channel leads to skeletal muscle hyperexcitability, resulting in involuntary muscle action potentials (myotonic discharges) seen clinically as muscle stiffness (myotonia). Stiffness affects the limb and facial muscles, though specific muscle involvement can vary between patients. With its incorporation into clinical practice in the early 1980s, the class of pharmacological agents known as calcineurin inhibitors (CNIs) quickly became the cornerstone of immunosuppressive therapy post-organ transplantation. However, its use is limited by irreversible kidney damage in the form of renal fibrosis. The molecular mechanism by which CNIs induce renal fibrosis remains to be better understood, and to date, there are no specific therapeutic strategies to mitigate this damage. Our research seeks to develop a long-lasting and high-quality en- gagement between the user and the social robot, which in turn requires a more sophisticated alignment of the user and the system than is currently commonly available. Close monitoring of inter- locutors’ states, and we argue their confusion state in particular, and adjusting dialogue policies based on this state of confusion is needed for successful joint activity. The imbalance in the gut microbiome plays a vital role in the progression of many diseases, including cancer, due to increased inflammation in the body. Since gut microbiome-induced inflammation can serve as a novel therapeutic strategy, there is an increasing need to identify novel approaches to investigate the effect of inflammation instigated by gut microbiome on cancer cells. Yttria-stabilized tetragonal zirconia polycrystal (Y-TZP) has gained popularity as the choice of material for dental prosthetics. Ivoclar Vivadent’s IPS e.max ZirCAD Prime dental ceramic incorporates a unique gradient technology that varies the yttria content over the thickness of the material. The top layer is composed of 5Y-TZP which is desired for its optical properties while the bottom layer is composed of a much stronger 3Y-TZP. In between the two layers, 5Y-TZP and 3Y-TZP are mixed to form a transition layer. Background
The COVID-19 pandemic of 2020-2022 caused a rapid shift away from face-to-face instruction and toward online instruction. Due to the emergency speed with which the changes were rolled out results varied, desirable data on the quality of the changes was often not kept, and the retrospective study of what happened and the outcomes thereof is ongoing at the time this case study was written.
Purpose/Hypothesis
