Organic pollutants such as dyes and volatile organic compounds which are widely used materials in industries and discharged daily to the aqueous environment can be harmful to human health. This present study proposed an environmentally friendly approach for silver-doped titanium dioxide (Ag-TiO2) synthesis as a photocatalyst material used as photocatalytic organic pollutant degradation. The synthesis of Ag-TiO2 was conducted by using dry and wet ball milling sol−gel method with the aqueous extract of turmeric powder under 6000C calcination. งานวิจัยนี้แสดงการผลิตเอนไซม์เอสเทอเรสที่มีค่าแอกติวิตีสูงสุด 9,200 ยูนิตต่อมิลลิลิตร จากเชื้อแอกติโนมัยซีส Thermobifida sp. เพื่อนำไปย่อยสลายเศษเส้นใยพอลิเอสเทอร์ที่ได้จากการซักผ้าสปันพอลิเอสเทอร์โดยศึกษาผลกระทบของพีเอชและอุณหภูมิต่อค่าแอกติวิตีของเอนไซม์ ศึกษาภาวะที่เหมาะสมสำหรับการย่อยสลายเศษเส้นใย ได้แก่ อุณหภูมิ พีเอช เวลา และความเข้มข้นของเอนไซม์ จากการทดลองพบว่า พีเอชและอุณหภูมิ ที่ทำให้เอนไซม์เอสเทอเรสมีค่าแอกติวิตีสัมพัทธ์สูงสุดคือ พีเอช 9 (ทริสไฮโดรคลอริกบัฟเฟอร์) และ อุณหภูมิ 55 องศาเซลเซียส และเป็นภาวะที่เอนไซม์เอสเทอเรสมีความเสถียร เมื่อนำเอนไซม์เอสเทอเรสที่ความเข้มข้น 25–100 เปอร์เซ็นต์ (ปริมาตรต่อปริมาตร) ไปย่อยสลายเศษเส้นใยพอลิเอสเทอร์ เป็นเวลา 1-7 วัน แล้วนำสารละลายหลังการย่อยสลายมาวิเคราะห์หาชนิด และปริมาณผลิตภัณฑ์ที่เกิดขึ้น (TPA, EG, BHET) รวมทั้งศึกษาสมบัติของเศษเส้นใยพอลิเอสเทอร์หลังการย่อยสลายโดยใช้เทคนิค SEM XRD และ FT-IR พบว่า สารละลายหลังการย่อยสลายเศษเส้นใยพอลิเอสเทอร์ 0.05 กรัม ด้วยเอนไซม์ 75 เปอร์เซ็นต์ (ปริมาตรต่อปริมาตร) ที่พีเอช 9 และ อุณหภูมิ 55 องศาเซลเซียส เป็นเวลา 5 วัน ที่ถูกวิเคราะห์ด้วยเทคนิค UV-Vis Spectrophotometry แสดงปริมาณ TPA ที่เกิดขึ้นสูงสุดที่ 49.05 ไมโครกรัมต่อมิลลิลิตร ในขณะที่การวิเคราะห์สารละลายหลังการย่อยสลายที่ภาวะข้างต้นโดยใช้เทคนิค HPLC-UV detector แสดงปริมาณ TPA ที่ 11.19 ไมโครกรัมต่อมิลลิลิตร และ BHET ที่ 13.27 ไมโครกรัมต่อมิลลิลิตร และการวิเคราะห์โดยใช้เทคนิค HPLC-RI detector พบปริมาณ EG ในสารละลายที่ 446.71 ไมโครกรัมต่อมิลลิลิตร ส่วนการวิเคราะห์เศษเส้นใยพอลิเอสเทอร์ด้วยเทคนิค SEM หลังการย่อยสลายด้วยเอนไซม์เอสเทอเรส 75 เปอร์เซ็นต์ (ปริมาตรต่อปริมาตร) ที่ภาวะข้างต้น พบว่า เศษเส้นใยถูกทำลายและมีพื้นผิวขรุขระเป็นบริเวณกว้างมากกว่าเศษเส้นใยที่ผ่านการย่อยสลายด้วยเอนไซม์ที่ภาวะอื่นๆ นอกจากนั้นยังพบว่าการวิเคราะห์เศษเส้นใยพอลิเอสเทอร์ก่อนและหลังการย่อยสลายด้วยเอนไซม์ด้วย FTIR ปรากฎพีกที่คล้ายคลึงกัน …

