Long-chain aldehyde compounds such as hexanal, heptanal, octanal and nonanal are regarded as potential biomarkers of many lung cancer disease and served as the important quality indicators of fat and oil products. As advantage of the hydrophobic tails of hydrocarbon chain and hydrophilic head of aldehyde, we expected that long-chained aldehydes might act as the induced-fit surfactants to form the completely self-assembling organized nanomicelle. Fluorescent probes are great tools for visualization and imaging of reactive oxygen species (ROS) in living cells. Although several probes perform well with green emission, however, to maximize light penetration for tissue imaging and minimize interference from auto-fluorescence, a probe with longer wavelength is desirable. In this study, three new boron dipyrromethene (BODIPY)-based fluorescent probes SB1, SB2 and SB4 have been developed for detection of ROS. Each probe has two styryl groups anchored to the BODIPY pyrrole rings to extend the emission wavelength into the red region (634–662 nm). It has been reported the spherical shape of oxidized carbon nanoparticulates shows higher ability to penetrate phospholipid bilayer membrane than tubular and sheet shapes. However, the effect of the particle size on this membrane penetration ability have never been investigated. In this work, we synthesized various sizes of oxidized carbon spheres through a four steps process. Firstly, we adjusted the surfactants and styrene concentrations to prepare polystyrene of various sizes. Then, the polystyrene particles of selected sizes were hypercrosslinked via Frieldel-Craft alkylation reaction. Novel triazolium, imidazolium, and benzimidazolium salts are known to possess anti-cancer properties and have potential as cancer therapeutics. This thesis describes the aqueous solubility and the differential activity of N,AT-bis-substituted-l,2,4-triazolium bromide salts and N/N'-bis(naphthylmethyl)imidazolium bromide salts against MDA-MB-468 breast cancer cells and PC-3 prostate carcinomas. Using a microscale technique, the partition coefficient (log P) of each salt was determined. The log P value was explored to determine the bioavailability of each derivative.

Quantitative assessment of biomarkers is essential in numerous contexts from decision-making in clinical situations to food quality monitoring to the interpretation of life-science research findings. However, appropriate quantitation techniques are not as widely addressed as detection methods. One of the major challenges in biomarker’s quantitation is the need to have a calibration for correlating a measured signal to a target amount. Regioisomers of the functional group of the main ligand (L) on a series of [Ru(phen)2L]2+and [Ru(bpy)2L]2+ complexes, where phen is 1,10 phenanthroline and bpy is 2,2′-bipyridine, were synthesised to investigate the interaction with deoxyribonucleic acid (DNA) as potential therapeutics. UV–Vis binding titrations, thermal denaturation and circular dichroism were used to evaluate their interaction with DNA. The conclusions indicated the significance of the auxiliary ligand; especially 1,10-phenanthroline has on the binding constants (Kb). The end of the human chromosome is protected by telomeres which contain a special tandem guanine-rich DNA sequence, 5’-TTAGGG. The length of telomeres is shortened during cell replications, and its length limits the replication capacity of cells. Telomerase is over-expressed in 85–90% of cancer cells, responsible for extending the telomere length in cancer cells. Guanine-rich DNA sequence can self-assemble into unique G-quadruplex structures that interfere with the extension of telomeres by telomerase. Metal N-heterocyclic carbene (NHC) complexes have recently gained much popularity due to their tunable, steric, and electronic properties. Applications for such versatile molecules include organocatalysis, olefin metathesis, sundry cyclization reactions, and materials chemistry. Redox active NHCs are of special interest due to their ability to alter the electronic properties of the metal centers they are ligated to. In the first chapter, Au(I)-NHC complexes were synthesized and tested for biological activity in human cancer cell lines. Ion-selective electrodes (ISEs) have extensively been used for food analysis, as medical diagnostic tools, and for some environmental applications. However, ISEs are hindered by the need of a bulky reference electrode and the pre-conditioning step of the sensor, which can often be lengthy. This work will discuss how the direct addition of target analyte into the cocktail sensing membrane can circumvent the pre-conditioning step. This dissertation is an integration of fundamental research and chemical education. It begins with two nonlinear spectroscopic studies of compounds important to the study of brain chemistry. In Chapter 2, we present a novel method using quantum mechanics for modelling ligand docking and the potential of nonlinear circular dichroism for elucidating the mechanism of cannabinoids docking to their receptor, a contribution to studies of varying psychological effects of cannabinoids. This dissertation is an investigation of the nanoscale characteristics and mechanism of electrolessly deposited silver metal seeded by gold nanoparticles. The process of growing seed-nanoparticles on a polymer surface was studied. Several bifunctional amines and organic reducing agents were used to explore how these chemical factors affect the size and distribution of gold nanoparticles formed at the interface. The nanoparticles were characterized by transmission electron microscopy (TEM) and high-angle annular dark-field scanning transmission electron microscopy (HAADF-STEM). Nucleic acid amplification tests (NAATs) enable sensitive detection of low density infections that microscopy and rapid diagnostic test are not capable of detecting. They enable quantitative and qualitative nucleic acid detection, genotype analysis, and single nucleotide polymorphism (SNP) detection. Current state of the art molecular probes used with NAATs includes molecular beacon (MB), Taqman and its variations. This work presents novel molecular probe designed from Spinach and Dapoxyl aptamers. CYP17A1 is a key steroidogenic enzyme known to conduct several distinct chemical transformations on multiple substrates. In its hydroxylase activity, this enzyme adds a hydroxyl group at the 17α position of both pregnenolone and progesterone at approximately equal rates. However, the subsequent 17,20 carbon–carbon scission reaction displays variable substrate specificity in the numerous CYP17A1 isozymes operating in vertebrates, manifesting as different Kd and kcat values when presented with 17α-hydroxypregnenlone (OHPREG) versus 17α-hydroxyprogesterone (OHPROG). Effective analytical techniques need to be developed to characterize the products of lignin degradation experiments to be able to generate renewable products from lignin. Mass spectrometry is an valuable analytical approach for lignin characterizaion, but it is hindered by lignin’s poor ionization efficiency, especially in the positive ion mode. The non-mevalonate pathway (MEP), which was discovered in the 1990’s, is essential for most bacteria, plants and apicomplexan parasites. The oxidation of lignin model compounds was studied in conventional solvents in parallel with oxidations in ionic liquid solvents. Catalyst systems were investigated in ionic liquid solvents to determine how reaction rates and the selectivity for benzylic carbon oxidation were affected. Oxidation rates were often lower in ionic liquids than in conventional solvents ‒ as indicated by lower conversion in a standard reaction time ‒ likely due, at least in part, to the higher viscosity of ionic liquids. Mechano chemical treatment of Indulin AT kraft lignin by ball milling with KOH and toluene produced significant carbonyl functionality, among other changes. …

