An interdisciplinary pop-up learning community (PLC) allowed students from various disciplines with different levels of content knowledge to discuss their perspectives and beliefs on climate change. The impact of this event on students was gauged by a survey from the Yale Project on Climate Change Communication. Furthermore, this project focuses on pre-service teachers who participated in the PLC. In order to investigate how pre-service teachers understand climate change and how they may or may not integrate these issues into their own instruction, individual interviews were conducted prior to and after the PLC to determine if the event had an impact …

The G-quadruplex (GQ) is a well-studied non-canonical DNA structure formed by G-rich sequences found at telomeres and gene promoters. Biological studies suggest that GQs may play roles in regulating gene expression, DNA replication, and DNA repair. Small molecule ligands were shown to alter GQ structure and stability and thereby serve as novel therapies, particularly against cancer. In this work, we investigate the interaction of a G-rich sequence, 5’-GGGTTGGGTTGGGTTGGG-3’ (T1), with a water-soluble porphyrin, N-methyl mesoporphyrin IX (NMM) via biophysical and X-ray crystallographic studies. As strains of bacteria continuously evolve and become resistant to modern antibiotics, there exists a need for the development of new treatments to combat bacterial infection. Staphylococcus aureus (S. aureus) is known to develop such resistance and remains a leading cause of invasive infections and biofilm formation on prosthetic medical devices. In 2018, Brumley and Spencer et al. isolated anaephenes A–C, natural products from a marine bacterium. Illicitly manufactured fentanyls (IMFs) have become a global and local epidemic and have recently caused an increase in the number of accidental exposures and overdoses in first responders and the community. The aim of the research is to explore a method to discriminate between IMFs and cutting agents in mock street samples using Raman Spectroscopy. Predominant Raman bands corresponding to the structural moieties of each individual illicit, licit drugs, and cutting agents were tracked, then used to discriminate between varying components in a mock street sample. Affinity capillary electrophoresis (ACE) is a separation technique that combines a biologically-related binding agent with the separating power and efficiency of capillary electrophoresis. This review will examine several classes of binding agents that have been used in ACE and applications that have been described for the resulting methods in clinical or pharmaceutical analysis. Binding agents that will be considered are antibodies, aptamers, lectins, serum proteins, carbohydrates, and enzymes. This thesis is composed to two studies, each of which relies on recent advances in time-dependent density functional theory (TD-DFT) to accurately model fluorescence spectra. The first study focuses on evaluating the fluorescence spectrum of polycyclic aromatic hydrocarbons (PAHs) having a molecular weight of 302. Three isomers are analyzed including dibenzo[a,j]fluoranthene, dibenzo[a,k]fluoranthene and dibenzo[a,l]fluoranthene. These isomers differ in the placement of one of six aromatic rings and in the amount that each molecule deviates from planarity. In this study, 9-anthraldehyde-N(4)-methylthiosemicarbazone (MeATSC) 1 and [Co(phen)2(O2CO)]Cl·6H2O 2 (where phen = 1,10-phenanthroline) were synthesized. [Co(phen)2(O2CO)]Cl·6H2O 2 was used to produce anhydrous [Co(phen)2(H2O)2(NO3)3 3. Carbohydrate-based low molecular weight gelators (LMWGs) are interesting compounds with a variety of applications. In this research, a library of nineteen carbamate derivatives of N-acetyl-D-glucosamine were synthesized and characterized, and several derivatives were found to be effective LMWGs. They formed gels in pump oils as well as mixtures of water with ethanol or water with DMSO. The structures of the carbamoyl chains played an important role in the gelation properties, short chain aliphatic derivatives and phenyl carbamates formed gels in more solvents than certain aromatic and dimeric carbamates. This innate ability is a driving force for the development of nature inspired, synthetic analogues that are robust and can catalyze a variety of organic transformations. Most graduate students attend graduate school to perform research and are then asked to teach for the first time without sufficient pedagogy training. Sulfur mustard is a highly toxic and dangerous vesicant that has been utilized as a chemical warfare agent (CWA) since World War I. Despite its extensive history, an effective antidote to sulfur mustard exposure still does not exist. The scorpionate ligand tris(pyrazolyl)borate (Tp) has contributed to the understanding of coordination and organometallic chemistry and has proven to be useful in the synthesis of metalloprotein models. Studies focusing on the similar ligand, tris(pyrazolyl)methane (Tpm), have recently been made possible with improved synthetic techniques. Characterization of the Tpm ligand and its metal derivatives, [TpmW(CO)3L]+ and [TpmMo(CO)3L]+, unveiled unexpected activity in the 1H NMR chemical shift of the methane hydrogen after the coordination of a seventh ligand. DNA serve as an ideal target where drugs such as cisplatin (cisPt) bind and exert their anticancer activities. CisPt is known to preferentially coordinate with DNA leading to formation of cisPt-deoxyguanosine (dGuo) adducts. The adducts distort the DNA structure and contribute to inhibition of DNA-mediated cellular functions and ultimately cancer cell death. Electrochemical and micro-imaging analysis of a commercial graphite-composite material is presented following electro-oxidation with β-methylumbelliferone. Charge-transfer surface modification was observed for the graphite electrode, presumed to have arisen from adsorbed interfacial umbelliferone moieties. The molecular permeability of the new surface towards a range of similar, yet size-variable (23 Å3–136 Å3) molecular redox probes is discussed. Red-shift fluorescence in confocal microscopy offers further support for the presence of a surface-bound umbelliferone layer. An SEM-platinum profiling technique was used as an imaging tool to map the umbelliferone surface and size-distribution of electroactive sites.

The binding of advanced glycation end products (AGEs) to the receptor for advanced glycation end products (RAGE) is an important feature of the RAGE signaling pathway that plays a role in the pathogenesis of diabetes. Under high glucose concentration, RAGE expression increases immensely from the formation of a Schiff base by glucose bounded to lysine. This triggers an inflammatory and immune response and upregulates the expression of RAGE and causes an accumulation of AGEs in the body. Herein, we developed a novel artificial tongue using machine learning and 12 nanoassemblies (2D-NAs) to identify and analyzed different kinds of milk beverages for quality control. This biomimetic sensor array was trained to “taste” different milk types as an “artificial tongue” which is the first time we demonstrated that this sensor array can be implemented to complex systems. Two-dimensional nanoparticles (2D-nps) and nine fluorescently labeled single stranded oligonucleotides (ssDNA) with different length and nucleobases were assembled to create 12 2D-NAs.