คอมพอสิตชีวภาพของพอลิแล็กทิกแอซิด/ยางธรรมชาติอิพ็อกซิไดส์/เส้นใยมะพร้าว, วรพร คิ้วเจริญ

วัตถุประสงค์ของงานวิจัยนี้ คือ การเตรียมคอมพอสิตชีวภาพจากพอลิแล็กทิกแอซิด/ยางธรรมชาติ อิพ็อกซิไดซ์ด้วยเส้นใยมะพร้าว โดยเริ่มจากยางธรรมชาติอิพ็อกซิไดซ์ที่มีระดับอิพ็อกซิเดชันปานกลาง (ร้อยละ 38 โดยโมลอิพ็อกซิเดชัน) ถูกเตรียมผ่านกระบวนการ ‘อินซิทู’ อิพ็อกซิเดชันของน้ำยางธรรมชาติ โดยใช้กรดฟอร์มิก 0.75 โมลาร์ และไฮโดรเจนเปอร์ออกไซด์ 0.75 โมลาร์ จากนั้นพอลิเมอร์ผสมพอลิแล็กทิกแอซิด/ยางธรรมชาติอิพ็อกซิไดซ์ที่อัตราส่วนต่าง ๆ (100/0, 90/10, 80/20, 70/30, 60/40, 50/50 โดยน้ำหนัก) ได้ถูกเตรียมโดยการผสมแบบหลอมเหลวด้วยเครื่องอัดรีดแบบเกลียวคู่ตามด้วยการฉีดแบบ จากการทดสอบ พบว่าความทนแรงกระแทกและการยืดตัว ณ จุดขาดได้รับการปรับปรุงด้วยการเติมยางธรรมชาติอิพ็อกซิไดซ์ที่ปริมาณร้อยละ 10 โดยน้ำหนัก หากแต่ความทนแรงดึง ยังส์มอดุลัส ความทนแรงดัดโค้ง เสถียรภาพทางความร้อนและการเกิดผลึกลดลง อีกทั้งความทนแรงกระแทกและการยืดตัว ณ จุดขาดลดลงเมื่อเติมยางธรรมชาติอิพ็อกซิไดซ์ปริมาณเพิ่มขึ้น จากผลของความทนแรงกระแทกและการยืดตัว ณ จุดขาด พอลิเมอร์ผสม พอลิแล็กทิกแอซิด/ยางธรรมชาติอิพ็อกซิไดซ์ที่อัตราส่วน 90/10 ถูกเลือกสำหรับเตรียมเป็นคอมพอสิตชีวภาพกับเส้นใยมะพร้าว 3 อัตราส่วน (5, 10 และ 20 ส่วนต่อเรซินร้อยส่วน) และทำการเปรียบเทียบเทคนิคการผสม ได้แก่ เครื่องอัดรีดแบบเกลียวคู่และเครื่องผสมแบบปิดตามด้วยการฉีดแบบ จากการทดสอบ พบว่า การผสมเส้นใยมะพร้าวที่ปริมาณ 5 ส่วนต่อเรซินร้อยส่วนด้วยเครื่องอัดรีดแบบเกลียวคู่ สามารถปรับปรุงความทนแรงดึง ยังส์มอดุลัส การยืดตัว ณ จุดขาด ความทนแรงดัดโค้งและการเกิดผลึกได้เล็กน้อย แต่เสถียรภาพทางความร้อนลดลง ขณะที่การผสมเส้นใยมะพร้าวทุกอัตราส่วนด้วยเครื่องผสมแบบปิดไม่สามารถปรับปรุงสมบัติเชิงกลและเสถียรภาพทางความร้อนอย่างมีนัยยะสำคัญได้เมื่อเปรียบเทียบกับพอลิแล็กทิกแอซิด/ยางธรรมชาติอิพ็อกซิไดซ์ที่อัตราส่วน 90/10 ที่ผสมด้วยเทคนิคเดียวกัน ถึงแม้ว่าการปรับปรุงนี้ขึ้นกับเทคนิคการผสม คอมพอสิตชีวภาพที่ผสมด้วยเครื่องผสมแบบปิดมีสมบัติเชิงกลและเสถียรภาพทางความร้อนที่ดีกว่าชิ้นงานที่ผสมด้วยเครื่องอัดรีดแบบเกลียวคู่ที่มีองค์ประกอบเดียวกัน นอกจากนี้ การเติมทั้งยางธรรมชาติอิพ็อกซิไดซ์และเส้นใยมะพร้าวสามารถปรับปรุงการย่อยสลายทางชีวภาพของชิ้นทดสอบได้

