The electrochemical activity of glycopyrrolate was studied. Different voltammetric techniques were applied in this study, including cyclic voltammetry (CV), differential pulse voltammetry (DPV), and linear sweep voltammetry (LSV). Glassy carbon and platinum working electrodes were utilized. CV indicated that glycopyrrolate has a reversible redox reaction on the working electrode surface, with an anodic peak current at approximately 1.05 V and a cathodic peak at 0.80 V. Several parameters that affect the sensitivity of these methods were optimized for the quantitation of glycopyrrolate. LSV showed a better correlation coefficient than DPV did, with a value of ca. 0.9990 in the range …

An enzymeless electrochemical sensor for detection of low amounts of H$_{2}$O$_{2}$ with the aid of Ag nanoparticles supported on conducting poly(vinylferrocene) (PVF) film was developed. Experimental results revealed that contribution of Ag nanoparticles led to remarkable improvement by means of reduction potential and reduction current. Influence of experimental parameters (i.e. polymeric film thickness, concentration of Ag precursor, immersion time in precursor solution, reduction time, and reduction potential) were investigated. The Ag/PVF-modified electrode system was characterized physically by scanning electron microscopy. Picornaviruses are small, positive-stranded RNA viruses, divided into twelve different genera. Members of the Picornaviridae family cause a wide range of human and animal diseases including the common cold, poliomyelitis, foot and mouth disease, and dilated cardiomyopathy. The picornavirus genome is replicated via a highly conserved mechanism involving a presumed cloverleaf structure located at the 5’ noncoding region of the virus genome. The 5’ cloverleaf consists of three stem loops (B, C, and D) and one stem (A), which interact with a variety of virus and host cell proteins during replication. Low molecular-weight gelators (LMWGs) are a fascinating and interesting class of compounds that can self-assemble and form supramolecular structures in appropriate solvents. The gelators form fibrous networks which entrap the solvent and result in a supramolecular gel. Carbohydrate-derived LMWGs are especially useful since they are more likely to be biocompatible and biodegradable. Dendritic gelators are branch-shaped molecules with precise molecular weight that are between small molecules and polymer gelators. Glycoclusters with sugar moieties at the periphery of the molecules are important classes of bioactive compounds due to the multivalency effect. Biochar is the carbon rich solid by-product of biomass pyrolysis. Interest in biochar can be broken down to several main categories: use as a carbon sequestration agent, use as a medium for the removal via adsorption of unwanted materials in wastewater, and as a soil amendment for the increase of cation exchange capacity (CEC). In order to generate a biochar which is stable enough for carbon sequestration, higher temperature pyrolysis must be used in order to ensure a lower O:C ratio in order to increase the half-life of biochar in soil. "The first courses freshman university students typically enroll in are the introductory science and math, courses that bridge from and build upon their prior educational experiences. These introductory courses often have large enrollment lectures coupled with supplemental sessions to teach using traditional educational practices, which may operate counter to the attitudes and culture of the students who take them. "NMR spectroscopy is a powerful analytical tool to obtain explicit information about molecular structures, conformations, dynamics, and functions. To extract the desired information most accurately, NMR reference standards and devices are needed for calibrating the spectrometer hardware, generating reference data for the NMR software, and testing the applicability of pulse sequences. In this dissertation, the CapPack (Capillary-tube Package) platform is introduced which provides well-defined external reference and calibration standards for liquid and solid-state NMR applications. "Novel small molecule inhibitors and breakers of advanced glycation end-products (AGEs) have been synthesized and their in vitro inhibitory activities have been studied by 13C NMR, fluorescence, and UV-visible spectroscopy. We have demonstrated that these AGE-inhibitors and AGE-breakers were found to be more effective in their sequestration of AGE intermediates, such as dehydroascorbate (DHA) than the state-of-the art phenacylthiazolium bromide (PTB)-based AGE-inhibitors and AGE-breakers. Citric acid and other polyphenolic antioxidants have substantial AGE inhibitory effects in the D-glucose/leucine or benzylamine Maillard reaction model systems, and thus they are suitable as dietary additives for ameliorating AGE-induced complications.

