A genetic means of identifying "pregnancy competent" oocytes is provided. The means comprises detecting the level of expression of one or more genes that are expressed at characteristic levels (upregulated or downregulated) in cumulus cells derived from pregnancy competent oocytes. This characteristic gene expression level, or pattern referred to herein as the "pregnancy signature", also can be used to identify subjects with underlying conditions that impair or prevent the development of a viable pregnancy, e.g., pre-menopausal condition, other hormonal dysfunction, ovarian dysfunction, ovarian cyst, cancer or other cell proliferation disorder, autoimmune disease and the like. In preferred embodiments the …

Missouri River Recovery Program: Reclaiming Lost Habitat: Real Estate Program Jan 2009

Missouri River Recovery Program: Reclaiming Lost Habitat: Real Estate Program

United States Army Corps of Engineers, Omaha District

Historically, the Missouri River continually reshaped itself by eroding banks in some areas and building up islands and sandbars in other areas. The river’s dynamic nature provided plenty of habitat for native species of animals and plants.

Efforts in the 1900s to manage the river by building dams, levees and a navigation channel provided many social and economic benefits to the nation but drastically reduced the amount of habitat available for the river’s native species. Today, lack of habitat is a major reason that several Missouri River species are endangered or threatened, such as the pallid sturgeon, least tern and …

Antiproliferation Properties Of Grain Sorghum Dry Distiller’S Grain Lipids In Caco-2 Cells, Richard Zbasnik, Timothy P. Carr, Curtis L. Weller, Keum Taek Hwang, Lijun Wang, Susan L. Cuppett, Vicki Schlegel

Department of Agricultural and Biological Systems Engineering: Faculty Publications

Antiproliferative properties of lipids extracted from grain sorghum (GS) dry distiller’s grain (DDG) were analyzed to determine the feasibility of developing GS coproducts as a source for human health dietary ingredients. The lipid extract of GSDDG was delivered to human colon carcinoma (Caco-2) cells by solubilizing 0−1000 μg/mL of GS-DDG lipids in 100 μg/mL increments with micelles. A significant reduction in cell viability (25−50%) resulted at treatment levels of 400−1000 μg/mL GS-DDG lipids (p < 0.05). Alternatively, total protein levels of cells treated with 400, 500, and 600 μg/mL of GS-DDG lipid were not significantly different from the control, indicating cell growth during the treatment period. Total cell counts for the control were not significantly different from the GS-DDG lipid treated cells, but dead cell counts increased by 10% for the latter sample with a concomitant increase of the intercellular protein lactate dehydrogenase leakage (30−40%) in the medium. Preliminary analysis by the fluorescence-activated cell method (FACs) demonstrated that nonviable cells were in either the early apoptotic, late apoptotic, or necrotic stage post-treatment with 400, 500, and 600 μg/mL GS-DDG lipids. Physiochemical characterization of the GS-DDG lipids used for the antiproliferation study showed the presence of vitamin E (predominantly γ-tocopherol), triacylglycerides (predominantly linoleic acid), policosanols, aldehydes, and sterols (predominantly campesterol and stigmasterol), each of which or as synergistic/additive group of constituents may be responsible for the antiproliferative effect.

Integration Of An Extended Octagonal Ring Transducer And Soil Coulterometer For Identifying Soil Compaction, Santosh Pitla, L. G. Wells, S. A. Shearer

Department of Agricultural and Biological Systems Engineering: Faculty Publications

The soil coulterometer is an “on‐the‐go” electro‐mechanical system which collects impedance force data at multiple depths using an oscillating coulter. During the initial testing (summer 2006), only vertical soil impedance force data was collected using a pressure sensor. To improve the performance of the coulterometer, an extended octagonal ring transducer was integrated into the system to collect both the horizontal and vertical impedance forces given by the soil. In the summer of 2007, data was collected using the revised sensor from a typical central Kentucky field setting in a 0.8‐ha (2‐acre) plot. Four passes were made with the coulterometer. Seventy …

N-Glycosylation Microheterogeneity And Site Occupancy Of An Asn-X-Cys Sequon In Plasma-Derived And Recombinant Protein C, Geun-Cheol Gil, William H. Velander, Kevin E. Van Cott

Kevin Van Cott Publications

Human protein C (hPC) is glycosylated at three Asn-X-Ser/Thr and one atypical Asn-X-Cys sequons. We have characterized the micro- and macro-heterogeneity of plasma-derived hPC and compared the glycosylation features with recombinant protein C (tg-PC) produced in a transgenic pig bioreactor from two animals having approximately tenfold different expression levels. The N-glycans of hPC are complex di- and tri-sialylated structures, and we measured 78% site occupancy at Asn-329 (the Asn-X-Cys sequon). The N-glycans of tg-PC are complex sialylated structures, but less branched and partially sialylated. The porcine mammary epithelial cells glycosylate the Asn-X-Cys sequon with a similar efficiency as human hepatocytes …

Development Of Measurement And Modeling Techniques To Quantify Atmospheric Deposition Of Persistent, Bioaccumulative And Toxic Chemicals In The Great Lakes, Mark D. Rowe

Dissertations, Master's Theses and Master's Reports - Open

Measurement and modeling techniques were developed to improve over-water gaseous air-water exchange measurements for persistent bioaccumulative and toxic chemicals (PBTs). Analytical methods were applied to atmospheric measurements of hexachlorobenzene (HCB), polychlorinated biphenyls (PCBs), and polybrominated diphenyl ethers (PBDEs). Additionally, the sampling and analytical methods are well suited to study semivolatile organic compounds (SOCs) in air with applications related to secondary organic aerosol formation, urban, and indoor air quality.