The ability to identify and quantify compounds in complex matrices is paramount for analysis of petroleum samples. Multidimensional gas chromatography (MDGC) is one of the most powerful techniques to obtain such capability. My forensic research resulted in the first peer-reviepaper to address the forensic challenges presented by 3D-printed polymer firearms. The work involved a systematic approach to the analysis of evidence stemming from 3D-printed firearms filling a critical void in current forensic knowledge. We used DART-MS to characterize the polymer evidence left behind by 3D-printed firearms as well as an evaluation of pre-existing firearm and toolmark techniques and fingerprint analysis. We demonstrated that 3D-printed firearms leave behind characteristic polymer residue on cartridge cases bullets and the receiving surface which can be identified using DART-MS. The following chapter of this thesis will cover the oxidation of guanine monophosphate (GMP). GMP has been linked to numerous conditions when it is oxidized or damaged. There have been two oxidation pathways suggested in association to the oxidation of GMP: EPT and MS-EPT. The ideal conditions for GMP’s oxidation is still be studied and collected. The study focuses on the acetate versus phosphate buffer, varying base concentration in phosphate buffer, and the varying concentration of ruthenium complex, a redox mediator.

The objective of the thesis is to develop regioselective methods for mono-fluorination of disubstituted triazoles, both 1,4- and 1,5-regioisomer, and isoxazoles. These strategies focus on one-pot direct access to final mono-fluorinated triazoles and isoxazoles. Eventually, these fluorinated substrates will be screened against various biological targets. Similarly, the optimization study of a regioselective method to generate 5-fluoro-1,4-disubstituted triazole was conducted. This reaction also utilized 1,3-dipolar cycloaddition to synthesize fluorinated triazole. However, difluoroalkene was used as a synthetic equivalent of fluoroalkyne for this method. This resulted into the inversion of polarity affording a different regioisomer. Here, fluoronitroalkene was identified as a synthetic …