Mycobacterium tuberculosis is the etiologic agent of tuberculosis. The demand for new chemotherapeutics with unique mechanisms of action to treat (multi)resistant strains is an urgent need. The objective of this work was to test the effect of manganese(II) and copper(II) phenanthroline/dicarboxylate complexes against M. tuberculosis. The use of plastics has spanned across almost all aspects of day to day life. Although their uses are invaluable, they contribute to the generation of a lot of waste products that end up in the environment and end up polluting natural habitats such as forests and the ocean. The discovery of the high-affinity, high-specificity folate receptor in mamalian kidney cells, coupled with the ability of folate to enter cells by folate receptor-mediated endocytosis and the subsequent elucidation of the folate receptor’s overexpression in specific cancer cell types; heralded the arrival of the area of chemotherapeutic folate targeting. The application of purely organic folate-based small-molecule drug conjugates that selectively target the folate receptor, which is over expressed in several diseases such as cancer, is well established. The measurement of pH is undertaken frequently in numerous settings for many applications. The common glass pH probe is almost ideal for measuring pH, and as such, it is used almost ubiquitously. However, glass is not ideal for all applications due to its relatively large size, fragility, need for recalibration and wet-storage. Therefore, much research has been undertaken on the use of metal oxides as an alternative for the measurement of pH.

Increased understanding of the importance of the affective domain in chemistry education research has led to the development and adaptation of instruments to measure chemistry-specific affective traits, including motivation. Many of these instruments are adapted from other fields by using the word ‘chemistry’ in place of other disciplines or more general ‘science’ wording. Psychometric evidence is then provided for the functioning of the new adapted instrument. When an instrument is adapted from general language to specific (e.g. replacing ‘science’ with ‘chemistry’), an opportunity exists to compare the functioning of the original instrument in the same context as the adapted instrument. …

Although paraCEST is a method with immense scope for generating image contrast in MRI, it suffers from the serious drawback of high detection limits. For a typical discrete paraCEST agent the detection limit is roughly an order of magnitude higher than that of a clinically used relaxation agent. One solution to this problem may be the incorporation of a large payload of paraCEST agents into a single macromolecular agent. The X-ray crystal structure of the dapE-encoded N-succinyl-l,l-diaminopimelic acid desuccinylase from Haemophilus influenzae (HiDapE) bound by the products of hydrolysis, succinic acid and l,l-DAP, was determined at 1.95 Å. Surprisingly, the structure bound to the products revealed that HiDapE undergoes a significant conformational change in which the catalytic domain rotates ∼50° and shifts ∼10.1 Å (as measured at the position of the Zn atoms) relative to the dimerization domain. This heretofore unobserved closed conformation revealed significant movements within the catalytic domain compared to that of wild-type HiDapE, which results in effectively closing off access to …

Speciation of copper in marine systems strongly influences the ability of copper to cause toxicity. Natural organic matter (NOM) contains many binding sites which provides a protective effect on copper toxicity. The purpose of this study was to characterize copper binding with NOM using fluorescence quenching techniques. Fluorescence quenching of NOM with copper was performed on nine sea water samples. The resulting stability con- stants and binding capacities were consistent with literature values of marine NOM, show- ing strong binding with log K values from 7.64 to 10.2 and binding capacities ranging from 15 to 3110 nmole mg C −1 …

