This thesis describes a bio-orthogonal chemistry-based approach for new methods of fluorescence labelling of RNA in live HeLa cells via an IEDDA reaction of TCO with tetrazine. The key step of the approach is synthesis of two nucleobase analogues which were modified with TCO and a masked 5′-phosphate group using aryl phosphonamidite chemistry. Synthesis of tubercidin and uridine analogues were considered in this investigation for incorporation into RNA in live HeLa cells. Photopolymerization is a well-known process with applications in a variety of industries. Photopolymers are formed by a reaction of monomers or oligomers, photoinitiators and in some cases crosslinkers. Upon irradiation with UV or visible light, monomers undergo conformational change to form polymers via a photochemical process. Several photopolymerization types will be discussed in this thesis. End products of photopolymerizations included epoxies, inks, coatings, photoresists, etc.

This thesis describes the synthesis of an oligonucleotide containing an unnatural neutralbackbone. The backbone was constructed using bio-orthogonal inverse electron demand Diels- Alder chemistry between trans-cyclooctene (TCO) and tetrazine (Tz). A TCO phosphoramidite was synthesized and attached to solid support for strand propagation. Uridine monomers containing either bis-TCO or bis-Tz were also synthesized. The oligonucleotide strand was formed by sequential ligation of the bis-Tz and bis-TCO monomers. The oligonucleotide was analyzed using mass spectrometry. Determining whether the origin of a bloodstain is human or non-human is an important capability of a forensic investigation. In their pioneering work, Bian et al. (Journal of biomedical optics, 2017) introduced a self-reference peak algorithm for the analysis of bloodstain Raman spectra and demonstrated a great potential of this approach for differentiating human and nonhuman blood. However, this work only used three non-human species in the creation of their model. Chapter 1: Dimers of naphthalenol derivatives were synthesized by photocatalysis, cobalt(II) acetylacetonate. Several functional groups were tolerant of its skeleton. This novel dimer was connected through a reversible sigma bond. Its reversibility was confirmed by chiral HPLC and 1H-NMR experiments. Through EPR and Stern-Volmer quenching experiments, we proposed the reaction mechanism. Sulfur (S)-containing amino acids are key sources of carbon, nitrogen, and sulfur involved in protein synthesis, protein function, and providing energy for microbial growth. Dissolved free and combined methionine is one of two S-containing amino acids incorporated into proteins and has been attributed to their stability and function. The oxidation of methionine has received considerable attention given its ubiquitous presence in most biological systems and has been associated with losses in protein function and pathological disorders. Oxidative stress may contribute to the pathology of many diseases, and endogenous thiols, especially glutathione (GSH) and its metabolites, play essential roles in the maintenance of normal redox status. In this research, a new ferromagnetic-recoverable core-shell Fe3O4@SiO2@CuO nanocomposite of a certain size (20-25 nm) has been synthesized based on Cu(II) complex coated on Fe3O4@SiO2@CuO nanoparticles by facile and fast solid state microwave irradiation method. The photocatalytic activity of the nanocomposite was investigated for degradation of methylene blue (MB) and methyl orange (MO) dye pollutants in aqueous media under solar light irradiation. The nanocomposite could destroy these dyes with high efficiency in short time. This study aims to derive an essential oil from Cystoseira crinita Duby, analyze the chemical composition of the essential oil, discover the antimicrobial activities of the oil and the extracts, and investigate the scolicidal activities of the extracts. The volatile organic compounds of Cystoseira crinita Duby were determined by GC/MS-FID using both hydro-distillation (HD) and solid-phase microextraction (SPME) methods. As a result of the essential oil analysis, 97.14% of 17 compounds and 93.13% of 19 compounds were elucidated. In this study, 7 new isoxazole compounds (8-14) were synthesized from the cyclization of chalcone compounds (1-7) containing different functional groups with hydroxylamine hydrochloride in alkaline medium. Tyrosinase and antioxidant properties of 8-14 were investigated. IC50 values for the tyrosinase enzyme inhibition of compounds 8, 11, 12, and 13 were varied between 61.47 ± 3.46 and 188.52 ± 5.85, while compounds 9, 10 and 14 did not show any inhibition effect. The oxidation of ethylbenzene (EB) using tert-butyl hydroperoxide as the oxidizing agent was carried out in presence of gold nanoparticles (3 nm) supported on zinc oxide in acetonitrile solution. A higher selectivity towards acetophenone (ACP) as the major product, and a moderate selectivity towards other products such as 1-phenylethanol (PE), benzaldehyde (BZL), and benzoic acid (BzA) were observed using the prepared Au/ZnO nanocatalysts at 100 °C for 24 h. In this paper, we have prepared peripherally tetra-({6-[3-(diethylamino)phenoxy]hexyl}oxy substituted cobalt(II), copper(II), manganese(III) phthalocyanines (3, 4, 5) and their water-soluble derivatives (3a, 4a, 5a). Then, in vitro α-glucosidase and cholinesterases inhibitory actions of the water-soluble 3a, 4a, 5a were examined using spectrophotometric methods. 