The margin of the western North Pacific Ocean releases redox-active elements like Mn, Co, and Ce into the water column to undergo further transformation through oxide formation, scavenging, and reductive dissolution. Near the margin, the upper ocean waters enriched in these elements are characterized by high dissolved oxygen, low salinity, and low temperature, and are a source of the North Pacific Intermediate Water. High dissolved concentrations are observed across the Western Subarctic Gyre, with a rapid decrease in concentrations away from the margin and across the subarctic-subtropical front. Soluble extractives in wood function to protect living trees from destructive agents and also contribute to wood color and fragrance. Some extractive components have biological activities with medical applications. They also play important roles in wood processing and related applications. To increase the knowledge of wood chemistry, maple and oak were extracted by water. Ultraviolet/visible (UV/vis) spectroscopy indicated the presence of a phenolic compound, resorcinol, in maple extractives having higher molecular mass and more aromatic components than oak extractives. Soil organic matter (SOM) constitutes a global reservoir of carbon that is more than twice that of either atmospheric carbon or aquatic carbon; however, the manner in which it forms from degraded plant biomass is poorly understood. Some have recently questioned whether plant biomass is involved directly in SOM formation and suggest that it is microbial carbon that constitutes the main source of stable SOM. Such a view implies that above and below ground plant biomass is rapidly decomposed and mineralized. These technologies are being developed in order to deliver therapeutic agents, such as genes, proteins, and drugs, to patients more efficiently. The following dissertation looks at addressing environmental contaminants in the environment and the integration of an active learning style in an introductory chemistry course. It begins with addressing the concern for chlorinated propanes and ethane in the environment and the importance of looking into environmental remediation applications. This research looks at incorporating vitamin B12 as an environmentally friendly catalyst in the presence of zero valent iron for the reduction of chlorinated propanes and ethane. Smart drug-design for antibody and nanomaterial-based therapies allows for optimization of drug efficacy and more efficient early-stage pre-clinical trials. The ideal drug must display maximum efficacy at target tissue sites, but to track and predict distribution to these sites, one must have a mechanistic understanding of the kinetics involved with the individual cells of the tissue itself. This process can be tracked through biological simulations coupled with in-vitro approaches, which result in a rapid and efficient in-depth understanding of drug transport within tissue vasculature and cellular environment. This study evaluated the stability of anthocyanins in six blueberry products (gummy, graham bar, oatmeal bar, rice krispy bar, ice pop and juice) prepared with freeze-dried wild blueberry powder during processing and over eight weeks storage. Total anthocyanins were determined by HPLC before processing and at day 0 and 2, 4, 6, and 8 weeks of storage. Thermal processing of gummy and graham bar products resulted in losses of anthocyanins (50 and 31%, respectively). An eight-week storage time also resulted in a decrease in anthocyanins (7 to 51%) in products stored at ambient temperature. The ice pop, which was stored …

The ability of plants to take in water and release oxygen into the atmosphere is crucial to the survival of life on Earth. During photosynthesis, water is oxidized to O2 (dioxygen) at the Oxygen Evolving Complex (OEC) of Photosystem II. Structurally, the OEC resembles a box with an open lid, consisting of metal atoms (four manganese and one calcium) bridged by oxygen atoms. The mechanism of action of this complex, however, is not well understood. The polyoxometalate P2W18O626-, the Wells-Dawson cluster, stabilized the ribosome sufficiently for the crystallographers to solve the phase problem and improve the structural resolution. In the following we characterize the interaction of the Wells-Dawson cluster with the ribosome small subunit. There are 14 different P2W18O626- clusters interacting with the ribosome, and the types of interactions range from one simple residue interaction to complex association of multiple sites including backbone interactions with a Wells-Dawson cluster. Although well-documented that bridging oxygen atoms are the main basic sites on other polyoxometalate interaction with most proteins reported, the W=O groups are the main sites of …

Methyl group transfer from S-adenosyl-l-methionine (AdoMet) to various substrates including DNA, proteins, and natural products (NPs), is accomplished by methyltransferases (MTs). Analogs of AdoMet, bearing an alternative S-alkyl group can be exploited, in the context of an array of wild-type MT-catalyzed reactions, to differentially alkylate DNA, proteins, and NPs. This technology provides a means to elucidate MT targets by the MT-mediated installation of chemoselective handles from AdoMet analogs to biologically relevant molecules and affords researchers a fresh route to diversify NP scaffolds by permitting the differential alkylation of chemical sites vulnerable to NP MTs that are unreactive to …

In this study, the hydroxyapatite powder is investigated for both of methylene blue and thymol blue in aqueous solution. The physical and chemical properties of the adsorbent were evaluated systematically using the different techniques including Microsoft Excel programming, linear regression model and also the coefficient of determination. Batch adsorption experiments were conducted to determine the effect of contact time, solution pH, initial dye concentrations, and also the adsorbent dosage on adsorption. The adsorption kinetic parameters confirmed the better fitting of pseudo-second order kinetic model for both of methylene blue and thymol blue. The isotherm data of methylene blue and thymol …