Single molecule studies can provide information of biological molecules which otherwise is lost in ensemble studies. A wide variety of fluorescence-based techniques are utilized for single molecule studies. While these tools have been widely applied for imaging soluble proteins, single molecule studies of transmembrane proteins are much more complicated. A primary reason for this is that, unlike membrane proteins, soluble proteins can be easily isolated from the cellular environment. A growing number of flavoprotein systems have been observed to bifurcate pairs of electrons. Flavin-based electron bifurcation (FBEB) results in products with greater reducing power than that of the reactants with less reducing power. Highly reducing electrons at low reduction midpoint potential are required for life processes of both aerobic and anaerobic metabolic processes. For electron bifurcation to function, the semiquinone (SQ) redox intermediate needs to be destabilized in the protein to suppress its ability to trap electrons. Carbon quantum dots (CQDs) are the latest generation of carbon nanomaterials in applications where fullerenes, carbon nanotubes, and graphene are abundantly used. With several attractive properties such as tunable optical property, edge-functionalization, and defect-rich chemical structure, CQDs have the potential to revolutionize optoelectronics, electro- and photocatalysis, and biomedical applications. Chemical modifications through the addition of heteroatoms, chemical reduction, and surface passivation are found to alter the band gap, spectral position, and emission pathways of CQDs. Despite extensive studies, fundamental understanding of structure-property relationship remains unclear due to the inhomogeneity in chemical structure and a complex emission mechanism for CQDs.

Currently, our understanding of alkalinity (AT) variability in highly dynamic environments such as coral reefs is limited by the dearth of AT measurements. Metal containing complexes have been used to catalyze various organic transformations for the past few decades. The success of several mononuclear catalysts led to transition metal catalysts used in pharmaceuticals, environmental, and industrial processes. While mononuclear complexes have been used extensively, bimetallic systems have received far less attention. Bimetallic or polynuclear sites are commonly found in metalloenzymes that perform elegant transformation in biological systems, underlying their significance. This study investigates the interactions of both purine (adenine and guanine) and pyrimidine (cytosine, thymine, and uracil) nucleobases with a pair of silver atoms (Ag2). Full geometry optimizations were performed on several structures of each nucleobase/Ag2 complex and the corresponding isolated monomers using the M06-2X density functional theory (DFT) method with a correlation consistent triple-ζ basis set augmented with diffuse functions on all atoms and a relativistic pseudopotential on Ag (aug-cc-pVTZ for H, C, N, and O and aug-cc-pVTZ-PP for Ag; denoted aVTZ). Here, we aim to further the potential use of surface-enhanced Raman spectroscopy (SERS) in analyzing hair dyes for use in forensic investigations. Current methods include light microscopy and thin-layer chromatography (TLC). However, these techniques can be destructive to the hair samples, do not have high discriminatory power, and have had problems with giving inconclusive results. Raman spectroscopy, on the other hand, is a fast, simple technique with a high discriminatory power. Previous work has shown that SERS can be used for in-situ analyses that is not destructive to the sample. This previous work also demonstrated that SERS could definitively differentiate …