We measured fluxes of methane, a suite of non-methane hydrocarbons (C2-C11), light alcohols, and carbon dioxide from oil and gas produced water storage and disposal ponds in Utah (Uinta Basin) and Wyoming (Upper Green River Basin) United States during 2013-2016. In this paper, we discuss the characteristics of produced water composition and air-water fluxes, with a focus on flux chamber measurements. Noncovalent interactions play an important role for the design of novel drugs, better catalysts, synthesis of complex supramolecular structures, and so on. To develop new materials, a well-founded knowledge of how to control the strength of these interactions is desirable. Despite the many investigations done so far, a quantitative assessment of the intrinsic strength of most types of noncovalent interactions is still missing. Multidrug resistant (MDR) Gram-negative bacteria represent a global health crisis. Traditional antibiotics create selective pressure and breed resistance, making our current drugs less and less effective. Targeting of bacterial virulence factors has gained interest as an alternative strategy to potentially circumvent this problem. Virulent Acinetobacter baumannii synthesize and secrete pre-acinetobactin, a small molecule metal chelator that scavenges life-sustaining iron and is critical for establishing infection. The work described herein sheds light on the unusual properties of a non-enzymatic isomerization from pre-acinetobactin to acinetobactin and proposes a siderophore-swapping mechanism by which A. baumannii expands its pH window for virulence. A series of square-planar metal (diimine)(dichalcogenolene) chromophoric complexes have been synthesized, characterized and studied spectroscopically. Studies conducted on diimineplatinum(II) dichalcogenolene complexes (LPtL’) using time resolved spectroscopic techniques such as transient absorption and emission, reveal a charge-separated excited state of the type dichalcogenolene → diimine charge-transfer (LL’CT) [(dichalcogenolene•+)Pt(diimine•-) with an open shell donor-acceptor biradical character. This makes the low-energy ligand-to-ligand charge-transfer (LL’CT), or the mixed-metal ligand-to-ligand charge-transfer (MMLL’CT), transitions of diimineplatinum(II) dichalcogenolene complexes particularly interesting to us in facilitating our knowledge of biradical charge-separated states. The vitreous humor of the eye is a biological hydrogel principally composed of fibrillary collagen interspersed with semi-flexible polyelectrolyte, hyaluronic acid (HA). Certain pathological conditions necessitate its removal and replacement. Current vitreous substitutes, such as silicone oils and perfluorocarbons, are hydrophobic, do not resemble the properties of the vitreous, and have known complications. An ideal vitreous substitute should have properties of the natural vitreous, perform its functions, and be biocompatible in the eye. New and advanced opportunities are arising for the synthesis and functionalization of membranes with selective separation, reactivity, and stimuli-responsive behavior. One such advancement is the integration of bio-based channels in membrane technologies. By a layer-by-layer (LbL) assembly of polyelectrolytes, outer membrane protein F trimers (OmpF) or “porins” from Escherichia coli with central pores ∼2 nm in diameter at their opening and ∼0.7 × 1.1 nm at their constricted region are immobilized within the pores of poly(vinylidene fluoride) microfiltration membranes, in contrast to traditional ruptured lipid bilayer or vesicle processes. Tryptophan 7-halogenase catalyzes chlorination of free tryptophan to 7-chlorotryptophan, which is the first step in the antibiotic pyrrolnitrin biosynthesis. Many biologically and pharmaceutically active natural products contain chlorine and thus, an understanding of the mechanism of its introduction into organic molecules is important. Whilst enzyme-catalyzed chlorination is accomplished with ease, it remains a difficult task for the chemists. Therefore, utilizing enzymes in the synthesis of chlorinated organic compounds is important, and providing atomistic mechanistic insights about the reaction mechanism of tryptophan 7-halogenase is vital and timely. In this work, we report the direct correlation of photoinduced carrier dynamics and electronic structure of CuInS2 (CIS) nanoparticles (NPs) using the combination of multiple spectroscopic techniques including steady-state X-ray absorption spectroscopy (XAS), optical transient absorption (OTA), and X-ray transient (XTA) absorption spectroscopy. XAS results show that CIS NPs contain a large amount of surface Cu atoms with ≪four-coordination, which is more severe in CIS NPs with shorter nucleation times, indicating the presence of more Cu defect states in CIS NPs with smaller size particles. Using the combination of OTA and XTA spectroscopy, we show that electrons are trapped …