Human endogenous retroviruses (HERVs), viral-associated sequences, are normal components of the human genome and account for 8–9% of our genome. These original provirus sequences can be transactivated to produce functional products. Several reactivated HERVs have been implicated in cancers and autoimmune diseases. An emerging body of literature supports a potential role of reactivated HERVs in viral diseases, in particular viral-associated neoplasms. Demystifying studies on the mechanism(s) of HERV reactivation could provide a new framework for the development of treatment and prevention strategies targeting virus-associated tumors. Organic aerosol affects human health and climate. These effects are largely determined by the composition of the organic aerosol, which is a complex mixture of species. Understanding the complexity of organic aerosol is critical to determining its effect on human health and climate. In this study, long range transported organic aerosol collected at the Pico Mountain Observatory was analyzed using ultrahigh resolution mass spectrometry. Organic aerosol transported in the free troposphere had an overall lower extent of oxidation than aerosol transported in the boundary layer. Oligonucleotide cross-linking agents are expected to become a new class of antisense drugs. They function by first hybridizing with a complementary mRNA and then form a covalent bond to permanently link the mRNA to the oligonucleotide agent. Because of covalent cross-linking, this new class of antisense drugs are expected to have much higher potency than existing ones.
Some oligonucleotide cross-linking agents have appeared in the literature, but they have various drawbacks, which include slow cross-linking rate and reversible cross-linking. Varied intracellular pH levels are critical for various physiological processes such as enzymatic activity, cell proliferation and apoptosis, ion transport, and muscle contraction. Cellular compartments, like lysosomes, must retain an acidic environment (pH ~ 4.5) to activate hydrolytic enzymes necessary for the breakdown of large biomolecules. Another cellular organelle, the mitochondria, provides the cell with energy and must retain an alkalis environment (pH ~ 8.0) for proper function. Substantial lysosomal and mitochondrial pH deviation is associated with cellular dysfunction and disease. In the present work, polyurethane resins have been synthesized using soybean oil based polyols (> 80% bio-renewable content) as a primary component. To overcome some of the limitations associated with soft and flexible structure of soy-based polyols, the resin has been designed with two unique set of functional groups –OH and acrylate, as a means of crosslinking sites. Such resin system can be cured by two independent cure mechanisms – (a) thermal (or ambient) curing of –OH groups by use of isocyanate cross-linkers, and (b) by UV-curing of acrylate functional groups using appropriate photoinitiators. Streams and rivers are‘active pipelines’where high rates of carbon (C) turnover can lead to globally importantemissions of carbon dioxide (CO2) and methane (CH4) from surface waters to the atmosphere. Streambed sedi-ments are particularly important in affecting stream chemistry, with rates of biogeochemical activity, and CO2and CH4concentrations far exceeding those in surface waters. Despite an increase in research on CO2and CH4instreambed sediments there is a lack of knowledge and insight on seasonal dynamics. Plasminogen activator inhibitor type-I (PAI-I ) is a serpin superfamily member protein that acts as the major inhibitor of tissue-type plasminogen activator (tPA) and urokinase-type plasminogen activator (uPA). At physiological levels, PAI-I plays an active role in regulating fibrinolysis, the process of normal blood clot degradation. Pathological levels of PAI-I have been linked to multiple health ailments: fibrosis, heart disease, cancer, and obesity. In recent years, PAI-1 has been the subject of targeted drug design. Surface chemistry is the study of the chemical and physical phenomena that transpire at interfaces such as liquid-solid, solid-gas, liquid-gas, and liquid-liquid. To study the reactions occurred at the surfaces, surface sensitive techniques come to play. Adsorption of arsenical compounds (liquid adsorbate), particularly arsenate (iAs) and dimethylarsinic acid (DMA), to the surface of iron bearing materials (solid adsorbent) that frequently are found in geosorbents such as hematite (Fe2O3) has been studied in this thesis.