A novel gas-phase cleanup method is described for use with thermal desorption methods for analysis of atmospheric SOCs using multicapillary denuders. The cleanup selectively removed hydrogen-bonding chemicals from samples, including much of …

Long-Range Pollution Transport: Trans-Atlantic Mechanisms And Lagrangian Modeling Methods, Robert Christopher Owen Jan 2009

Long-Range Pollution Transport: Trans-Atlantic Mechanisms And Lagrangian Modeling Methods, Robert Christopher Owen

Dissertations, Master's Theses and Master's Reports - Open

Over the past several decades, it has become apparent that anthropogenic activities have resulted in the large-scale enhancement of the levels of many trace gases throughout the troposphere. More recently, attention has been given to the transport pathway taken by these emissions as they are dispersed throughout the atmosphere. The transport pathway determines the physical characteristics of emissions plumes and therefore plays an important role in the chemical transformations that can occur downwind of source regions. For example, the production of ozone (O3) is strongly dependent upon the transport its precursors undergo. O3 can initially be formed …

Efficient and effective analysis of the growing genomic databases requires the development of adequate computational tools. We introduce a fast method based on the suffix tree data structure for predicting novel targets of hypoxia-inducible factor 1 (HIF-1) from huge genome databases. The suffix tree data structure has two powerful applications here: one is to extract unknown patterns from multiple strings/sequences in linear time; the other is to search multiple strings/sequences using multiple patterns in linear time. Using 15 known HIF-1 target gene sequences as a training set, we extracted 105 common patterns that all occur in the 15 training genes …

An Experimental And Numerical Investigation Of Flapping And Plunging Wings, Taylor A. Swanson

Doctoral Dissertations

"Micro air vehicles, or MAVs, are of current interest for a multitude of uses to which they, being small, unmanned vehicles, are uniquely suited. Among the proposed uses are exploration, reconnaissance, and communications. They can be deployed inside buildings, where their small size, hovering capability, and maneuverability, are important factors. Due to their small size, they operate at low Reynolds numbers where conventional flying mechanisms are not advantageous. Thus, attempts have been made to learn from natural flyers like insects and birds. Natural flight is accomplished by flapping wings, and this idea has been proposed for certain types of MAVs …

Spectroscopic Characterizations Of Organic/Inorganic Nanocomposites, Jayesh Rasikalal Govani

Open Access Theses & Dissertations

Inorganic potassium dihydrogen phosphate (KDP) is widely known for its value as a nonlinear optical material. In this study, pure and l-arginine-doped KDP single crystals were grown by the slow solvent evaporation technique and further subjected to infrared absorption and Raman studies for the confirmation of chemical group functionalization and possible bonding between the organic and inorganic materials. The appearance in the infrared absorption spectra of additional vibrational lines, which mostly originate from disturbed N-H, C-H, and C-N bonds of the l-arginine-doped salt, confirm the interaction between KDP and the organic material. This affirmation is supported by more evidence from …

Regeneration Of Damaged Growth Plate Using Igf-I Plasmid-Releasing Porous Plga Scaffolds, Nirmal Ravi

University of Kentucky Doctoral Dissertations

Growth plate injuries account for 15-30% of long bone fractures in children. About 10% of these result in significant growth disturbances due to formation of a boney bar. If not treated correctly, this can lead to life-lasting consequences of limb length inequalities and angular deformities. Current treatments for growth plate injuries include removal of boney bar and insertion of fat, silicone, bone cement, etc.. This treatment y is inadequate, leaving almost half of these patients with continued deformities. This dissertation reports characterization of a DNA–containing porous poly(lactic-co-glycolic acid) (PLGA) scaffold system, chondrogenesis using insulin-like growth factor I (IGF-I) plasmid-releasing scaffolds …

Renewable Energy Resources And Technologies Applicable To Ireland, Fergal O'Rourke, Fergal Boyle, Anthony Reynolds

Articles

The energy consumed in Ireland is primarily achieved by the combustion of fossil fuels. Ireland’s only indigenous fossil fuel is peat; all other fossil fuels are imported. As well as fossil fuels continually becoming more expensive, their use as an energy source also has a negative impact on the environment. Ireland’s energy consumption can be separated into 3 divisions; transportation, electricity generation and heat energy. Ireland however has a vast range of high quality renewable energy resources. Ireland has set a target of 33% of its electricity will be generated from renewable sources by 2020[1]. The use of biomass, wind …