4a had the highest inhibitory effects among the tested compounds against α-glucosidase due to IC50 values. 4a and 5a had 40 fold higher inhibitory effects than the positive control. For cholinesterases, the compounds showed significant inhibitory actions that of galantamine which was used as a positive control. This work describes the electrochemical investigation of a promising antiviral agent, favipiravir (FAV) utilizing a nonmodified glassy carbon (GC) electrode, along with a unique voltammetric approach that can determine FAV with a good degree of accuracy, speed, and cost-effectiveness. Several 5-(alkylsulfonyl)-1-substituted-2-(1H-pyrrol-2-yl)-1H-benzo[d]imidazole derivatives were synthesized and their antioxidant activities were investigated using lipid peroxidation (LPO) and 7-ethoxyresorufin O-deethylase (EROD) assays. Docking analysis with Human NAD[P]H-Quinone oxidoreductase 1 (NQO1) was also performed to gather thorough information about these compounds that have antioxidant activities. Moreover, their molecular descriptors and ADME properties were calculated using the SwissADME online program. As a result, most of our compounds possessed better affinity and created ample interactions with NQO1. The most potent compound 5j had LP inhibition value of 3.73 nmol/mg/min. In this study, cotton spunlace nonwoven fabrics with four different basis weights were coated with graphene oxide (GO) by conventional dip coating method which is one of the most simple and effortless process in textile industry. The graphene oxide coated cotton nonwoven fabrics were immersed in an aqueous sodium dithionite solution in order to obtain reduced graphene oxide (RGO) coated cotton spunlace nonwoven fabrics. Chemical absorption of CO2 into aqueous amine solutions using a nonstirred bubble column was experimentally investigated. The performance of CO2 absorption of four different primary and secondary amines including monoethanolamine (MEA), piperazine (PZ), 2-piperidineethanol (2PE), and homopiperazine (HPZ) were compared. In this study, the synthesis of dimeric disulfide-Schiff bases was carried out using two methods. The structures of the obtained Schiff bases were elucidated by various spectroscopic methods as well as elemental analysis. The Schiff base derivative compounds (3a-6) were screened for in vitro antibacterial activity against multidrug-resistant microorganisms using microdilution method. All the tested compounds showed varying inhibition zones against the pathogens. According to MIC results, the compound 2 was shown strong inhibitory activity against all the tested microorganisms compared to antibiotics. The essential oil (EO) of Cota tinctoria var. tinctoria was analyzed using GC-FID / MS. A total of 51 compounds were determined from this taxon, accounting for 99.79% in hydrodistillation. Monoterpenes were the primary chemical class for the volatile organic compounds in the EO (36.1%, 13 compounds). Borneol (18.1%), camphor (14.9%), and β-pinene (11.3%) were the major components in the EO of C. tinctoria var. tinctoria. The antimicrobial activities of EO and n-hexane, acetonitrile, methanol, and water solvent extracts of the taxon were screened in vitro against ten microorganisms. In this study, olive oil residue (OR) biomass was pyrolyzed in the presence of bulk MgO (B-MgO), nano-MgO (N-MgO), bulk ZnO (B-ZnO)), and nano-ZnO (N- ZnO) metal oxides at different temperatures (400, 600, and 800 ºC). Significant results were obtained in terms of synthesis gas formation and CO2 reduction. Coordination-driven self-assembly of nickel (II) ions with equimolar amounts of 1, 10 phenanthroline, and benzoic acid was used to create homonuclear nickel (II) MOF-type supramolecule. Single crystal X-ray diffraction, scanning electron microscopy, differential scanning calorimetry, photoluminescence measurement, FT-IR, UV-visible spectroscopy, and nitrogen physisorption measurement were used to characterize the compound. The single crystal X-ray diffraction study reveals that the synthesized compound is two-dimensional with pores in the structure.
